Abstract:3-(R)-3-(adenin-9'-yl)-4-(R)-hydroxy-5-(R)-hydroxymethyltetrahydrofuran was synthesized by the regeoselective substitution of 3,4-epoxy-(trans)-5-(R)-dimethoxymethyltetrahydrofuran which was obtained by 4 steps from L-xylose.

Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:Two synthetic strategies were developed to obtain isonucleosides 2a and 2b. Starting from the known compound 4, an extension of one carbon unit at sugar 6-terminal was achieved by Wittig reaction and Stannyl-desulfonylation reaction. After oxidation of the double bond, the isonucleosides with elongated side chain 2a and 2b were synthesized. For the synthesis of isonucleosides containing different bases, an epoxide intermediate approach was developed. Isonucleosides 2a and 2b were synthesized by regioselective epoxide opening reaction of 18 in good yield.

Abstract:A new synthesis of the tetrasubstituted tetrahydrofuran fragment of the marine secondary metabolites amphidinolides X and Y is described. Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:A novel efficient HFIP-catalyzed synthesis of structurally diverse 2,2-difluoro-3-hydroxy-1,4-diketone derivatives from readily available glyoxal monohydrates and difluoroenoxysilanes is described. This convenient protocol is induced by the distinctive fluorine effect of the reactants and the fluoroalcohol catalyst, which represents the first application of fluoroalcohol catalysis in a Mukaiyama aldol reaction.

Abstract:2,5:3,4-Dianhydro-L-talofuranose dimethylacetal 7 was synthesized from D-glucose in 7 steps. A series of 4-deoxy-4-nucleobase-2,5-anhydro-L-mannitols 13-16 were synthesized regioselectively from 7 in good yields. 6-O-p-Tolylsulfonyl-2,5:3,4-dianhydro-L-talofuranose dimethylacetal 8 reacted with uracil or thymine to give the corresponding isonucleosides 17 and 19, but in the case of reaction of 8 with adenine, reformation of tetrahydrofuran ring took place, giving 4-(S)-adenyl-5-(R)-[1'-(R)-hydroxy-2',2'-dimethoxyl] ethyl-2,3-dihydrofuran 21 and 4-deoxy-4-adenyl-2,5:3,6-dianhydro-L-mannofuranose dimethylacetal 22. Abstract:A general route to either cis- or trans-2, 5-disubstituted tetrahydrofurans was described by using the nucleophilic addition of organolithium derivatives to tricyclic lactones, followed by a highly stereocontrolled acid-assisted reduction with sodium cyanoborohydride of the hemiketals formed. The stereoselectivity observed can be rationalized by the preferential approach of the hydride on the less hindered face of an oxonium ion intermediate. The cis- or trans-2, 5-disubstituted tetrahydrofurans were provided by the hot decomposition and then hydrogenation of tricycle compounds produced.

Abstract:A series of novel alkyl substituted fructose-based oxadiazoles were synthesized and their cytotoxic activities toward tumor cells were investigated. We studied the reaction mechanism and the stereochemistry of the reaction. Tautomerization between isomers 2 and 3 was observed in solution. The tautomerization was accelerated by heating or in the presence of acetic acid. An intermediate 6 during the heterocyclization was isolated, and two different pathways for the heterocyclization were suggested. On the basis of these findings, an efficient method, with the assistance of microwave irradiation was developed for the synthesis of 4 and 5. The yields were satisfactory and no by-products were found. We also proposed that the (R/S)-configurations of oxadiazoles were determined by the E/Z configurations of hydrazones.

Abstract:A convenient,high-yield synthesis of 1-O-acetyl-3,5-di-o-(p-chlorobenzoyl)-2-deoxy-α,β-D-ribofuranoside from 2-deoxy-D-ribose was reported.The product was a very important medical intermediate.Its anomers were separated by column chromatography.The optimum conditions were as follows: methyl-3,5-di-o-(p-chlorobenzoyl)-2-deoxy-α,β-D-ribofuranoside:acetic anhydride,1∶4.4;2 mL conc.sulfuric acid;temperature of solution when adding conc.sulfuric acid,(-2°C);hydrolysis temperature,(6°C).The yield was above 84.14%.The separated α and β anomers were characterized by()~1H NMR and melting point.

Abstract:A TFA-catalyzed dehydrofluorinative cyclization of 2,2-difluoro-3-hydroxy-1,4-diketones has been developed in the presence of amines under mild conditions in which the difluorinated substrates are readily prepared according to our reported literature. This protocol provides a rapid construction of fluoro 3(2H)-furanones in good to excellent yields with good functional group tolerance. Easy scale-up synthesis also shows a practical advantage.

Abstract:In order to optimize the synthesis and separation of anomers of 1-(diethyl)-2,5-anhydro-D-ribofuranosyl phosphonate,they were synthesized by Wadsworth-Emmons reactions in which two different protected D-ribose derivatives were used as the starting materials.In method A,2,3-O-isopropylidene-5-O-pivaloyl-D-ribofuranose reacted with tetraethyl methylenediphosphonate(TEMDP) to give 1-(diethyl)-2,3-O-isopropylidene-5-O-pivaloyl-D-ribofuranosyl phosphonate.Sodium ethoxide was used to remove the pivaloyl group in 81% yield.The α and β anomers were isolated and then hydrolyzed under acidic condition to afford the desired products in 94% and 91% yields,respectively.In method B,the D-ribose 1-phosphonates were obtained through the reaction of 2,3,5-tri-O-benzyl-D-ribose with TEMDP in 70% yield.After separating the anomers,the desired products were obtained through deprotection of α and β anomers with Pd/C and H_2 in 93% and 91% yields,respectively.The products were characterized by ~1HNMR to verify the feasibility of our synthesis.

Abstract:Objective To simplify the synthesis of 3-aryl-2,2-disubtituted dihydrobenzofurans.Methods It involves a selective cross McMurry reaction coupling of a 2-hydroxyphenyl-aryl-methanone with an aldehyde as a pivotal step and a sequential acid-promoted cyclization of the resulting ortho-vinylphenols.Results The structure of 3-aryl-2,2-disubtituted dihydrobenzofurans was confirmed by IR,1H-NMR,13C-NMR and HRMS(EI).An array of 3-aryl-2,2-disubtituted dihydrobenzofurans were conveniently and efficiently obtained with two-step total yields of 89%~98%.Conclusion The advantages of this synthetic approach are as follows: the reagents are readily available,the conditions are mild and the operations are simple,and the preparations are suitable for scale-up.

Abstract:Stereocontrolled synthesis of rare sugar derivatives, namely α-D-allulofuranosides, was achieved using d -fructose, one of the most abundant carbohydrates in nature. The following are the key steps of the α-D-allulofuranosides' synthesis. (1) An α-selective glycosidation reaction of 1,3,4,6-tetra- O -benzoylated d -fructofuranosyl donor to obtain α-D-fructofuranosides with 98 %–75 % isolated yields. (2) A regioselective 1,4,6-tri-O-pivaloylation reaction of the tetraol of α-D-fructofuranosides with the C3-hydroxy group remaining intact. (3) The oxidation of the C3-hydroxy group followed by the stereoselective reduction of the C3-carbonyl group. Primary and secondary alcohols and sugars can be used as glycosyl acceptors and aglycones for the following pivaloylation and stereoinversion reactions to obtain α-D-allulofuranosides.

A Low Cost Synthesis Of(e)-4-hydroxynon-2-enal

Abstract:The resin-catalyzed selective hydrolysis of 1,1,4,4-tetramethoxybut-2-ene,which was formed from the reaction of methanol and furan,resulted in the formation of 4,4-dimethoxybut-2-enal only.Key of this low cost technic is that no butendial was found in the hydrolysis product.Addition of pentylmagnesium bromide to 4,4-dimethoxybut-2-enal gave 1,1-dimethoxy-4-hydroxynon-2-ene,resin-catalyzed hydrolysis of which afforded the final product(E)-4-hydroxynon-2-enal in overall yield of 8.7% from furan.Structures of the final product and intermediates were characterized by ~1HNMR and ~(13)CNMR.

Abstract:2-Amino-7-hydroxytetralin monohydrate was synthesized from 7-methoxy-2-tetralone.7-Methoxy-2-tetralone reacted with benzylamine,then sodium borohydride was added for reduction and HCl was introduced for acidification to obtain hydrochloride of the benzyl-protecting amine.Hydrogenation of the hydrochloride was carried out with the existence of 5% Pd/C,and the hydrochloride of 2-amino-7-methoxytetralin was obtained.The salt was treated with alkali to obtain 2-amino-7-methoxytetralin,and the chiral mixture of 2-amino-7-methoxytetralin was separated by reacting with mandelic acid in absolute alcohol.The separated salt of the isomer with mandelic acid was neutralized with solution of NaOH,then hydrolyzed separately in solution of 48% hydrobromic acid,and the 2-amino-7-hydroxytetralin monohydrate was obtained separately after neutralizing with ammonia.The yields of both of the two isomers were up to 30%.

Abstract:We have developed a facile and effective synthesis of 3-iodofurans from 3-monosubstituted 1,2-allenyl perfluoroalkyl ketones or 2-hydroxy-4-iodo-2,5-dihydrofurans from 3,3-disubstituted 1,2-allenyl perfluoroalkyl ketones.
