Single-Molecule Fingerprint Spectrum for Biological Nanopore Sensing

Abstract:Nanopore technique has been developed as a powerful approach for single-molecule detection. Current nanopore data analysis method mainly involves time-domian process including the amplitude, translocation duration and interval time of the current events. However, more abundant information has been hidden from the noise which lacks quantitative method to extract them appropriately. In view of the advantages that there are intrinsic characteristics in noise during the analytes traversing through the nanopore, here, we present a mutidimensional data analysis strategy to obtain the single-molecule fingerprint frequency spectrum from current traces by Hilbert-Huang Transform. The underlying physical and chemical process was explored behind this spectrum when poly(dA)4 was in the aerolysin nanopore. The crosslink was established between the frequency value and a series of characteristic variables which included the properties of the analytes, the nanopore integrity, the amino acids residue of aerolysin nanopore at mutant site. The results have demonstrated that the existence of analytes inside the pore, the reversible protonation of amino acids residue in pore lumen and the number of carriers would cause great changes of this single-molecule fingerprint frequency spectrum. The pearson correlation coefficient has also showed that the spectrum has mainly associated with the conductance of nanopore without DNA, while it correlates more with the properties of mutant amino acids with the DNA inside. These results indicate that the non-covalent interaction between the analytes and nanopore may be reflected in it. This spectrum gives multidimensional feature of data and could by further utilized in the application of peptide sequencing.
