Abstract:Two arrays of behenoyl (22 : 0) based amphiphilic lipids with sugar alcohols as a flexible linker were synthesized through acylation of sugar alcohol mono-behenates with diacetyl tartaric anhydride or acetoxy succinic anhydride and some of the resulting products are capable of yielding stable and uniform solid lipid nanoparticles (SLNs) (mean diameter similar to 100 nm, PDI < 0.2) without any additive surfactant, displaying excellent dual functions (as both a solid lipid excipient and emulsifier).

Abstract:We synthesized a duplex cyclodextrin with two aliphatic links and functionalized with eight pentyl ethers orientated towards the centre of the duplex. We found that this molecule self-assembled in water to form stable vesicles with a diameter within the range 1–10μm.

Abstract:Two single-chain glycolipids, octadecyl-beta -D-glucopyranoside(1) and 2-octadecanamido-2-deoxy-D-glucopyranose(2) were synthesized and their dispersing properties in water were studied. Incorporation of 1 into the phospholipid liposomes could inhibit the aggregation of liposomes and improve the molecular packing in the bilayer membranes.

Abstract:The objective of the present study was to improve the stability of (water-in-oil-in-water) double emulsions by dual gelation, including the gelation of lipid phase through oleogels and the gelation of continuous aqueous phase through hydrogels. Glycerol monostearate (GMS) could be used to form the oleogels and improve the stability of primary emulsions and double emulsions through reinforcing oil film. However, the redundant GMS crystals in both the oil phase and the continuous phase could cause the destabilization of double emulsions. The double emulsions reinforced by oleogels were successfully encapsulated in Ca-alginate hydrogel beads, and this encapsulation would not influence the integrity of double emulsions. The redundant GMS crystals might cause the leakage of the inner aqueous phase in the encapsulation process, and this leakage was more likely to happen during the mixing step, rather than the crosslinking step. Ca-alginate hydrogel beads were conducive to the confinement of the encapsulated oil globules.

Abstract:The Lewis x –Lewis x interaction has been increasingly studied, using a variety of techniques including nuclear magnetic resonance spectroscopy, mass spectrometry, vesicle adhesion, atomic force microscopy, and surface plasmon resonance spectroscopy. However, the detailed molecular mechanism of these weak, divalent cation dependent interactions remains unclear, and new models are needed to probe the nature of this phenomenon in term of key roles of the different hydroxyl groups on Lewis x trisaccharide determinant involved in the Lewis x –Lewis x interaction. An interesting solution is to synthesize a series of Lewis x pentaosyl glycosphingolipid derivatives in which one of the eight hydroxyl groups of Lewis x trisaccharide is replaced by a hydrogen atom, and to test the adhesion induced by interaction of these derivatives, in order to gain insight into the functions played by the hydroxyl groups of the Lewis x trisaccharide. This article describes the synthesis of 3d-deoxy and 4d-deoxy Lewis x pentaosyl glycosphingolipids, to be used for study of the Lewis x –Lewis x interaction.

Abstract:Three single chain amphiphilic cholesterol derivatives with amino acid as hydrophilic groups, I, II and III, were synthesized. When they were hydrated in water, they can all form lamellae structures as evidenced by DSC measurement. The lamellae states of II and III can be converted to vesicles upon ultrasonication of the diluted aqueous solutions. The vesicles showed very slow release rate of the encapsulated water-soluble dyes, indicating that they are stable and belong to the least permeable vesicles.

Abstract:A lithocholic acid derivative having high compatibility with phospholipid was synthesized and incorporated to the phospholipid bilayer membrane. A significant influence of it on the molecular packing and the stability enhancement of the vesicle suspension were clarified by a fluorescence depolarization method and a turbidity measurement, respectively.

Abstract:The properties of vesicles formed from the self-assembly of amphiphilic molecules can mimic the functionality of the natural lipid membranes. In this study, the self-assembly process of the amphiphilic structures formed by the interaction between ultra-small fatty acids [FM, Cn (n = 4-8)] and amino acids (AAs) to generate vesicles under aqueous conditions were investigated in detail, along with the corresponding dynamic vesiculation mechanisms. Our results showed that the molar ratio of FAs/AAs and the chain length of FM largely affected the structural characteristics and dispersion of vesicles. The detailed information about the entire size distributions and morphology of obtained vesicles were explored by the cryogenic transmission electron microscopy (Cryo-EM). Fourier transform infrared (FT-IR) spectra and quantum calculations suggested that the intermolecular hydrogen bond and electrostatic interactions between ultra-small molecules (FM and AM) during the aggregation processes were responsible for the formation of vesicles, where the hydrogen-bonding effect was dominant. Our findings shed new light on the effective and simple preparation of biological vesicles via ultra-small molecules self-assembly in aqueous solutions, which may have potential applications in vesicle physiology and drug delivery systems, and also get a mature understanding of the fundamental intermolecular interactions in life process.

Abstract:Supramolecular glycolipid has been fabricated based on ternary complex system, which is composed of naphthyl glucosamine (GlcNap), alkyl viologen (RV8), and cucurbit[8]uril (CB[8]). CB[8] plays a key role to connect the other two building blocks together, and the process is driven by host-enhanced charge transfer interaction. Compared with the classic glycolipids, supramolecular glycolipids display redox responsiveness. Supramolecular glycolipids are able to self-assemble in water to form spherical aggregations, such as vesicle like structure. In addition, spherical aggregations can specifically interact with Concanavalin A, indicating that the carbohydrate groups are available on the surfaces of the aggregates.

Abstract:Stable vesicles were formulated based on the self-assembling of L-alpha-phosphatidylcholine (PC) and mannosylerythritol lipid-A (MEL-A) in a manner of weak or non-cooperative interactions. Their structural characterization, stability and encapsulation properties were evaluated. The results showed that the predominant average diameters varied ranging from 200 nm to 700 nm with MEL-A/PC ratio changed. In case of the ratio at 5:5, 6:4 or 7:3, the vesicles performed the smaller size or turbidity and larger entrapment efficiency for hydrophilic molecules. Anthocyanins were successfully encapsulated using these vesicles, which was confirmed by the formation of egg-shaped structures with the core size less than 500 nm and an encapsulation efficiency of 54.9 +/- 1.6%. Compared with free anthocyanins, the encapsulated anthocyanins displayed higher retention rates when exposed to storage and simulated gastrointestinal fluid conditions, and their antioxidant capacity after simulated intestinal digestion was enhanced by 3-3.5 times because of protection of vesicle encapsulation and possible synergic effects.

Abstract:Three single chain lithocholic acid derivatives with a steroidal moiety in the center of the hydrocarbon chain and two head groups at both ends of the chain were synthesized and characterized. Due to their high compatibility with phospholipids, they can be incorporated homogeneously into the phospholipid bilayer membrane. The resulting mixed vesicles showed high stability against aggregation by monitoring the turbidity change in the vesicle suspensions. The entire membrane-spanning packing thus locates the steroidal moiety at the center of the bilayer. The interaction of the steroidal moiety with Lipid molecules at the center of the bilayer resulted in higher mobility of lipid molecules below the T-c of the host phospholipid membrane as clarified by temperature-dependent H-1 NMR, fluorescence depolarization anisotropy, and differential scanning calorimetry (DSC). The results were compared with those of cholesterol, whose steroidal moiety is located near the surface of the membrane.
Preparation and Properties of Vesicles from Condensable Amphiphilic Amino Acids

Abstract:Three double-chain amphiphiles with amino acid groups as hydrophilic moiety were synthesized. These amphiphiles can be easily dispersed in buffer solution to form transparent dispersion. Examination of the dispersion by transmission electron microscopy (TEM) showed the formation of stable vesicular aggregates, which was also confirmed by the ability to encapsulate water-soluble dyes. Since amino acid groups are located on the surface of the vesicles, water-soluble carbodiimide can induce the condensation of these groups to form peptide. The phase transition temperatures of these vesicles were estimated by differential scanning calorimetry (DSC), and a decrease of phase transition temperature was observed after polycondensation due to the disturbance of the ordered arrangement of the hydrophobic chains. The leakage rate of the vesicles before and after condensation was studied by monitoring the increase of fluorescence intensity of water-soluble dye. These vesicles belong to the least permeable ones and the leakage rate can be controlled by varying the degree of condensation or the temperature.

Abstract:Stable unilamellar vesicles were formed in water under appropriate pH from dispersions of N-(O,O-di-n-hexadecyl)phosphorylalanine, an amphiphilic N-phosphorylamino acid. We found that condensation occurred in the vesicle solution after incubated at 40 °C, which may contribute to the stability of the vesicular system. Dipeptide derivative in the vesicle solution was identified by electrospray ionization mass spectrometry (ESI-MS), which suggests the peptide formation without any coupling reagents. Hydrogen bond and electrostatic interactions play important roles in the process of vesicle formation, while the suitable orientation and packing of the amphiphilic molecules at the vesicle/water interface together with certain conformational freedom in the vesicular bilayer are considered to be most favorable for the condensation in ordered systems as vesicles.
Enhancing Stability of Liposome by Synergistic Effect of Polymer Multiple-units

Abstract:We synthesized a water soluble glycol chitosan polymer grafted with cholesterol through substitution reaction, and the polymer was applied to form a polymer-liposome complex (PLC). According to dynamic light scattering (DLS) and transmission electron microscope (TEM) characterization, the diameter of the PLC was about 100 nm and the morphology was similar to the plain liposome. The cholesterol group can induce the polymer backbone to attach to the surface of the liposome membrane and the integration cability of the glycol chitosan-cholesterol (GCC) was characterized by laser confocal scanning microscope (LCSM). The zeta potential of the liposome containing negative charged phospholipid was - 23.2 mV. With integration of the glycol chitosan-cholesterol, the zeta potential significantly increased to - 1.78 mV, which indicated that the attached water-soluble polymer can shield the surface charge of the membrane. By encapsulating fluorescent molecules in the liposome or mixing fluorescent phospholipid during the lipid preparation process, fluorescence resonance energy transfer (FRET) experiment was introduced to test the stability of the PLC. Charge-induced liposome fusion occurred by adding positively charged ions or liposome into negatively charged liposome solution. The PLC has shown the ability to resist the fusion process which can keep the integrity and monodispersity of the liposome. However, the plain liposome was observed in aggregates visible to the naked eyes after adding hetero-charged ions in several minutes. The FRET variation results indicated that the addition of the GCC can slow down the hetero-charged liposome fusion process. In addition, the PLC can also protect against the liposome leakage induced by surfactant, which resulted from synergistic effect of polymer multiple-units. Incubated with 20% FBS under 37 degrees C, the PLC showed well stability in 3 days, while the PDI and the size of the plain liposome were increased obviously during incubation. The preparation process of this polymer-liposome complex strategy is simple and fast, and it has shown good stability in in vitro experiments. This system has potential applications in long circulation drug delivery. [GRAPHICS] .

Abstract:Cyclic-phospholipids-based vesicles can play a role in facilitating the chemical evolution of protocells from the structurally simple to the functionally more complex form. Here, we present a protocol for preparing decanoic acid-derived cyclic phospholipid and glyceryl-diester phosphate-containing vesicles. We describe steps for sample preparation, equilibration, and image acquisition using confocal microscopy. This protocol has the potential for preparing a wide variety of these phospholipid-based artificial cell constructs. Hybrid polymer/liquid vesicles as new particles for drug delivery and cell mimics

Abstract:Hybrid copolymer/lipid vesicle are recently developed self-assembled structures that could present biocompatibility and biofunctionality of liposomes, as well as robustness, low permeability and functionality variability conferred by the copolymer chains. However, to date, physical and molecular parameters governing copolymer/lipid phase separation in these hybrid membranes are not well understood. In this work, we studied in detail the formation and phase separation in the membranes of both Giant Unilamellar Hybrid Vesicles(GHUVs) and Large Unilamellar Hybrid Vesicles (LHUVs) obtained from the mixture of phospholipids in the fluid (liquid disordered) or gel state (solid ordered) with various copolymers based on poly(dimethylsiloxane) (PDMS) and poly(ethylene glycol) (PEO) with different architectures (grafted, triblock) and molar masses. For GHUVs, phase separation at the micron scale and nanoscale was evaluated through confocal microscopy, and Fluorescence lifetime imaging microscopy technique (FLIM) respectively, where as acombination of Small angle neutron scattering (SANS), Cryo-transmission electron microscopy (Cryo-TEM) and Time-resolved Forster resonance energy transfer (TR-FRET) techniques was used for LHUVs. We demonstrate that the lipid/polymer fraction, lipid physical state, and the line tension at lipid polymer/lipid boundaries which can be finely modulated by the molar mass and architecture of the copolymer are important factors that govern the formation and structuration of hybrid vesicles. We also evidence that elasticity ofthe hybrid membrane can be modulated via the lipid polymer composition, through the use of micropipettes techniques.

Abstract:Two amphiphilic monomers from aspartic acid, N-methacryloyl dihexadecyl L-aspartate (A16-MA) and N-itaconyl dioctadecyl L-aspartate (A16-I) were synthesized. Three kinds of amphiphilic copolymers were obtained by copolymerization of A16-MA with hydroxyethyl methacrylate (HEMA) and 3-sulfopropyl methacrylate potassium salt (SPMAP) respectively and of A16-I with HEMA. The aggregation behavior of the two monomers and three copolymers were examined in an aqueous system. A16-MA and its copolymers with HEMA are very difficult to disperse in water to form regular aggregates; however, copolymers from A16-MA and SPMAP can be dispersed in aqueous media as stable vesicles. Both A16-I and its copolymers with HEMA can be easily dispersed in aqueous medium and form vesicular assemblies. These bilayer membranes undergo a typical gel-to-liquid crystal phase transition as confirmed by differential scanning calorimetry. The phase transition temperatures of the copolymer vesicles shift to slightly higher temperatures with a broader phase transition temperature range. The permeability of A16-I vesicles was studied by monitoring the release of encapsulated fluorescent probes; however, the encapsulated probes cannot be retained for very long in the vesicles prepared from amphiphilic copolymers.
