Abstract:To find out the efficient peptide of selenium-rich silkworm pupas in inducing the apoptosis of hepatoma cells, selenium-containing proteins were isolated and characterized. One of the two major proteins in selenium-rich silkworm pupas was identified to be a new protein by peptide mass fingerprinting on matrix assisted laser desorption ionization - time of flight - mass spectrometry. Amino acid sequences of peptides digested by trypsin from the new protein were determined by capillary liquid chromatography electrospray ionization - quadrupole/time of flight - mass spectrometry and searched with Mascot in NCBI database. Results showed that the six major peptides from the protein were also new peptides that could not be found in the database up to date

Abstract:According to the large-scale sequencing of cDNA library from silkworm pupae, the cDNA of a novel gene with blank research background was identified and temporarily named Bm-X. The length of this cDNA is 778 bp. We obtained its ORF for further study by bioinformatics analysis. It is 444 bp and encodes 147 amino acid residues, with a predicted molecular weight (MW) of 16.4 kD and an isoelectric point (pI) of 3.69. In this study, we successfully constructed a recombinant plasmid pET-28a(+)-Bm-X and expressed it in Escherichia coli. We used the fusion protein rBm-X which purified by Niaffinity chromatography to produce polyclonal antibodies against Bm-X for Western blot analysis. The analysis revealed that Bm-X was expressed in the larval midgut, the epidermis and the silk gland. In addition, the subcellular localization analysis of silkworm ovary epithelial cells (BmN cells) showed that Bm-X protein was located both in cytoplasm and nucleus, and the signal was stronger in cytoplasm than in nucleus. Our findings indicate that Bm-X gene is a novel species-specificity gene and its expression product can be detected in tissues of the fifth silkworm instar larvae and BmN cells.

Abstract:Human growth hormone (hGH) is a peptide hormone secreted by eosinophils of the human anterior pituitary, and a regulatory factor for a variety of metabolic pathways. A 30-kD protein from the pupa stage of silkworm was detected by Western blotting and confirmed by immunoprecipitation based on its ability to bind to anti-hGH antibody. This protein, named BmhGH-like protein, was purified from fresh silkworm pupas through low-temperature homogenization, filtration, and centrifugation to remove large impurity particles. The supernatants were precipitated, resuspended, and passed through a molecular sieve. Further purification by affinity chromatography and two-dimensional electrophoresis resulted in pure protein for analysis by MS MALDI-TOF-MS analysis. An alignment with predicted proteins indicated that BmhGH-like protein consisted of two lipoproteins, which we named hGH-L1 and hGH-L2. These proteins belong to the β-trefoil superfamily, with β domains similar to the spatial structure of hGH. Assays with K562 cells demonstrated that these proteins could promote cell division in vitro. To further validate the growth-promoting effects, hGH-L2 was cloned from pupa cDNA to create recombinant silkworm baculovirus vBmNPV-hGH-L2, which was used to infect silkworm BmN cells at low titer. Flow cytometric analysis demonstrated that the protein shortened the G0/G1 phase of the cells, and enabled the cells to rapidly traverse the G1/S phase transition point to enter S phase and promote cell division. Discovery of hGH-like protein in silkworm will once again arouse people's interest in the potential medicinal value of silkworm and establish the basis for the development of new hormone drugs.

Abstract:A synthetic gene encoding a chimeric silklike protein was constructed that combined a polyalanine encoding region (Ala)(18), a sequence slightly longer than the (Ala)(12-13) found in the silk fibroin from the wild silkworm Samia cynthia ricini, and a sequence encoding GVGAGYGAGAGYGVGAGYGAGVGYGAGAGY, found in the silk fibroin from the silkworm Bombyx mori. A tetramer of the chimeric repeat sequence encoding a similar to29 kDa protein was expressed as a fusion protein in Escherichia coli. In comparison to S. c. ricini silk, the chimeric protein demonstrated improved solubility because it could be dissolved in 8 M urea. The purified protein assumed an alpha-helical structure based on solid-state C-13 CP/MAS NMR and was less prone to conformational transition to a beta-sheet, unlike native silk proteins from S. c. ricini. Model peptides representing the crystalline region of S. c. ricini silk fibroin, (Ala)(12) and (Ala)(18), formed beta-sheet structures. Therefore, the solubility and structural transitions of the chimeric protein were significantly altered through the formation of this chimeric silk. This experimental strategy to the study of silk structure and function can be used to develop an improved understanding of the contributions of protein domains in repetitive silkworm and spider silk sequences to structure development and structural transitions.

Abstract:A Bombyx mori (B. mori) cDNA was isolated from silkworm pupae cDNA library encoding a homologue of translationally controlled tumor protein (BmTCTPk). BmTCTPk was expressed in E. coli; SDS-PAGE and Western blot showed the molecular weight of recombinant and native BmTCTPk is approximately 28 and 25 kDa, respectively; they are larger than the theoretical molecular weight. Immunohistochemical studies showed that BmTCTPk is uniformly distributed throughout the cytoplasm of BmN cells. In silkworm pupae, BmTCTPk is expressed in the midgut wall, the midgut cavity, and some fat body tissues lying between the midgut wall and body wall. Western blot and ELISAs performed on total protein extracts isolated from silkworm pupae at different development stages showed that, although BmTCTPk is expressed during all pupae stages, its expression level increases dramatically during late pupae stages, suggesting that BmTCTPk may play an important role during the developmental transition from pupa to imago.

Abstract:Leucine-rich repeat (LRR) proteins play important roles in the transduction of cellular signals and activation of defense responses. By scanning the cDNA library of silkworm (Bombyx mori) pupae constructed in our laboratory, we identified a 1557 bp gene that encodes a protein homologous to the death-associated small cytoplasmic leucine-rich protein, which was named as BmDSCLP. The full-length gene (GenBank accession no. FJ602779) contained a 642 bp open reading frame (ORF) encoding 213 amino acid residues. The ORF of this gene was inserted into the prokaryotic expression vector pET-28a(+) to construct a recombinant expression plasmid and the fusion protein was expressed in Escherichia coli BL21(DE3) cells. The fusion protein was purified by Ni-affinity chromatography and fast protein liquid chromatography (FPLC) and its size was then, determined by liquid chromatography-mass spectrometry (LC/MS/MS) and found to be 27.74 kD. Polyclonal antibodies were raised by subcutaneous injection of the recombinant protein into New Zealand white rabbits and the titer reached 1:12800. Analysis of the subcellular localization of the BmDSCLP protein revealed that, the protein was localized in both the cytoplasm and nucleus, but the amount in the former was slightly higher than that in the latter. In addition, real-time fluorescence quantification polymerase chain reaction studies were conducted to investigate BmDSCLP transcription at different developmental stages and in different tissues of the fifth instar larva. The results indicated that, BmDSCLP is widely transcribed in different stages and tissues of the silkworm. Analysis of stage-specific transcription patterns indicated that, the transcriptional level of BmDSCLP was highest in adults and lowest in eggs. Analysis of tissue-specific transcription patterns revealed that, the transcriptional level of BmDSCLP was highest in genital organs and lowest in silk glands. These results suggest that BmDSCLP plays important roles in the reproductive development of B. mori.

Abstract:[Objective] In screening the pupa cDNA library, a novel gene Bm595 was identified, the tissue distribution and localization of Bm595 were analyzed in silkworm. [Method] The recombinant plasmid was transformed into E.coli BL 21 which were induced with LPTG The purified fusion protein was used to immune a rabbit and the polyclonal antibody was prepared. The tissue distribution of Bm595 was analyzed by real-time PCR and Western blot. [Result] The quantitative real-time PCR results indicated that the transcription level of Bm595 was the highest in the fifth instar larva and the lowest in egg. The quantity of Bm595transcription from high to low was testis, silk gland, gut, head, ovary, fat body, epidermis, and air tube, respectively. Western blot results showed that Bm595 was highly expressed in testis, moderately expressed in silk gland and head. Bm595 also existed in fat body. Bm595 was mainly localized in nucleus by irnmunocytochemistry. [Conclusion] A preliminary analysis on Bm595 was made at the mRNA, protein expression and cell level which laid a good foundation for the further studies on Bm595 function.

Abstract:An unique new Bombyx mori cDNA was identified from the pupal cDNA library constructed in our laboratory.Sequence analysis revealed that the full-length cDNA is 1 011 bp long with 200 bp 5′-UTR and 136 bp 3′-UTR.This gene codes for a protein with 224 amino acids which is rich in cysteine at the N-terminus and was thus named BmCRP(Bombyx mori cysteine-rich protein) gene(GenBank accession number DN985186.1).This protein has such secondary structures as α-helix,β-pleated sheet and random coil and possesses 5 functional sites including cAMP-and cGMP-dependent protein kinase phosphorylation site.The ORF of the gene was cloned into pET-28a(+) vector and transformed into E.coli BL21(DE3).Recombinant protein was expressed successfully in E.coli BL21(DE3) induced by IPTG with the final concentration of 1 mM.Polyclonal antibodies were generated by immunizing New Zealand rabbit with purified recombinant protein.Western blotting analysis showed that the antibody could bind His-BmCRP specifically.Expression of the protein was observed in various developmental stages including egg,larva,pupa and moth,among which pupa had the highest expression level.The expression was also observed in tissues such as epidermis,brain,ovary,testes and Malpighian tubule of the fifth instar larvae.Real-time quantitative PCR results indicated that the expression level of BmCRP mRNA in silkworm egg,larva,pupa and moth increased gradually;and that in epidermis,brain,ovary,testis,midgut,Malpighian tubule,trachea,silk gland and fat body of the fifth instar larvae decreased gradually.Immunocytochemistry in Bm5 cells showed that BmCRP existed in the nucleus and cytoplasm.The fluorescent signal from nucleus was stronger than that from cytoplasm.These results laid a good foundation for further study on BmCRP′s structure and function in silkworm.

Abstract:The peritrophic membrane lines the gut of most insects at one or more stages of their life cycles. It facilitates the digestive processes in the guts and protects from invasion by pathogens or food particles. In the current study, a novel PM protein, designated as BmMtch, was identified from the silkworm, Bombyx mori. The open reading frame of BmMtch is 888 bp in length, encoding 295 amino acid residues consisting of two domains (Mito_carr domains) and three transmembrane regions. They are localized on the 11th chromosome as single copy with one exon only. Quantitative real time PCR analysis (qRT-PCR) revealed that BmMtch was mainly expressed in larval fat bodies, Malpighian tubules, testis and ovaries, and could be detected through all stages of the life cycle of silkworm. Immuno-fluorescence analysis indicated that BmMtch was localized within the goblet cell of larval midgut. Western blotting analysis showed that BmMtch were detected in total proteins of PM and larval midgut. The characteristics of BmMtch indicated that BmMtch represents a novel member of insect PM proteins, without chitin-binding domains.
A Proteomic Analysis to the Substance in Middle Silk Gland Lumen of Silkworm Varieties with High Reelability

Abstract:Cocoon filament reelability is one of the important economic traits of silkworm(Bombyx mori).To find specific proteins related to high cocoon filament reelability,the silkworm strain B84(yellow cocoon color) with high cocoon filament reelability and Fengyi(white cocoon color) with normal cocoon filament reelability were used as materials to conduct proteomic analysis and identification on the substance in middle silk gland lumen from the mature larvae of these two silkworm strains by utilizing two-dimensional electrophoresis(2-DE) and matrix assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS).341 and 363 protein spots were detected from the substance in middle silk gland lumen of Fengyi and B84 respectively.These proteins included a large number of functional proteins which have no relationship with sericin.For instance,there were serine protease inhibitors that have protective effect to silk fibroin.In the substance of middle silk gland lumen of B84,apart from carotenoid binding proteins that are involved in cocoon color formation,specific Sericin 2 protein and carbohydrate metabolism related enzymes were also identified.It was inferred that these proteins might be the underlying reason for high cocoon filament reelability of B84 due to the existence of Sericin 2 in cocoon shell as well and the influence of glycan chain on protein adhesion.
In Silico Identification of Silkworm Selenoproteomes

Abstract:Selenium (Se) is an essential trace element in vivo. Its biological function is mainly exerted through selenoproteins. Selenocysteine (Sec), the active site of selenoproteins, is incorporated into the protein at an in-frame TGA codon under the guidance of Sec insertion sequence (SECIS) element in the 3′-untranslated region (UTR) of the gene. In this work, a method was developed and a series of programs were edited by PERL language to in silico identify selenoproteomes from the genome of domesticated silkworm (Bombyx mori). Out of 18510 annotated genes, 6348 was terminated with TGA codons, 249 containing both in-frame TGAs and SECIS elements in the 3′-UTRs. Alignments of those selenoprotein candidates with their cysteine (Cys)-containing homologs revealed that 52 genes had TGA/Cys pairs and similar flanking regions around the in-frame TGAs. Restricted by the pattems of SECIS elements only 5 genes were screened out to fully meet the requirements for selenoproteins. Among them glutathione S-transferase (GST) has been reported as a microbial selenoprotein, the other four are novel selenoproteins annotated as CG6024, CG5195, ATP-binding cassette transporter subfamily A (ABCA), and nuclear VCP-like protein. Derived from the general properties of GST, ABCA and VCP, silkworm selenoproteins may play important roles in redox regulation, Se storage and transportation, as well as cell apoptosis.
[Characterization and Stage-Specific Change of Proteins During the Embryonic Development of Silkworm Bombyx Mori].

Abstract:The gene expression pattern was analysed employing protein two-dimensional electrophoresis and amino acid sequencing during the embryonic development of silkworm Bombyx mory. The 2D-PAGE pattern of the silkworm embryonic proteins had little change in the long developmental period from the Critical Stage until the Head Pigmentation Stage of silkworm embryo. The percentage of matched proteins that found between the spots in the 2D-PAGE pattern of the Critical Stage of silkworm embryo and the spots in the 2D-PAGE pattern of the Head Pigmentation Stage of silkworm embryo was 63%. Silkworm Egg-specific Protein and 30K Protein were in large amount. However, 2D-PAGE pattern of the silkworm embryonic proteins showed considerably varied in shorter developmental stage from the Head Pigmentation Stage to the Body Pigmentation Stage and until the Newly Hatched Larva. The percentage of matched proteins that found the spots in the 2D-PAGE pattern of each developmental stage of silkworm embryo became lesser. The protein spots that the isoelectric points lying on the acidic side greatly increased, the Silkworm Egg-specific Protein and 30K Protein were gradually disappeared and the gene that resembles BmLSP's, Enolase's and Superoxide dismutase's gene was activated and expressed with the embryonic development.

Abstract:Sexual dimorphism is initialed by the components of the sex determination pathway and is most evident in gonads and germ cells. Although striking dimorphic expressions have been detected at the transcriptional level between the silkworm larval testis and the ovary, the sex-dimorphic expressions at the protein level, have not yet been well characterized. The proteome of silkworm larval gonads was investigated using a shotgun-based identification. A total of 286 and 205 nonredundant proteins were identified from the silkworm testis and ovary, respectively, with a false discovery rate (FDR) lower than 1%. Only 40 and 16 proteins were previously identified, and 246 and 189 proteins were newly identified in the silkworm testis and the ovary, respectively. The gametogenesis mechanism of silkworm was demonstrated using the protein expression profile and bioinformatics analysis. Cellular retinoic acid binding protein (CRABP) showed to be highly abundant in testis, while tubulins were abundant in ovary. Several homologies of Drosophila essential proteins for gametogenesis were identified in silkworm, such as male meiotic arrest gene product ALY and VISMAY in testis, and maternal mRNA localization protein exuperantia and SQUID in ovary. The gene ontology (GO) annotation and pathway analysis provide system-level insights into the sexual dimorphism and gametogenesis.
A Comparison of the Protein Composition of the Middle Silk Gland Cells of the Fifth Instar Larvae of Silkworm

Abstract:Research of the protein composition of different parts of the middle silk gland (MSG) cells of the fifth instar larvae of silkworm (Bombyx mori L.) are useful to find the functional protein concerned with the composition and secretion of the sericin protein. Using the protein two dimensional electrophoresis (2D-PAGE) and the image analysis technology, a significant differences of the protein composition among the anterior, middle and posterior parts of the MSG cells of silkworm were found. Twenty specific proteins that expressed only in anterior part of the MSG cells were likely to relate with the composition and secretion of 4 kinds of sericin proteins, which are Ser2A, Ser2B S4 and S5. Twenty-two specific proteins that expressed only in middle part of the MSG cells were likely to relate with the composition and secretion of 4 kinds of sericin proteins, which are Ser1B, Ser1C, Ser1D and S3. Twenty-seven specific proteins that expressed only in posterior part of the MSG cells were likely to relate with the composition and secretion of 2 kinds of sericin proteins, which are Ser1A and S3. Moreover, fifty-one specific proteins which expressed in middle and posterior, but not in anterior part of MSG cells and twenty proteins whose volume in middle and posterior part of MSG cells were higher than that in anterior part likely participated in the synthesis of the sericin protein of the corresponding parts.

Abstract:The material was silkworm after reeling silk.Protein of silkworm( PSP) was extracted and determined the amino acid and protein pupa molecular weight distribution of PSP. The experimental results showed that,the optimum technological conditions of extracted the PSP was that solid-to-liquid ratio was 1∶3,extraction times were 3,pH was 3.0,in this process the protein extraction rate was 53.0%.Finally,PSP was rich in 18 kinds of amino acids and amino acids content conform to the provisions of high quality protein standards of the FAO/WHO,and the molecular weight distribution from 500～100000u. The silkworm protein after purification would be further used in some fields likes pharmaceutical,food and cosmetics and so on.

Abstract:Synopsis Although the ecdysteroid of the silkworm had been studied for decades, the proteome of the prothoracic gland, the primary source of ecdysteroid hormones, has not been studied previously. In the present paper, we utilized a proteomic approach to investigate the fifth instar prothoracic gland during the growth and development of the silkworm, Bombyx mori L. The two-dimensional electrophoresis results showed that the majority of proteins were acidic proteins, especially concentrated in the area of 25-65 kDa, with pl values of between 4 and 7, and the difference was not distinct. When compared with Qiufeng (Japanese strain), the interspecific distinction was larger than the intraspecific distinction, and 19 particular spots, excized from the third, fifth and ninth days of p5O (Chinese strain) and Qiufeng were subjected to MALDI-TOF-MS (matrix-assisted laser-desorption ionization-time-of-flight MS) analysis. We sorted them into seven catagories: energetics and/or metabolism, storage proteins, protection, lipid metabolism, signal transduction, cell function and unknown function proteins. Of these proteins, arginine methyltransferase is discussed as playing an important role in regulating the activation of ecdysteroidogenesis via transcription or translation.
Protein Databank for Several Tissues Derived from Five Instar of Silkworm

Abstract:We attempted to construct protein databank of silkworm (Bombyxmori L.) for clarifying the gene expression and post-translational modification. Proteins from silkworm body wall, fat body and middle intestines were separated by two-dimensionalelectrophoresis. The N-terminal amino acid sequences of 40 proteins and their homology to proteins from other organisms were determined. 58.5% proteins were high homologous to the already reported proteins in D. Melanogaster and 36.5% to those in other organisms. Only 5% of the proteins were homologous to the known protein in silkworm. The N-terminal sequences of 27 proteins were first found in silkworm, and all these data were registered in SWISS-PROT through the Internet. The results suggested that the information of gene expression and post-translational modification could be obtained by the information of protein databank.

Abstract:In silkworm moth the colleterial gland markedly enlarged due to the secretion and accumulation of glue like substances before adult emergence. However, the Ng mutant female moth only secreted little glue-like substance and laid loose eggs naturally. In the present experiment, it was extracted the proteins of secretory part of the variety E981 and its Ng mutant line and analyzed by two-dimensional electrophoresis. More than 700 protein spots were resolved both in two samples and most of the proteins were distributed in the area from 30 kD to 70 kD and pH 4 - 8. Through the comparison and analysis, it was found that 4 proteins were only expressed in E981 and 2 proteins were only expressed in Ng mutant. Furthermore, there are about 29 proteins were expressed higher in 981 and about 15 proteins expressed volume were higher in Ng mutant. These differential proteins may be have some relations with the Ng mutant form and directly lead to the Ng mutant can't secret the glue-like substance.

Abstract:The silkworm middle silk gland (MSG) is the sericin synthesis and secretion unique sub-organ. The molecular mechanisms of regulating MSG protein synthesis are largely unknown. Here, we performed shotgun proteomic analysis on the three MSG subsections: the anterior (MSG-A), middle (MSG-M), and posterior (MSG-P) regions. The results showed that more strongly expressed proteins in the MSG-A were involved in multiple processes, such as silk gland development and silk protein protection. The proteins that were highly expressed in the MSG-M were enriched in the ribosome pathway. MSG-P proteins with stronger expression were mainly involved in the oxidative phosphorylation and citrate cycle pathways. These results suggest that the MSG-M is the most active region in the sericin synthesis. Furthermore, comparing the proteome of the MSG with the posterior silk gland (PSG) revealed that the specific and highly expressed proteins in the MSG were primarily involved in the ribosome and aminoacyl-tRNA biosynthesis pathways. These results indicate that silk protein synthesis is much more active as a result of the enhancement of translation-related pathways in the MSG. These results also suggest that enhancing ribosome biogenesis is important to the efficient synthesis of silk proteins.

Abstract:Qualitative and quantitative differences in proteins expressed in the middle silkglands of male and female silkworm larvae that differ in silk colour were investigated by high resolution two-dimensional polyacrylamide gel electrophoresis (2-D PAGE), followed by computer assisted image analysis. About 1000 protein spots were resolved in both the sexes and most proteins were shown to be distributed in the area from 15 kDa to 70 kDa and pH 4–8. It was found that some proteins displayed higher expression in yellow cocoon, while two proteins were only expressed in female silkworm silkgland tissue through the comparison and analysis by two-D software. These proteins especially existed in female silkworm middle silkgland tissue of yellow cocoon. Furthermore, these proteins might be involved in the expression of cocoon colour phenotype
