Abstract:Luminescent gold nanocrystals (AuNCs) are a recently-developed material with potential optic, electronic and biological applications. They also demonstrate energy transfer (ET) acceptor/sensitization properties which have been ascribed to Förster resonance energy transfer (FRET) and, to a lesser extent, nanosurface energy transfer (NSET). Here, we investigate AuNC acceptor interactions with three structurally/functionally-distinct donor classes including organic dyes, metal chelates and semiconductor quantum dots (QDs). Donor quenching was observed for every donor-acceptor pair although AuNC sensitization was only observed from metal-chelates and QDs. FRET theory dramatically underestimated the observed energy transfer while NSET-based damping models provided better fits but could not reproduce the experimental data. We consider additional factors including AuNC magnetic dipoles, density of excited-states, dephasing time, and enhanced intersystem crossing that can also influence ET. Cumulatively, data suggests that AuNC sensitization is not by classical FRET or NSET and we provide a simplified distance-independent ET model to fit such experimental data.

Abstract:Herein we report that few-atom silver nanoclusters (Ag NCs) can be effective energy acceptors for upconversion phosphors (UCPs). A luminescence resonance energy transfer (LRET) probe for biothiols was constructed by decorating UCPs with dithiol-stabilized Ag NCs. Owing to the unique properties of ultrasmall NCs, properties which bridge the gap between those of small molecules and those of nanoparticles, the use of approximately 1.9 nm Ag NCs as energy acceptors endows the probe with high energy-transfer efficiency, good biocompatibility, and flexibility. The UCP-Ag NC nanoprobe enables rapid and robust target assay in solutions. It was also uploaded into living cells and used to detect intracellular biothiol levels with high discrimination. Moreover, the probe shows transportability in vivo and can be used for tissue imaging. The facile growth of few-atom metal NCs on diverse templates may enable the development of various nanoprobes combining UCPs and metal NCs.

Abstract:Recent booming advances in luminescent gold nanoclusters (AuNCs), have prompted the development of novel fluorescent sensors. The luminescent AuNCs possess unique and intriguing physical and chemical properties including responsive photoluminescence and peroxide-like activity, providing abundant potentials for sensing strategy design. As of now, a wide variety of chem-/bio-sensors based on AuNCs have been developed and reviewed according to varied analytes. In this review, from a different point of view, we follow the route of how those sensors realize their functions and focus on the actual roles AuNCs play, in order to hierarchically and logically display the recent progress in the sensing applications of AuNCs. This review not only opens new windows to understand the development of sensors based on AuNCs but can also inspire broader and deeper utilization of luminescent nanomaterials.

Abstract:Fluorescent gold nanoclusters (AuNCs) have recently emerged as a novel kind of promising fluorescent probes for high-performance sensors and bioimaging because of their ultrasmall size (<3 nm), strong luminescence, good photostability, and excellent biocompatibility. Over the past decade, we have witnessed growing popularity of AuNCs in analytical applications and enormous efforts have been devoted to their development. In this review, we provide an update on recent advances in the development of AuNCs in terms of physicochemical properties, synthesis strategies, and bioapplications. The optical, electrochemical, catalytical, and solvatochromic properties of AuNCs are first summarized, which are followed by different ligands or template-assisted controllable synthetic methods. Afterwards functionalization of AuNCs is described in terms of ligand exchange, bioconjugation, and noncovalent interaction. We then focus on the applications of AuNCs as fluorescent probes for detection of metal ions, inorganic anions, small biomolecules, proteins, nucleic acids, drug molecules, pH, and temperature. We also summarize the usage of metal NCs in cellular and in vivo targeting and imaging. Finally, we conclude with a brief look at the future challenges and prospects of the development of AuNCs.

Energy Level Engineering in Gold Nanoclusters for Exceptionally Bright NIR Electrochemiluminescence at a Low Trigger Potential

Abstract:Electrochemiluminescence (ECL) is a widely used light output mechanism from electrochemical excitation. Understanding the intrinsic essence for ideal ECL generation remains a fundamental challenge. Here, based on the molecular orbital theory, we reported an energy level engineering strategy to regulate the ECL performance by using ligand-protected gold nanoclusters (AuNCs) as luminophores and N,N-diisopropylethylamine (DIPEA) as a coreactant. The energy level matching between the AuNCs and DIPEA effectively promoted their electron transfer reactions, thus improving the excitation efficiency and reducing the trigger potential. Simultaneously, the narrow band gap of the AuNCs further enabled enhanced emission efficiency. Using the energy level engineering theory developed here, a dual-enhanced strategy was proposed, and β-CD-AuNCs were designed to further verify this mechanism. The β-CD-AuNCs/DIPEA system resulted in highly stable near-infrared ECL with an unprecedented ECL efficiency (145-fold higher than that of the classic Ru(bpy)32+/tetra-n-butylammonium perchlorate system) and a low trigger potential of 0.48 V. A visual NIR-ECL based on this ECL system was successfully realized by an infrared camera. This work provides an original mechanistic understanding for designing efficient ECL systems, which promises to be a harbinger for broad applicability of this strategy for other ECL systems and ECL sensing platforms.

Abstract:Atomically precise gold nanoclusters (Au NCs) have high specific surface area and abundant unsaturated active sites. Traditionally, Au NCs are employed as thermocatalysts for multielectron transfer redox catalysis. Meanwhile, Au NCs also exhibit discrete energy levels, tunable photophysical and electrochemical properties, including visible to near infrared absorption, microsecond long-lived excited-state lifetime, and redox chemistry. In recent years, Au NCs are increasingly employed as visible to near infrared photocatalysts for their high photocatalytic activity and unique selectivity. This review focuses on the photophysical properties of a variety of Au NCs and their employment as photocatalysts in photocatalytic reactions and related applications including solar energy conversion and photodynamic therapies.

Abstract:Gold nanoclusters (Au NCs) exhibit broad fluorescent spectra from visible to near-infrared regions and good enzyme-mimicking catalytic activities. Combined with excellent stability and exceptional biocompatibility, the Au NCs have been widely exploited in biomedicine such as biocatalysis and bioimaging. Especially, the long fluorescence lifetime and large Stokes shift attribute Au NCs to good probes for fluorescence sensing and biological detection. In this review, we systematically summarized the molecular structure and fluorescence properties of Au NCs and highlighted the advances in fluorescence sensing and biological detection. The Au NCs display high sensitivity and specificity in detecting iodine ions, metal ions, and reactive oxygen species, as well as certain diseases based on the fluorescence activities of Au NCs. We also proposed several points to improve the practicability and accelerate the clinical translation of the Au NCs.

Abstract:Ultrasmall gold nanoclusters (NCs) have been engineered as a new kind of functional material due to their excellent photoluminescence properties. However, the synthesis of highly luminescent water-soluble nanoclusters with near-infrared (NIR) emission remains limited. Herein, we developed a pH-regulated strategy to facilitate the construction of self-assemblies with enhanced luminescence based on aggregation-induced emission (AIE) strategy. Using 2-mercaptobenzoic acid (MBA) as reductant and stabilizer, the original weakly luminescent AuNCs exhibited intense emission by adjusting pH controllably. The formation of compact organized nanostructures could effectively restrict the rotation and vibration of capping ligands by non-covalent interactions, which reduced the nonradiative relaxation from excited states and finally improved the emission properties of AuNCs. Moreover, the assemblies possess many intriguing features including bright NIR luminescence and excellent biocompatibility, which could be used as luminous probes in biological molecules sensing (tyrosinase (TYR) and dopamine (DA)) and promising candidates for cell imaging. This study provides a simple and feasible strategy for developing metal NCs-based smart optical materials in the field of bioscience.

Abstract:Gold nanoclusters (AuNCs) are a class of novel luminescent nanomaterials that exhibit unique properties of ultra-small size, featuring strong anti-photo-bleaching ability, substantial Stokes shift, good biocompatibility, and low toxicity. Various biomolecules have been developed as templates or ligands to protect AuNCs with enhanced stability and luminescent properties for biomedical applications. In this review, the synthesis of AuNCs based on biomolecules including amino acids, peptides, proteins and DNA are summarized. Owing to the advantages of biomolecule-protected AuNCs, they have been employed extensively for diverse applications. The biological applications, particularly in bioimaging, biosensing, disease therapy and biocatalysis have been described in detail herein. Finally, current challenges and future potential prospects of bio-templated AuNCs in biological research are briefly discussed.

Abstract:Abstract Metal nanoclusters (NCs) have been developed as a new class of luminescent nanomaterials with potential applications in various fields. However, for most of the metal NCs reported so far, the relatively low photoluminescence quantum yield (QY) in aqueous solution hinders their applications. Here, we describe the utilization of bis-Schiff base linkages to restrict intramolecular motion of surface motifs at the single-cluster level. Based on Au 22 (SG) 18 (SG: glutathione) NCs, an intracluster cross-linking system was constructed with 2,6-pyridinedicarboxaldehyde (PDA), and water-soluble gold NCs with luminescence QY up to 48% were obtained. The proposed approach for achieving high emission efficiency can be extended to other luminescent gold NCs with core-shell structure. Our results also show that the content of surface-bound Au(I)-SG complexes has a significant impact on the PDA-induced luminescence enhancement, and a high ratio of Au(I)-SG will be beneficial to increasing the photoluminescence intensity of gold NCs.

Revealing the Mechanism of Photoluminescence from Single Gold Nanospheres by Defocused Imaging

Abstract:The mechanism for the photoluminescence (PL) emission from gold nanoparticles has attracted considerable attention for many years. However, there is an important gap between small nanoclusters (similar to 2 nm) and larger plasmonic particles (similar to 50 nm). In this work, using defocused imaging technique, we investigate the PL properties of gold nanospheres (15-20 nm in diameter) on a single particle level. Photoblinking and photobleaching phenomena are both observed. We notice that although these nanospheres can support surface plasmon resonance at similar to 515 nm, they emit at similar to 630 nm (excited by 532 nm), which is obviously plasmon-independent. The observed defocused images (DIs) exhibit isotropy first and then either transform into anisotropy or vanish rapidly. Surprisingly, the Dls can change their emission pattern within one single nanosphere during the tracing time. All these PL properties suggest a multiple-dipole emission model. High-resolution transmission electron microscopy images demonstrate that these nanospheres are polycrystalline containing multiple small crystal domains (similar to 3 nm). We believe that these small domains divide the particle into different clusters and give rise to the photoblinking behavior and rotary DIs. The presented PL mechanism links gold nanoclusters and single-crystal nanorods, which could be helpful in understanding the origins of photoblinking and the luminescent properties from metallic nanoparticles. In addition, these water-soluble gold nanospheres provide new opportunities for biological labels and light-emitting sources in nanophotonics.
Unveiling the photoluminescence dynamics of gold nanoclusters with fluorescence correlation spectroscopy

Abstract:Gold nanoclusters (AuNCs) have captured significant interest for their photoluminescent properties; however, their rapid photodynamics remain elusive while probed by ensemble-averaging spectroscopy techniques. To address this challenge, we use fluorescence correlation spectroscopy (FCS) to uncover the photoluminescence dynamics of colloidal Au18(SG)14 nanoclusters. Our FCS analysis reveals the photoluminescence (PL) brightness per nanocluster, elucidating the impact of photoexcitation saturation and ligand interactions. Unlike DNA-encapsulated silver nanoclusters, the gold counterparts notably exhibit minimal blinking, with moderate amplitudes and 0.2 ms characteristic times. Our data also clearly reveal the occurrence of photon antibunching in the PL emission, showcasing the quantum nature of the PL process, with each AuNC acting as an individual quantum source. Using zero-mode waveguide nanoapertures, we achieve a 16-fold enhancement of the PL brightness of individual AuNCs. This constitutes an important enabling proof-of-concept for tailoring emission properties through nanophotonics. Overall, our study bridges the gap between ensemble-averaged techniques and single-molecule spectroscopy, offering new insights into AuNC photodynamics for biosensing and imaging applications.

Abstract:Localized surface plasmon resonance coupling effects (LSPR) have attracted much attention due to their interesting properties. This Letter demonstrates significant photoluminescence (PL) enhancement in the Au NS/CH3NH3PbI3-xClx heterostructures via the LSPR coupling. The observed PL emission enhancement is mainly attributed to the hot electron energy transfer effect related to the LSPR coupling. For the energy transfer effect, photo-generated electrons will be directly extracted into Au SPs, rather than relaxed into exciton states. This energy transfer process is much faster than the diffusion and relaxation time of free electrons, and may provide new ideas on the design of high-efficiency solar cells and ultrafast response photodetectors.

Abstract:Surface-protecting ligands, as a major component of metal nanoclusters (MNCs), can dominate molecular characteristics, performance behaviors, and biological properties of MNCs, which brings diversity and flexibility to the nanoclusters and largely promotes their applications in optics, electricity, magnetism, catalysis, biology, and other fields. We report herein the design of a new kind of water-soluble luminescent gold nanoclusters (AuNCs) for enzyme-activatable charge transfer (CT) based on the ligand engineering of AuNCs with 6-mercaptopurine ribonucleoside (MPR). This elaborately designed cluster, Au 5 (MPR) 2 , can form a stable intramolecular CT state after light excitation, and exhibits long-lived color-tunable phosphorescence. After the cleavage by purine nucleoside phosphorylase (PNP), the CT triplet state can be easily directed to a low-lying energy level, leading to a bathochromic shift of the emission band accompanied by weaker and shorter-lived luminescence. Remarkably, these ligand-engineered AuNCs show high affinity towards PNP as well as decent performance for analyzing and visualizing enzyme activity and related drugs. The work of this paper provides a good example for diversifying physicochemical properties and application scenarios of MNCs by rational ligand engineering, which will facilitate future interest and new strategies to precisely engineer solution-based nanocluster materials.

Abstract:Photoluminescent (PL) gold nanoclusters (AuNCs) show many advantages over conventional semiconductor quantum dots, however, their application potential is limited by their relatively low absorption cross-section and quantum yield. Plasmonic enhancement is a common strategy for improving the performance of weak fluorophores, yet in the case of AuNCs this method is still poorly explored. Here a robust synthetic approach to a compact plasmonic nanostructure enhancing significantly the PL of AuNCs is presented. Two gold nanostructures, AuNCs and plasmonic gold nanorods (AuNRs), are assembled in a compact core-shell nanostructure with tunable geometry and optical properties. The unprecedented degree of control over the structural parameters of the nanostructure allows to study the effects of several parameters, such as excitation wavelength, AuNC-AuNR distance, and relative loading of AuNCs per single AuNR. Consequently, a more general method to measure and evaluate enhancement independently of the absolute particle concentrations is introduced. The highest PL intensity enhancement is obtained when the excitation wavelength matches the strong longitudinal plasmonic band of the AuNRs and when the separation distance between AuNCs and AuNRs decreases to 5 nm. The results presented are relevant for the application of AuNCs in optoelectronic devices and bioimaging.

Abstract:With the rapid development of materials technology, fluorescent gold nanoclusters (AuNCs) are emerging as novel functional materials for diagnostic applications including the detection of biomarkers and bioimaging due to the advantages of their ultra-small size, tunable emissions, size-dependent fluorescence and excellent biocompatibility. In this review, we introduced the synthetic methods, and physical and chemical properties of AuNCs. Subsequently, we described the AuNCs-based design strategies for the detection of biomarkers including small molecules, DNA and proteins. The applications of AuNCs for tumor imaging in vitro and in vivo were also presented. Finally, we discussed the challenges and potential solutions of AuNCs-based nanosensors.

Abstract:Research on noble metal nanoclusters (MNCs) (elements with filled electron d-band) is boosting ahead because of the extensive and extraordinary chemical, optical, and physical properties of these materials. Because of the ultrasmall size of the MNCs (typically within 1-3 nm), they can be applied in areas of nearly all possible scientific domains. The greatest advantage of MNCs is the tunability that can be imposed, not only on their structures, but also on their chemical, physical, and biological properties. Nowadays, the MNCs are very effectively used as energy donors and acceptors under suitable conditions and hence cater as energy harvesters in solar cells, semiconductors, and biomarkers. In addition, ultrafast photoinduced electron transfer (PET) can be practised using MNCs under various circumstances. Herein, we have focused on the energy harvesting phenomenon of the Au, Ag, and Cu-based MNCs and elaborated on the different ways to apply them.

Abstract:The Förster Resonance Energy Transfer (FRET) is a widely used mechanism harnessed for sensing applications. For temporal stability of the energy transfer between donor and acceptor molecules their photostability and maintenance of the coupling geometry are important. A donor‐acceptor system was created from silicon nanoclusters (Si NCs) and silver nanoclusters (dihydrolopic acid (DHLA)‐protected Ag29 NCs). Optical spectroscopy and lifetime measurements helped to understand the energy transfer process between the Si NCs and Ag29 NCs. Compared with donor‐acceptor pairs based on gold nanocluster and organic dyes, the FRET pair formed between Si NCs and Ag29 (DHLA)12 NCs reaches a coequal efficiency of 60%.
