Abstract:Due to the unique effect of fluorine atoms, the efficient construction of high-value alkyl fluorides has attracted significant interest in modern drug development. However, enantioselective catalytic strategies for the efficient assembly of highly functionalized chiral C(sp3)-F scaffolds from simple starting materials have been underutilized. Herein, we demonstrate a nickel-catalyzed radical transfer strategy for the efficient, modular, asymmetric hydrogenation and hydroalkylation of alkenyl fluorides with primary, secondary, and tertiary alkyl halides under mild conditions. The transformation provides facile access to various structurally complex secondary and tertiary α-fluoro amide products from readily available starting materials with excellent substrate compatibility and distinct selectivity. Furthermore, the utility of this method is demonstrated by late-stage modifications and product derivatizations. Detailed mechanistic studies and DFT calculations have been conducted, showing that the rate-determining step for asymmetric hydrogenation reaction is NiH-HAT toward alkenyl fluorides and the stereo-determining step is alcohol coordination to Ni-enolates followed by a barrierless protonation. The mechanism for the asymmetric hydroalkylation reaction is also delivered in this investigation.

Abstract:This study presents a single-atom Pt catalyst that achieves efficient C-F bond activation, which is a challenging reaction in both chemical synthesis and environmental remediation of recalcitrant fluorinated hydrocarbons. Up to 1.6 wt % Pt was loaded as a single-atom on SiC substrate (Pt-1/SiC) using a facile, scalable wet-chemical method developed based on anchor-site and photoreduction techniques. The high catalytic activity of Pt-1/SiC for hydrodefluorination of perfluorooctanoic acid, which is a select perfluorinated pollutant of significant environmental concern, is attributed to the effective hydrogen spillover from isolated Pt onto the SiC surface where the resulting Si-H bond further redistributes with the C-F bond to accomplish hydrodefluorination.

Abstract:A new fluoroamide-driven intermolecular hydrogen atom transfer (HAT) strategy for copper-catalyzed intermolecular 1,2-alkylarylation of alkenes with carbazates and various arene nucleophiles, such as indoles, pyrroles and 1,3,5-trimethoxybenzene, is described. By manipulating N-(tert-butyl)-N-fluoro-3,5-bis(trifluoromethyl)ben-zenesulfon-amide (NFBS) as the intermolecular HAT reagent, this reaction enables the generation of the carbazate nitrogen-centered radicals through intermolecular HAT followed by decomposition to form the alkyl radicals, radical addition across the alkene, single electron oxidation and nucleophilic coupling cascades to achieve the alkene 1,2-alkylarylation.

Abstract:In the modern era of organic synthesis, mechanisms centering on radical intermediates have become increasingly impact-ful in unlocking novel reactivity. Among all, hydrogen atom transfer (HAT) represents one of the most fundamental chemical reaction steps and has found applications in designing practical transformations. Herein, we present a detailed case study on selective hydrodefluorination of trifluoromethylarenes utilizing N-heterocyclic carbene boranes (NHC-boranes) as the HAT donor. Under the optimal conditions featuring an acridine-based photocatalyst, complete selectivity for mono-hydrodefluorination was achieved across a wide array of substrates. Comprehensive mechanistic studies com-bining experimental and computational approaches disproved a chain process involving a fluorine atom transfer but ra-ther pointed to a HAT non-chain mechanism, where the key step involves the difluorobenzylic radical abstracting a hy-drogen atom from the NHC-borane to generate a boryl radical in a polarity-matched fashion. Evaluation of a selection of Lewis base-ligated boranes revealed molecular descriptors critical to the outcomes of this reaction, and a classification model is built to explain the structure-reactivity relationship and how various elementary steps can be influenced. These results collectively provide valuable information for future reaction design to elevate the utility of boranes in organic radical chemistry.

Abstract:The renaissance of catalytic hydrogen atom transfer from a metal hydride (MHAT) offers advanced tools for practicing sophisticated radical chemistry on simple olefins. While 3d transition metals like cobalt, iron, and manganese have been extensively studied in catalytic MHAT, the potential of copper remains unexplored. This is due to the polar reactivity exhibited by classical nucleophilic Cu(I) hydrides. Here we report the first examples of catalytic hydrofunctionalization reactions enabled by copper-mediated MHAT. The Cu-MHAT process produces alkyl radicals in high chemo- and regioselectivity, which are subsequently captured by Cu(II) species to undergo coupling reactions with an exceptionally broad scope of oxygen-, nitrogen-, halogen-, and carbon-based nucleophiles. The distinct reactivity and selectivity observed in Cu-MHAT system is attributed to the involvement of Cu(II)–H species, a hydrogen atom donor that possesses substantially altered charge distribution and weakened bonding in contrast to conventional Cu(I) hydrides. Preliminary results suggest potential extension to asymmetric catalysis and radical polymerization are viable. This work opens up new opportunities for MHAT chemistry by going beyond the common oxidation states.

Abstract:We report the hydrofluorination of unactivated alkenes using N-fluorobenzenesulfonimide as a fluorination reagent. The reaction produces exclusively Markovnikov hydrofluorination products under mild conditions. It is not affected by air or moisture and uses an inexpensive and readily available nickel compound as a catalyst. Distinct from the previously reported ionic pathway of the NiH-catalyzed hydrofunctionalization of olefins, this reaction was shown to proceed through a hydrogen atom transfer pathway.

Abstract:Here, we reported for the first time a mechanistically distinctive cobalt-catalyzed Markovnikov-type sequential semihydrogenation/hydrohydrazidation of aliphatic terminal alkynes in one pot. A cobalt hydride species was employed as two roles for both a unique metal-catalyzed Markovnikov-type insertion of the aliphatic terminal alkynes and then metal-catalyzed hydrogen atom transfer of alkenes. This operationally simple protocol exhibits excellent functional group tolerance and step economy. The hydrazone products could be easily transferred to various valuable amine derivatives.

Abstract:With halogen-atom transfer as an effective tool, a novel catalytic enantioselective protocol to generate chiral trifluoromethylated alkynes has been established by a cooperative photoredox and nickel catalysis system, providing a straightforward and modular route to access this type of product in good yields and enantioselectivities. The halogen-atom transfer process is essential for the reaction and this novel strategy offers another promising way to utilize alkyl halides with highly negative reduction potentials. It firstly expands nickel-catalyzed asymmetric reductive cross-couplings of organohalides from the traditional single-electron transfer to halogen-atom transfer.

Abstract:There is ongoing interest in catalysis with the earth-abundant metal copper (Cu). This density functional theory (DFT) study explores the detailed workings of the first CuH-catalyzed hydroarylation of alkenes with polyfluoroarenes involving C–F bond functionalization. The CuH complex undergoes Markovnikov hydrocupration with the aryl alkene substrate to give a Cu(I) benzyl intermediate, and the regioselectivity is due to a combination of orbital and dispersion interactions induced by the aryl alkene substrate. The benzyl ligand allows a 1,3-cupratropic shift for the Cu(I) complex to have an exposed carbonanionic atom facilitating nucleophilic aromatic substitution to the polyfluoroarene substrate. The distinct regioselectivity of para-C–F substitution is controlled by the noncovalent C(sp3)–H/π and C(sp2)–H●●●O interactions. These mechanistic insights can have implications for the development of new transitional metal-catalyzed hydroarylation and C–F functionalization reactions.

Abstract:N-centered radicals mediated remote C(sp3)–H functionalization via HAT processes have been successfully applied in the difunctionalization of alkenes, serving as an elegant and robust method to convert readily available alkenes into various functionalized molecules. However, HAT strategy-enabled difunctionalization of alkenes using electrophiles as functionalizing reagents remains underexplored. In this study, we report a nickel-catalyzed regioselective reductive three-component 1,2-alkylarylation of alkenes with O-oxalate hydroxamic acid esters and aryl iodides. This radical addition/cross-coupling cascade reaction involves amidyl radical-triggered intramolecular 1,5-HAT and nickel-catalyzed reductive coupling processes under mild reaction conditions with good coupling efficiency. Additionally, this approach can be extended to the reductive 1,2-alkylarylation of alkynes, providing an efficient method for the synthesis of multi-substituted alkenes from easily accessible starting materials.

Abstract:The synthesis of deuterated gem-difluoroalkenes by selective deuterodefluorination of β-CF3-cinnamates with nickel catalyst has been reported for the first time. Commercial available deuterated formic acid is a cheap and convenient deuterium source. The nickel-catalyst showed high selectivity on monodefluorination and avoided competitive reactions such as multiple defluorination or hydrogenation.

Abstract:Transition-metal-catalyzed difunctionalization of olefins constitutes a fertile synthetic platform for rapid access to complex molecules from bulk chemicals. However, substrates featuring fluoroalkyl substituents are rarely employed because of facile β-fluoride elimination pathways. Herein, we report a hydroalkylation of trifluoromethylalkenes with alkyl halides under nickel catalysis that enables the rapid construction of 1,1,1-trifluoropropane derivatives. The common β-fluoride elimination pathway is suppressed by identifying a competent proton donor that favors a protonolysis process. Also, unactivated alkenes could be readily employed as the alkyl donor when using a Ni/hydrosilane catalytic system.

Abstract:Within the context of Ni photoredox catalysis, halogen atom photoelimination from Ni has emerged as a fruitful strategy for enabling hydrogen atom transfer (HAT)-mediated C(sp3)-H functionalization within the context of Ni photoredox catalysis. Despite the numerous synthetic transformations invoking this paradigm, a unified mechanistic hypothesis that is consistent with experimental findings on the catalytic systems and accounts for halogen radical formation and facile C(sp2)–C(sp3) bond formation remains elusive. We employ kinetic analysis, organometallic synthesis, and computational investigations to deci-pher the mechanism of a prototypical Ni-catalyzed photochemical C(sp3)–H arylation reaction. Our findings revise the previ-ous mechanistic proposals, first by examining the relevance of SET and EnT processes from Ni intermediates relevant to the HAT-based arylation reaction. Our investigation highlights the ability for blue light to promote efficient Ni–C(sp2) bond homolysis from cationic NiIII and C(sp2)–C(sp3) reductive elimination from bipyridyl NiII complexes. However interesting, the rates and selectivities of these processes do not account for the productive catalytic pathway. Instead, our studies support a mechanism that involves halogen atom evolution from in situ generated NiII dihalide intermediates, radical capture by a NiII(aryl)(halide) resting state, and a key C–C bond formation from NiIII. Oxidative addition to NiI, as opposed to Ni0, and rapid NiIII/NiI comproportionation play key roles in this process. The findings presented herein offer fundamental insight into the reactivity of Ni in the broader context of catalysis.

Abstract:An iron/chromium system (Fe(OAc)2, CpCr(CO)3H) catalyzes the preparation of β,γ- or γ,δ-unsaturated amides from 1,4,2-dioxazol-5-ones. An acyl nitrenoid iron complex seems likely to be responsible for C-H activation. A cascade of three H• transfer steps appears to be involved: (i) the abstraction of H• from a remote C-H bond by the nitrenoid N, (ii) the transfer of H• from Cr to N, and (iii) the abstraction of H• from a radical substituent by the Cr•. The observed kinetic isotope effects are consistent with the proposed mechanism if nitrenoid formation is the rate-determining step. The Fe/Cr catalysts can also desaturate substituted 1,4,2-dioxazol-5-ones to 3,5-dienamides.

Abstract:This density functional theory (DFT) study explores the reaction mechanism of nickel-catalyzed hydroarylation of unactivated alkenes that selectively generates branched (Markovnikov) products. Extensive DFT calculations have revealed a novel and plausible mechanism via hydronickelation, which differs from the originally proposed pathway through carbonickelation. The N-heterocyclic carbene-anchored catalyst IPrNiOi-Pr favors β-hydride elimination from the alkoxide ligand over oxidative addition with carbon electrophiles. This chemoselectivity results in the formation of the Ni( I ) hydride complex IPrNiH, initiating a hydronickelation pathway involving alkene into Ni–H insertion, PhOTf oxidative addition, and alkyl–phenyl reductive elimination. The β-hydride elimination represents the rate-limiting step, while the oxidative addition of PhOTf determines the regioselectivity, which is sterically controlled. This DFT study demonstrates a robust theoretical–experimental synergy and provides new mechanistic insights valuable for reaction development.

Abstract:Herein, we described a nickel-catalyzed monofluoroalkenylation through defluorinative reductive cross-coupling of gem-difluoroalkenes with alkyl halides. Key to the success of this strategy is the combination of C- F cleavage with alkyl halides activation. This reaction enables the convenient synthesis of a large variety of functionalized monofluoroalkenes under mild reaction conditions with broad functional group compatibility and excellent Z-selectivity. The combination of Ni catalysis with (Bpin)(2)/K3PO4 as terminal reductant promoted the efficient C(sp(2))-C(sp(3)) formation especially the generation of all-carbon quatemary centers with high chemoselectivity.

Abstract:DFT calculations have been conducted to elucidate the mechanistic details of a novel Ni-catalyzed hydrocarboxylation reaction of alkynes, in which formic acid is atom-economically used through a catalytic CO recycling manner. On the basis of our theoretical investigations, the bisphosphine (dppbz, 1,2-bis(diphenylphosphino)benzene) ligated nickel monocarbonyl complex (dppbz)NiCO was located as the active catalytic species for this process, and such a carbonyl ligand is found to be critical for the final reductive elimination. Our studies also revealed the addition of H to alkynes proceeds via a proton transfer process directly from formic acid (i.e., outer-sphere pathway) rather than through a proposed hydrometalation process (i.e., direct hydride shift from Ni-H). The thermal decomposition of formic anhydrides was found to be vital to a successful reaction, and its barrier must be slightly higher than the energetic span of the Ni catalytic cycle. Fast release of CO can poison the Ni catalyst, so that the reaction would be shut down. Other intriguing experimental observations, such as ligand effect, regioselectivity, and extraordinary compatibility of C-X (X = halogen) bonds, are also discussed in this article.
