Novel Tablets With Heterogeneous Distributions Of Release-Retarding Polymer Fabricated By 3dp

Abstract:A novel complex tablet was designed to furnish zero-order drug release profiles and was fabricated directly from the CAD models using three-dimensional printing technique (3DP). The tablets were in donut shape and had heterogeneous distributions of the release-retarding polymer in their three regions. Environmental scanning electron microscopy showed that the two dense regions, which had low porosity but high binding intensity, were symmetric around the middle loose region, which had high porosity but poor binding effect. Fluorescence tracing technique showed that the concentration of release-retarding polymer in the dense regions was about four times that in the loose region. In vitro dissolution tests showed that up to 97% of the drug in the tablets was released in a linear manner before leveling off to completion. However, the surface finish of the tablets was relatively rough and needed to be further improved. 3DP has advantages in the fabrication of complex tablets and can offer strategies for the development of novel dosage forms.
