Abstract:ACO is one of the rate-limiting enzymes of ethylene biosynthesis in plant. The expression of ACO gene help us to study how endogen ethylene effect on fruit ripening. In this research, we used virus-induced gene silencing technique to silencing the ACO gene in peach. The gene-silencing vector carrying the fragment of ACO gene had been constructed before this study. The modified virus vector carrying the fragment of ACO gene was transformed into Agrobacterium GV3101 and infiltrated peach fruit to silencing the ACO gene. After 6 days of the infection, the mRNA expression level of ACO gene in peach was too low to detected; Ethylene production of peach decreased 8.94 mu l. kg(-1) with significant differences compared with control; And the ACO activity in gene silencing peach decreased 150.95 U. g(-1). h(-1) than that in control; ACC content of peach increased 0.009 nmol. g(-1) with significant differences. Our results provide a scientific basis for prolonging storage period of preach by inhabiting the ethylene-biosynthetic pathway.
