Abstract:Objective: Epstein-Barr virus (EBV) is associated with arcinogenesis of many tumors, and EBV encoded latent membrane proteinl (LMP1) is an oncoprotein. The molecular mechanism of LMP1 involving has not been elucidated. This study was designed to investigate that LMP1 regulates the expression of Survivin through NFkappaB and AP-1 signaling pathways. Methods: The RTPCR and Western-Blotting assay were used, and a tetracycline-regulated LMP1-expressing nasopharyngeal carcinoma cell line HNE2 (Tet-on-LMP1-HNE2) serviced as cell model. The expressions of Survivin RNA and protein were detected after inducing the expression of LMP1 or/and blocking the activation of NFkappaB and AP-1 respectively. Results: The expression of Survivin RNA and protein increased along with expression of LMP1. Expression of Survivin RNA and protein reduced when the activation of NFkappaB and AP-1 was blocked. Survivin expression when blocking the activation of NFkappaB and AP-1 was lower than that of blocking NFkappaB and/or AP-1. Conclusion: LMP1 may trigger the expression of Survivin via NFkappaB and AP-1 signaling pathway, which is involved in carcinogenesis of tumor.
