Synthesis, characterization, DFT studies, and in silico biological evaluation of novel dipeptide-sulphonamide derivatives against Helicobacter pylori receptors

Abstract:In the recent era, scientific interest has been piqued by Helicobacter pylori , a bacterium that resides in the human stomach and has been linked to a range of gastrointestinal disorders including gastritis, peptic ulcers, and an increased risk of gastric cancer. In this study, we conducted synthesis, characterization, DFT-based investigations, and in silico biological assessments of three novel dipeptide-sulphonamide derivatives to evaluate their potential as drug candidates targeting H. pylori receptors. The results of the vibrational energy distribution study were compared to the experimental vibrational frequencies, and they were found to be in good agreement. Investigation of intramolecular charge transfer interactions and stabilization energies was conducted through natural bond orbital (NBO) analysis. The analysis revealed a range of stabilization energies, with compound C exhibiting the highest energy of 657.15 kcal/mol, followed by compound A at 448.97 kcal/mol, and compound B at 418.97 kcal/mol. This order of stability correlates with their reactivity, as evidenced by energy values of 7.6693 eV for compound C, 8.4154 eV for compound A, and 8.9295 eV for compound B. These findings suggest that compound C is the most reactive among the studied species. The molecular docking results revealed that the ligands; N -(2-(Isobutylamino)-2-oxoethyl)-3-methyl-2-(4-methylphenylsulphonamido) pentanamide (A ), N -(2-(Isobutylamino)-2-oxoethyl)-3-methyl-2-(phenylsulphonamido) pentanamide (B), and N- (2(Isobutylamino)2-oxoethyl)3-methyl-2(4-nitrophenylsulphonamido) pentanamide (C) exhibited excellent selectivity and reactivity with the protein 2QV3 compared to the standard lansoprazole. The findings clearly show that these novel dipeptide-sulphonamides could represent a new class of potential anti-ulcer agents and, with further research, could be used as new anti-ulcer medications.
