Abstract:The present study is to investigate the role of [Ca2+]i in the apoptosis and cell cycle arrest of SGC-7901 and HepG2 induced by paclitaxel. In vitro experiments, MTT assay and flow cytometer (FCM) was used to determine the cell growth inhibitory rate and cell cycle distribution and apoptosis rate. Fluo-3/AM and laser confocal scanning microscopy (LCSM) to observe the change of [Ca2+]i in cells. The rate of positive cell (PT) of calcineurin (CaN) and Cytochrome-C were examined by immunofluorescence staining and FCM. The results showed paclitaxel inhibited the growth SGC-7901 cells and HepG2 cells in a dose-dependent manner. G(2)/M cell cycle block and apoptosis of SGC-7901 cells and HepG2 in treated group were induced by paclitaxel. Intracellular [Ca2+]i significantly increased and the rate of positive cell of calcineurin and cytochrome-C increased simultaneity in treaded group. Paclitaxel is able to arrest the cell cycle and induce the apoptosis in SGC-7901 cells and HepG2 by increasing the concentration of Intracellular [Ca2+]i, upgrade of the expression of CaN and improvement the activation of CaN. Ca2+ signal pathway plays the key role in rest of cell cycle and apoptosis of SGC-7901 cells and HepG2 cells.
