Abstract:In vivo optical imaging of trace biomarkers in residual microtumors holds significant promise for cancer prognosis but poses a formidable challenge. Here, a novel hydrogel sensor is designed for ultrasensitive and specific imaging of the elusive biomarker. This hydrogel sensor seamlessly integrates a molecular beacon nanoprobe with fibroblasts, offering both high tissue retention capability and an impressive signal-to-noise ratio for imaging. Signal amplification is accomplished through exonuclease I-mediated biomarker recycling. The resulting hydrogel sensor sensitively detects the biomarker carcinoembryonic antigen with a detection limit of 1.8 pg mL-1 in test tubes. Moreover, it successfully identifies residual cancer nodules with a median diameter of less than 2 mm in mice bearing partially removed primary triple-negative breast carcinomas (4T1). Notably, this hydrogel sensor is proven effective for the sensitive diagnosis of invasive tumors in post-surgical mice with infiltrating 4T1 cells, leveraging the role of fibroblasts in locally enriching tumor cells. Furthermore, the residual microtumor is rapidly photothermal ablation by polydopamine-based nanoprobe under the guidance of visualization, achieving approximate to 100% suppression of tumor recurrence and lung metastasis. This work offers a promising alternative strategy for visually detecting residual microtumors, potentially enhancing the prognosis of cancer patients following surgical interventions. A postoperative implantable hydrogel sensor integrating a molecular beacon (MB) nanoprobe and fibroblasts is designed for visual detection and elimination of residual microtumors by ultrasensitive in situ monitoring of trace biomarkers, whose amplification of the signal is achieved by fibroblast-mediated local enrichment of tumor cells and spatially selective release of exonuclease I (exo I)-triggered biomarker recycling.image

Abstract:Although the cure rate of cancer is very low, the survival rate of patients can be improved if it is diagnosed early and treated in time. Polarity, as a significant parameter of tumor microenvironment, is bound up with the development of cancer. Unfortunately, there is currently no report about tumor-targeted and polarity-specific NIR fluorescent probe for accurate cancer diagnosis. Here, a polarity-specific near-infrared (NIR) fluorescent probe named CX-P based on coumarin-xanthene dye is reported. The probe has the advantages of long emission wavelength (>650 nm) and outstanding tumor targeting ability. CX-P is highly sensitive to polarity, and has a linear range from Delta integral = 0.284 (30% water) to Delta integral = 0.321 (100% water). Moreover, the probe can actively target cancer cells and display strong red fluorescence, while the fluorescence in normal cells is negligible. CX-P is also used to real-time monitor the polarity changes after the treatment of cancer cells with LPS, sucrose and taxol. Based on its NIR characteristics, CX-P is applied to real-time imaging of tumor-bearing mice and the significant tumor targeting ability is further confirmed. As far as we know, this is the first tumor-targeting and polarity -specific NIR fluorescent probe, which will offer a powerful means for accurate cancer diagnosis in the future.

Abstract: Cells are the basic units of life. All organisms are made up of cells. Thus, developing sensing platforms for probing the chemistry and physics in or at a living cell is one of the basic goals in understanding the intricate processes that ultimately contribute to life and life processes. Moreover, in the field of medicine, each cell type has a unique molecular signature that distinguishes between healthy and diseased tissues [1]. Cancerous cells are differentiated from noncancerous ones on the basis of intracellular or extracellular (cell surface) biomarkers. In most cases, the distinctions between normal vs. tumor and benign vs. metastatic cells are often subtle. Thus, the identification of cellular signatures for early cancer cell detection is a major hurdle for cancer therapy. The earlier these signatures can be established, the more effectively they can be treated [2]. In this regard, technological systems that provide sensors with high sensitivity, selectivity, and stability are in high demand to identify the unique cellular characteristics such as proteins and nucleic acids in and on living cells at early disease states, providing the prospects of better health and more effective therapy [3].

Abstract:Abstract With the annual increases in the morbidity and mortality rates of tumors, the use of biomarkers for early diagnosis and real-time monitoring of tumor cells is of great importance. Biomarkers used for tumor cell detection in body fluids include circulating tumor cells, nucleic acids, protein markers, and extracellular vesicles. Among them, circulating tumor cells, circulating tumor DNA, and exosomes have high potential for the prediction, diagnosis, and prognosis of tumor diseases due to the large amount of valuable information on tumor characteristics and evolution; in addition, in situ monitoring of telomerase and miRNA in living cells has been the topic of extensive research to understand tumor development in real time. Various techniques, such as enzyme-linked immunosorbent assays, immunoblotting, and mass spectrometry, have been widely used for the detection of these markers. Among them, the detection of tumor cell markers in body fluids based on electrochemical biosensors and fluorescence signal analysis is highly preferred because of its high sensitivity, rapid detection and portable operation. Herein, we summarize recent research progress in the detection of tumor cell biomarkers in body fluids using electrochemical and fluorescence biosensors, outline the current research status of in situ fluorescence monitoring and the analysis of tumor markers in living cells, and discuss the technical challenges for their practical clinical application to provide a reference for the development of new tumor marker detection methods.

Abstract:Early detection of lung and ovarian cancers relies heavily on identifying tumor biomarkers, but current methods require large blood samples and complex genetic testing. This study presents a novel photonic-metamaterial-based biosensing approach that leverages near-field radiative enhancement to detect cancer biomarkers (CA 125, CEA, and CYFRA 21-1) with high sensitivity. By utilizing structured photonic metamaterials, we optimize specific wavelengths to identify these biomarkers in interstitial fluid, which can be easily collected via minimally invasive microneedle arrays. Integrating near-field interactions with wavelength-selective metamaterials amplifies the thermal response at the nanoscale, allowing for the detection of deficient concentrations of biomarkers. This photonic metamaterial technique provides a faster, more accessible, and affordable alternative to conventional blood-based methods, significantly improving early detection and monitoring of cancer. Ultimately, this approach offers a transformative tool for clinical and research applications in cancer diagnostics.

Abstract:The construction of biosensors for specific, sensitive, and rapid detection of tumor biomarkers significantly contributes to biomedical research and early cancer diagnosis. However, conventional assays often involve large sample consumption and poor sensitivity, limiting their further application in real samples. In recent years, single-molecule biosensing has emerged as a robust tool for detecting and characterizing biomarkers due to its unique advantages including simplicity, low sample consumption, ultra-high sensitivity, and rapid assay time. This review summarizes the recent advances in the construction of single-molecule biosensors for the measurement of various tumor biomarkers, including DNAs, DNA modifications, RNAs, and enzymes. We give a comprehensive review about the working principles and practical applications of these single-molecule biosensors. Additionally, we discuss the challenges and limitations of current single-molecule biosensors, and highlight the future directions.

Abstract:Background: Cancer is one of most dangerous diseases that seriously threaten human health, while tumor biomarkers provide important information for clinical diagnosis and treatment of cancers. Given the low abundance of tumor biomarkers in the bodily fluids at the early stage of cancers, it is particularly important to develop bio sensing methods for accurate measurement of tumor biomarkers with high sensitivity. Objective: Nowadays, gold nanoparticles (AuNPs) that have remarkable physical and chemical properties are extensively used in the design of biosensing strategies. In this context, we mainly review the research progress of AuNPs-based biosensing methods for tumor-related biomedical applications in bodily fluids in recent years. Results: Optical, electrochemical and mass spectrometric biosensing methods using AuNPs are widely used for excellent performances in the assay of tumor biomarkers. Conclusion: The existing methods demonstrate high clinical value, while challenges and expectation of biosensing method in tumor-related biomedical application are also discussed.

Abstract:Single cell analysis is essential for understanding the heterogeneity, behaviors of cells, and diversity of target analyte in different subcellular regions. Nucleolin (NCL) is a multifunctional protein that is markedly overexpressed in most of the cancer cells. The variant expression levels of NCL in subcellular regions have a marked influence on cancer proliferation and treatments. However, the specificity of available methods to identify the cancer biomarkers is limited because of the high level of subcellular matrix effect. Herein, we proposed a novel technique to increase both the molecular and spectral specificity of cancer diagnosis by using aptamers affinity based portable nanopipette with distinctive surface-enhanced Raman scattering (SERS) activities. The aptamers-functionalized gold-coated nanopipette was used to capture target, while p-mercaptobenzonitrile (MBN) and complementary DNA modified Ag nanoparticles (AgNPs) worked as Raman reporter to produce SERS signal. The SERS signal of Raman nanotag was lost upon NCL capturing via modified DNA aptamers on nanoprobe, which further helped to verify the specificity of nanoprobe. For proof of concept, NCL protein was specifically extracted from different cell lines by aptamers modified SERS active nanoprobe. The nanoprobes manifested specifically good affinity for NCL with a dissociation constant Kd of 36 nM and provided a 1000-fold higher specificity against other competing proteins. Furthermore, the Raman reporter moiety has a vibrational frequency in the spectroscopically silent region (1800-2300 cm-1) with a negligible matrix effect from cell analysis. The subcellular localization and spatial distribution of NCL were successfully achieved in various types of cells, including MCF-7A, HeLa, and MCF-10A cells. This type of probing technique for single cell analysis could lead to the development of a new perspective in cancer diagnosis and treatment at the cellular level.

Abstract:Cellular biomarkers mainly contain proteins, nucleic acids, glycans and many small molecules including metabolites, reactive oxygen species and other cellular chemical entities. The detection and mapping of the key cellular biomarkers can effectively help us to understand important cellular mechanisms associated with physiological and pathological processes, which extremely promote the development of clinical diagnosis and diseases treatment. Surface-enhanced Raman scattering (SERS) possesses high sensitivity and is free from the influence of strong self-fluorescence in living systems as well as the photobleaching of the dyes. It holds the rich and narrow chemical fingerprint spectra for multiplexed detection, and has become a powerful tool to detect and map the cellular biomarkers. In this review, we present an overview of recent advances in the detection and mapping of different classes of cellular biomarkers based on SERS sensing. These advances fully confirm that the SERS-based sensors and sensing methods have great potentials for the exploration of biological mechanisms and clinical applications. Additionally, we also discuss the limitations of present reserch and outlook the future developments of the SERS technology in this field.

Abstract:In situ measurement of cellular metabolites is still a challenge in biology. Conventional methods, such as mass spectrometry or fluorescence microscopy, would either destruct the sample or introduce strong perturbations to the functions of target molecules. Here, we present multiplex stimulated Raman scattering (SRS) imaging cytometry as a label-free single-cell analysis platform with chemical specifity, and high-throughput capabilities. Cellular compartments such as lipid droplets, endoplasmic reticulum, and nuclei are seperated from the cytoplasm. Based on these chemical segmentations, 260 features from both morphology and molecular composition were generated and analyzed for each cell. Using SRS imaging cytometry, we studied the metabolic responses of human pancreatic cancer cells under stress by starvation and chemotherapy drug treatments. We unveiled lipid-facilitated protrusion as a metabolic marker for stress-resistant cancer cells through statistical analysis of thousands of cells. Our findings also demonstrate the potential of targeting lipid metabolism for selective treatment of starvation-resistant and chemotherapy-resistant cancers. These results highlight our SRS imaging cytometry as a powerful label-free tool for biological discoveries with a high-throughput, high-content capacity.

Abstract:Because of insufficient information, a single biomarker is not sufficient for early diagnosis of cancer, whereas sensitive and selective detection of multiple biomolecules can significantly reduce analysis time, sample size, and accurately perform cell screening in early cancer. Therefore, the development of a noninvasive strategy that can simultaneously quantify multiple biomarkers (i.e., nucleic acids, proteins, and small molecules) in a single cell is particularly important. Herein, a universal sensing system (functional DNA@mesoporous silica nanoparticles (MSN)-Black Hole Quencher-rhodamine 6G (RhB), FDSBR), which is based on the combination of functionalized DNA and smart responsive nanomaterial, was successfully constructed. After incubation with the cells, different types of targets trigger the strand displacement reaction to release the fluorophore-labeled nucleic acids as the output signals to reflect the intracellular level of the telomerase and adenosine triphosphate (ATP), respectively. Simultaneously, intracellular miR-21 can be clearly indicated by the restored fluorescence of RhB when the caged double-stranded DNA was substituted into single-stranded DNA to open the pore. The concentrations of intracellular telomerase, miR-21, and ATP were identified successfully in three cell lines at the single-cell level. The results show that the contents of three biomolecules have significant differences in the three model cell lines and provide a promising route for developing innovative early disease diagnosis and cell screening assay.

Abstract:<p>Cancer cells alter their metabolism to promote rapid proliferation, resulting in significant amounts of glucose to be used for aerobic glycolysis in the tumor microenvironment. Due to the spatial mismatch between electrodes and cells, existing methods for evaluating cancer cell metabolism lack kinetic and microenvironmental information. In this paper, we present a hollow fiber structure loaded with sensing elements as a long-term cellular metabolism monitoring platform. The unique gradient porous structure allowed the glucose sensor to be close to cultured cells but not directly in contact to prevent adverse effects. The liquid exchange channels in the porous fiber structure ensured continuous and real-time monitoring of glucose concentration changes in the culture media. Experimental results showed high electrochemical sensitivity and stability for continuously monitoring the glucose consumption rate. Furthermore, a continuous three-day long test quantified the change in glucose consumption in response to the anticancer drug Osimertinib. In addition to traditional endpoint cell counting and analysis, this hollow fiber sensing structure provided a real-time monitoring tool for cell metabolism. Such continuous monitoring of the cell metabolism microenvironment improves in-vitro toxicology models for personalized medicine and cancer therapy.</p>

Abstract:Nucleic acid-based cancer biomarkers (NABCBs) have emerged as a notable and continuously captivating research focal point within the domain of oncology. As a result, NABCBs have garnered sustained and steadfast attention from the realms of scientific inquiry and medical practice alike. This comprehensive review embarks on a systematic exploration, delving into the classifications, properties, and functions of NABCBs. Subsequently, it navigates the expanse of clinical applications, where NABCBs assume roles in cancer diagnosis, prognosis, targeted therapy, and immunotherapy. This underscores their pivotal role in translating biomolecular signatures into tangible diagnostic tools. Moreover, an exploration into the strategies underpinning the development of NABCB-based biosensors, such as coordination, crosslinking, fluorescent labeling, and so on, was discussed in details. This segment accentuates the potential of NABCBs to underpin diagnostic and monitoring paradigms, thereby offering a cutting-edge technological foundation for the realization of real-time assessments of early cancer diagnosis and treatment efficacy. In addition, this review embarks on a thorough exploration of these challenges, contributing to an enriched comprehension of the existing status and limitations of NABCBs within the practical clinical milieu. Abstract:The analysis of in situ single cells in tissue is crucial for the study of tumors. However, simultaneous analysis of multiple biomolecules for single cells by fluorescence immunohistochemistry is challenging. Spontaneous Raman spectroscopy provides biochemical fingerprints of individual cells in a label-free and nondestructive manner, yet it is not well applied to single cells in tissue. Here, we develop an in situ single-cell spontaneous Raman spectroscopy (SRS) system that can localize a single cell in tissue with fluorescence imaging, while obtaining the single-cell Raman spectra for molecular analysis. Our system is verified by measuring the standard polystyrene beads of different sizes. Measurements of in situ single macrophages in clinical cancerous and para-cancerous tissues are then performed. By using discriminant analysis, it is shown that CD68 + macrophages in cancerous and para-cancerous tissues are classified with an accuracy of 98.3 %. The molecular content of single macrophages in tissue is analyzed by using multivariate curve resolution-alternating least squares method. We find that the lipid content of tumor-associated macrophages in cancerous tissues is higher while both the contents of protein and nucleic acid are lower than those in para-cancerous tissues, which is challenging to achieve by conventional immunohistochemistry. Our in situ single-cell SRS system has the capability for the obtaining of molecular information of a localized single cell in tissue, which may find potential applications for clinical cancer diagnosis.

Abstract:A novel, effective, and label-free electrochemical sensor was constructed for investigating the interactions between cancer cells and molecules, based on targeted cancer cells immobilized on a bilayer architecture of N-doped graphene-Pt nanoparticles-chitosan (NGR-Pt-CS) and polyaniline (PANI). The interactions between folic acid (FA, positive control) and dimethyl sulfoxide (DMSO, negative control) and the choice of targeted cells, HepG2 and A549 cells, were investigated by measuring the current change of the sensor to [Fe(CN)6]3-/4- before and after interactions, and the binding constants were calculated to be 1.37 × 105 and 1.92 × 105 M-1 by sensing kinetics. Furthermore, 18 main components from Aidi injection (ADI) were studied to screen compounds that have interactions with different targeted cancer cells including HepG2 and A549 cells. The potential target groups of the interactions between screened active compounds and targeted cancer cells were analyzed through computer-aided molecular docking. In this sensing system, molecules did not require electrochemical activity, and different targeted cancer cells could be immobilized on the modified electrode surface, truly reflecting the categories and numbers of targets. Additionally, the proposed sensor specifically circumvented the current paradigm in most cells-based electrochemical sensors for screening drugs, in which the changes in cell behavior induced by drugs are monitored. This study provided a novel, simple, and generally applicable method for exploring the interaction of molecules with cancer cells and screening multitarget drugs.

Abstract:Recently, we have developed a series of robust, nanobiotechnology-based electrochemical biosensing approaches with high sensitivity, and reproducibility toward the simultaneous multiplex detection and classification of both acute myeloid leukemia and acute lymphocytic leukemia cells, quantitative evaluation of DR4/DR5 expression on leukemia cell surface, dual-channel detection of matrixmetalloproteinase-2 (MMP-2) in human serum as well as ultrasensitive circulating tumor cells (CTCs) electrochemical cytosensing using Fe3O4@AuPd nanocages as the nonenzymatic nanoelectrocatalysts . Self-powered biosensor based on enzyme biofuel cell (EBFC) was another kind of super-sensitive self-contained sensing system which was powered by EBFC, meanwhile, the output of EBFC was directly related to the analyte concentration. The system was capable of detecting cancer cells, quantifying two types of cancer-related biomarkers (miR-21 and miR-141). Furthermore, we have developed a self-powered cascading integrated system for tumor diagnosis, therapy, and prognosis supervision. These versatile electrochemical biosensing platforms hold great promise as new point-of-care diagnostic tools for early detections and are believed to be of great clinical value for the evaluation of therapeutic effects on patients after radiation therapy or drug treatment, moreover, provide potential opportunity for developing economical preclinical theranostic platform.
Multicolor Cocktail for Breast Cancer Multiplex Phenotype Targeting and Diagnosis Using Bioorthogonal Surface-Enhanced Raman Scattering Nanoprobes.

Abstract:Early precise diagnosis of cancers is crucial to realize more effective therapeutic interventions with minimal toxic effects. Cancer phenotypes may also alter greatly among patients and within individuals over therapeutic process. The identification and characterization of specific biomarkers expressed on tumor cells are highly demanded for diagnosis and treatment, but remain ongoing challenge. Herein, we designed three new bioorthogonal surface enhanced Raman scattering (SERS) nanoprobes and successfully applied the cocktail of them for MDA-MB-231 and MCF-7 breast cancer multiplex phenotype detection. The SERS nanoprobes containing Raman reporters with diynl, azide or cyano moieties demonstrated apparent Raman shift peaks in 2205 cm-1, 2120 cm-1 and 2230 cm-1, respectively, in biologically Raman-silent region. Three target ligands including oligonucleotide aptamer (AS1411), arginine-glycine-aspatic acid (RGD) peptide and homing cell adhesion molecule antibody (anti-CD44) were respectively conjugated to the nanoprobes for active recognition capability. The cocktail of the nanoprobes manifested minimal cytotoxicity and simultaneously multiplex phenotype imaging of MDA-MB-231 and MCF-7 cells. Quantitative measurement of cellular uptake by in-ductively coupled plasma mass spectrometry (ICP-MS) verified that MDA-MB-231 cells harbored much higher expres-sion level of CD44 receptor than MCF-7 cells. For in vivo SERS detection, Raman shift peaks of 2120 cm-1, 2205 cm-1 and 2230 cm-1 in micro tumor were clearly observed, representing the existence of three specific biomarkers of nucleolin, integrin αvβ3 and CD44 reporter, which could be used for early cancer phenotype identification. The biodistribution results indicated that target ligand modified nanoprobes exhibited much more accumulation in tumors than those nanoprobes without target ligands. The multicolor cocktail of bioorthogonal SERS nanoprobes offers an attractive and insightful strategy for early cancer multiplex phenotype targeting and diagnosis in vivo with non-invasion, low cross-talk, unique spectral-molecular signature, high sensitivity and negligible background interference.

Abstract:Early screening and diagnosis are the most effective ways to prevent the occurrence and progression of cancers, thus many biosensing strategies have been developed to achieve economic, rapid, and effective detection of various cancer biomarkers. Recently, functional peptides have been gaining increasing attention in cancer-related biosensing due to their advantageous features of a simple structure, ease of synthesis and modification, high stability, and good biorecognition, self-assembly and antifouling capabilities. Functional peptides can not only act as recognition ligands or enzyme substrates for the selective identification of different cancer biomarkers but also function as interfacial materials or self-assembly units to improve the biosensing performances. In this review, we summarize the recent advances in functional peptide-based biosensing of cancer biomarkers according to the used techniques and the roles of peptides. Particular attention is focused on the use of electrochemical and optical techniques, both of which are the most commonly used techniques in the field of biosensing. The challenges and promising prospects of functional peptide-based biosensors in clinical diagnosis are also discussed.
