Immune Toxicity Of Small Size Titanium Dioxide Nanoparticles On Rats Pulmonary Alveolar Macrophages Via Intra-Tracheal Instillation In Vivo

Abstract:[Objective] To explore the, effect of 5 nm-TiO2 nanoparticles on immune function of pulmonary alveolar macrophages in rats: [Methods] 0.5, 5.0, 50 mg/kg body weight of 5nm-TiO2 and 300nm-TiO2 suspensions was administrated via intra-tracheal instillation in rats. One weeks later, the rats were sacrificed and the pulmonary alveolar macrophages were isolated for assays of morphology, phagocytosis function, ACP; NO, Fc receptor expression, MHC-class II molecules expression. [Results] The light microscope and electron microscope showed that TiO2 nanoparticles was dose-dependent increasing inside the PAMs and induced the impaired subcellular structures such as mitochondrion. There were a significant increase in aspects of PAM phagocytosis, ACP. activity and NO, level and a significant decrease in aspects of the chemotactic ability; EA rosettes formation, and the expression of MHC-class H molecules in rats. PAMs exposed with TiO2. particles when compared with;those in the controls (P<0.05). Compared with the 300nm-TiO2 particles, exposure groups, the PAMs showed significantly decreased ALP activity. PAM phagocytosis; chemotactic ability and NO level in the 5nm-TiO2 particles exposure groups (P<0.05). And the PAM chemotactic, ACP activity; EA rosette quantity and MHC-class II molecules expression were decreased acid the NO; level was increased in a dose-dependent fashion in the 5nm-TiO2 groups: [Conclusion] Small size TiO2 nanoparticles could induce more severe damage to the PAMs and inhibition immune function of PAMs associated with-non-specific, immunity and specific immunity at some extent, and then decrease. the immune response and immune defense of host.
