Abstract:The effects were compared of three 2' chlorophenyl-benzodiazepines (triazolam, clonazepam and lorazepam) and three corresponding 2' deschloro-phenyl-derivatives (alprazolam, nitrazepam and oxazepam, respectively) on the incidence of six ethological elements in both timid and aggressive singly-housed male mice, treated with drugs in paired interactions with untreated non-aggressive males. Alprazolam and oxazepam reduced defensive upright postures and escapes at doses which did not reduce rearing and actually increased walking, while their chlorinated counterparts (triazolam and lorazepam, respectively) decreased incidence of defenses and escapes mostly at doses decreasing locomotor acts involving a similar movement (rears and walks, respectively). Alprazolam and oxazepam also reduced attacks at doses not reducing rears, in contrast to triazolam and lorazepam which reduced attacks only at doses suppressing rearing. Nitrazepam stimulated sniffing partners much more than its chlorinated counterpart clonazepam. The 2' deschloro-phenyl-benzodiazepines were more potent in reducing defensive-escape activities than attacks or locomotion. Yet, none of the benzodiazepines tested produced a complete inhibition of timid defensive-escape behavior at non-sedative doses. The present study suggests that 2' deschloro-phenyl-benzodiazepines are less "sedative" with respect to their "anxiolytic" activity.
Zaleplon and triazolam in humans: acute behavioral effects and abuse potential

Abstract:Zaleplon, a pyrazolopyrimidine that is under development as a hypnotic, produces its pharmacological effects at the benzodiazepine-recognition site on the GABA(A) benzodiazepine-receptor complex. Unlike most benzodiazepines, zaleplon binds selectively to the BZ1 (omega1) subtype of the benzodiazepine receptor. The present study compared the acute subject-rated effects, performance-impairing effects, and abuse potential of zaleplon and triazolam, a triazolobenzodiazepine hypnotic, in 14 healthy volunteers with histories of drug abuse. Zaleplon (25, 50, and 75 mg), triazolam (0.25, 0.5, and 0.75 mg) and placebo were administered orally in this double-blind, crossover study. Zaleplon and triazolam produced comparable dose-related effects on several subject-rated drug-effect questionnaires. Zaleplon and triazolam also produced comparable dose-dependent decrements on several performance tasks including balance, circular lights, digit-enter and recall, DSST, picture recall/recognition and repeated acquisition. Same-day and next-day subject-rated measures reflecting abuse potential (e.g., drug liking, good effects, and monetary street value) also suggest that zaleplon and triazolam were comparable. The only notable between-drug difference observed in the present study was that the time-action function of zaleplon differed from that of triazolam. The onset time, time to maximum drug effect, and duration of action were shorter with zaleplon than triazolam. Thus, despite its non-benzodiazepine structure and unique benzodiazepine-receptor binding profile, the behavioral pharmacological profile of zaleplon is similar to that of triazolam.
Behavioral effects of triazolam and pregnanolone combinations: reinforcing and sedative-motor effects in female rhesus monkeys

Abstract:Introduction: Benzodiazepines (BZs) are prescribed as anxiolytics, but their use is limited by side effects including abuse liability and daytime drowsiness. Neuroactive steroids are compounds that, like BZs, modulate the effects of GABA at the GABAA receptor. In a previous study, combinations of the BZ triazolam and neuroactive steroid pregnanolone produced supra-additive (i.e., greater than expected effects based on the drugs alone) anxiolytic effects but infra-additive (i.e., lower than expected effects based on the drugs alone) reinforcing effects in male rhesus monkeys, suggestive of an improved therapeutic window. Methods: Female rhesus monkeys (n=4) self-administered triazolam, pregnanolone, and triazolam-pregnanolone combinations intravenously under a progressive-ratio schedule. In order to assess characteristic sedative-motor effects of BZ-neuroactive steroid combinations, female rhesus monkeys (n=4) were administered triazolam, pregnanolone, and triazolam-pregnanolone combinations. Trained observers, blinded to condition, scored the occurrence of species-typical and drug-induced behaviors. Results: In contrast to our previous study with males, triazolam-pregnanolone combinations had primarily supra-additive reinforcing effects in three monkeys but infra-additive reinforcing effects in one monkey. Scores for deep sedation (i.e., defined as atypical loose-limbed posture, eyes closed, does not respond to external stimuli) and observable ataxia (any slip, trip, fall, or loss of balance) were significantly increased by both triazolam and pregnanolone. When combined, triazolam-pregnanolone combinations had supra-additive effects for inducing deep sedation, whereas observable ataxia was attenuated, likely due to the occurrence of robust sedative effects. Discussion: These results suggest that significant sex differences exist in self-administration of BZ-neuroactive steroid combinations, with females likely to show enhanced sensitivity to reinforcing effects compared with males. Moreover, supra-additive sedative effects occurred for females, demonstrating a higher likelihood of this adverse effect when these drug classes are combined.

Abstract:The benzodiazepines are psychoactive drugs with wide therapeutic applications as anxiolytics, anticonvulsants, and muscle relaxants (Zbinden 8: Randall 1967). Their site and mechanism of action have recently become the subject of extensive investigations because of the availability of tritium-labelled diazepam and flunitrazepam with high specific activity. Using these ligands. in vitro and in vivo binding studies have indicated that a specific high affinity binding site, the 'benzodiazepine receptor', in the brain may be relevant to the pharmacological action of the drugs (Mohler & Okada 1977; Squires & Braestrup 1977; Tallman et al 1979). Triazolobenzodiazepines. like alprazolam and triazolam, have been found to be both anxiolytic and hypnotic in man (Fabre & McLendon 1979; Chatwin &John 1976) in whom these drugs are metabolized to hydroxylated compounds and benzophenones, which are excreted in urine (Eberts et al 1980, 1981). The activity of alprazolam. triazolam and their metabolites on the benzodiazepine receptor is not known. We have undertaken to determine the activity of these compounds on benzodiazepine receptors by using [3H]flunitrazepam ([?H]FNZ) binding to crude rat brain membrane preparations. Abstract:Journal of Psychopharmacology, Ahead of Print. Background:Benzodiazepines bind to γ-aminobutyric acid type A (GABAA) receptor subtypes identified by different α subunits (i.e., α1GABAA, α2GABAA, α3GABAA, and α5GABAA). Sedative-motor effects of benzodiazepines are thought to involve α1GABAA and α3GABAA subtypes.Aims:We evaluated observable measures of sedative-motor effects and species-typical behaviors in monkeys following acute administration of novel GABAkines (positive allosteric modulators of GABAA receptors), with varying degrees of selective efficacy at different GABAA receptor subtypes. We predicted that the induction of sedative-motor effects would depend on the degree of α1GABAA and α3GABAA efficacy.Methods:Adult female rhesus monkeys (N = 4) were implanted with chronic indwelling i.v. catheters. Quantitative behavioral observation was conducted by trained observers following administration of multiple doses of the conventional benzodiazepine alprazolam and the GABAkines MP-III-80 (preferential efficacy at α2/α3/α5GABAA subtypes), KRM-II-81, MP-III-24 (both with preferential efficacy for α2/α3GABAA subtypes), and MP-III-22 (preferential potency and efficacy for α5GABAA subtypes).Results:As with alprazolam, all GABAkines induced significant levels of mild sedation ("rest/sleep posture"). Deep sedation was observed with alprazolam, MP-III-80, and MP-III-22; motoric effects (observable ataxia) were obtained with alprazolam, KRM-II-81, and MP-III-22 only. Surprisingly, the order of potency for rest/sleep posture was significantly associated only with potency at α5GABAA subtypes.Conclusions:GABAkines with preferential efficacy at α2/α3GABAA and/or α5GABAA subtypes engendered sedative-motor effects in monkeys, although only compounds with α5GABAA activity engendered deep sedation. Moreover, the significant relationship between potency obtained with in vitro electrophysiology data and the rest/sleep posture measure suggests a role for the α5GABAA subtype in this milder form of sedation.

Abstract:A series of 16 1-phenyl-1H-1,2,3-triazoles with substituents at both the 4- and 5-positions of the triazole ring were synthesized, and a total of 49 compounds, including previously reported 4- or 5-monosubstituted analogues, were examined for their ability to inhibit the specific binding of [(3)H]4'-ethynyl-4-n-propylbicycloorthobenzoate (EBOB), a non-competitive antagonist, to human homo-oligomeric beta3 and hetero-oligomeric alpha1beta2gamma2 gamma-aminobutyric acid (GABA) receptors. Among all tested compounds, the 4-n-propyl-5-chloromethyl analogue of 1-(2,6-dichloro-4-trifluoromethylphenyl)-1H-1,2,3-triazole showed the highest level of affinity for both beta3 and alpha1beta2gamma2 receptors, with K(i) values of 659pM and 266nM, respectively. Most of the tested compounds showed selectivity for beta3 over alpha1beta2gamma2 receptors. Among all 1-phenyl-1H-1,2,3-triazoles, the 4-n-propyl-5-ethyl analogue exhibited the highest (>1133-fold) selectivity, followed by the 4-n-propyl-5-methyl analogue of 1-(2,6-dibromo-4-trifluoromethylphenyl)-1H-1,2,3-triazole with a >671-fold selectivity. The 2,6-dichloro plus 4-trifluoromethyl substitution pattern on the benzene ring was found to be important for the high affinity for both beta3 and alpha1beta2gamma2 receptors. Comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) provided similar contour maps, revealing that an electronegative substituent at the 4-position of the benzene ring, a compact, hydrophobic substituent at the 4-position of the triazole ring, and a small, electronegative substituent at the 5-position of the triazole ring play significant roles for the high potency in beta3 receptors. Molecular docking studies suggested that the putative binding sites for 1-phenyl-1H-1,2,3-triazole antagonists are located in the channel-lining 2'-6' region of the second transmembrane segment of beta3 and alpha1beta2gamma2 receptors. A difference in the hydrophobic environment at the 2' position might underlie the selectivity of 1-phenyl-1H-1,2,3-triazoles for beta3 over alpha1beta2gamma2 receptors. The compounds that had high affinity for beta3 receptors with homology to insect GABA receptors showed insecticidal activity against houseflies with LD(50) values in the pmol/fly range. The information obtained in the present study should prove helpful for the discovery of selective insect control chemicals.
Diazepam Produces Disinhibitory Cognitive Effects in Male Volunteers

Abstract:Rationale Diazepam has well known amnestic and sedative effects but effects on fronto-executive function remain largely uninvestigated, especially on neuropsychologically validated tests of risk taking and orbitofrontal cortex function. Objectives We aimed to determine the impact of diazepam on a variety of executive tasks. Methods The effects of 5, 10 and 20 mg of diazepam on a battery of neuropsychological tests were investigated using a randomised, double blind, placebo-controlled design. Seventy-five adult men were recruited. The Rogers et al. ( 1999b ) test of risk-taking was given along with tasks from the CANTAB battery. Results Diazepam impaired performance on the Tower of London test of planning, without influencing visual pattern recognition memory. Subjects who had taken diazepam made more risky choices on the risk-taking task. On two speeded reaction time tasks diazepam impaired discrimination sensitivity and increased the bias to respond. Conclusions In contrast to the well-known sedative effects of diazepam, we demonstrate disinhibitory effects on two speeded reaction time tasks. Our results show that diazepam can impair performance on reaction time tasks both by impairing sensitivity and by increasing the bias to respond. Furthermore diazepam impaired performance on tests of planning and risky decision making that depend predominantly on dorsolateral and orbitofrontal regions of the prefrontal cortex, respectively.
Contrasting Effects of Clonidine and Diazepam on Tests of Working Memory and Planning

Abstract:Theα2 adrenoceptor has recently been implicated in working memory (WM), a function dependent on the integrity of the prefrontal cortex. Using a double-blind, placebo-controlled design, the present investigation examines the effects of two doses (1.5 µg/kg and 2.5 µg/kg) of the mixedα1/α2 adrenoceptor agonist clonidine (CLO) on performance of various computerised tests of WM and planning in healthy, young volunteers. These are compared to the effects produced by two doses (5 mg and 10 mg) of diazepam (DZP) on largely the same set of neuropsychological tests in a comparable set of subjects. Administration of CLO resulted in impulsivity of responding in a planning task, as well as differential dose-dependent effects on two analogous tests of spatial and visual WM. The nature of these effects were suggestive of mnemonic, rather than executive, dysfunction. Conversely, DZP produced specific deficits on tests of spatial WM and planning very similar to those seen following lesions to the frontal lobes. Therefore, these two sedative drugs produce doubly dissociable, dose-dependent effects on different aspects of cognitive function.
Self-Administration of Benzodiazepines in Drug-Naïve Rhesus Monkeys: Role of GABAA Receptor Subtypes

Abstract:ID 87806 Poster Board 496 Benzodiazepines (BZs) are safe and effective therapeutics for anxiety when used as directed, however, BZs have unwanted side effects like potential for misuse, physical dependence, and toxicity when combined with other substances. BZs bind to a site on the GABA A receptor located at the interface of the alpha and gamma subunit, with alpha 1, alpha 2, and alpha 3 subunit-containing receptors likely involved in their reinforcing effects. Furthermore, compounds that lack efficacy at the alpha 1 subunit-containing GABA A receptor (alpha 1 sparing compounds) appear to have reduced addiction potential, though their reinforcing effects are dependent upon past drug experience. Choice procedures were used to evaluate whether subjects without prior drug experience would self-administer alpha 1 sparing compounds when substituted for food or when combined with food. Two female and two male rhesus monkeys chose between a fixed food amount on one option (1 or 2 pellets/delivery) and on the other option, in different conditions, between increasing food amounts (0-4 pellets/delivery), increasing drug doses (0-0.1 mg/kg/injection), or increasing food amounts combined with a fixed drug dose (0-0.1 mg/kg/injection). Three alpha 1 sparing compounds with various levels of functional selectivity at alpha 2, alpha 3, and alpha 5 subunit-containing GABA A receptors (a2GABA A , a3GABA A , a5GABA A , respectively) were evaluated. The compounds YT-III-31 (0-0.1 mg/kg/injection), MP-III-24 (0-0.1 mg/kg/injection), and MP-III-80 (0-0.1 mg/kg/injection), with functional selectivity for a3GABA A , a2/3GABA A , and a2/3/5GABA A receptors, respectively, were not self-administered on their own, and failed to reliably produce enhancement of food choice. These results suggest that alpha 1 sparing compounds do not function as reinforcers on their own in drug naïve subjects and is similar to outcomes obtained with cocaine-experienced subjects. However, unlike with cocaine experience, alpha 1 sparing compounds did not reliably enhance choice of another reinforcer (i.e., food). These experiments are currently being replicated with a nonselective BZ, midazolam, and the alpha 1 preferring compound, zolpidem, in the same subjects. From a therapeutic perspective, alpha 1 sparing compounds may hold promise as treatments for anxiety with reduced potential for misuse and in the treatment of BZ use disorder. Effects of Scopolamine on Delayed-Matching-To-Sample and Paired Associates Tests of Visual Memory and Learning in Human Subjects: Comparison with Diazepam and Implications for Dementia

Abstract:Two experiments examined dose-related effects of 200, 400 and 600 μg scopolamine (n = 24, SC) and 5 and 10 mg diazepam (n = 6, PO) on parallel tests of visual memory and learning taken from the CANTAB battery. Scopolamine significantly impaired accuracy of performance on a delayed matching to sample test of visual recognition memory in a dose-and delay-dependent manner, but had only marginal decremental effects on a test of visuospatial paired associates learning. Scopolamine significantly lengthened decision times in a visual search matching to sample task at the 400 and 600 μg doses, without significantly affecting accuracy. The drug also impaired performance on tests of spatial (on accuracy and response time measures) and pattern (on response time only) memory. Most of the deleterious effects on scopolamine were removed by covariance analyses with indices of subjective sedation, but the effects of delayed matching accuracy and latency remained. By contrast, diazepam significantly impaired paired associates learning but affected delayed matching to sample in a delay-independent manner. These results suggest that scopolamine can produce selective deficits in tests of short-term visual recognition memory which do not depend on overall impairments in arousal and which contrast with deficits in visual associative learning produced by diazepam. They have implications for the pharmacological modelling of dementia and memory disorders in man and for the neurochemical substrates of the short-term recognition memory and associative learning for visual stimuli.
Clonidine and Diazepam Have Differential-Effects on Tests of Attention and Learning

Abstract:The noradrenergic system has repeatedly been implicated in the mediation of attentional processes. Using a double-blind, placebo-controlled design, the present investigation examines the effects of two doses (1.5 µg/kg and 2.5 µg/kg) of theα2 adreno ceptor agonist clonidine (CLO) on performance of various computerised tests of attention and learning in healthy, young volunteers. These are compared to the effects produced by two doses (5 mg and 10 mg) of diazepam (DZP) on largely the same set of neuropsychological tests in a comparable set of subjects. Both doses of CLO were found to impair performance of the RVIP test of sustained attention, while the higher dose alone improved visuo-spatial learning. Conversely, the higher dose of DZP produced profound deficits on visuo-spatial learning, and impaired attentional set-shifting. This study suggests a role for theα2 adrenoceptor in selective attention, and for the benzodiazepine receptor in specific cognitive processes mediated by discrete cortical regions.
Delayed-Response Deficit Induced By Local Injection Of The Alpha(2)-Adrenergic Antagonist Yohimbine Into The Dorsolateral Prefrontal Cortex In Young-Adult Monkeys

Abstract:Two young adult monkeys (Macaca mullata) were trained to perform a delayed-response task that required the monkeys to remember a cued spatial position (left or right) over a delay interval and then to make a response to the cued position. Local injection of the ��2-adrenergic antagonist yohimbine (10 ��g in 2 ��l saline) into the dorsolateral prefrontal cortex (Walker's area 46 and area 9) impaired the performance of the delayed-response task, and it was without effect on the performance of the task if there was no delay between the cue and choice signals. The main performing error after injection of yohimbine was that the monkeys responded to uncued position with higher rate. Local injection of the ��1-adrenergic antagonist prazosin (10 ��g in 2 ��l saline) or the ��-adrenergic antagonist propranolol (10 ��g in 2 ��l saline) into the same cortical areas induced no significant effect on the performance of the task. The present study suggests that prefrontal ��2-adrenoceptors play an important role in the spatial working memory in young adult monkeys. ? 1994 Academic Press, Inc.

Abstract:GABAA (γ-aminobutyric acidA) receptors are molecular substrates for the regulation of vigilance, anxiety, muscle tension, epileptogenic activity and memory functions, which is evident from the spectrum of actions elicited by clinically effective drugs acting at their modulatory benzodiazepine-binding site. Here we show, by introducing a histidine-to-arginine point mutation at position 101 of the murine α1-subunit gene, that α1-type GABAA receptors, which are mainly expressed in cortical areas and thalamus1, are rendered insensitive to allosteric modulation by benzodiazepine-site ligands, whilst regulation by the physiological neurotransmitter γ-aminobutyric acid is preserved. α1(H101R) mice failed to show the sedative, amnesic and partly the anticonvulsant action of diazepam. In contrast, the anxiolytic-like, myorelaxant, motor-impairing and ethanol-potentiating effects were fully retained, and are attributed to the nonmutated GABAA receptors found in the limbic system (α2, α5), in monoaminergic neurons (α3) and in motoneurons (α2, α5)1. Thus, benzodiazepine-induced behavioural responses are mediated by specific GABAA receptor subtypes in distinct neuronal circuits, which is of interest for drug design.

Abstract:Benzodiazepines are commonly used drugs to treat anxiety in crime witnesses. These increase GABA inhibitory effects, which impairs aversive memory encoding and consolidation. Eyewitness memory is essential in justice. However, memory is malleable leading to false memories that could cause a selection of an innocent in a lineup. Here, we studied whether a low dose of Clonazepam impairs memory encoding as well as consolidation of faces and narrative of the event. We performed two experiments using a double-blind and between subject design (N = 216). Day 1: subjects watched a crime video and received Clonazepam 0.25 mg (CLZ group) or placebo (PLC group) before (Exp. 1) or after the video (Exp. 2) to assess the effect on encoding and consolidation. One week later, the memory was assessed using a present and absent target lineup and asking for a free recall. Regarding encoding, we found that in the CLZ group memory was impaired in the free recall task, while no differences were found for recognition memory. Regarding consolidation, we did not observe memory measures that were affected by this dose of benzodiazepines. The results suggest that while some aspects of eyewitness memory could be modulated even with low doses of benzodiazepine, others could not be affected. More studies should be performed with higher doses of CLZ similar to those administered in real life. These results are relevant in the judicial field to assess the reliability of the eyewitness elections under the effects of this drug.

Abstract:Behavioral flexibility, which allows organisms to adapt their actions in response to environmental changes, is impaired in a number of neuropsychiatric conditions, including obsessive-compulsive disorder and addiction. Studies in human subjects and monkeys have reported correlations between individual differences in dopamine D2-type receptor (D2R) levels in the caudate nucleus and performance in a discrimination reversal task, in which established contingent relationships between abstract stimuli and rewards (or punishments) are reversed. Global genetic deletion of the D2R in mice disrupts reversal performance, indicating a likely causal role for this receptor in supporting flexible behaviors. To directly examine the specific role of caudate D2-type receptors in reversal performance, the D2/3/4R agonist quinpirole was infused via chronic indwelling cannulae into the medial caudate of male and female marmoset monkeys performing a touchscreen-based serial discrimination reversal task. Given prior evidence for dose-dependent effects of quinpirole and other dopaminergic drugs, a full dose-response curve was established. Individually, marmosets displayed marked differences in behavioral sensitivity to specific doses of intra-caudate quinpirole. Collectively, they exhibited a behaviorally specific bi-phasic deficit in reversal learning, being consistently impaired at both relatively low and high doses of quinpirole. However, intermediate doses of intra-caudate quinpirole produced significant improvement in reversal performance. These data support previous human and monkey neuroimaging studies by providing causal evidence of a U-shaped function describing how dopamine modulates cognitive flexibility in the primate striatum.
Thalamus Segmentation from MRI Images by Lagrangian Surface Flow

Abstract:In this paper, we present a new thalamus segmentation method for MRI images based on a new type of deformable model-Lagrangian surface flow. Given a MRI image, the user can interactively initialize a seed model within region of interest. The model will then start to grow according to both boundary and region information based on the principle of variational analysis. The deformation will stop when an equilibrium state is achieved. Our experiments demonstrate that the new method is robust to image noise and inhomogeneity and will not get stuck into local minima or leak from spurious edge gaps
Idazoxan Potentiates Rather Than Antagonizes Some of the Cognitive Effects of Clonidine

Abstract:Several investigations have revealed substantial influences of pharmacological manipulation of central noradrenergic activity upon performance in cognitive tests sensitive to frontal lobe dysfunction. They suggest a significant role for the noradrenergic coeruleo-cortical projection in cognitive function but conflicting findings and the complex pharmacology of adrenoceptor agents make it difficult to be precise about underlying mechanisms. In order to clarify these we have compared the effects of an α1/α2-adrenoceptor agonist, clonidine, an α2-adrenoceptor antagonist, idazoxan, and these agents in combination. Three groups of healthy volunteers were used to investigate the effects of these noradrenergic manipulations upon performance of tasks from the CANTAB test battery known to be sensitive to frontal lobe dysfunction. Previously reported effects of clonidine upon sustained visual attention and upon session-to-session improvement were replicated. Furthermore, idazoxan inhibited the hypotensive effect of clonidine. Idazoxan had no overall effect on performance of any of the tests but did inhibit session-to-session improvement in performance of a planning task, attentional set shifting and sustained visual attention. Rather than leading to the anticipated mutual antagonism of effects, combining clonidine and idazoxan led to a wider and more striking range of cognitive impairments. These results are discussed alongside findings which support a role for imidazoline (I1) receptors in blood pressure control, where clonidine and idazoxan are antagonistic, and evidence of less potent antagonism at somato-dendritic α2-adrenoceptors in the locus coeruleus.

Abstract:The mass spectrometric behaviour of four cis- and trans-la, 3-disubstituted -1,1 -dichloro-4-formyl-1a,2,3,4-tetrahydro-1H-azirino[1, 2-a] [1,5] benzodiazepines has been studied with the aid of mass-analysed ion kinetic energy spectrometry and exact mass measurements under electron impact ioniza-tion. All compounds show a tendency to eliminate a chlorine atom from the aziridine ring, and then eliminate a neutral propene or styrene from the diazepine ring to yield azirino[ 1, 2-6][1,3]benzimidazole ions. These azmno[1,2-a] [1,5]-benzodiazepines can also eliminate HCl, or Cl plus HG simultaneously to undergo a ring enlargement rearrangement to yield 1,6-benzodiazocine ions, which further lose small molecular fragments, propyne or phenylacetylene, with rearrangement to give quinoxaline ions.
