Abstract:In the post-genome era, transcriptomics has become an inspiring field in life science. It reflects the dynamics of genome expression in specific time point and specific space. Transcriptomics research helps to explore the complexity and heterogeneity of asthma on genetic susceptibility, pathologic difference and medication responses. RNA next-generation sequence (RNA-Seq) is one of transcriptomic measurements which is superior to traditional ones like microarray, SAGE and MPSS, because of its higher sensitivity, broader dynamic range, capacity for novel transcript discovery. Developments in asthma research using RNA-seq technique are discussed as follows. Bronchial epithelial cells, airway smooth muscle cells and immune cells are mostly used target cells in these basic research. The interactions between host immune systems with colonizing microbiota are a hot research spot. Elucidating the diversity of GC response of asthmatics and finding novel pharmaceutical targets are major issue in this area. Single cell RNA-seq is a cutting-edge of transcriptomic study. It will enable transcriptome profiling from bulk population to thousands of individual cells, and further expand our understanding of lung allergic diseases such as asthma.
