Cell Apoptosis Induced By Lanthanum Chloride In Hela Cells Via Caspase-3 Activation

Abstract:Objective To explore the role and molecular mechanism of lanthanum chloride on inducing apoptosis and inhibiting cell growth in cervical cancer cells in vitro, which will provide a theoretical basis for medical use of rare earth compounds. Methods The anti-tumor activity of lanthanum chloride was detected by morphological observation and MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. Distribution of cell cycle and apoptosis were analyzed using flow cytometry (FCM). The expression of activated protein of Caspase-3 was measured by Western Blot. Results Lanthanum chloride can induce Hela cell apoptosis and inhibit the cell growth dose-dependently. Morphological observation showed that with the increase of lanthanum chloride dosage, the cell density decreased, cytoplasmic particles increased, colors deepen, the cells' gap increased, a few cells became round and floating, detached cells gradually increased; The results were further confirmed by the observation using laser confocal microscope, which showed typical characteristics of apoptosis: condensation and margination of nuclear chromatin, the smaller nucleus, degraded nuclear chromatin, the gradually breakdown nuclear envelopes, and finally, the completely broken nuclei. FCM analysis showed that the cell ratio of G0/G1 phase increased and that of S phase decreased significantly with the increase of lanthanum chloride doses. Lanthanum treatment increased the cell apoptosis rate significantly. 5 mu M of Lanthanum chloride was enough to trigger apoptosis and the effects was dose-dependent. Western blot analysis proved that lanthanum chloride can up-regulate the activated Caspase-3 (Cleaved Caspase-3, ASP175) expression dose-dependently. Conclusion Lanthanum chloride is capable of inhibiting the cell growth of human cervical cancer Hela cells and inducing cell apoptosis via Caspase pathway.
