Abstract:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.

Abstract:Multi-walled carbon nanotubes (MWCNTs) were homogeneously dispersed in silk fibroin (SF) solutions at different compositions, and a simple solvent-casting method was used to fabricate SF/MWCNT films. Structure, viscosity, and mechanical properties of the SF/MWCNT nanocomposites were characterized by FTIR (Fourier Transform Infrared Spectroscopy), viscometry, and tensile testing. Fibroblast cells were used to examine cell viability and attachment to nanocomposite films. Compared to a pure SF film, adding just 0.5 % (w/w) of MWCNT to the SF matrix could enhance the Young’s modulus and ultimate tensile strength by approximately 24 % and 39 %, respectively. In addition, with increasing MWCNTs content, the percentage of β -sheet structure increased significantly. When the content of MWCNTs increased from 0 to 1 % (w/w), the β -sheet content improved from 17.8 % to 53.6 %. Furthermore, adding up to 1 % (w/w) MWCNT increased the viscosity of the SF solution ten fold. From a biological point of view, no toxic effect was observed on fibroblast cells in the presence of MWCNTs after 3 and 7 days. This investigation suggests that SF/MWCNT nanocomposite films with improved mechanical properties and biocompatibility are well-suited as novel biomaterials.

Abstract:Using genetic engineering methods, we attempted to produce novel silk-like proteins with new function by combining several functional sequences selected from fibroin of Bombyx mori (B.mori), Samia Cynthia ricini (S.c.ricini) and spider silks or by inducing cell adhesive sequence or calcium binding sequence into silk proteins. The secondary structure of these silk-like proteins was characterized with solid state NMR. Cell adhesion assay indicated that silk-like proteins have higher cell activity. Mineralization of fibroin protein was improved with induction of calcium binding sequence. Nanofiber formation of silk-like proteins was achieved using electrospinning. Fiber was formed from silk-like proteins. These silk-like proteins might be candidates to meet requirement in the field of biomaterials.

Abstract:Electrospinning is a very effective method to fabricate nanofibers. This technique has been achieved more attention due to its cheap cost, simple device and obtaining nanofibers similar to extracellular matrix.Silk fibroin(SF) is a natural protein with good biodegradability and low immunogenicity, Therefore, it can be widely applied in tissue engineering. In this paper, we summarize the research of electrospun silk fibrion nanofibers in regenerative medicine. Furthermore, the future research work of SF nanofibers was suggested.

The introduction of nanoparticles brings many optimized and new functionalities to silk fibroin, such as mechanical, biological, thermal, electrical, florescent and magnetical properties. Abstract:Silk has a long history of use in medicine as sutures. To address the requirements of a mechanically robust and biocompatible material, basic research to clarify the role of repeated sequences in silk fibroin in its structures and properties seems important as well as the development of a processing technique suitable for the preparation of fibers with excellent mechanical properties. In this study, three silk-like protein analogs were constructed from two regions selected from among the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(2), the crystalline region of Samia cynthia ricini silk fibroin, (Ala)(12,) the crystalline region of spider dragline silk fibroin, (Ala)(6), and the Gly-rich region of spider silk fibroin, (GGA)(4). The silk-like protein analog constructed from the crystalline regions of the spider dragline silk and B. mori silk fibroins, (A(6)SCS)(8), that constructed from the crystalline regions of the S. c.ricini and B. mori silk fibroins, (A(12)SGS)(4), that constructed from and the crystalline region of S. c.ricini silk fibroin and the glycine-rich region of spider dragline silk fibroin, (A(12)SGS)(4),were expressed their molecular weights being about 36.0 kDa, 17.0 kDa and 17.5 kDa, respectively in E. coli by means of genetic engineering technologies. (A(12)-SCS)(4) and (A(12)SGS)(4) undergo a structural transition from alpha-helix to beta-sheet on a change in the solvent treatment from trifluoroacetic acid (TFA) to formic acid (FA). However, (A(6)SCS)(4) takes on the P-sheet structure predominantly on TFA treatment and FA treatment. Structural analysis was performed on model peptides selected from spider dragline and S. c.ricini silks by means of C-13 CP/MAS NAIR.

Abstract:The unique amino acid composition and spatial structure of silk fibroin make it an excellent biomaterial with perfect biocompatibility and biodegradability.Silk fibroin nanofibers and composite fibroin nanofibers prepared via high voltage electrostatic spinning technology are conferred further superior property for application as biomedical materials.This review systematically introduces advances in application and modification of the electrospun silk fibroin nanofibers and composite fibroin nanofibers in bone tissue engineering,in repair of cartilage,wound,nerve and blood vessel,and in controlled release of drugs in recent years,based on which key innovations for future development of electrospun silk fibroin nanofiber materials are briefly discussed.

Abstract:A variety of artificial spinning methods have been applied to produce regenerated silk fibers; however, how to spin regenerated silk fibers that retain the advantages of natural silks in terms of structural hierarchy and mechanical properties remains challenging. Here, we show a bioinspired approach to spin regenerated silk fibers. First, we develop a nematic silk microfibril solution, highly viscous and stable, by partially dissolving silk fibers into microfibrils. This solution maintains the hierarchical structures in natural silks and serves as spinning dope. It is then spun into regenerated silk fibers by direct extrusion in the air, offering a useful route to generate polymorphic and hierarchical regenerated silk fibers with physical properties beyond natural fiber construction. The materials maintain the structural hierarchy and mechanical properties of natural silks, including a modulus of 11 ± 4 GPa, even higher than natural spider silk. It can further be functionalized with a conductive silk/carbon nanotube coating, responsive to changes in humidity and temperature.

Abstract:Silk fibroin (SF) is a structural protein derived from natural silkworm silks. Materials fabricated based on SF usually inherit extraordinary physical and biological properties, including high mechanical strength, toughness, optical transparency, tailorable biodegradability, and biocompatibility. Therefore, SF has attracted interest in the development of sustainable biodevices, especially for emergent bio-electronic technologies. To expand the function of current silk devices, the SF characteristic sequence has been used to synthesize recombinant silk proteins that benefit from SF and other functional peptides, such as stimuli-responsive elastin peptides. In addition to genetic engineering methods, innovated chemistry modification approaches and improved material processing techniques have also been developed for fabricating advanced silk materials with tailored chemical features and nanostructures. Herein, this review summarizes various methods to synthesize functional silk-based materials from different perspectives. This review also highlights the recent advances in the applications of natural and recombinant silks in tissue regeneration, soft robotics, and biosensors, using B. mori SF and silk-elastin-like proteins (SELPs) as examples.

Abstract:The high crystallinity and sophisticated hierarchical architecture of native animal silk endow it with comprehensive mechanical properties that are superior to those of most synthetic fibers. However, these features also make the direct exfoliation of silk nanofibrils (SNFs) highly challenging. On the other hand, silk-based materials prepared by conventional method (i.e., through silk fibroin aqueous solution) are usually weak, so the preparation methods based on SNFs have attracted much attention in recent years. Herein, a facile and environmentally friendly route is developed to directly exfoliate SNFs from natural Bombyx mori silkworm silk via ammonium persulfate oxidation followed by ultrasonication. The obtained SNFs have a high yield (nearly 40%) and are well dispersed in water in a wide pH range, so they can act as a good starting material to prepare subsequent silk-based materials. The main application of the SNFs demonstrated in this article is a curcumin (Cur)/SNFs hydrogel as wound dressing. In vivo experimental results show that the Cur/SNFs hydrogel significantly enhanced the healing rate of wounds on diabetic mice. Therefore, the preparation method developed in this study provides an efficient way to produce SNFs, which have great potential for a wide range of applications, including as wound dressings for diabetics. Abstract:Animal silks are built from pure protein components and their mechanical performance, such as strength and toughness, often exceed most engineered materials. The secret to this success is their unique nanoarchitectures that are formed through the hierarchical self-assembly of silk proteins. This natural material fabrication process in sharp contrast to the production of artificial silk materials, which usually are directly constructed as bulk structures from silk fibroin (SF) molecular. In recent years, with the aim of understanding and building better silk materials, a variety of fabrication strategies have been designed to control nanostructures of silks or to create functional materials from silk nanoscale building blocks. These emerging fabrication strategies offer an opportunity to tailor the structure of SF at the nanoscale and provide a promising route to produce structurally and functionally optimized silk nanomaterials. Here, we review the critical roles of silk nanoarchitectures on property and function of natural silk fibers, outline the strategies of utilization of these silk nanobuilding blocks, and we provide a critical summary of state of the art in the field to create silk nanoarchitectures and to generate silk-based nanocomponents. Further, such insights suggest templates to consider for other materials systems.

Abstract:The silk nanofibrils were isolated from wasted silk fibers to create biocompatible silk fibroin nanocomposites with high performance. Regenerated silk fibroin (SF) as a biomaterial has attracted increasing attention due to its outstanding biocompatibility and processibility. However, it faces great challenges in practical application because of the deterioration of the intrinsic mechanical properties of natural silk during the regeneration process. Natural silk nanofibrils (SNFs) has a typical hierarchical structure and maintains excellent mechanical properties of natural silk. Based on principle of homogeneous reinforced composites, herein, the SNFs were isolated as a reinforcing material and regenerated SF as matrix material to fabricate high‐performance SF nanocomposites. The SNFs with diameters ranging from 100 to 600 nm were prepared with yields of 87%. They not only maintained the crystallinity of natural silk, but also remained high stability in acid and alkaline solutions. Thanks to homogenous reinforcement of the SNFs, the tensile stress of the reinforced film was up to 2.2 MPa, the strain was 129%, and the water absorption was 50% in wet state. In vitro, the reinforced films supported adhesion and proliferation of human umbilical vein endothelial cells, which has potential in the application of biomedical dressings. Highlights Natural silk nanofibrils (SNFs) were exfoliated using a facile approach. The SNFs reinforced the silk nanocomposites significantly. The silk nanocomposites showed remarkable cytocompatibility. Homogeneous reinforcement is useful strategy to fabricate nanocomposites.

Abstract:Two silklike proteins, [TGRGDSPAGG(GAGA GS)(3)AS](5) (FS5) and [TGRGDSPA-(GVPGV)(2)GG(GAGA GS)(3)AS](8) (FES8) were designed to demonstrate the superior performance as biomaterials of silklike proteins. The former protein consists of the crystalline domain sequence, (GAGAGS)n from Bombyx mori silk fibroin and cell-adhesive sequence TGRGDSPA coming from fibronectin-containing RGD triplet. The additional sequence (GVPGV)(n) from elastin was included in the latter protein. The considerably higher cell-adhesion activities of these proteins for NHDF and VERO cells were observed by comparing with those of silklike materials without RGD sequences and also the crystalline fraction of B. mori silk fibroin. This tendency was independent of the treatments, 4.5M LiClO4 or formic acid (FA), on silklike proteins. Their activities are also higher than those of commercial Fibronectin F (TM) for NHDF cell. Their structural characterization was studied using C-13 solid-state NMR. Although the overlapped peaks in usual C-13 CP/MAS NMR spectra make the detailed structural analysis difficult, the methyl resonance regions observed using dipolar dephasing NMR were very useful for the analysis. The presence of both random coil and P-sheet structures was observed in these proteins clearly. The content of P-sheet structure in both proteins increases after FA treatment when compared with the lyophilized samples. The production of electrospun nanofibers from their hexafluoroacetone solution was also tried. The silklike protein FES8 could prepare nonwoven silk fibers although FS5 could not. Abstract:Silk fibroin (SF) is a natural protein largely used in the textile industry but also in biomedicine, catalysis, and other materials applications. SF is biocompatible, biodegradable, and possesses high tensile strength. Moreover, it is a versatile compound that can be formed into different materials at the macro, micro- and nano-scales, such as nanofibers, nanoparticles, hydrogels, microspheres, and other formats. Silk can be further integrated into emerging and promising additive manufacturing techniques like bioprinting, stereolithography or digital light processing 3D printing. As such, the development of methodologies for the functionalization of silk materials provide added value. Inorganic nanoparticles (INPs) have interesting and unexpected properties differing from bulk materials. These properties include better catalysis efficiency (better surface/volume ratio and consequently decreased quantify of catalyst), antibacterial activity, fluorescence properties, and UV-radiation protection or superparamagnetic behavior depending on the metal used. Given the promising results and performance of INPs, their use in many different procedures has been growing. Therefore, combining the useful properties of silk fibroin materials with those from INPs is increasingly relevant in many applications. Two main methodologies have been used in the literature to form silk-based bionanocomposites: in situ synthesis of INPs in silk materials, or the addition of preformed INPs to silk materials. This work presents an overview of current silk nanocomposites developed by these two main methodologies. An evaluation of overall INP characteristics and their distribution within the material is presented for each approach. Finally, an outlook is provided about the potential applications of these resultant nanocomposite materials.

Abstract:A ubiquitous feature of natural silk fibers is the presence of well-organized mesostructures, including microfibrils, nanofibrils, and nanoparticles. These mesoscale building blocks are typically well organized into sophisticated arrangements and contribute robust mechanical performance and functions as part of natural silk fibers. However, it remains a major challenge to directly isolate these mesostructures for engineering applications. Here, an environmentally friendly and scalable "partial dissolution and physical dispersion" strategy is developed to exfoliate silk fibers into different mesostructures, including microfibrils, nanofibrils, nanorods, and nanoparticles. On the basis of the advantages of these mesosilks in tunable sizes, sharp size distributions, high modulus, excellent redispersibility, as well as versatile processability, the applications of these mesosilks in electronic and environmental fields are further explored, including water treatment, recycling organic solvent, paper sensors, and nanofertilizers. These explorations open a new avenue for silk fiber applications while also providing a pathway to help address critical issues in electronic and environmental fields.

Abstract:Silk nanofibers (SNF) have great applications in high-performance functional nanocomposites due to their excellent mechanical properties, biocompatibility, and degradability. However, the preparation of SNF by traditional methods often requires the use of some environmentally harmful or toxic reagents, limiting its application in green chemistry. In this paper, we successfully prepared SNF using natural silk as raw material and solvent stripping technology by adjusting the solvent concentration and solution ratio (the diameter of about 120 nm). Using the above SNFs as raw materials, SNF membranes were prepared by vacuum filtration technology. In addition, we prepared an SNF/MXene nanocomposite material with excellent humidity sensitivity by simply coating MXene nanosheets with silk fibers. The conductivity of the material can approach 1400.6 S m−1 with excellent mechanical strength (51.34 MPa). The SNF/MXene nanocomposite material with high mechanical properties, high conductivity, and green degradability can be potentially applied in the field of electromagnetic interference (EMI) shielding, providing a feasible approach for the development of functional nanocomposite materials.

Abstract:Silk Fibroin Nanoparticles (SFNs) have become a reliable and effective biomaterial for nanoencapsulation in several fields such as food, biocosmetics, and medical products due to their physicochemical characteristics. Recently, it has also been developed for application in biomaterials and regenerative medicine, also for cellular nanoencapsulation, and drug delivery systems. Silk fibroin is a natural biomaterial relatively not expensive, biocompatible, eco-friendly, and non-toxic FDA-approved protein derived from the Bombyx mori silkworm cocoon. Fibroin has recently been investigated in the drug field carrier for controlled release. Their application has also become widespread in regenerating therapy as a support for regenerating tissues, as well as in nanomedicine. Furthermore, SFNs have been studied as a carrier for single bioactive molecules such as resveratrol, quercetin, or curcumin for volatile compounds, and for vegetal oils in the form of emulsions. The nanoencapsulation of bioactive compounds in a biopolymer of silk fibroin can be efficiently protected from harmful environmental agents like light, oxygen, or water. The present review reveals that silk fibroin nanoparticles constitute a useful biomaterial to protect, stabilize, and deliver bioactive components. This is a promising combination in safety food, biocosmetic products, nanomedicine, and healthcare.
