Abstract:Neuronal growth inhibitory factor (GIF) of porcine brain, was isolated and purified by a similar procedure which was used on the isolation of human and bovine GIF. The native porcine protein with stoichiometry of 4Cu+, 3Zn2+ was obtained for the first time. The kinetics of zinc transfer from Cu4Zn3MT-3 to apo-carbonic anhydrase were studied, and zinc transfer rate constants and thermodynamic parameters were obtained. It is found that like other MTs, porcine Cu4Zn3MT-3 can also transfer its zinc atom to apoCA, even much easier than other MTs. A possible association mechanism has been proposed, the formation of Cu4Zn3MT3-apoCA complex may be the rate-determining step. The obtained data indicate besides its neuronal growth inhibitory function, GIF might play a role in cellular Zn homeostasis in brain.

Abstract:Neuronal growth inhibitory factor (GIF), known also as metallothionein-III (MT-III), was the first validated to be capable of inhibiting growth of neuronal cells in nervous system, its beta-domain being functional. GIF functional di-domain (GIFbeta- beta) was constructed to study the structure and function of GIF. N terminal beta-domain and C terminal beta-domain cDNAs were amplified by PCR, inserted into vector pGEX-4T-1 and expressed in Escherichia coli, as carboxyl terminal extension of glutathione-S-transferase (GST), by IPTG induction. After digestion by thrombin, the fusion protein was isolated by passing through a glutathione-Sepharose 4B affinity chromatography column and was purified by gel fit ration on Sephacryl-S100. About 60 mg protein per liter of bacterial cell culture was achieved. The results of SDS-PAGE, amino acid composition, molecular mass, the ratio of metal/protein and sulfhydryl group/protein showed that the purified protein was the GIFbeta- beta. Circular dichroism (CD) spectroscopy show GIFbeta- beta has characteristic metal-sulfhydryl clusters of metallothionein family. Inhibitory activities detected by the MTT reduction assay are: GIF > GIFbeta-beta > GIF beta-domain.

Recent studies on the structure of human GIF, carried out using NMR and molecular dynamics simulation techniques, have been summarized. By studying a series of protein-engineered mutants of GIF, we showed that the bioactivity of GIF is modulated by multiple factors, including the unique TCPCP motif-induced characteristic conformation, the solvent accessibility and dynamics of the metal-thiolate cluster, and the domain-domain interactions.

Abstract:Human metallothionein-3 (hMT3), also named as human neuronal growth inhibitory factor (hGIF), can inhibit the outgrowth of embryonic cortical neurons in the presence of brain extracts. In order to systematically study the structure–property–reactivity–function relationship of hGIF, our laboratory designed a series of mutants and studied their structure, property, reactivity and functions by a series of chemical and biological tools including UV spectroscopy, CD spectroscopy, NMR, chemical reaction and primary neuronal culture assays. In summary, we concluded that the bioactivity of hGIF was regulated by multiple factors, including the 6CPCP9 motif, an additional threonine insert at sequence position 5, domain–domain interactions, the structure and stability of the metal–thiolate cluster and the linker. Our studies provide more and more evidences which revealed that the bioactivity of hGIF is mainly related to the essential metal release and its characteristic conformation.

Abstract:Growth inhibitory factor (GIF), renamed as metallothionein-3, is a 68-amino acids brain specific member of the metallothionein family. GIF plays comprehensively physiologic roles and is deemed to be implicated in Alzheimer's disease. When supplemented with Alzheimer's brain extracts, GIF demonstrates its in vitro growth inhibitory activity on cultured neuron; but the underlying molecular mechanism remains unknown. To probe this mystery, the yeast two-hybrid system was employed to screen a human brain cDNA library for GIF-interacting proteins. From 4.1 x 10(6) transformants of library screening, a C-terminal fragment of small G-protein Rab3a was obtained, harboring 87 amino acids that interacted with GIF in yeast. The full-length Rab3a was amplified by polymerase chain reaction ( PCR) using human total placenta cDNA as template ana concomitantly cloned. In yeast two-hybrid test, full-length Rab3a also interacted with GIF. Through coimmunoprecipitation and Western blotting experiments, interaction between GIF and Rab3a was confirmed in mammalian cell. Furthermore, the result indicated that Rab3a interacted with GIF in a GTP-binding form.

Abstract:Neuronal growth inhibitory factor(GIF),the brain-specific member of the metallothionein(MT)family,was the first validated neuronal growth inhibitor,and it uniquely exhibits the growth inhibitory activity on in vitro cultured neuron cell when supplemented with Alzheimer's brain extracts.Rab3a is one of the v-SNARE proteins and plays an important role by Ca2+-dependent in the regulation of neurotransmitter release.Objective we use immunohistochemistry method to prove the existence of the interaction between the two proteins.Methods To find the localization and colocalization of GIF and Rab3a,we use ABC Kit to staining tissues and double label immunofluorescence by confocal laser scanning microscopy.Results GIF and Rab3a were located and colocated in the cerebral cortex and hippocampi of the brain.Conclusions These results indicate that the interaction of GIF and Rab3a is exist through they colocated in the cerebral cortex and hippocampi of the brain.

Abstract:Neuronal growth inhibitory factor (GIF), a brain-specific metallothionein protein (Metallothionein- Ⅲ), can inhibit the growth of neuronal cell. Though exhibiting the 70% same sequence, GIF distinguishes from othet MT isoforms by inhibiting neuronal survival in vitro. It was reported that MT- Ⅲ and its mRNA were greatly reduced in Alzheimer's disease (AD) brain. For further research, the monoclonal antibody of GIF was prepared and the interaction between GIF, a-domain. β-domain, peptides and McAb of GIF were studied with ELISA. The epitope of the MT- Ⅲ was studied. The results show that the epitope of GIF is closely related to GIF's conformation.

Abstract:Metallothinein-3 (MT3), also named neuronal growth inhibitory factor (GIF), is attractive by its distinct neuronal growth inhibitory activity, which is not shared by other MT isoforms. The polypeptide chain of GIF is folded into two individual domains, which are connected by a highly conserved linker, KKS. In order to figure out the significance of the conserved segment, we constructed several mutants of human GIF (hGIF), including the K31/32A mutant, the K31/32E mutant and the KKS-SP mutant by site-directed mutagenesis. pH titration and DTNB reaction exhibited that all the three mutations made the β-domain lower in stability and looser. More significantly, change of KKS to SP also altered the general backbone conformation and metal–thiolate cluster geometry. Notably, bioassay results showed that the bioactivity of the K31/32A mutant and the K31/32E mutant decreased obviously, while the KKS-SP mutant lost inhibitory activity completely. Based on these results, we proposed that the KKS linker was a crucial factor in modulating the stability and the solvent accessibility of the Cd3S9 cluster in the β-domain through domain–domain interactions, thus was indispensable to the biological activity of hGIF.
Neuronal Growth‐inhibitory Factor (metallothionein‐3): a Unique Metalloprotein

Abstract:Metallothionein (MT) was first identified in 1957 by M. Margosch and B. Vallee as ‘a cadmium protein from equine kidney cortex’. In fact, the small protein was a Cu, Zn-containing protein, with an array of conserved cysteine residues. Since its discovery, intensive study has been focused on this protein, but its functions remain elusive. In 1991, a new small protein, which exhibited neuronal growth-inhibitory activity, was isolated by Y. Ushida et al. from healthy human brain; this protein was subsequently named neuronal growth-inhibitory factor (GIF). It was later found that this protein showed high sequence similarity ( 70%) to MT, together with conserved cysteines and metal content, and it was renamed MT3. The neuronal growth-inhibitory activity of this protein is unique among members of the MT family. In their paper, Ushida et al. wrote that: ‘‘... we do not know the mechanism of growth inhibitory action of GIF, especially why GIF but not MT has growth inhibitory activity, despite [their] homologous sequences.’’ To answer this question we have been carrying out a study of structure–property–reactivity–function relationships. Owing to the conformational flexibility of its polypeptide chain, there is no GIF crystal structure available as yet. The solution structure of the a-domain of rat and human Cd-GIF has been determined by means of NMR spectroscopy, but the structure of the b-domain could not be solved because of insufficient NOE connectivities. However, a model of the b-domain structure of GIF was constructed on the basis of molecular dynamic simulation, and a systematical mutagenesis study has been performed. This has revealed that the particular conformation of TCPCP(5–9) in the b-domain, the exposure of the metal–thiolate clusters, the dynamics of EAAEAE(55–60) in the a-domain and the interdomain interactions are closely connected to the bioactivity of human GIF, as discussed in the first minireview of the series. The two-domain architecture of proteins in the MT family gives rise to their specific properties, such as binding to metals for detoxification of pollutants and homeostasis of essential metals, and a supply of thiols (cysteines) for scavenging free radicals. What then are the differences between GIF and MT1 ⁄ 2 that confer on GIF the specific growth-inhibitory activity? In the second minireview, Faller and colleagues have produced a detailed summary of possible differences in the cluster structure between GIF and MT1 ⁄ 2. The higher dynamics and greater exposure of the metal–thiolate clusters to the outside environment in GIF is thought to result in the high reactivity of this protein. I welcome the controversy introduced in respect of previous studies. In line with Confucian philosophy ‘ (jian ting ze ming)’, contrary results and views can look opposite but sometimes they are complementary, reflecting the same truth from different aspects. Some of the functions of MTs have been uncovered, but the real roles played by GIF are still not clear. In the final minireview, the possible biological functions are discussed by Chung and coworkers. Taking into account the expression and secretion of GIF in brain, and combining studies of neurodegenerative diseases with those of the injured brain, they describe how GIF has neuroprotective functions when administered to the injured brain, as opposed to its neuro-inhibitory actions upon cultured neurons. These authors conclude that ‘the studies to date have only begun to investigate and understand the physiological role of GIF within the brain. Further studies are required to determine the function of GIF within the normal, injured and neurodegenerative brain.’
Molecular Cloning and Identification and Biological Activity of GIF-interacting Protein(s)

Abstract:In order to examine the elusive functional mechanism of GIF (Neuronal growth inhibitory factor, GIF) and elucidate the possible relationship between GIF and Alzheimer's disease, we constructed bait1 plasmid (pHyblex-GIF) by cloning GIF cDNA directly in frame with plasmid pHyblex, and used the yeast two-hybrid system to screen Alzheimer's disease human brain cDNA library and found the GIF-interacting proteins. The final results from coimmunoprecipitation and western blotting experiments confirmed that interacting proteins specifically binds to GIF. After sequencing the nucleotide of the putative positive plasmids and searching for homologues, we found that one of these is the part of human nuclear dUTPase protein sequence. Then the dUTPase genes are cloned into pGEX-4T-1, the fusion expression vector of GST,and highly expressed in E. coli BL21. The proteins dUTPase and GIF were purified and obtained by affinity chromatography, thrombin digestion and gel filtration on Sephacryl S100. It demonstrated that the proteins dUTPase and GIF had the growth inhibitory activity on co-cultured neuron in vitro. The inhibitory curve was very similar to the GIF. It's possible that dUTPase is one of the proteins interacting with GIF in Alzheimer's disease human brain extracts.

Abstract:We have purified and characterized the growth inhibitory factor (GIF) that is abundant in the normal human brain, but greatly reduced in the Alzheimer's disease (AD) brain. GIF inhibited survival and neurite formation of cortical neurons in vitro. Purified GIF is a 68 amino acid small protein, and its amino acid sequence is 70% identical to that of human metallothionein II with a 1 amino acid insert and a unique 6 amino acid insert in the NH2-terminal and the COOH-terminal portions, respectively. The antibodies to the unique sequence of GIF revealed a distinct subset of astrocytes in the gray matter that appears to be closely associated with neuronal perikarya and dendrites. In the AD cortex, the number of GIF-positive astrocytes was drastically reduced, suggesting that GIF is down-regulated in the subset of astrocytes during AD.

It is known that the alpha-beta domain-domain interaction of hGIF is crucial to the neuron growth inhibitory bioactivity although the exact mechanism is not clear. Herein, the beta(MT3)-beta(MT3) mutant and the hGIF-truncated Delta33-35 mutant were constructed, and their biochemical properties were characterized by pH titration, EDTA, and DTNB reactions. Their inhibitory activity toward neuron survival and neurite extension was also examined. We found that the Delta33-35 mutant alpha-domain containing beta-domain-like M(3)S(9) cluster exhibits the function of alpha-domain with M(4)S(11) cluster in hGIF. These results showed that the stability and solvent accessibility of the metal-thiolate cluster in beta-domain is very significant to the neuronal growth inhibitory activity of hGIF and also indicated that the particular primary structure of alpha-domain is pivotal to domain-domain interaction in hGIF.

Abstract:Neural growth inhibitory factor (GIF), a member of metallothionein family (metallothionein-3, MT3), was well known by its distinct neural growth inhibitory activity, which is not shown by other MT isoforms. However, till now, people still did not know clearly how GIF exerts its biological functions. Since it has been reported that GIF might serve as NO scavenger and was related to the release of zinc, our study was focused on the interaction of GIF and NO. By studying the reactions of human GIF and human MT1g with SNOC-a type of NO donor, it was found that GIF was more reactive than MT-1g toward SNOC. In order to further figure out if the high reactivity of GIF in this reaction resulted from the acid-base catalysis, several mutants were constructed: E23K, E41G/E43A, E23K/E41G/E43A. By studying their basic properties and the reactions toward SNOC, it was found that the S-nitrosylation of GIF was not only related to the acid-base catalysis, but also to the accessibility of metal-thiolate clusters.

Abstract:We examined the protective effect of growth inhibitory factor (GIF) against zinc-induced neuronal death in rat hippocampal neurons. In an in vitro cell culture system, 300 microM Zn(2+) readily induced death of hippocampal neuronal cells, which was characterized by massive necrosis and a minor degree of apoptosis. Neither the addition of recombinant GIF nor Rab3A alone could rescue these cells from death. However, the combination of GIF with Rab3A could significantly enhance the survival of the hippocampal neurons. This result was supported by both Annexin -V FITC/propidium dual staining and chromosomal DNA analysis. These findings suggest that GIF may inhibit Zn(2+)-induced neuronal death via its interaction with Rab3A.

Abstract:The zinc transfer reactions from Zn-7-MT-I, Zn-7-MT-II, Zn-4 alpha fragment (MT-I) and Zn-4-alpha fragment (MT-II) to ape-carbonic anhydrase have been studied. In each reaction, no more than one zinc ion per molecule is involved in metal transfer. Zn-7-MT-I and Zn-7-MT-II donate zinc to apo-carbonic anhydrase and de novo constitute it at a comparable efficiency, while Zn-7-MT-II exhibits a little faster rate. Surprisingly, Zinc is released from Zn-4-alpha fragment (MT-II) with a much faster rate than from Zn-4-alpha fragment (MT-I), whose rate is close to that of Zn-7-MT-I. The reason for the difference is still unknown. Introducing complex compounds into this system may give rise to an effect on the reaction. The transfer from Zn-7-MT-II in the presence of reduced glutathione shows little difference compare to the control, suggesting that the reduced glutathione is not involved in zinc transfer process. However, glutathione disulfide does accelerate this zinc transfer reaction remarkably, indicating that the oxidative factors contribute to zinc release from metallothioneins.

The Role of Thr5 in Human Neuron Growth Inhibitory Factor

Abstract:GIF, a member of the metallothionein (MT) family (assigned as MT3), is a neuron growth inhibitory factor that inhibits neuron outgrowth in Alzheimer’s disease. The conserved Thr5 is one of the main differences between GIF and other members in the MT family. However, natural sheep GIF has an unusual Ala5, casting doubt on the role of common Thr5. We constructed a series of human GIF mutants at site 5, and characterized their biochemical properties by UV spectroscopy, circular dichroism spectroscopy, EDTA reaction, 5,5′-dithiobis(2-nitrobenzoic acid) (DTNB) reaction, and pH titration. Their inhibitory activity toward neuron survival and neurite extension was also examined. Interestingly, the T5A mutant exhibited distinct metal thiolate activity in the EDTA and DTNB reactions, and also lost its bioactivity. Meanwhile, the T5S mutant had similar biochemical properties and biological activity as wild-type human GIF, indicating the hydroxyl group on the Thr5 was critical to the bioactivity of human GIF. We suggest the hydroxyl group in human GIF may help stabilize the biologically active conformation. On the other hand, lack of the hydroxyl group in sheep GIF may be partially compensated by its abnormal structure.
