Abstract:Isoquercitrin (quercetin 3-o-beta-glucoside) can be obtained on a large scale by directional hydrolysis of rutin with hesperidinase, which has more physiologically potential application than rutin in a wide range of biological activities. A rapid and inexpensive HPLC method in reversed phase mode has been developed for the quantitative analysis of rutin and its enzymatic hydrolysis products. Separation in HPLC-UV was on an Alltima C-18 column (250mmx4.6mm i.d.; 5 mu m) with a mobile phase consisting acetonitrile: 0.02% phosphoric acid solution (20:80 v/v), at a flow rate of 1.0 mL.min(-1), detected at 360nm. Regression equations of rutin and isoquercitrin were y = 0.0000002x-19287 (r(2)=0.9973) and y= 0.0000002x-4648 (r(2)=0.9990), which showed good linear relationships between the peak area of each marker and concentration, respectively. The method has good repeatability and precision, the intraday and interday RSD for both retention time and peak area was less than 1.10%. The mean recoveries of rutin and isoquercitrin in samples, measured at three concentration levels, were 96.6 and 98.9%, their RSD were 2.1% and 1.4% (n = 3), respectively. The biotransformation conversion rate of isoquercitrin was the highest when crude hesperidinase was used as biocatalyst. Aspergillus niger LJ-11 was selected as producer and it was used to obtain the crude hesperidinase in submerged fermentation with added rutin. The results indicated that the developed assay method was rapid, accurate, reliable and could be readily utilized as a quantitative analysis tool for investigation of isoquercitrin from enzymatic biosynthesis products of rutin.
