Abstract:Oleanolic acid (OA) has been used to treat liver disorders, but whether it can attenuate hepatic ischemia-reperfusion- (IR-) associated liver dysfunction remains unexplored. In the present study, 160 male Sprague-Dawley rats were equally divided into five groups: group SH received neither hepatic IR nor drugs; group IR received hepatic IR without drugs; group CM and group OA received 0.5% sodium carboxymethylcellulose and 100 mg/kg OA, intragastrically, once a day for seven days before the hepatic IR, respectively; on the basis of treatment in group OA, group OA+wortmannin further received 15 μg/kg of PI3K inhibitor wortmannin, intraperitoneally, 30 min before the hepatic IR. Then each group was equally divided into four subgroups according to four time points (preoperation, 0 h, 3 h, and 6 h after reperfusion). Serum ALT activity, IL-1β concentration, and hepatic phosphorylation of PI3K, Akt, and GSK-3β protein expression were serially studied. We found that OA pretreatment improved histological status and decreased serum ALT and IL-1β levels. It also increased p-PI3K, p-Akt, and p-GSK-3β protein expression at all the four time points. Prophylactic wortmannin partially reversed OA's protective effects. The data indicate that OA pretreatment protects liver from IR injury during the acute phase partially through PI3K/Akt-mediated inactivation of GSK-3β.
Preconditioning and Postconditioning Reduce Hepatic Ischemia-Reperfusion Injury in Rats

Abstract:BACKGROUND:Ischemia-reperfusion injury occurs when ischemic tissues or organs suffer from further functional and structural damage when their blood supply recovers. This study aimed to contrast the protective effects of ischemic preconditioning and ischemic postconditioning in hepatic ischemia-reperfusion injury in rats.METHODS:Thirty-two healthy male Wistar rats were randomly divided into four groups: sham-operated (SO), ischemia-reperfusion (IR), ischemic preconditioning (I-pre), and ischemic postconditioning (I-post). Blood samples and hepatic tissue were taken from all groups after the experiments.RESULTS:There were significant differences between the IR, I-pre and I-post groups in alanine aminotransferase and aspartate aminotransferase levels, NF-kappaB p65 expression, apoptosis index and superoxide dismutase activity in hepatic tissue. There were no significant differences between the I-pre and I-post groups.CONCLUSIONS:Ischemic postconditioning and ischemic preconditioning reduce hepatic ischemia-reperfusion injury, but in clinical practice the former is a more appropriate choice.
Effect of Oleanolic Acid Pretreatment on Hepatic Ischemia-Reperfusion Injury in Rats

Abstract:Objective To investigate the effect of oleanolic acid pretreatment on hepatic ischemiareperfusion (I/R) injury in rats. Methods One hundred and twenty-eight male SD rats weighing 230-250 g were randomly divided into 4 groups (n = 32 each): sham operation group (group S), I/R group, 0.5% sodium carboxymethyl cellulose group (group CMC) and oleanolic acid preconditioning (group OA). Partial liver ischemia was produced by clamping hepatic portal vein and hepatic arteries for 60 min with atraumatic mini-clamp, followed by 12 h of reperfusion in group I/R, CMC and OA. Oleanolic acid suspension 100 mg/kg was infused intragastrically in group OA, while the equal volume of 0.5% CMC-Na (in group CMC) and drinking water (in group S and I/R) was infused intragastrically instead once a day for 7 days, and then hepatic I/R was performed at day 8. The left liver was removed and blood sample was taken from inferior vena cava at 0, 3, 6 and 12 h ofreperfusion for determination of serum alanine amino transferase (ALT) activity, superoxide dismutase (SOD)activity, malondialdehyde (MDA) and glutathione (GSH) content, and expression of phospho-phosphatidylinositol3-kinase (p-PI3K), Akt, p-Akt, Bcl-2, Bax, p-Bad and Bad in the liver, and microscopic examination. Results Serum ALT activity and MDA content in the liver were significantly increased, SOD activity and GSH content in the liver were significantly decreased, expression of p-PI3K, p-Akt, Bax, Bad and p-Bad was up-regulated, and Bcl-2 expression was down-regulated during reperfusion in group I/R, CMC and OA as compared with group S (P ＜0.05). Compared with group I/R, serum ALT activity and MDA content in the liver were significantly decreased, SOD activity and GSH content in the liver were significantly increased, expression of p-PI3K, p-Akt,Bcl-2 and p-Bad was up-regulated, and expression of Bad and Bax was down-regulated during reperfusion in group OA (P ＜ 0.05), but no significant change was found in the indexes mentioned above in group CMC (P ＞ 0.05).Serum ALT activity and MDA content in the liver were significantly lower, SOD activity and GSH content in the liver were significantly higher, expression of p-PI3K, p-Akt, Bcl-2 and p-Bad was significantly higher, and expression of Bad and Bax was significantly lower during reperfusion in group OA than in group CMC (P ＜ 0.05).The pathological changes in the liver were milder in group OA than in group I/R. Conclusion Oleanolic acid pretreatment can alleviate hepatic I/R injury by activating PI3K/Akt signaling pathway and inhibiting apoptosis.
Influence of Oleanolic Acid Pretreatment to the PI3K-AKT-GSK-3β Signaling Pathway During Rat's HIRI

Abstract:Objective:It is proved that the pretreatment of oleanolic acid(OA)increases rat's hepatic glycogen and alleviates the hepatic ischemic reperfusion injury(HIRI),but the mechanism is still not quite clear.This research is focused on the influence of OA pretreatment to rat's PI3K-AKT-GSK-3β signaling pathway around its HIRI.Methods:Total of 128 male Sprague-Dawley(SD)rats wereindvidedinto shamgroup(SH),ischemic repedusion group(IR),0.5%sodium carboxymethycellulose(CMC-Na)group(CM),OA+0.5%CMC-Na group(OA)equally.Before operation all rats got intragastric administration one time a day for Seven days,OA group with OA(100 mg/kg)+0.5% CMC-Na,SH group and IR group with water of the same volume,CM group with 0.5% CMC-Na of the same volume.At the 8th day,rats were suffered from segmental(70%)hepatic ischemia for 60 min and were followed by different periods of reperfusion.In the 8th day before the operation,the hepatic tissu were reperfused at the point of 0 h,3 h,and 6 h. The protein expres aloha of p-PI3K(p85),p-AKT(ser473),AKT,P-GSK-3β(ser9) and GSK-3β were evaluated.Results:Before operation,at the point of oh,the protein expressions of p-PI3K(p85),p-AKT(ser473)and p-GSK-3β(ser9)in the OA group increased much more remarkablv than other three groups(P＜0.05),while the protein expresions of SH group,IR group and CM group were no differences(P＞0.05).At the reperfusion point of 3 h,6 h,the protein expressions of p-PI3K(p85),p-AKT(set473)and p-GSK-3β(set9)in the OA group increased much mole remarkably than other three groups(P＜0.05),while the protein expressions of SH group were lower than those of otherthree groups(P＜0.05).There was no difference between IR and CM group in the 3 proteins expression(P＞0.05).The protein expressions of AKT and GSK-3β in all the groups at all the points were no differences(P＞0.05).Conclusion:The pretreated OA increases the protein expressions of p-PI3K(p85),p-AKT(aer473)and pGSK-3β(ser9)before and after HIRI,activating the signaling pathway of hepatic PI3K-AKT-GSK-3β,which may be one of the mechanisms of OA alleviating HIRI.

Abstract:Hepatic ischemia reperfusion (IR) injury (IRI) occurs during liver transplantation, hepatectomy, and hemorrhagic shock. Oleanolic acid (OA) is a natural compound with antioxidant and anti-inflammatory activity that has been used to treat liver disorders in clinical practice for several years. Here, we investigated the effects and underlying mechanisms of OA in hepatic IRI. A 60-minute partial (70%) hepatic, warm, ischemic reperfusion model was established in BALB/c mice, and two doses (30 and 60mg/kg) of OA were administered intragastrically for 7 consecutive days prior to hepatic IR. Orbital blood and liver specimens were collected at 2, 8, and 24h after IR. The results showed that OA preconditioning significantly alleviated hepatic injury, as evidenced by decreased alanine aminotransferase and aspartate aminotransferase levels; improved histology, inhibition of JNK phosphorylation, and high mobility group box 1 (HMGB1); and tumor necrosis factor-alpha downregulation in hepatic IR mice. OA upregulated Bcl-2 and downregulated caspase-3, caspase-9, Bax, Beclin 1, and LC3, which play crucial roles in the regulation of apoptosis and autophagy. These findings highlighted the protective effects of OA against hepatic IRI mediated by the inhibition of apoptosis and autophagy and the release of HMGB1, which acted as a late inflammatory mediator in hepatic IRI.
Oleanolic acid alleviating ischemia-reperfusion injury in rat severe steatotic liver via KEAP1/NRF2/ARE

Abstract:Severe steatosis in donor livers is contraindicated for transplantation due to the high risk of ischemia-reperfusion injury (IRI). Although Ho-1 gene-modified bone marrow mesenchymal stem cells (HO-1/BMMSCs) can mitigate IRI, the role of gut microbiota and metabolites in this protection remains unclear. This study aimed to explore how gut microbiota and metabolites contribute to HO-1/BMMSCs-mediated protection against IRI in severe steatotic livers. Using rat models and cellular models (IAR20 and THLE-2 cells) of steatotic liver IRI, this study revealed that ischemia-reperfusion led to significant liver and intestinal damage, heightened immune responses, impaired liver function, and altered gut microbiota and metabolite profiles in rats with severe steatosis, which were partially reversed by HO-1/BMMSCs transplantation. Integrated microbiome and metabolome analyses identified gut microbial metabolite oleanolic acid as a potential protective agent against IRI. Experimental validation showed that oleanolic acid administration alone alleviated IRI and inhibited ferroptosis in both rat and cellular models. Network pharmacology and molecular docking implicated KEAP1/NRF2 pathway as a potential target of oleanolic acid. Indeed, OA experimentally upregulated NRF2 activity, which underlies its inhibition of ferroptosis and protection against IRI. The gut microbial metabolite OA protects against IRI in severe steatotic liver by promoting NRF2 expression and activity, thereby inhibiting ferroptosis.
Effects of Pretreatment with Oleanolic Acid on Glycogen Content in Rots During Hepatic Ischemic Reperfusion

And changes of histological pathology were also observed by light microscope. Results:Before operation, glycongen content of group D increased compared with other groups (P 0.05). After reperfusion glycogen content in grup A decreased at 0,3,6 h compared with group B,C and D (P 0.05);glycongen content in group D increased compared with group B and C respec- tively (P 0.05). At 0 h after reperfusion,lactate level of group D increased compared with other groups (P 0.05);lactate level of group B and C increased compared with group A (P 0.05). At 3,6 h after reperfusion,the activity of ALT of group B,C and D increased compared with group A(P 0.05);the activity of ALT in group D decreased compared with group B and C respectively(P 0.05);group D had relieved morphologic changes compared with group B and C. Conclusion:OA pretreatment could increase glycogen content,also provide the rat liver with significant tolerance to IR injury. The mechanism may be related to the increased level of energy during IR.
Effect of Oleanolic Acid on Oxygen Free Radicals in Liver Ischemia-Reperfusion Injury in Rats

Abstract:Objective To investigate the effects of oleanolic acid (OA) on oxygen free radicals in liver ischemia-reperfusion(IR) injury in rats. Methods One hundred and twenty-eight male SD rats were randomly divided into four groups of sham operation(SH), IR, sodium carboxymethycellulose(CMC-Na), and oleanolic acid (OA). Rats I-R model was established by segmental (70%) hepatic ischemia for 60 min, followed by reperfusion for different times. The animals were sacrificed at 0, 3, 6 or 12 h after reperfusion, respectively. The activity of serum ALT, content of malondialdehyde (MDA), activity of superoxide dismutase(SOD) and content of glutathione (GSH) were measured. Results At 3 h, 6 h or 12 h after reperfusion, the activity of ALT and the content of MDA were significantly higher in group IR, CMC-Na and OA than those in group SH and the levels of SOD and GSH were lower in group IR,CMC-Na and OA than those in group SH(P<0.05). The activity of ALT and the content of MDA were significantly lower but SOD and GSH were higher in group OA than those in group IR and CMC-Na at 3 h, 6 h, or 12 after reperfusion(P<0.05). Conclusion OA can protect the liver from IR injury, which is related to the inhibition of production and release of oxygen free radicals.

Abstract:OBJECTIVE:To observe the effect of ischemic postconditioning on phosphatidylinositol-3-OH kinase (PI3K) and extracellular signal-regulated protein kinase 1/2(ERK1/2) in rats after hepatic ischemia reperfusion in rats and investigate the mechanism of ischemic postcoditioning of the liver.METHODS:Three cycles of 1 min-off-1 min-on ischemic postconditioning regime were used in a rat model of 70% hepatic ischemia-reperfusion injury. The changes in the liver function, hepatocyte apoptosis, phosphorylation of Akt and ERK1/2 were assessed in rats treated with sham operation, LY294002+sham operation (LY+S), PD98059+sham operation (PD+S), ischemia reperfusion (IR), ischemic postconditioning (IPO), LY294002+ ischemic postconditioning (LY+IPO), or PD98059+ischemic postconditioning (PD+IPO).RESULTS:Ischemic postconditioning significantly alleviated hepatic ischemia-reperfusion-induced liver function injury and hepatocyte apoptosis and increased phosphorylation of Akt and ERK1/2. LY294002 and PD98059 antagonized the effects of ischemic postconditioning in the liver.CONCLUSION:Activation of PI3K and ERK1/2 may mediate the protective effect of ischemic postconditioning against hepatic ischemia- reperfusion injury in rats.
Influence of Oleanolic Acid Pretreatment to the IKK/I-κB/NF-κB Signaling Pathway During Rat's HIRI

Abstract:Objective:To investigate the influence of oleanolic acid(OA) pretreatment to rat's IKK/I-κB/NF-κB signaling pathway around its hepatic ischemic/reperfusion injury(HIRI).Methods:One hundred and twenty eight male Sprague-Dawley(SD) rats were randomly divided into sham group(SH),ischemic/reperfusion group(IR),0.5% sodium carboxymethycellulose(CMC-Na) group(CM) and OA+0.5% CMC-Na group(OA).Before the operation,the rats of each group received intragastric administration of corresponding solution once a day for seven days(OA group with 100 mg/kg of OA dissolved in 0.5% CMC-Na,SH group and IR group with water of the same volume,CM group with 0.5% CMC-Na of the same volume).At the 8th day,rats were suffered from segmental(70%) hepatic ischemia for 60 min and then followed with different periods of reperfusion.The hepatic tissue were obtained at 0 h,3 h,and 6 h after the reperfusion.The expression of IKK2 and I-κBα in the cytoplasm and NF-κB p65 in the nucleus were evaluated by the method of Western blotting.Results:There was no IKK2 expression in the cytoplasm,and only few of NF-κB p65 was detected in the nucleus before operation or 0h after reperfusion in each group.Meanwhile,the volume of I-κBα detected in the cytoplasm has no significant difference among four groups at the same point of time.The expression of IKK2 in the cytoplasm and NF-κB p65 in the nucleus in the SH group was much lower than those in other three groups(P 0.05),while those proteins detected in OA group is less than those in CM group and IR group(P 0.05)at 3h or 6 h after reperfusion.At these two points of time,the expression of I-κBα in SH group was higher than that in the other three groups(P 0.05),and this protein detected in OA group is more than that in CM group and IR group(P 0.05).The expressions of IKK2 and I-κBα in the cytoplasm and NF-κB p65 in nucleus between CM group and IR group had no significant difference at each point of time(P 0.05).Conclusion:HIRI would increase the expression of IKK2 in the cytoplasm,leading to the phosphorylation of I-κB,activating NF-κB and making it to translocate into the nucleus,and then the activated NF-κB which could mediate inflammatory response eventually caused damage of liver cells.The pretreatment of OA can inhibit the activating of the signaling pathway of IKK/I-κB/NF-κB,which may be one of the mechanisms for OA alleviating HIRI.

Abstract:BACKGROUND:Recently research demonstrated that primary non function of transplanted hepar is the main reason for early death after hepatic transplantation,so as to ischemia-reperfusion injury for poor graft function.How to relieve or eliminate the ischemia-reperfusion injury is the hot topic in clinic research.OBJECTIVE:To observe the expression of ERK,JNK,and P38 in MAPK signal transduction pathway of hepatic ischemia-reperfusion injury and hepatic ischemic postconditioning settings.DESIGN,TIME AND SETTING:The randomized controlled experiment of animals was performed in the Experimental Animal Center of Affiliated Xiangya Hospital of Central South University from May to October 2007.MATERIALS:A total of 102 Wistar rats were randomly divided into the sham operation(n=6),ischemia-reperfusion(n=48) and ischemic postconditioning(n=48) groups.The animals in the ischemia-reperfusion and ischemic postconditioning groups were assigned into subgroups treated with various time(0,0.5,1,2,4,8,12,24 hours),with 6 animals in each subgroup.METHODS:The orthotopic partial hepatic ischemia-reperfusion injury animal model was used.120 Wistar rats were randomly divided into ischemia reperfusion,ischemic postconditioning and sham operation groups.Liver tissues were harvested at 0,0.5,1,2,4,8,12 and 24 hours after reperfusion.MAIN OUTCOME MEASURES:The phosphorylation P38,JNK and ERK were detected by immunohistochemistry and semi-quantitative analysis.RESULTS:p-ERK and p-JNK increased slightly after ischemia,increased significantly at 30 minutes until 4 hours,peaked at 2 hours after reperfusion.Compared with the ischemia reperfusion group,the expression of p-ERK,p-JNK in the treated group was higher than that of the ischemia reperfusion group at 1,2 and 4 hours after reperfusion(P < 0.05).The expression of p-P38 increased slightly after ischemia,significant increased at 30 minutes,peaked at 1 hour,and then decreased at 2 hours after reperfusion,which remained a ischemia level.Compared with the ischemia reperfusion group,the expression of p-P38 in the treated group was higher,in particular,at 1 hour after reperfusion(P < 0.05).CONCLUSION:The ischemia preconditioning can relieve the liver injury in ischemia-reperfusion by increase the levels of ERK,P38,and decrease JNK phosphorylation level.

Abstract:Hepatic ischemia-reperfusion (I/R) injury is a serious complication in patients who have undergone hepatic surgery such as orthotopic liver transplantation and partial hepatectomy. Recently, a new cytoprotective agent, ursodeoxycholyl lysophosphatidylethanolamide (UDCA-LPE), was reported to protect against hepatic I/R injury. However, the protective mechanism of UDCA-LPE is not fully understood. Therefore, we conducted this study to explore its underlying mechanism. We used liquid chromatography-tandem mass spectrometry (LC-MS/MS) to analyze the liver lipid metabolism changes in mice during I/R. KEGG enrichment indicated that UDCA-LPE is likely to exert its protective role by regulating fatty acid (FA) metabolism. Further analysis found that UDCA-LPE significantly increased the ratio of oleic acid (OA) to palmitic acid (PA). We found that mice pretreated with OA improved tolerance to hepatic I/R injury. In addition, the phosphorylation level of AKT was markedly upregulated during oxidative stress to promote p65 nuclear translocation, triggering an inflammatory response that exacerbated cell damage and OA treatment significantly inhibited this process. Notably, OA was found to inhibit H2O2-induced oxidative stress, inflammation, and cell death in HepG2 cells. Furthermore, we found that OA supplementation to the medium did not result in a significant increase in intracellular OA, but marked increase in the ratio of OA to PA, which may be an important mechanism for the inflammatory response induced by oxidative stress during I/R. Finally, we demonstrated that OA increased the level of autophagy in HepG2 cells, which may be one of the protective mechanisms against oxidative stress. Collectively, this study revealed that FA metabolism functionally determines the oxidative stress-related inflammation caused by hepatic I/R. We hypothesize that OA treatment may be a promising strategy for preventing and treating I/R-induced liver damage.

Abstract:Objective To explore the effect and mechanism ofα-Lipoic acid on hepatic ischemi- a-reperfusion injury in rats.Methods The models of liver partial ischemia-reperfusion were established in male Wistar rats.The rats were randomly divided into a control group (group A) and an experimental group (group B) and administered either saline orα-Lipoic acid 15 rain before the ischemia.GPT,GSH and GSH-PX were examined at various time points after reperfusion (1,3,6 and 12 h) in serum of the rats with or withoutα-Lipoic acid administration.The expression of NF-κB p65 and MIP-2 mRNA was detected by using immunohistochemistry and RT-PCR respectively in ischemic liver.Results Compared with group B,the ALT level and the activity of GSH-PX in serum,the expression of NF-κB p65 and MIP-2 mR- NA in iscbemic liver of the rats in group A were significantly increased (P<0.01),while GSH in serum was decreased (P<0.01).Conclusionα-Lipoic acid may preteet rats from liver ischemia-reperfusion injury through cleaning oxygen free radicals,increasing GSH production,and reducing NF-κB activation and GSH consumption.
Optimized Postconditioning Algorithm Protects Liver Graft after Liver Transplantation in Rats.

Abstract:BACKGROUND:Ischemia reperfusion injury (IRI) causes postoperative complications and influences the outcome of the patients undergoing liver surgery and transplantation. Postconditioning (PostC) is a known manual conditioning to decrease the hepatic IRI. Here we aimed to optimize the applicable PostC protocols and investigate the potential protective mechanism.METHODS:Thirty Sprague-Dawley rats were randomly divided into 3 groups: the sham group (n = 5), standard orthotopic liver transplantation group (OLT, n = 5), PostC group (OLT followed by clamping and re-opening the portal vein for different time intervals, n = 20). PostC group was then subdivided into 4 groups according to the different time intervals: (10 s × 3, 10 s × 6, 30 s × 3, 60 s × 3, n = 5 in each subgroup). Liver function, histopathology, malondialdehyde (MDA), myeloperoxidase (MPO), expressions of p-Akt and endoplasmic reticulum stress (ERS) related genes were evaluated.RESULTS:Compared to the OLT group, the grafts subjected to PostC algorithm (without significant prolonging the total ischemic time) especially with short stimulus and more cycles (10 s × 6) showed significant alleviation of morphological damage and graft function. Besides, the production of reactive oxidative agents (MDA) and neutrophil infiltration (MPO) were significantly depressed by PostC algorithm. Most of ERS related genes were down-regulated by PostC (10 s × 6), especially ATF4, Casp12, hspa4, ATF6 and ELF2, while p-Akt was up-regulated.CONCLUSIONS:PostC algorithm, especially 10 s × 6 algorithm, showed to be effective against rat liver graft IRI. These protective effects may be associated with its antioxidant, inhibition of ERS and activation of p-Akt expression of reperfusion injury salvage kinase pathway.

Abstract:Background Ischemia-reperfusion injury (IRI) is a serious complication of liver surgery, especially for extended hepatectomy and liver transplantation. The aim of this study was to evaluate the protective effect of combined ischemic preconditioning (IPC) and salvianolic acid-B (Sal-B) pretreatment against IRI-induced hepatocellular injury. Methods Sixty male Wistar rats weighing around 200 g were randomized into five groups ( n = 12): sham group: only anesthesia and laparotomy; IR group: 90 min sustained ischemia by blocking the left portal vessels; IPC group: 10 min ischemia and 10 min reperfusion prior to the sustained ischemia; Sal-B group: 10 mg/kg injection of Sal-B intravenously 10 min prior to the sustained ischemia; IPC + Sal-B group: same IPC procedure as in IPC group, but proceeded by intravenous administration of Sal-B 10 min prior to sustained ischemia. After 5 h of reperfusion, serum levels of ALT and AST were measured; the amount of malondialdehyde (MDA) and adenine nucleotides in liver tissue was determined; the expression of Bcl-2 and caspase-3 was detected by immunofluorescent and western blotting techniques; the severity of apoptosis and pathological alterations was evaluated by TUNEL and H&E staining, respectively. Results The serum aminotransferases, hepatic MDA concentration, and apoptotic index in groups IPC, Sal-B, and IPC + Sal-B were significantly lower than those in the IR group ( P < 0.001), while the IPC + Sal-B group had the lowest values among these groups ( P < 0.05). Compared with the IR group, groups IPC and Sal-B not only had statistically higher ATP levels and energy charge (EC) values ( P < 0.01), but also had upregulated Bcl-2 expression and downregulated cleaved caspase-3 expression in liver tissue. All these effects were further augmented in the IPC + Sal-B group. Liver histopathological findings were consistent with these results. Conclusions Based on these results, the combined IPC and Sal-B pretreatment had a synergistically protective effect on liver tissue against IRI, which might be due to decreased post-ischemic oxidative stress, improved energy metabolism, and reduced hepatocellular apoptosis.

Abstract:Objective To investigate the protective effect of propotol against hepatic ischemiareperfusion injury in rats on mitochondrial permeability transition pore (MPTP) and the mechanism of GSK-3β.Methods Thirty SD rats were randomly assigned into five groups (n =6):sham operation group (S group),ischemia reperfusion group (I/R group),CsA pretreatment group (C group),propofol pretreatment group (P group),and propofol plus atractyloside pretreatment group (A + P group).Nauta liver ischemia-reperfusion rat model was used.Liver lobes were subjected to warm ischemia for 60min and then reperfusion for 120 min.In P group,propofol [12 mg/(kg · h)] was administered in the femoral vein for 30 min before ischemia until the end of reperfusion.In C group,CsA (2 mg/kg) was administered in the femoral vein for 20min before ischemia.In A + P group,20 μmol/kg of atractyloside was given through the femoral vein 10min before the injection of propofol.Rats were sacrificed at the end of reperfusion,and venous blood and hepatic tissue specimens from the same part of ischemia were obtained from different groups.Results Compared with S group,the AST and ALT levels were increased significantly,mitochondrial swelling were increased and mitochondrial membrane potential were decreased significantly in I/R group and A + P group.Casepase-3 were increased significantly and p-GSK3β Ser9 were decreased significantly in I/R group and A + P group.Compared with I/R group,the content of AST and ALT were decreased significantly,mitochondrial swelling were decreased and mitochondrial membrane potential were increased significantly,casepase-3 release were decreased significantly and p-GSK3β Ser9 were increased significantly in P group and C group.GSK-3β in each group displayed no significant difference.Conclusions Propofol can significantly reduce hepatic ischemia-reperfusion injury.The protective effect of propofol may be achieved via the inhibition of GSK-3β activation,increased p-GSK-3β Ser9 level,suppressing MPTP opening and decreasing hepatocytes apoptosis.
Protective Effects of Gastrodin Pretreatment on Mouse Hepatic Ischemia-Reperfusion Occurring Through Antioxidant and Anti-Apoptotic Mechanisms

Abstract:Hepatic ischemia-reperfusion injury (HIRI) often occurs following surgical procedures such as liver resection and transplantation. However, despite its clinical prominence, to the best of our knowledge, there remain no effective strategies to treat HIRI. Therefore, the aim of present study was to identify therapeutic agents that can exert beneficial effects against HIRI. The present study found that following hepatic IR modeling in mice, gastrodin (Gas) pretreatment improved the IR outcomes in terms of the serum biochemical indexes (alanine transaminase and aspartate transaminase), tissue biochemical indexes (superoxide dismutase, malondialdehyde and reduced glutathione content) and tissue pathology (H&E staining). In addition, compared with those in the IR + vehicle group, the IR + Gas group showed upregulated expression levels of nuclear erythroid 2-related factor 2, heme oxygenase 1 and Bcl-2 as detected by western blotting and reverse transcription-quantitative PCR. The mRNA and protein expression levels of Bax and caspase-3 were downregulated in the IR + Gas group compared with the IR + vehicle group. Concurrently, no significant differences were observed in the parameters between the Sham + vehicle and the Sham + Gas groups, indicating that Gas pretreatment may not cause liver damage. In conclusion, the findings of the present study revealed that Gas pretreatment exerted a protective effect in HIRI through both antioxidant and anti-apoptotic mechanisms.
