Preparation Of The Temperature Sensitive Hemicellulose-Based Hydrogels Via Click Chemistry

Abstract:In this paper, the hemicelluloses (HCB) isolated from corncobs were modified by click chemistry. The introduction of azide functionalities into the backbone of the polysaccharide was done in DMF/LiCl at 100 degrees C for 24h in presence of sodium azide and the pre-modified hemicellulose with 2-bromoisobutyl bromide (BiBB) through esterification reaction which was processed under ambient temperature for 24 h. The chemical structure of the products obtained in each step was precisely characterized by FTIR as well as NMR. Moreover, the achieved degree of substitution of azide groups in the hemicelluloses which was characterized by elemental analysis was up to 0.23. Alkyne-terminated functionalized NIPAAm-co-HEMA (ANCH) copolymer was then grafted onto the synthesized azide-functionalized hemicelluloses (HBN) with different DS of the azide groups varying from 0.08 to 0.23 by copper-catalyzed azide-alkyne cycloaddition (CuAAC), a typical reaction of click chemistry, at 80 degrees C for 8h to form the novel temperature sensitive hemicellulose-based hydrogels (HNG).Their temperature sensitivity and morphology were investigated by DSC and SEM. From the results, it can be seen that the LCST of the hydrogels were increased with the increase of the DS of the azide groups in BEN, whereas, the pore size of the hydrogels was decreased with increasing the azide groups' DS. Comparing with those reported photo-crosslinking and free radical copolymerization methods, click chemistry could not only shorten the formation time of such hydrogels, but also endow them with more uniform pore structure due to its features of high stereospecificity, selectivity and high efficiency. Besides, the drug loading and releasing behavior of the hydrogels were discussed as well.
