Abstract:AIM: To observe the effects of different steamed times of Polygonum miltiflorum Thunb. on swelling of acute inflammation of external ear in mice and find the best steamed time.METHODS: ① The experiment was conducted in the Department of Pharmacology, Medical College, Yanbian University from June to October 2004. Eighty-one male and female mice of either sex of Kunming were selected and randomly divided into nine groups with 9 mice in each group: saline group, raw Polygonum miltiflorum Thunb. group, 0,2,4,6,8,10 hours black bean juice mixing with steamed Polygonum miltiflorum Thunb. and indomethacin group. ② Polygonum miltiflorum Thunb. 10 kg was weighted, and 5 kg was put into 1.25 kg of black bean juice, compaining for 15 hours was soaked. Then 0.5 kg of soaked Polygonum miltiflorum Thunb. was weighted solely and was steamed for 0, 2, 4, 6, 8, 10 hours. ③ Saline group: The mice in saline group were infused with saline 20 ml/kg and fed normally per day. Those in the Polygonum miltiflorum Thunb. group were given 10 g/kg dosage, and the volume of gastric perfusion was 20 mL/kg. Those in the 0,2,4,6,8,10 hours black bean juice mixing with steamed group were treated with 10 g/kg dosage with the 20 mL/kg volume of gastric perfusion. Those in the indomethacin group were given 7.5 mg/kg dosage with the 0.2 mL/10 g volume of gastric perfusion. The mice in every administration group were treated with gastric perfusion once a day for 3 days. ④ The method of inflammatory and swelling of the earflaps of mice: The two sides earflaps of mice was induced by 20 (l xylene and croton 1 hour after the last administration, respectively. Mice were killed after 2 or 4 hour (the best inflammation time with xylene and croton) of inflammation. The ear of left or right was selected at the same part, weighting and degree of swelling (degree of swelling = mass of earflap after inflammation-mass of earflap before inflammation) was calculated. ⑤ Differences of measurement data were compared with single factor analysis of variance. RESULTS: Totally 81Kunming mice were involved in the result analysis. ① Degree of swelling of external ear induced by xylene at 2 hours after inflammatory of mice in the every black bean juice mixing with steamed Polygonum miltiflorum Thunb. group, raw Polygonum miltiflorum Thunb. group and indomethacin group was significantly lower than that in the saline group (P < 0.01). With the prolongation of steamed time in the every black bean juice mixing with steamed Polygonum miltiflorum Thunb. group, the degree of swelling of external ear decreased. The degree of swelling of external ear was the lowest in the 10 hours black bean juice mixing with steamed Polygonum miltiflorum Thunb. group. Effect of 8 and 10 hours black bean juice mixing with steamed Polygonum miltiflorum Thunb. on degree of swelling of external ear induced by xylene was equal to that in the indomethacin group (P > 0.05). ② The degree of swelling of the earflaps in each black bean juice mixing with steamed Polygonum miltiflorum Thunb. group, raw Polygonum miltiflorum Thunb. group and indomethacin group after 4 hours of inflammation due to croton was lower than that in the saline group (P < 0.05-0.01). With the elongation of steamed time, the degree of swelling of earflaps decreased. Degree of 10-hour black bean juice mixing with steamed Polygonum miltiflorum Thunb. was the lowest. There was no significant difference of each black bean juice mixing with steamed Polygonum miltiflorum Thunb. group on the anti-inflammatory action induced by croton oil, as compared with indomethacin group (P > 0.05).CONCLUSION: The black bean juice mixing with steamed Polygonum miltiflorum Thunb. of different steamed times have effect of anti-inflammation. Steamed for 10 hours with black bean has the best time for the anti-inflammatory action.
