Effects Of Methyltestosterone On Ar And Igf-Iea Gene Expression In Porcine Satellite Cell Cultures

Abstract:Androgens enhance muscle growth in a number of species; however, the mechanism by which is not known. The goal of this study was to determine if treatment of porcine satellite cell cultures with androgens directly affects the level of mRNA for androgen receptor (AR) and IGF-IEa. Porcine satellite cell cultures were established from the longissimus muscles of male piglet and then treated with various concentrations of methyltestosterone. The expressions of AR and IGF-IEa genes were studied using real-time quantitative RT-PCR. Relative AR mRNA levels were significantly increased at 10(-8) and 10(-7) M methyltestosterone in nonfused porcine satellite cell cultures, and were significantly increased at 10(-9), 10(-7) and 10(-6) M methyltestosterone in fused porcine satellite cell cultures. Relative IGF-IEa mRNA levels were significantly increased at 10(-7) M methyltestosterone in nonfused porcine satellite cell cultures, and were significantly increased at 10(-9) and 10(-6) M methyltestosterone in fused porcine satellite cell cultures. In summary, treatment of porcine satellite cell cultures with methyltestosterone partly increased AR and IGF-IEa mRNA level, however, we did not find close correlation between mRNA level and methyltestosterone concentrations.
