Effects Of Mutations In Mitochondrial Cytochrome C Oxidase Genes On Beta-Amyloid Protein Secretion And Neurofibrillary Tangles Formation And Neuronal Apoptosis In Alzheimer'S Disease Cybrids

Abstract:Objectives: To investigate the effects of mutations in mitochondrial cytochrome C oxidase genes on tangles(NFT) formation and neuronal apoptosis in Alzheimer's disease cytoplastic hybrid(hybrid) cell lines. Methods: Four AD hybrid cell lines ad four control hybrid cell lines were cultured in DMEM or serum free medium. The production of reactive oxygen species (ROS) was determined by fluorophotometry assay with DCF-DA and Hoechst3342. The vitality of cells and the percentage of apoptotic cells were detected by MTT method and TUNEL assay respectively. Media from the cells were quantified for human Abeta1-40 level by solid phase sandwich ELISA and the expression of NFT was determined by immunocytochemistry staining with rabbit (polyclonal) anti-human NFT antiserum. Results: As compared with the control cybrids, ROS production and Abeta(1-40) secretion and the ratio of apoptotic cells were significantly increased (P<0.01) and the vitality of cells was statistically depressed (P<0.01) in AD cybrids. The control hybrid cell lines were not NFT immunoreactivity. In contrast, the AD hybrid cell lines were NFT strong immuno reactivity. Conclusions: Expression of mtDNA COX mutations in AD hybrids contributed to mitochondrial oxidative phosphorylation dysfunction leading to the elevated ROS production, which, in turn, induced Abeta oversecretion, NFT formation and neuronal death, suggesting a primary etiologic event that results in AD.
