Abstract:Apolipoprotein E (APOE) is a risk factor for late-onset Alzheimer's disease (AD). APOE4/4 homozygosity increases AD risk by >10-fold. Although an association between the APOE ε4allele and increased risk has been established, the mechanism by which APOE4 increases disease risk is poorly understood. Here, we hypothesize that APOE4 contributes to disease risk through both cell autonomous and non-cell autonomous mechanisms. We have optimized protocols to develop pure populations of human induced pluripotent stem cells (iPSCs)-derived astrocytes and neurons, determined by immunohistochemistry and global gene expression. For molecular and functional characterization we have established a co-culture system of astrocytes and neurons and measured their activities by the multi-electrode array (MEA), inflammatory and calcium-based assays. APOE4/4 or APOE3/3 iPSCs are being engineered to generate isogenic lines using CRISPR/Cas9 genome-editing. We will then use our differentiation and co-culture strategy to test the impact of APOE genotype in isogenic lines on neuronal and astrocyte gene expression and function. We have developed a protocol to efficiently and reproducibly generate pure populations of astrocytes and neurons from iPSC-derived neuroprogenitor cells (NPCs). Our iPSC-astrocytes are positive for astrocyte markers including S100B, GFAP, GLAST and ALdh1L1. Quantitative PCR confirmed decreased expression of neuronal markers such as TUJ1, MAP2, RELN and CACNA1C. RNAseq comparisons show that iPSC-astrocytes cluster with human fetal astrocytes and not with the source NPCs or matched NPC-derived neurons. Using the MEA platform, we have shown that co-culture of iPSC-excitatory neurons with iPSC-astrocytes is sufficient to improve the neuronal firing of co-cultured iPSC-neurons. iPSC-astrocytes secrete IL-6 in response to treatment with poly I:C, LPS or Ab42 and have constant waves of calcium signals in a manner similar to primary astrocytes. This established co-culture system will be applied to astrocytes and neurons differentiated from APOE4/4 and isogenic APOE3/3iPSC lines. Our optimized differentiation protocol gives consistent results across multiple lines, and significant changes are observed in neuronal activity as a consequence of co-culture of neurons with astrocytes. We are currently co-culturing astrocytes and neurons of different APOE genotypes to determine the impact of APOE4/4 astrocytes on isogenic APOE3/3 neuronal gene expression and activity.
