Abstract:Oenococcus oeni (O. oeni) induces malolactic fermentation to improve wine quality. However, the molecular basis of mechanisms involved in its freeze-drying resistance remains unclear. In this work, O. oeni SD-2a without freeze-drying (No-FD), with freeze-drying (FD) and freeze-drying with monosodium glutamate (MSG) (FD-MSG) were investigated. Flow cytometry results showed severe cell damage in FD cells and less membrane transmissibility in FD-MSG cells. Meanwhile, extracellular polymeric substances (EPS) were detected in FD and FD-MSG cells by scanning electron microscopy. Bioinformatic analysis revealed that varying proteins were involved in carbon, lipid and nucleic acids metabolism, stress response, oxidoreductase activity and signal sensing. Among the identified proteins, the highlighted proteins were those involved in polysaccharides production and signal sensing for freeze-drying resistance and cyclopropane fatty acid metabolism for MSG addition.

Abstract:OBJECTIVE Several cryoprotectants were employed to study the protective effect on the freeze-drying process of the oleanolic acid-loaded nanosuspensions (OLA-LS) in order to select the optimum formulation. METHOD The protective effect was evaluated by measuring the mean particle size of samples before and after freeze-drying process. RESULT Sucrose with the concentration of 10% was selected as the optimum cryoprotectant. The average size of excellent sample was 236.3 nm (versus 211.2 nm of fresh one), and a much higher polydispersity index of 0.242 (versus 0.180). CONCLUSION The optimum lyophilized powder could be obtained with suitable type of cryoprotectant with appropriate concentration and proper freeze-drying conditions.

Abstract:Ethanol acclimations have been proven to be beneficial for enhancing the viability of lyophilized Oenococcus oeni. To explore the relevant mechanisms, cell integrity, protein patterns, morphology, exopolysaccharides (EPS) content of lyophilized O. oeni SD-2a with ethanol acclimations were investigated in this research. After freeze-drying, the improvement of cell integrity was found in all ethanol acclimated cells. Comparative proteomics combined with bioinformatics revealed the important role of central carbohydrate metabolism, polysaccharides production, cell envelope biosynthesis and stress responses related proteins with ethanol acclimations in freeze-dried O. oeni SD-2a. Extracellular polymeric substances were observed in mild ethanol (5% and 8%) adapted cells using scanning electron microscope. In addition, mild ethanol (5% and 8%) adapted cells showed higher EPS content. Transcriptional analysis showed that citrate lysis gene over-expressed in all ethanol acclimated freeze-dried cells. Therefore, citrate was added in culture medium and it is highlighted that the addition of citrate was useful for the growth of freeze-dried O. oeni SD-2a. These results provide new insights of molecular mechanisms regulated by ethanol acclimation and improve our understanding of biological characteristics involved in freeze-dried O. oeni.

Abstract:Malolactic fermentation (MLF) is an important process in wine production. To achieve successful MLF, expanding interest in ready-to-use Oenococcus oeni starter cultures has drawn greater emphasis on developing starter production and preservation methods. Growth phase, protective agents, rehydration media and stress pretreatments were the main factors to affect the survival of O. oeni SD-2a after freeze-drying. It was found that O. oeni SD-2a cells in the early stationary phase survived better than those in the mid-log phase after freeze-drying. Protective agents play an important role in the conservation of viability. Sodium glutamate (2.5%) was the best protectant, giving the cell viability 69.5%. The addition of polysaccharides and disaccharides in suspension media also improved the cell viability significantly. Rehydration is a critical step in recovery after freeze-drying. When freeze-dried O. oeni SD-2a was rehydrated in acidic tomato broth (ATB) medium, the highest viability was obtained. Rehydration in the tested disaccharide solutions decreased the cell viability obviously. During stress treatments, O. oeni SD-2a treated with 8% ethanol resulted in the highest freeze-drying survival rate (82%), the treatment with pH 3.5 or 3.2 also notably increased freeze-drying viability.

Abstract:To achieve successful malolactic fermentation in wine making, expanding interest in ready-to-use concentrated Oenococcus oeni starter culture has placed greater emphasis on developing starter production and preservation methods. The effect of growth factors on the survival of the O. oeni cells after lyophilization and its relationship with the membrane fatty acid composition were investigated. O. oeni cells in the early stationary phase survived better after freeze-drying than those in the mid-exponential phase, and those early stationary phase O. oeni cells cultured in designed ATB, FMATB and MATB growth media with different pH buffering capabilities exhibited different freeze-drying viabilities. Results concerning both membrane fatty acid composition and freeze-drying viability of O. oeni cells grown in those three media at initial culture pH 4.8, 4.0, 3.5, 3.2 were subjected to correlation analysis, indicating a decrease of the growth pH improved the freeze-drying survival of O. oeni cells, which was correlated with the enrichment of C19cyc11 and the decrease of C16:0 in the membrane lipids. A better understanding of the mechanisms of O. oeni resistance to lyophilization could rationalize their exploitation to prepare commercial starter cultures.

Abstract:Enterococcus faecium is one strain of normal microflora of human and animal intestinal tracts.The bacterial colony has regulative function to intestines.The application of Enterococcus faecium in probiotics is highly limited due to the number of vial bacteria and storage stability.In this study,the optimal cryoprotectant formula for Enterococcus faecium was explored using orthogonal array design and storage stability was tested at 4 ℃ and 37 ℃.The optimal cryoprotectant formula was composed of 5 g/100 mL skim milk,1 g/100 mL sucrose and 5 g/100 mL maltodextrin.The survival rate of Enterococcus faecium using this optimal formula in sealed aluminum foil bag with temperature of 37 ℃ and relative humidity of 75% was up to 97.90% after the storage for 4 weeks.

Abstract:The effects of different factors (including cell age, enzyme concentration, reaction temperature and osmotic stabilizers) on the preparation and regeneration of Oenococcus oeni SD-2a protoplast were studied by single-factor and orthogonal tests.. The results showed that the optimal conditions for protoplast preparation and regeneration were as follows: SD-2a cell age was 34 h, enzyme concentration was 1 mg/mL, enzymolysis temperature was 37℃, 0.8 mol/L KCl was used as osmotic stabilizer, and 0.8 mol/L KCl was also added in regeneration medium. Under above conditions, the formation ratio of protoplast times the regeneration ratio of protoplast is 10.65%.

Abstract:The protective agent formula of freeze-drying for Lactobacillus plantarum NCU116 was optimized by single factor experiment and a three-factor Box-Behnken design.The results of single factor experiment indicated that trehalose,D-sorbitol and the skim milk had significant effect to the survival of freeze-drying cells.The critical factors selected for this investigation were the skim milk,trehalose,and D-sorbitol,the cell survival rate used as response value.The optimal formula was skim milk concentration 9.24%,trehalose concentration 4.47% and D-sorbitol concentration 13.42%.The efficiency of protective agent formulation optimzed has been tested.The result showed that the cells survival rate reached 91.76%±1.82% in freeze-drying process,it more in line with the theoretical prediction.

Abstract:To establish an efficient and stable method for protein extraction from Oenococcus oeni,collected bacteria were used as raw materials and protein was extracted after treatment with ultrasonic,repeated freeze and thaw and glass-beads broken,respectively.Efficiency of different methods was evaluated with indexes,such as rate of bacteria broken,concentration of extracted protein and pattern of SDS-PAGE of protein.It was showed that treatment with ultrasonic led to significant broken rate of Oenococcus oeni cells,high concentration of extracted protein.And the method was stable,reproducible and easy to handle.

Abstract:In order to minimize the damage of cell of Lactobacillus casei subsp. rhamnosus719 during freeze-drying, this study was undertaken to choose proper cryoprotectance and optimize the cryoprotectance. The effects of the cryoprotectances on the survival rate of this strain were investigated. The result of single-factor test showed that, the effect of seven cryoprotectances follows the order as that: D-Trehalose, Skim Milk, Sucrose, Glycerol, Soluble starch, MnSO4, Vc. The best composition was: 10 % D-Trehalose +10 % Skim Milk,in which the survival rate of the bacteria was 100 % and the amount of live bacteria in freeze-dried power was 1011 g-1.

Abstract:Freeze drying has a deleterious effect on the viability of microorganisms. In front of this difficulty, the present study adopts response surface methodology to optimize the chemical compositions of protective agents to seek for maximum viability of Bifidobacterium longum BIOMA 5920 during freeze-drying. Through the comparative analysis of single protectant, the complex protective agents show better effect on the Bifidobacterium viability. Human-like collagen (HLC), trehalose and glycerol are confirmed as significant factors by Box-Behnken Design. The optimized formula for these three variables is tested as follows: HLC 1.23%, trehalose 11.50% and glycerol 4.65%. Under this formula, the viability is 88.23%, 39.67% higher in comparison to the control. The viable count is 1.07×109 cfu·g−1, greatly exceeding the minimum viable count requirement (106 cfu·g−1).

Abstract:OBJECTIVE:To achieve the high quality of malo-lactic starter cultures, we investigated the effect of three culture media on the direct inoculation viability, freeze-drying viability and membrane fatty acid composition of Oenococcus Oeni SD-2a.METHODS:We monitored the bacterial growth and change in medium pH when O. oeni SD-2a cells were cultured in ATB, FMATB and MATB media. O. oeni SD-2a cells in early stationary phase were harvested, and subjected to direct inoculation experiments and freeze-drying processes. Then we determined inoculation and freeze-drying viability. Membrane fatty acid composition of those corresponding O. oeni SD-2a cells was determined by GC/MS method.RESULTS:The results showed ATB medium without supplementation of DL-malate had weak pH buffering capability. Compared with FMATB and MATB, O. oeni cells cultured in ATB increased inoculation viability and freeze-drying viability. Concerning the membrane fatty acid composition, it was observed that ATB medium increased distinctly the relative concentration of lactobacillic acid (C19cyc11) and U/S (the unsaturated: saturated fatty acid) ratio in cell membrane lipid composition of O. oeni SD-2a.CONCLUSION:The increased resistance to wine stressor and freeze-drying was probably a result of the cross protection conferred by self acid stress response induced in ATB medium, which might be related with changes in membrane fatty acid composition of O.oeniSD-2a. Therefore, ATB medium was more suitable for preparation of O. oeni SD-2a commercial starter cultures.

Abstract:During the vintages of 1999 and 2000, isolation and screening of Oenococcus oeni from the. main wine producing regions of China were carried out. After conformed the isolates on malolactic fermentation (MLF) ability, the 127 isolates were tested for their capacity to resist to the individual stress condition of low pH, high ethanol and SO2 content. According to the screening results, the eight selected strains'growth under the combination of external stress physical conditions of ethanol %(v/v), pH and SO2 (mg center dot L-1) i.e. ethanol X pH X SO2 were also tested, the results showed that there were four finally selected strains had good adaptation to adverse conditions. Especially, the strain O. oeni SD-2a had much high adaptability to stress environments than commercial strain O. oeni 31DH.

Abstract:To develop cryopreservation for Oenococcus oeni.We investigated the influence of the age and concentration of the strain in the protective medium, rate of freezing, temperature of thawing and the protective medium on the number of viable cells after cryopreservation by colony counting.We got the highest number of viable cells after cryopreservation by collecting cells in the early stationary growth phase, keeping their concentration in the protective medium at 10(9) CFU/mL, transferring cells into liquid nitrogen directly and thawing in a water bath at 37 degrees C. The ratio of viable cells was above 99% for 21 of the Oenococcus oeni after storing in liquid nitrogen for 6 months.We The storage of Oenococcus oeni in liquid nitrogen is convenient for strains preserved in culture collections.

Abstract:Malolactic fermentation (MLF) by Oenococcus oeni is an important process for the production of high-quality wine, and the preparation of a high cell viability starter is of great significance to this process. The key way to obtain a high cell viability starter is to select an effective lyoprotectant. In this study, by optimizing the ratio of lyoprotectant and improving its pH, prefreezing temperature, and period of the growth phase, the number of viable bacteria of O. oeni was 1.08 +/- 0.04 X 10(11) CFU/g. MLF experiments on the lab scale and the pilot scale were carried out. Organic acids, fermented aroma volatiles, and sensory scores of samples during MLF were analyzed, and the results showed that there was no significant difference between the lab-scale and pilot-scale MLF. Both O. oeni ES-1 and commercial starters successfully completed MLF, but the aroma volatile type, aroma volatile concentration, and sensory score of O. oeni ES-1 were excellent. In addition, the fermentation capacity of O. oeni ES-1 in the pilot scale was studied first.

Abstract:To improve the preparation technology of direct vat set in industrial production, the liquid nitrogen cryogenic treatment was used in the preparation ofLactobacillus bulgaricus B9 starter. The optimal formula of freeze protectant, which was determined with screening different kinds of freeze protectant by single-factor experiments and optimizing the concentration of preferred freeze protectant by orthogonal test were skim milk powder 10%, Glcerol 3%, Trehalose 5%, Vc 0.075%(m:m). The viability of bacteria cells can reach 92% after freezing with our freeze protectant.
