Abstract:Hantzsch 1,4-dihydropyridine derivatives which contain 1,4-dihydropyridine structure of natural reduced nicotinamide adenine dinucleotide[NAD(P) H] coenzyme and possess a high biological activity as a class of useful drugs, can be chosen as models of NAD(P) H to react with NO or its donor to mimic the oxidation of NAD (P) H by NO in vivo. Herein, 4-substituted derivatives of Hantzsch 1,4-clihydropyridine were treated by 2-nitro-2-nitrosopropane to give the corresponding pyridine derivatives. Replacement of HEH by N-d-HEH and 4,4'-2d-HEH to react with 2-nitro-2-nitrosopropane gave the observed kinetic isotope effects of 1.03 and 1.78. The study of the redox potentials(cyclic voltametry) of the two reactants indicated that the oxidation of HEH by 2-nitro-2-nitrosopropane initiated by one-electron transfer was extremely unfavorable in thermodynamics. In addition, the ineffectiveness of p-dinitrobenzene on the reaction can also support it. These results indicate that the oxidation of HEH by 2-nitro-2-nitrosopropane is initiated by nitrosation to give the corresponding N-nitroso compound, which is subsequently subjected to two steps of homolytic cleavage to afford the aromatized Hantzsch pyridine. We believe that the present work will stimulate the investigations of the chemical features of C-nitroso compounds and its role in biological and medicinal chemistry.
Energetics of Multistep Versus One-step Hydride Transfer Reactions of Reduced Nicotinamide Adenine Dinucleotide (NADH) Models with Organic Cations and P-Quinones.

Abstract:Free energy changes of each elementary step involved in the formal hydride transfer (H(-)(T)) reactions (including the so-called "one-step" H(-)(T) and "multistep" H(-)(T) mechanisms) of the reduced nicotinamide adenine dinucleotide (NADH) models with various cations and quinones (17) were investigated either by direct thermodynamic measurements or by calculations from thermochemical cycles. Based on the energetic data thus derived, combined with kinetic observations (particularly kinetic isotope effects), the mechanistic characteristics of the NADH-mediated reductions were systematically analyzed. Practical guidelines that resulted from these analyses suggest that a mutistep mechanism (e(-)-H(*) or e(-)-H(+)-e(-)) would be followed if the energy gap of the initial electron transfer [DeltaG(e(-)(T))] between the NADH model compound and the reducing substrate is considerably smaller than the empirical critical limit of 1.0 V for an endothermic e(-)(T). In contrast, if DeltaG(e(-)(T)) is much greater than 1.0 V, a concerted one-step H(-)(T) may take place. The guideline also suggests that a "hybrid" mechanism is possible if the DeltaG(e(-)(T)) is in an intermdiate situation.

Abstract:Nicotinamide adenine dinucleotide (NADH) and nicotinamide adenine dinucleotide phosphate (NADPH), as two kinds of well-known cofactor, are widely used in the most of enzymatic redox reactions, playing an important role in industrial catalysis. In general, supply of NAD(P)H is a major challenged factor in redox fermentation systems due to its high cost and low stability, which have stimulated the development of NADH regeneration systems in recent years. Until now, a series of NAD(P)H regeneration systems have been developed. This review focuses primarily on new approaches of NAD(P)H cofactor regeneration in the biosynthesis systems, such as single cell in vivo NADH regeneration system, double cell coupling NADH regeneration system, in vitro enzyme-coupled NADH regeneration system, microbial cell surface display NADH regeneration system. Finally, the prospect and tendency of NADH regeneration are discussed.

Abstract:Nicotinamide adenine dinucleotide (NAD+) is a redox active molecule that is universally found in biology. Despite the importance and simplicity of this molecule, few reports exist that investigate which molecular features are important for the activity of this ribodinucleotide. By exploiting the nonenzymatic reduction and oxidation of NAD+ by pyruvate and methylene blue, respectively, we were able to identify key molecular features necessary for the intrinsic activity of NAD+ through kinetic analysis. Such features may explain how NAD+ could have been selected early during the emergence of life. Simpler molecules, such as nicotinamide, that lack an anomeric carbon are incapable of accepting electrons from pyruvate. The phosphate moiety inhibits activity in the absence of metal ions but facilitates activity at physiological pH and model prebiotic conditions by recruiting catalytic Mg2+. Reduction proceeds through consecutive single electron transfer events. Of the derivatives tested, including nicotinamide mononucleotide, nicotinamide riboside, 3‐(aminocarbonyl)‐1‐(2,3‐dihydroxypropyl)pyridinium, 1‐methylnicotinamide, and nicotinamide, only NAD+ and nicotinamide mononucleotide would be capable of efficiently accepting and donating electrons from and to pyruvate within a nonenzymatic electron transport chain. The data are consistent with early metabolic chemistry exploiting NAD+ or nicotinamide mononucleotide and not simpler molecules.

Abstract:Hydride transfer mechanisms of the reductions of xanthylium ion by NAD(P)H analogues (i.e. BNAH, HEH and AcrH2) were investigated. Both the kinetic observations and an analysis of thermodynamic driving forces for each mechanistic step in all the possible mechanisms indicate that the reductions are initiated by a rate-determining electron transfer, followed by a fast hydrogen atom abstraction. The mechanism of the reductions of 9-phenylxanthylium and triphenylmethylium ions by BNAH were also investigated and are similarly discussed. © 1997 John Wiley & Sons, Ltd.

Abstract:The reactions of 9-fluorenylidenemalononitrile (FDCN) and 1,1-diphenyl-2,2-dicyanoethylene (DPCN) with Hantzsch ester (HEH), N-methyl Hantzsch ester (Me-HEH), and 1-benzyl-1,4-dihydronicotinamide (BNAH) in oxygen-saturated acetonitrile have been studied. The aerobic reactions with HEH give solely reduction products. However, reactions with Me-HEH and BNAH not only result in reduction products, but also give varying amounts of oxidation products. The amount of oxidation product appears to be related to the electronic character and bulkiness of reactants. We propose that all these reactions follow a general mechanism of concerted electron-hydrogen atom transfer mechanism. If the electron-transfer complex is very tight, only ‘concerted hydride transfer reaction’ occurs. However, if the electron-transfer complex is not so tight, oxygen can capture the radicaloid intermediate to result in oxidation products.

Abstract:Several 1-benzyl-1,4-dihydronicotinamide derivatives, which are important NADH model compounds were studied theoretically in acetonitrile. The performances of various DFT methods including B3LYP, B1B95, B3PW91, MPW1B95, MPWKCIS, and M06 were tested to calculate the redox potentials. The first theoretical protocol to predict the redox potentials of these derivatives is B1B95, whose reliability has been tested against almost all the available experimental data. Strikingly, the mean absolute derivations (MAD) and root mean square (RMS) error of the current theoretical model equal 0.015 and 0.017, respectively. By using this method, the important thermodynamic properties of BNAHs were investigated and the mechanisms of hydride transfer progress were explained. Besides, para-substituents that have a big effect on these redox potentials of BNAH were systematically studied and carefully demonstrated.

Abstract:Nucleotide-derived cofactors could function as a missing link between the informational and the metabolic part at life′s emergence. One well-known example is nicotinamide dinucleotide (NAD), one of the evolutionarily most conserved redox cofactors found in metabolism. Here, we propose that the role of these cofactors could even extend to missing links between geo- and biochemistry. We show NAD can be reduced under close-to nature conditions with nickel-iron-alloys found in water-rock-interaction settings rich in hydrogen (serpentinizing systems) and that nicotinamide mononucleotide (NMN), a precursor molecule to NAD, has different properties regarding reduction specificity and sensitivity than NAD. The additional adenosine monophosphate (AMP) ″tail″ of the dinucleotide, a shared trait between many organic cofactors, seems to play a crucial mechanistic role in preventing overreduction of the nicotinamide-bearing nucleotide. This specificity is also connected to the used transition metals. While the combination of nickel and iron promotes the reduction of NAD to 1,4-NADH most efficiently, in the case of NMN, the presence of nickel leads to the accumulation of overreduction products. Testing the reducing abilities of both NADH and NMNH under abiotic conditions showed that both molecules act as equally effective, soluble hydride donors in non-enzymatic, proto-metabolic stages of life′s emergence.

New Insight into the Mechanism of NADH Model Oxidation by Metal Ions in Nonalkaline Media

Abstract:For a long time, it has been controversial that the three-step (e-H+-e) or two-step (e-H•) mechanism was used for the oxidation of nicotinamide adenine dinucleotide coenzyme (NADH) and its models by metal ions in nonalkaline media. The latter mechanism has been accepted by the majority of researchers. In this work, 1-benzyl-1,4-dihydronicotinamide (BNAH) and 1-phenyl-l,4-dihydronicotinamide are used as NADH models and ferrocenium (Fc+) metal ion as an electron acceptor. The kinetics for oxidation of the NADH models by Fc+ in pure acetonitrile was monitored by using UV-vis absorption and a quadratic relationship between kobs and the concentrations of NADH models was found for the first time. The rate expression of the reactions developed according to the three-step mechanism is quite consistent with the quadratic curves. The rate constants, thermodynamic driving forces, and kinetic isotope effects of each elementary step for the reactions were estimated. All results supported the three-step mechanism. The intrinsic kinetic barriers of the proton transfer from BNAH+• to BNAH and the hydrogen-atom transfer from BNAH+• to BNAH+• were estimated by using Zhu equation; the results showed that the former is 11.8 kcal/mol and the latter is larger than 24.3 kcal/mol. It is the large intrinsic kinetic barrier of the hydrogen-atom transfer that makes the reactions choose the three-step rather than two-step mechanism. Further investigation of the factors affecting the intrinsic kinetic barrier of chemical reactions indicated that the large intrinsic kinetic barrier of the hydrogen-atom transfer originated from the repulsion of positive charges between BNAH+• and BNAH+•. The greatest contribution of this work is the discovery of the quadratic dependence of kobs on the concentrations of the NADH models, which is inconsistent with the conventional viewpoint of the "two-step mechanism" on the oxidation of NADH and its models by metal ions in the nonalkaline media.

Abstract:The reductions of N-arylfluorenimines by Hantzsch ester in the presence and absence of Mg2+ have been studied, The similar reductions by BNAH have been compared. The results show that (1) Mg2+ plays a role of electrophilic catalyst; (2) the stronger reduction ability of Hantzsch ester than BNAH is the result of electrostatic effect caused by its 3,5-carbonyl oxygens which decreases the energy of the transition state more effectively; (3) one-step hydride transfer mechanism has been proposed.

Abstract:Cations 2-methyl-5-nitroisoquinolinium (IQ+), tropylium (T+), and xanthylium (Xn+) were treated by an NAD(P)H model 1-(p-substituted phenyl)-1.4-dihydronicotinamide series (1) in buffered aqueous solution to give the corresponding reduced products by accepting hydride. Effects of the 4-substituents of 1 on the reaction rates were investigated. Hammett's linear free energy relationship analysis on the three reactions of 1 provides the reaction constants of -0.48, -2.2, and -1.4 with IQ+, T+, and Xn+ as the hydride acceptors, respectively. Comparison of the present reactions with the reaction examples whose mechanisms are well-known, such as the reaction of 1 with a one-electron oxidant Fe(CN)6(-3), shows that the active site of 1 in the oxidation with IQ+ is at the 4-position on the dihydropyridine ring but that the active site of 1 in the oxidations with T+ and Xn+ is at the 1-position, which is in agreement with the results from the Brønsted-type linear analysis and the relation studies of the logarithm of the second-order rate constants with the oxidation potentials of the hydride donors. According to the dependence of the reaction mechanism on the active site of 1, a conclusion can be made that the reaction of 1 with IQ+ proceeds by direct one-step hydride transfer mechanism, but the reactions of 1 with T+ and Xn+ would take place via multistep hydride transfer mechanism initiated by one-electron transfer.

Abstract:A series of compounds, o-bromomethylbenzylidene-malononitrile (1), dimethyl o-bromomethyl-benzylidenemalonate (2) and methyl α-cyano-o-bromomethylcinnamate (3) were reduced in the dark and under irradiation by an NAD(P)H model, 1-benzyl-1,4-dihydronicotinamide (BNAH). Two different mechanisms were found, i.e. one-step hydride transfer (polar pathway) and multi-step sequence initiated by single electron transfer (SET pathway). The effect of electron-withdrawing groups on the reactivity of the substrates were discussed in terms of Hammett substituent constants, 13C NMR chemical shift values and cyclic voltammetric redox potentials, and their correlations.

Abstract:This study shows the importance of resolving catalytic performance in the regeneration of the reduced form of nicotinamide adenine dinucleotide (NADH) through activity measurements based on NAD+ conversion and the closure of mass balance via byproduct quantification. This approach is applied to assess the performance of supported platinum catalysts with varying points of zero charge, utilizing H2 as a reductant. It was found that Pt/SiO2, which exhibits a net negative charge under the reaction conditions, outperforms the neutral Pt/C and positively charged Pt/MgO because of the favorable electrostatic attraction between the catalyst surface and positively charged (+1) nicotinamide ring. Abstract:The reduction of quinones by BNAH is related with the presence or absent of EWG; in the case of the weak or strong EWG, the SET and HT mechanisms in a two‐step reaction were observed; while in the absent of EWG, the hydrogen coupled electron transfer (HCET) was observed as the main mechanism. Although it is well known that coenzyme NAD(P)H is involved in anabolic and catabolic reactions in the living organism, there is still significant controversy over the reaction mechanism involved in this biochemical transformation. Thus, 1‐benzyl‐1,4‐dihydronicotinamide was used as a NAD(P)H model in the reduction reaction of 1,4‐benzoquinone (Q), 2,3,5,6‐tetrachloro‐1,4‐benzoquinone, and 2,3‐dicyano‐1,4‐benzoquinone in acetonitrile medium. The kinetic calculations support that formal hydride transfer is the main mechanism promoting Q reduction, while the two‐step process dominates 2,3‐dicyano‐1,4‐benzoquinone reduction. Interestingly, only the single‐electron transfer mechanism takes place when 2,3,5,6‐tetrachloro‐1,4‐benzoquinone is used, affording the corresponding semiquinone derivative as the main product. This mechanistic behavior is related to the presence or absence of electron‐withdrawing groups in the quinones used. Furthermore, the kinetic study results showed that calculated reaction rate constants are in close agreement with experimental results. The results support that formal hydride transfer on the reduction reaction of Q by 1‐benzyl‐1,4‐dihydronicotinamide in acetonitrile proceeds through a hydrogen coupled electron transfer mechanism. This theoretical analysis provides valuable knowledge that can be extrapolated to study the reduction of quinones performed by NADH and NADPH in physiological media.

Abstract:Potential energy surfaces for rotations of ribose about the nicotinatnide N-C bonds in NAD4 and NADH have been evaluated from ab initie molecular orbital calculations with the 3-21G and 6-3 IG In the optimal conformation of NAD the glycosiic C-O bond is near the plane of the nicotinamide ring. By contrast, the lowest energy conformer of NADH has the glycosidic C-O bond nearly perpendicular to the dihydronieotinamide ring. The redox potential of the NAD/NADH couple is a function of the ribose orientation. There is boat-like puckering of the 1,4-dihydropyridine ring, and the direction of the puckering is anti to the glycosidie bond. The geometry of the transition structure for hydride transfer is similar to that of NADH. When NADH is in the anti conformation, the transfer of the pro-R hydrogen is preferred, and when NADH is in the syn conformation, the transfer of the pro-S hydrogen is preferred.
