Abstract:Rh(III)-catalyzed C-H functionalization of 7-arylpyrazolo[1,5-a]pyrimidines was developed wherein the pyrazolo[1,5-a]pyrimidine moiety is reported for the first time to direct the C-H bond activation. Various 7-arylpyrazolo[1,5-a]pyrimidines underwent smooth C-H amidation with alkyl-, aryl-, and heteroaryl-substituted dioxazolones to afford the products in moderate to good yields. Mechanistic studies suggest that a six-membered rhodacycle intermediate involving N1 might play a key role in the regioselective catalytic cycle.

Abstract:Aromatization-driven deconstruction and functionalization of spiro dihydroquinazolinones via dual photoredox/nickel catalysis is developed. The aromatization effect was introduced to synergistically drive unstrained cyclic C-C bond cleavage, with the aim of overcoming the ring-size limitation of nitrogen-centered radical induced deconstruction of carbocycles. Herein, we demonstrate the synergistic photoredox/nickel catalyzed deconstructive cross-coupling of spiro dihydroquinazolinones with organic halides. Remarkably, structurally diverse organic halides including aryl, alkenyl, alkynyl, and alkyl bromides were compatible for the coupling. In addition, this protocol is also characterized by its mild and redox-neutral conditions, excellent functional group compatibility, high atom economy, and easy scalability. A telescoped procedure involving condensation and ring-opening/coupling was found to be accessible. This work provides a complementary strategy to the existing radical-mediated C-C bond cleavage of unstrained carbocycles.

Abstract:The pyridine-boryl radical chemistry has witnessed significant advancements in the last decade, enabling an array of novel synthetic applications. In this context, density functional theory calculations provide a computational mechanistic analysis of the pyridine/B2pin2-mediated cascade synthesis of indolizines. The radical-radical cross-coupling pathway is dissected to be energetically more feasible than the conventionally assumed Minisci-type radical addition route. Furthermore, the computations reveal that a 1,4-O[B] migration step leads to the formation of zwitterionic intermediates, whose cyclization then generates the indolizine scaffolds. H-shuttles, generated from water, alcohol, or amine molecules, are found to play pivotal roles in facilitating the elimination or aromatization processes.

Abstract:Understanding the reaction mechanism and regioselectivity control is critical for the rational synthesis of functionalized boron clusters. Herein we report a computational mechanistic study on the phenyliodo diacetate-mediated B-H functionalization of the amide-substituted closo-dodecaborate cluster. A concerted iodination-deprotonation process was identified for the key B-H bond iodination, which is the mechanistic basis for the exceptional regioselectivity of ortho-B-H functionalization. This mechanistic model highlights a distinctive reaction channel for iodine(iii)-promoted transformations, which paves the way for future design of directing groups in closo-dodecaborate derivatization using this strategy.

Abstract:Dearomatic reactions are very important to build alicyclic moieties and expand the structural diversity. Muto and Yamaguchi recently investigated the Pd-catalyzed dearomative allylic reactions of allyl borates with benzyl phosphates to give final compounds. Herein, the calculations were conducted to explore the reaction mechanism and selectivity, and the results clarified that the dearomatic product through carbon-carbon bond formation between eta(3)-exo-(naphthyl)methyl para-carbon atom and allyl terminal carbon atom (TS3 = 7.3 kcal/mol) is energetically more favorable than the other carbon-carbon bond formation paths to afford aromatic and dearomatic compounds.

Abstract:Copper-catalyzed radical transformations establish a powerful toolkit to construct versatile complex organic compounds. The copper-mediated bond formation step of radicals plays a critical role in controlling chemo- and stereoselectivity of copper-catalyzed radical transformation reactions. This bond formation step involves three possible pathways: ion-type bond formation, radical substitution, and reductive elimination. This review highlights the recent advances in theoretical studies on mechanisms and controlling models on the selectivity of Cu-mediated radical-involved bond formation, providing a general mechanistic comprehension of this key elementary step in copper catalysis.

Abstract:Dearomatization reactions provide a rapid approach to construct complicated molecules that are difficult to synthesize by traditional methods from simple aromatic compounds. Herein, we report an efficient dearomative [3+2] cycloaddition reaction of 2-alkynyl pyridines with diarylcyclopropenones, leading to the synthesis of densely functionalized indolizinones in moderate to good yields under metal-free conditions. In addition, this strategy can also be employed in dearomative cyclization of isoquinolines to access a variety of benzo-fused indolizinones. Abstract:Ruthenium-catalyzed arylation of ortho C-H bonds directed by a bidentate 8-aminoquinoline moiety not only is important to construct new biaryl derivates but also merges important research areas. In this study, the density functional theory (DFT) method M11-L was employed to predict the mechanism of this C-H bond arylation reaction. The computational results indicate that the initial step for this reaction is catalyst loading by electrophilic deprotonation to generate a substrate-coordinated Ru(II) intermediate, which is the key compound in the complete catalytic cycle. The catalytic cycle includes electrophilic deprotonation by carbonate, oxidative addition of bromobenzene, reductive elimination to form a new aryl aryl bond, proton transfer to release the product, and ligand exchange to regenerate the initial Ru(II) intermediate. Theoretical calculations suggest that the oxidative addition of bromobenzene is the rate determining step of the whole catalytic cycle, and the apparent activation free energy is 32.7 kcal/mol. The ligand effect was considered in DFT calculations, and the calculated results agree well with experimental observations.

Abstract:An efficient palladium-catalyzed post-Ugi dearomative cyclization has been achieved, enabling the rapid assembly of diverse benzoazepinespiroindolenine derivatives with excellent chemoselectivity and good diastereoselectivity. This methodology can controllably introduce functionality and complexity into the dearomatized alicyclic products in a cost-effective and step-economical manner with a wide range of substrate scope. The highly precise dearomatization of complex Ugi adducts opens the door to address formidable chemoselectivity issues among nucleophilic sites, and complements the functional group compatibility of the classical dearomatization protocols. Density functional theory (DFT) calculations shed light on the reaction mechanism as well as the origin of chemoselectivity.

Abstract:Radical chain reactions leading to C-C bond formation are widely used in organic synthesis, and initiation of the radical chain process usually requires thermolabile radical initiators. Recent studies on transition-metal-free cross-coupling reactions between aryl halides and arenes have demonstrated an unprecedented initiation system for radical chain reactions, where the combination of simple organic additives and a base was used in place of conventional radical initiators. Among them, the combination of N,N'-dimethylethylenediamine (DMEDA) and t-BuOK is one of the most efficient and representative reaction systems, and the radical initiation mechanism of this system has attracted considerable research interest. In this study, through the combination of kinetic studies, deuterium labeling experiments, and DFT calculations, the radical initiation mechanism of the diamine-promoted cross-coupling reaction was carefully reinvestigated. In light of the present study, a mechanistic network of radical initiation in the DMEDA/t-BuOK system was revealed, which differs dramatically from the previously realized single radical initiation pathway. In this mechanism, the diamine acts as a hydrogen atom donor and plays a dual role as both "radical amplifier" and "radical regulator" to initiate the radical chain process as well as to control the concentration of reactive radical species. This represents a rare example of a structurally simple molecule playing such a subtle role in the radical chain reaction system. The present study sheds some light on the novel radical initiation mode hi transition metal-free cross-coupling reactions following a base-promoted homolytic aromatic substitution (BHAS) mechanism, and may also help to understand the mechanism of relevant reactions.

Abstract:Developing pragmatic strategies for accessing functional benzofuran‐2‐ones from 3‐([1,1'‐biphenyl]‐2‐yl)benzofuran remains an enduring challenge. Herein, we have achieved a highly discriminating electrochemical oxidative dearomative spiroannulation of 3‐([1,1'‐biphenyl]‐2‐yl)benzofuran, culminating in the synthesis of 2H‐spiro[benzofuran‐3,9'‐fluoren]‐2‐one derivatives. By harnessing the electrophilic intermediates of benzofuryl radical cations supported by DFT calculations, we attain exceptional regioselectivity while eliminating the need for stoichiometric oxidants. Mechanistic investigations reveal a sequence of events involving the benzofuran radical cation, encompassing the capture of H2O, nucleophilic arene attack, and subsequent deprotonation, ultimately yielding the final benzofuran‐2‐ones.

Abstract:Dearomative functionalization of heteroarenes offers an attractive and sustainable approach for the rapid construction of complex 3D heterocyclic scaffolds from planar structures. Despite progress in this field, dearomative amination of heteroarenes via a radical anion intermediate remains a challenge. Here, we report a photoredox-catalyzed dearomative hydroamination of heteroarenes with hydrazodiformates under mild and transition-metal-free reaction conditions. Various benzofurans and benzothiophenes can efficiently participate in this transformation. A series of mechanistic experiments revealed that heteroaryl radical anions are the crucial intermediates, generated through photo-induced electron transfer between the excited phenolate photocatalyst and heteroarenes.

Abstract:Herein, we report the Pd(0)-catalyzed intermolecular asymmetric dearomative [3 + 2] annulation of phenols with vinyl cyclopropanes via in situ generated ortho-quinone methide intermediates. A series of highly functionalized spiro-[5,6] bicycles which bear three contiguous stereogenic centers including one all-carbon quaternary were obtained with excellent stereoselectivities. Density functional theory (DFT) calculations indicate that the reactions were controlled by thermodynamics.

Abstract:Dearomative reactions allow for concise assembly of structurally complex, highly functionalized three‐dimensional molecules by breaking the plane of two‐dimensional aromatic starting materials. With the growing interest in N‐heterocyclic carbene (NHC) organocatalysis as a powerful strategy for synthesizing diversiform molecules, mainly by polarity‐reversal (umpolung) approaches, NHC‐catalyzed dearomatization reactions have recently been successfully used for the construction of the skeletons of natural products and drugs. By generating various key intermediates (i.e., Breslow intermediates, acyl azolium intermediates, homoenolate intermediates, enolate intermediates, and azolium dienolate intermediates), NHCs can readily destroy aromaticity by electrophilic or nucleophilic interactions with aromatic compounds. In this overview, progress in NHC‐catalyzed dearomative reactions is summarized; the reactions are grouped according to reaction modes and categories of intermediates, and representative mechanisms are provided. We expect this review to serve as a comprehensive reference for work in the emerging field of organocatalytic dearomatization.

Abstract:Abstract Aromatic [5,5]-rearrangement can in principle be an ideal protocol to access dearomative compounds. However, the lack of competent [5,5]-rearrangement impedes the advance of the protocol. In this Article, we showcase the power of [5,5]-rearrangement recently developed in our laboratory for constructing an intriguing dearomative sulfonium specie which features versatile and unique reactivities to perform nucleophilic 1,2- and 1,4-addition and cyclization, thus achieving dearomative di- and trifunctionalization of easily accessible aryl sulfoxides. Impressively, the dearomatization products can be readily converted to sulfur-removed cyclohexenones, naphthalenones, bicyclic cyclohexadienones, and multi-substituted benzenes. Mechanistic studies shed light on the key intermediates and the remarkable chemo-, regio- and stereoselectivities of the reactions.

A Theoretical Study on Reductive Debromination of Polybrominated Diphenyl Ethers

Abstract:Recent progress has been made in the reductive debromination of polybrominated diphenyl ethers (PBDEs) by nanoscale zero-valent iron (nZVI). To better understand the mechanism of this reaction, seven selected BDE congeners and their anions were investigated at the density functional theory (DFT) level using four different methods, including B3LYP/6-31G(d), B3LYP/6-31+G(d), B3LYP/6-31G(d,p) and B3LYP/6-311G(d,p). The cleaved C–Br bonds observed in the equilibrium structures of anionic PBDEs were adopted as the probe of the susceptible debromination position of PBDEs in the presence of nZVI, and the proposed major reaction pathways based on our calculations can satisfactorily conform to the reported experimental results. The debromination preference is theoretically evaluated as meta-Br > ortho-Br > para-Br. In addition, both the calculated frontier orbital energies and adiabatic electronic affinities were found to be highly related to their experimental reductive debromination rate constants. The highest linear regression coefficient was observed in the case using the energy of lowest unoccupied molecular orbital as the molecular descriptor obtained from B3LYP/6-31G(d) (R2 = 0.961, n = 7) or B3LYP/6-31G(d,p) (R2 = 0.961, n = 7). The results clearly showed the evidence of an electron transfer mechanism associated with this reductive debromination reaction.

Abstract:Density functional theory investigations revealed that the pyridine-boryl radical generated in situ using 4-cyanopyridine and bis(pinacolato)diboron could be used as a bifunctional "reagent", which serves as not only a pyridine precursor but also a boryl radical. With the unique reactivity of such radicals, 4-substituted pyridine derivatives could be synthesized using α,β-unsaturated ketones and 4-cyanopyridine via a novel radical addition/C-C coupling mechanism. Several controlled experiments were conducted to provide supportive evidence for the proposed mechanism. In addition to enones, the scope could be extended to a wide range of boryl radical acceptors, including various aldehydes and ketones, aryl imines and alkynones. Lastly, this transformation was applied in the late-stage modification of a complicated pharmaceutical molecule.

Abstract:Diradical chemistry, typified by the cycloaromatization of enediynes or enyne‐allenes, have been extensively explored due to the involved intriguing biological activity and unique mechanistic actions. Because of the essential Myers‐Saito cycloaromatization involved in the maleimide‐assisted rearrangement and cycloaromatization (MARACA) mechanism disclosed in our recent work, the important object of diradical/zwitterion dichotomy and the reactivity of thermally induced σ,π‐diradicals were investigated in this work through the combination of experimental and computational studies. Deuterium incorporation experiments demonstrated that the polar product was afforded from the MARACA strategy, in which the diradical and zwitterion reactivities from the cycloaromatization step could both lead to the closed‐shell product via the subsequent nucleophilic addition reaction and protonation. Using the density functional theory calculations, the unusual reactivity of the heterosymmetric diradicals to closed‐shell zwitterions was examined, and the addition of carbonyl moiety to α,3‐dehydrotoluene was allowed to occur via a nonplanar cyclic allene transition structure with a small barrier of 9.1 kcal/mol. The observed nonplanar geometry is essential for the necessarily symmetry‐breaking action, resulted in the continuous transformation from open‐shell diradical to closed‐shell zwitterion species during the addition process.

Abstract:Abstract Vinyldiazo compounds are one of the most important synthons in the construction of a cyclic ring. Most photochemical transformations of vinyldiazo compounds are mainly focusing on utilization of their C═C bond site, while reactions taking place at terminal nitrogen atom are largely unexplored. Herein, a photocatalytic cascade radical cyclization of LBRs with vinyldiazo reagents through sequential B─N/C─N bond formation is described. The reaction starts with the addition of LBRs (Lewis base–boryl radicals) at diazo site, followed by intramolecular radical cyclization to access a wide range of important boron‐handled pyrazoles in good to excellent yields. Control experiments, together with detailed mechanism studies well explain the observed reactivity. Further studies demonstrate the utility of this approach for applications in pharmaceutical and agrochemical research.
