Abstract:The prognosis for severe periodontal diseases, a major cause of tooth loss in adults is generally poor, and conventional therapeutic strategies often fail ot regenerate soft and hard tissues in ther periodontium. Thus, it remains a major challenge and goal for clincicans to restore both fuchitonal and esthetic periodontal tissue.s With continuous breakthroughs in tissue engineetring, current regenerative therapies are rapiddly developing. Four elements including blood supply cells scaffolds and molecular mediators are essential components to reach these goals. Among them, growth factors (GFs) are important molecular mediators with thranslational feasibiligy. A number of animal experiments revealed that certain GFs are promising candidates that facilitate tissue regenration thrugh increases in cell migration, survival, and differentiation of various cell atypes. Recombinant human platelet-derived growth factor (rhPDGF) and reombinatnt human fibroblast growsth factor 2 (rhFGF-2) are tow GFs that have been invstigated in a number of clincial trials for periodontal regeneration. Although over a decade has passed since they were first introduced, long-standing quesiton regardign their efficacy, indications and appropriated concentrerations still exist. Today, evidence-based analysis derived from randomized clincial treials (RCTs) provides powerful and quanititative daTA to understand therir therapeutic characteristic.s This chapter discuss the biologic background and clincial uses 0.30% rhFGF-2 and 0.3 mg/mL rhPDGF-BB as GFs for periodontal bone regenereation. Furthermore guidelines regarding their clincial applicateion and case reports are provided to demonstrate their use in regenerative periodontoloyg.
