Abstract:Under climatic conditions dry and hot river-valley regions of southwest China,which are typical of the south subtropical climate,four grapevine varieties,Merlot,Fox,Chardonnay and Gamay,began their physiological dormancy in early October,middle October,early November and middle November,respectively;they began their ecological dormancy(bud breaking rate50%) approximately in early December,middle December,early January in the follwing year,and middle December,respectively.During grapevine dormancy,the starch content and total sugar of the young shoots tended to change in the following manner.With deepened grapevine dormancy,the starch content increased while the total sugar decreased;with broken dormancy,the total sugar increased while the starch content decreased;And thus the two showed opposite tendencies.The results provided the theoretical basis for breaking grapevine dormancy and then making rain-avoiding grapevine production.
Variation of Active Oxygen Metabolism, Anti-oxidation System in Grapevine Buds Tissue During Bud Dormancy Inducing and Releasing

Abstract:This study aimed to investigate the bud dormancy mechanisms of wine grape during natural and breaking dormancy from the following physiological and biochemical aspects:water status,active oxygen and ant-oxidation system,phenols and the related enzymes.The results show that:(1)Chardonnay which is an early maturity variety comes into deep dormancy on Dec.20th,and bud dormancy starts to release gradually from Jan.29th;the late Cabernet Sauvignon's deep dormancy stage is from Jan.9th to Feb.18th and bud dormancy starts to release gradually.Cyanamid treatment enhanced dormancy release.(2)The contents of active oxygen species O-·2 and H2O2 had a gradual increase in the early dormancy stage,reaching the peak value at the deep end-dormancy,then decreased after bud dormancy release.SOD activity decreased steadily during the whole period of end-dormancy,and then increased after the dormancy release.POD and CAT activities kept increasing during the early dormancy stage,then the POD value started to drop after the dormancy release and CAT maintained a high activity until bud sprouting.ASA-GSH cycle was relatively weak at the early stage of end-dormancy,and then it was activated gradually during the dormancy release.During the breaking dormancy,the content of active oxygen,the activities of anti-oxidation system enzymes and the contents of antioxidants are changed,all of these are better for dormancy releasing.
Effects of Chemicals on Dormancy Breaking of Wine Grape Cultivars under Dry-Hot Climate in Southwestern China

Abstract:In the winter from the year 2001 to 2002, dormant buds of four grapevine cultivars grown in Panzhihua were. treated immediately after pruning with dormancy-breaking chemicals. The data of next year's sprouting percentage indicate that the best time to treat is at the lowest temperature in winter and (1.0%H2CN2+Tween 80) has the best effects on improving sprouting percentage. Moreover, four wine grapevine cultivars with different maturity phases have different effects on improving sprouting percentage, when treated with dormancy-breaking chemicals. From the first to the last, the list of cultivars is early-mid ripe Gamay, mid-late ripe Merlot, early ripe Chardonnay, late ripe Fox according to the improved sprouting percentage.
Effects of Dormancy Breaking Chemicals on Fructification of Wine Grapevine under Dry-Hot Climate in Southwest China

Abstract:【Objective】 The study was done to know the influence of chemical treatments on grapevine fructification enhancement.【Method】 In December 2003 to January 2004 and December 2004 to January 2005,dormant buds of four grapevine cultivars grown in Panzhihua were treated immediately after pruning with dormancy breaking chemicals.The data of next year's fructification coefficient were investigated and analyzed.【Result】 The result indicated that the best treat time to improve fructification was 01-10 and(1.0%H2CN2+1.0%Tween 80) had the best effects of improving fructification coefficients.Moreover,four wine grapevine cultivars in experiment had different responses to dormancy-breaking chemicals.In treated grapevines,only fructification coefficients of Merlot and Gamay were increased.The highest improving extent of fructification coefficient in Merlot was above 30% and that of Gamay was about 7%.Dormancy-breaking chemicals had no effect on improving fructification for Chardonnay and would decrease fructification of Fox.【Conclusion】 The treat time of 01-10 is the best and the chemical of(1.0%H2CN2+ 1.0%Tween 80) is the best agent to improve the fructification of wine grapes in Panzhihua-Xichang Area.
Comparative Transcriptome Analysis Provides Insight into Differentially Expressed Genes Related to Bud Dormancy in Grapevine (vitis Vinifera)

Abstract:Bud dormancy is an important adaptive strategy of perennial woody plants that benefits survival of a species under unfavorable conditions. Grapevine is one of the most important fruit species with wide distribution all over the world. There are two kind of buds, latent buds and prompt buds, along shoots of grapevine, which show different phenotypes in terms of dormancy. To better understand the molecular mechanisms behind this, the transcriptomes of grapevine (Vitis vinifera) latent buds and prompt buds were analyzed using RNA-sequencing (RNA-seq) technology and compared. Of all the genes detected, 4864 were identified as differentially expressed genes (DEGs), with 2613 being upregulated and 2251 being downregulated in latent buds than in prompt buds. Gene Ontology (GO) enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed that these DEGs were mostly associated with biological process, single-organism transport, lipid metabolic process and transporter activity, and also involved in biosynthesis of secondary metabolites, metabolism of phytohormone, flavonoids, phenylpropanoid, sugar and fatty acid. Lots of transcription factors (TFs), especially some members of MTh, TCP, and DAM, were identified to be candidates regulating grapevine bud dormancy. Results of transcriptome profiling were validated using real-time PCR in 27 selected genes. The results revealed that interactions of phytohormone, sugars and flavonoids and their regulation to the cell-cycle genes via the mediation of TFs may contribute greatly to grapevine bud dormancy. Consequently, this study provides abundant genetic resources and lays a foundation for further research on the molecular mechanisms underlying bud dormancy in grapevine.

Abstract:This article studied the variation of water and carbohydrate compounds of grape winter buds during bud dormancy,taking as samples the grapevine varieties Vitis vinifera L.cv Cabernet Sauvignon and Vitis vinifera L.cv Chardonnay.The results indicated that in the climate conditions of Shaanxi Yangling Region,the two grapevine varieties,Cabernet Sauvignon and Chardonnay reached the deep dormancy respectively at December 20 and January 9,and then started the stage of breaking dormancy.Along with the deepening of bud dormancy,the amount of total water,free water and carbohydrate compounds in winter buds of two varieties decreased,but the amount increased in the stage of breaking dormancy.On the other hand,the amount of bound water,the rate of bound water/free water and soluble sugar in winter buds showed an opposite tendency.With the application of Dormex (hydrogen cynamide),free water amount in winter buds increased while bound water decreased and the effect was more obvious on Cabernet Sauvignon than Chardonnay.But the variations of total water,soluble sugar and starch contents are not significant during bud dormancy.It can be concluded that water content and condition,changes of soluble sugar and starch in grapevine winter buds appear notable relevance with the process of bud dormancy and release,and the application of Dormex (hydrogen cynamide) was effective to break the grapevine bud dormancy.

Effects of budbreaking chemicals on sprouting percentage of winemaking grape varieties under dry-hot climate in Southwest of China

Abstract:【Objective】 The research is to study the nitrogen metabolism in Cabernet Sauvignon during the period of dormancy.【Method】Taking Cabernet Sauvignon as experimental materials,this research was conducted to investigate the variation of nitrogenous compounds and relative enzymatic activity in the buds,phloem and xylem of one-year-old cane,and roots during the period of dormancy induction and release in Yangling region.Measure the indexes of the nitrides,total nitrogen,total protein,soluble protein content,nitrogen metabolism-related enzyme activity,nitrate reductase activity,protease activity were determined,and the proteins in various grape organs at dormant phase were analyzed by using SDS-polyacrylamide gel electrophoresis.【Result】The contents of total nitrogen,protein and soluble protein in different parts of grape from high to low were budsrootsphloemxylem,especially total nitrogen and protein in buds increased obviously.Contents of soluble proteins decreased gradually during the dormant induction,and increased gradually during the dormant release.Nitrate reductase activity of different dormant organs or tissues from high to low were budsrootsphloemxylem.From dormancy to germinating stage,nitrate reductase(NR)activity generally decreased.In different organs of grapevine,glutamine synzyme activity in buds was maximal,during the period of deep dormancy,the glutamine synzyme activity was lowered to a minimum,the general trend of glutamine synzyme activity in buds was rising with two peaks during the dormant period.In the early dormancy,protease activity decreased with the deepening of sleep,and increased with the lifting of dormancy and germination(except buds).A large number of proteins emerged in the period of grape dormancy.【Conclusion】The buds,one-year old canes phloem,xylem and nitrogen compounds content,type and enzyme activity of the root changed before and after dormancy in the natural process of dormancy and germination.

Abstract:To investigate dormancy and release mechanisms of grape seeds,we compared seed morphologi-cal characteristics and germination behavior of different grape varieties and their dormancy release under different conditions including an alternative temperature change simulation for a season cycle.The grape va-rieties tested included species introduced from three distribution centers(America,Europe-Asia and East-Asia)and their cross breeds.Seed dormancy of all varieties was attributed to physiological dormancy,but varied in degree of dormancy.The dormancy-releasing requirements of different varieties also differed,correlating to their genetic backgrounds.As a whole,seeds of Europe-Asia species and East-Asia species ex-hibited a lower degree of dormancy,compared to a relatively high degree of dormancy for American species.For Europe-Asia species,East-Asia species and their cross breeds,seed dormancy was effectively broken by cold stratification(5oC)and partially broken by alternative temperature stratification viz.20oC(14 h)/10oC(10 h)and 30oC(14 h)/20oC(10 h)for two months.As for American species and cross breeds with Europe-Asia species,seed dormancy could only be broken by more than six-month cold stratification;neither alternative temperature stratification nor warm stratification(25oC)affected seed dormancy release.Experi-ments simulating seasonal alternative changes further showed that seed dormancy degrees were related to their proveniences and adaptations to environment.Our results provide basic data for germplasm introduction and breeding of grapes.
Influence of Cyanamide on Polyphenol, Polyphenol Oxidase and Free Phenol in Different Places of Grapevine During Dormancy Induction and Release

Abstract:A research was conducted to investigate the regularity variation of total phenolics,activities of PPO and PAL in dormant grapevine buds.At the same time,six mono-phenolics were detected with switching detection wavelength in HPLC for dormant buds,phloem and xylem of one-year-old grapevine canes treated with cyananmide.It indicated that during the early dormancy stage,PPO,PAL activities and total phenolic content were increasing with the accumulation of low temperature,PAL activity and total phenolic slightly decreased and showed a similar trend during the dormancy release.On the whole,PPO activity was kept at a high level in this stage.No different effects were detected on the treatment with cyanamide.During the Pinot Noirs whole dormancy stage,except for trans-resveratrol,the contents of gallic acid,protocatechuic acid,(+)-catechin,epicatechin,(-)-epicatechin-3-O-gallate showed a downward trend in phloem;the contents of gallic acid,protocatechuic acid,epicatechin showed an upward trend in xylem and trans-resveratrol showed a downward trend,(+)-catechin showed a W-shape trend,(-)-epicatechin-3-O-gallate showed no obvious change;in winter bud,the contents of gallic acid,(+)-catechin showed an upward trend,protocatechuic acid,epicatechin,(-)-epicatechin-3-O-gallate and trans-resveratrol showed a downward trend,but protocatechuic acid,epicatechin,(-)-epicatechin-3-O-gallate(-)-epicatechin showed no change before bud break and dropped dramatically after budding.It was inferred gallic acid might promote the dormancy releasing,while trans-resveratrol might inhibit the dormancy releasing.
The VvWRKY37 Regulates Bud Break in Grape Vine Through ABA-Mediated Signaling Pathways

Abstract:Dormancy is a common survival strategy in plants to temporarily suspend visible growth under unsuitable conditions. The elaborate mechanism underlying bud break in perennial woody plants is gradually illustrated. Here, we identified a grape vine WRKY transcription factor, VvWRKY37 , which was highly expressed in dormant buds. It was particularly induced by the application of exogenous abscisic acid, and depressed on exposure to gibberellin and low temperature (4°C) stress at the transcript level. The yeast one-hybrid assay confirmed that VvWRKY37 had a transcriptional activity. Ectopic over-expression of VvWRKY37 significantly delayed bud break of transgenic poplar plants. As an ABA-inducible gene, VvWRKY37 also depressed the expression of ABA catabolic gene CYP707A s and enhanced the accumulation of endogenous ABA in transgenic poplar plants. The molecular pieces of evidence showed that VvWRKY37 preferentially recognized and bound W-box 5′-G/CATTGACT/C/G-3′ cis -element in vitro . Additionally, VvABI5 and VvABF2 acted as the upstream transcriptional activators of VvWRKY37 via protein-DNA interactions. Taken together, our findings provided valuable insights into a new regulatory mechanism of WRKY TF by which it modulates bud break through ABA-mediated signaling pathways.

Abstract:BackgroundBud dormancy is an important biological phenomenon of perennial plants that enables them to survive under harsh environmental circumstances. Grape (Vitis vinifera) is one of the most grown fruit crop worldwide; however, underlying mechanisms involved in grape bud dormancy are not yet clear. This work was aimed to explore the underlying molecular mechanism regulating bud dormancy in grape.ResultsWe have performed transcriptome and differential transcript expression analyses of “Shine Muscat” grape buds using the Illumina RNA-seq system. Comparisons of transcript expression levels among three stages of dormancy, paradormancy (PD) vs endodormancy (ED), summer buds (SB) vs ED and SB vs PD, resulted in the detection of 8949, 9780 and 3938 differentially expressed transcripts, respectively. Out of approximately 78 million high-quality generated reads, 6096 transcripts were differentially expressed (log2 ratio ≥ 1, FDR ≤ 0.001). Grape reference genome was used for alignment of sequence reads and to measure the expression level of transcripts. Furthermore, findings obtained were then compared using two different databases; Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG), to annotate the transcript descriptions and to assign a pathway to each transcript. KEGG analysis revealed that secondary metabolites biosynthesis and plant hormone signaling was found most enriched out of the 127 total pathways. In the comparisons of the PD vs ED and SB vs ED stages of grape buds, the gibberellin (GA) and abscisic acid (ABA) pathways were found to be the most enriched. The ABA and GA pathways were further analyzed to observe the expression pattern of differentially expressed transcripts. Transcripts related to the PP2C family (ABA pathway) were found to be up-regulated in the PD vs ED comparison and down-regulated in the SB vs ED and SB vs PD comparisons. GID1 family transcripts (GA pathway) were up-regulated while DELLA family transcripts were down-regulated during the three dormancy stages. Differentially expressed transcripts (DEGs) related to redox activity were abundant in the GO biological process category. RT-qPCR assay results for 12 selected transcripts validated the data obtained by RNA-seq.ConclusionAt this stage, taking into account the results obtained so far, it is possible to put forward a hypothesis for the molecular mechanism underlying grape bud dormancy, which may pave the way for ultimate improvements in the grape industry.

Abstract:The experiment on breaking dormancy of Chinese wild grapes with Ca(CN)2 was conducted. The results show that 5-10% Ca(CN)2 had optimal effect on breaking dormancy of clones Liuba-7, Liuba-1, Jiangxi-1 (green down, ) and Tonghua-3, but 20% Ca(CN)2 was optimal for Pingli-7. When the same concentration of Ca(CN) 2 (15%) was applied to 10 clones of Chinese wild grapes, the results show that only the budding rate of Lueyang-4 of V. qinlingenesis had been increased, but the rest had been little affected, some even been inhibited.

Abstract:Bud dormancy is one of the most important defensive mechanisms through which plants resist cold stress during harsh winter weather. DAM, Dof, and WRKY have been reported to be involved in many biological processes, including bud dormancy. In the present study, grapevine (Vitis vinifera) and other thirteen plants (six woody plants and seven herbaceous plants) were analyzed for the quantity, sequence structure, and evolution patterns of their DAM, Dof, and WRKY gene family members. Moreover, the expression of VvDAM, VvDof, and VvWRKY genes was also investigated. Thus, 51 DAM, 1,205 WRKY, and 489 Dof genes were isolated from selected genomes, while 5 DAM, 114 WRKY, and 50 Dof duplicate gene pairs were identified in 10 genomes. Moreover, WGD and segmental duplication events were associated with the majority of the expansions of Dof and WRKY gene families. The VvDAM, VvDof, and VvWRKY genes significantly differentially expressed throughout bud dormancy outnumbered those significantly differentially expressed throughout fruit development or under abiotic stresses. Interestingly, multiple stress responsive genes were identified, such as VvDAM (VIT_00s0313g00070), two VvDof genes (VIT_18s0001g11310 and VIT_02s0025g02250), and two VvWRKY genes (VIT_07s0031g01710 and VIT_11s0052g00450). These data provide candidate genes for molecular biology research investigating bud dormancy and responses to abiotic stresses (namely salt, drought, copper, and waterlogging).
