Abstract:A chemical investigation on the marine sponge Dysidea sp. resulted in the isolation of a series of diketopiperazines, including two new compounds, dysidines A ( 1 ) and B ( 2 ) as well as six known ones ( 3 – 8 ). Their structures with absolute configurations were determined on the basis of UV, IR, HRMS, NMR and calculated ECD method. Additionally, the cytotoxic, anti-inflammatory, antibacterial and antiviral activities of 1 – 8 were also tested. However, none of them exhibited significant bioactivities.

Abstract:A new diketopiperazine (DKP) derivative, (6R,3Z)-3-benzylidene-6-isobutyl-1-methyl piperazine-2,5-dione (1), as well as five known DKPs 2-6 was isolated from a deep sea-derived Streptomyces sp. SCSIO 04496. The structure of 1 was elucidated using a combination of 1D and 2D NMR, HR-ESI-MS and chiral-phase HPLC techniques. Compounds 1-6 did not show cytotoxic activity at a concentration of 100 μM in bioactivity assay.

Abstract:Four dimeric diketopiperazine stereoisomers (1-4) including two new ones (1-2) had been isolated from the culture broth of one marine-derived fungus Aspergillus sp. Z3, which was found in the gut of a marine isopod Ligia exotica. The planner structures and absolute configurations of the new compounds were determined by combination of NMR, HRESIMS, electronic circular dichroism calculation, Marfey's method as well as single-crystal X-ray diffraction. Their cytotoxicity against the prostate cancer PC3 cell line was assayed by the MTT method.

Abstract:Five new diketopiperazine alkaloids, brevicompanines D-H (3-7), together with two known analogs, allobrevicompanine B (1) and fructigenine B (2), were isolated from a deep ocean sediment derived fungus Penicillium sp. Their structures were established by spectroscopic methods including 2D NMR and chiral HPLC analysis. Compounds 4 and 7 inhibited lipopolysaccharide (LPS)-induced nitric oxide production in BV2 microglial cells.

Abstract:Three new 2,5-diketopiperazines, speramide C ( 1 ), 3,21- epi -taichunamide F ( 2 ), and 2- epi -amoenamide C ( 3 ), along with four known analogs ( 4 – 7 ), were obtained from the sponge-derived fungus Aspergillus sclerotiorum GDST-2013-0501 collected from the South China Sea. The chemical structures of new compounds were elucidated by analyzing NMR and MS spectroscopy data, and their absolute configurations were determined by electronic circular dichroism (ECD) calculations. Compound 1 represents the first prenylated indole alkaloid with an ethylene oxide ring at the isopentenyl side chain. Compound 4 displayed DNA topoisomerase I inhibitory activity and antibacterial activity against Staphylococcus epidermidis . The low cytotoxic or non-cytotoxic compound 4 displayed DNA topoisomerase I inhibitory activity, which could provide a starting point for the development of antitumor agents.

Abstract:To investigate the secondary metabolites from the marine sponge Hyrtios sp.of South China Sea,seven compounds were isolated from the dichloromethane extraction of Hyrtios sp.by Silica gel,ODS and HPLC.Their structures were identified by spectroscopic analysis and comparison with the literature as smenospongimine (1),smenospongiarine (2),sesquiter-penyl-amyl-amino-hydroxy-quinone (3),smenospongidine (4),dictyoceratin C (5),dictyoceratin A (6) and polyfibrospongol A (7),respectively.Compounds 1,2,3,4,5,6,7 were isolated from the genus of Hyrtios for the first time.

Abstract:Three new spiro-sesquiterpenoids, myrmekiones A-C (1-3), were isolated from the marine sponge Myrmekioderma sp. collected from the South China Sea. The structures of 1-3 were experimentally illuminated though comprehensive NMR spectra, X-ray diffraction analysis and calculated ECD. These three compounds possessed a special spiro skeleton. Compound 1 was characterized by a chamigrane-type structure, it is the first time to obtain the single-crystal of this type of oil compounds. 2 and 3 were a pair of diastereoisomers that possessed an acorane skeleton. This study expands the chemical diversity of marine origin spiro-metabolites.

Abstract:The gut actinobacteria of marine-inhabited fish is one of the most important reservoirs of novel natural products. Currently, the Streptomyces sp. MNU FJ-36 was isolated from the intestinal fabric of Katsuwonus sp. and determined by 16S rRNA analysis. From the cultures of the S. sp. MNU FJ-36, three new 2,5-diketopiperazines (2,5-DKPs) were discovered and identified as 3-(3-hydroxy-4-methoxybenzyl)-6-isobutyl-2,5-diketopiperazine (1), 3-(1,3-benzodioxol-5-ylmethyl)-6-isobutyl-2,5-diketopiperazine (2) and 3-(1,3-benzodioxol-5-ylmethyl)-6-isopropyl-2,5-diketopiperazine (3). Their structures were elucidated on the basis of spectroscopic data analysis. All the compounds were also evaluated for their inhibitory activity against P388, A-549 and HCT-116 cell lines with the MTT assay.

Abstract:Four new homoverrucosane-type diterpenes (1-4) and two known analogs (5-6) were isolated from the marine sponge Halichondria sp. Their structures including absolute configurations were determined by combination of spectroscopic analyses, theoretical calculations and comparison with data of literatures. The cytotoxicity against human multiple myeloma cell line RPMI-8266 of compounds 1-4 was evaluated, but none showed activity (IC50 > 10 mu M). It was the first report that homoverrucosanes isolated from the marine sponge Halichondria sp. (C) 2020 Published by Elsevier Ltd.

Abstract:OBJECTIVE:To study the chemical constituents of the marine sponge Rhabdastrella aff. distincta.METHOD:Compounds were obtained by normal phase silica gel, Sephadex LH-20, reverse phase chromatogrophy and methylation method. These structures were determined on the basis of IR, MS, NMR spectral analysis.RESULT:Eight compounds were isolated from bioactive CH3Cl fraction and identified as geoditin A (1), geoditin B (2), stellettin A (5), stellettin B (4), stellettin C (3), stellettinE (7), rhabdastrellic acid A (8) and the methyl ester of stellettin E (6).CONCLUSION:All these compounds were obtained from this species for the first time.

Abstract:One new quinazoline-containing diketopiperazine (1), along with 24 known compounds including nine alkaloids (2-9, and 25), thirteen lactones (10-22), aspterric acid (23), and catechol (24), were isolated from the marine sponge-derived Penicillium sp. SCSIO41043. Their planar structures were unequivocally elucidated by spectroscopic analysis. The absolute configuration of 1 was determined by a comparison of reported and experimental electronic circular dichroism (ECD) spectra. Compound 16 was found to notably inhibit the growth of five pathogenic bacteria and fungi with MIC values ranging from 0.5-16.0 μg/mL. Compounds 7, 17, 20, and 22 demonstrated moderate activity against Micrococcus luteus with MIC values ranging from 35.6 to 71.1 μg/mL. Moreover, 1-3 displayed different degrees of antioxidant activity with EC50 values of 0.98, 0.60, 0.46 mg/mL, respectively.

Abstract:One new 2,5-DKP derivative O-dihydroxycyclopenol (1) and seven known congeners 2-8 were isolated from the marine fungus Penicillium sp. ZJUT-34 cultured on rice medium. The planar structure of 1 was established by extensive spectroscopic analysis, including 1D, 2D NMR and HR-ESI-MS, while the relative configuration of 1 was determined by quantum chemical calculation. In the QS inhibitory assay, 1 significantly inhibited the production of violacein in Chromobacterium violaceum ATCC12472 (20.65%) at a concentration of 6.25 mu g/mL without affecting the growth of the strain, as compared with norharmane (22.14%), a quorum sensing inhibitor (QSI) identified in our previous study. It represented the first report on the QS inhibitory activity of the seven-membered 2,5-DKPs. In addition, compounds 1-8 were subjected to antibacterial assay against six pathogenic bacteria Compound 8 exhibited comparable antibacterial activity against Enterococcus faecalis FA2-2 (MIC = 96 mu g/mL) with the positive control gentamicin (MIC = 80 mu g/mL).

Abstract:Three new diketopiperazine derivatives (DKPs), saroclazines A–C (1–3) along with three known DKPs (4–6) were isolated from mangrove-derived fungi Sarocladium kiliense HDN11-84. Saroclazines A–B (1 and 2) possessed a free amide structure, which was first found in sulfur-containing aromatic DKPs. Their structures were elucidated by NMR, HRESIMS and X-ray. The cytotoxic activity of new compounds (1–3) was tested against HeLa cell lines, among which compound 2 showed an IC50 value of 4.2 µM.

Abstract:Two new polyketides (1-2), four new norterpenoids (3-6), together with five known compounds (7-11) were isolated from the marine sponge Diacarnus megaspinorhabdosa. The new structures were determined by extensive spectroscopic analyses, ECD spectra comparison, and Mosher's method. The cytotoxicities towards six cancer cell lines together with the antimicrobial activity against methicillinresistant Staphylococcus aureus, Canadida albicans, and Escherichia coli were valuated. Compound 10 showed cytotoxicities towards cancer cell lines SW480, NCl-H460, PC-9, and HepG2, with IC50 3.7, 7.8, 6.2 and 8.0 mu M respectively. (C) 2020 Elsevier Ltd. All rights reserved.
