Abstract:Endothelin-1 is a key regulator of vascular tone and blood pressure in health and disease. We have recently found that ET-1 production in human microvascular endothelial cells (HMECs) can be promoted by angiotensin II (Ang II) through a novel mechanism involving octamer-binding transcription factor-1 (Oct-1), NADPH oxidase-2 (NOX2), and superoxide anions. As the formation of bioactive ET-1 also depends on endothelin-converting enzyme-1 (ECE-1), we investigated the transcriptional regulation of the ECE1 gene. We found that exposure of HMECs to Ang II resulted in a concentration- and time-dependent increase in ECE1 mRNA expression. Pharmacological inhibition of ECE-1 reduced Ang II-stimulated ET-1 release to baseline values. The effect of Ang II on ECE1 mRNA expression was associated with Oct-1 binding to the ECE1 promoter, resulting in its increased activity. Consequently, the Ang II-stimulated increase in ECE1 mRNA expression could be prevented by siRNA-mediated Oct-1 inhibition. It could also be abolished by silencing the NOX2 gene and neutralizing superoxide anions with superoxide dismutase. In mice fed a high-fat diet, cardiac expression of Ece1 mRNA increased in wild-type mice but not in Nox2-deficient animals. It can be concluded that Ang II engages Oct-1, NOX2, and superoxide anions to stimulate ECE1 expression in the endothelium.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore through which mechanism angiotensin II (Ang II) induces the expression of endothelin - converting enzyme - 1 (ECE - 1) in endothelial cells. Specifically, the study aims to reveal the role of reactive oxygen species (ROS) derived from NADPH oxidase 2 (NOX2) in this process, and whether this role is related to the activation of octamer - binding transcription factor 1 (Oct - 1). ### Research Background - **Endothelin - 1 (ET - 1)**: ET - 1 is a potent and long - lasting vasoconstrictor peptide, which plays a crucial role in maintaining vascular tone, blood pressure and tissue perfusion. However, if the level of ET - 1 is dysregulated, it may lead to pathological effects such as systemic hypertension, pulmonary hypertension, heart failure and chronic kidney disease. - **ECE - 1**: ECE - 1 is a key enzyme that converts the precursor protein prepro - ET1 into mature and biologically active ET - 1. The expression and activity of ECE - 1 are regulated by multiple factors, but the specific mechanisms are not yet fully understood. ### Research Objectives - **Explore the effect of Ang II on ECE - 1 expression**: Researchers have found that Ang II can promote the production of ET - 1 through NOX2 - derived superoxide anions and the activation of Oct - 1. This study further explores whether Ang II regulates the expression of ECE - 1 through the same mechanism. - **Verify the roles of NOX2 and Oct - 1 in ECE - 1 expression**: Through pharmacological inhibition and gene silencing experiments, verify the specific roles of NOX2 and Oct - 1 in Ang II - induced ECE - 1 expression. ### Main Findings - **The necessity of ECE - 1 in Ang II - induced ET - 1 production**: Pretreatment of human microvascular endothelial cells (HMECs) with the ECE - 1 inhibitor CGS35066 showed that blocking ECE - 1 could completely inhibit Ang II - induced ET - 1 release. - **The concentration - and time - dependent effects of Ang II on ECE1 mRNA expression**: Ang II increases the expression of ECE1 mRNA in a concentration - and time - dependent manner, with the maximum effect occurring at an Ang II concentration of 1 µmol/L and peaking at 3 hours. - **Responsiveness of the ECE1 promoter region**: Through partial deletion of the ECE1 promoter region, it was found that the fragment containing positions - 1108 to - 609 is crucial for the response to Ang II. This fragment contains a potential high - affinity Oct - 1 binding site. - **The role of NOX2 - derived superoxide anions**: NOX2 gene silencing and superoxide anion neutralization experiments showed that NOX2 - derived superoxide anions play a key role in Ang II - induced ECE1 expression. - **Animal experiment verification**: In the hearts of mice fed a high - fat diet, the expression of Ece1 mRNA was significantly increased in wild - type mice, but not in Nox2 - knockout mice, further supporting the role of NOX2 in this process. ### Conclusions - **Ang II regulates the expression of ECE - 1 through NOX2 and Oct - 1**: The research results show that Ang II promotes the expression of ECE - 1 through NOX2 - derived superoxide anions and the activation of Oct - 1, thereby ensuring the effective production and activity of ET - 1. - **Potential clinical significance**: This mechanism may play a role in multiple pathological conditions, especially in metabolic disorders caused by high - fat diets, providing a theoretical basis for the development of new treatment strategies.
