Abstract:Ultra-high pressure sterilization technology is currently one of the popular non-thermal sterilization technologies of agricultural products(food).As one of the alternative heat sterilization technologies,it has the most industrialization potential.In recent years,this technology has become a hot area of agricultural products(food) sterilization technology,and the sterilization mechanism and influencing factors of sterilization effect are constantly have new discoveries.On the basis of summary and analysis of domestic and foreign research reports,this paper reviewed the mechanism research status of microorganisms impacted by high pressure from morphological,chemical reaction,gene mechanism and cell membrane.Experiments research status of factors influencing germicidal efficacy were also reviewed in this paper.Theoretical guidance is provided for its further research.

In details, the popular rules resulted from laser irradiating microorganism experiments were described and the operating mechanism of the interaction between laser and microorganism was discussed. On this basis, the application study of laser irradiating microorganism technology for mutagenesis was also discussed. The future progress of this field was forecasted and some useful advices were given in the end also.

Abstract:Heavy ion biotechnology has received much attention since the mid-1980s,when the first mutagenic effect of heavy ion beams was found in rice.Since then,plant breeding is one of the focuses in heavy ion biotechnology.In this paper we outlined the recent developments in plant breeding by heavy ion beams.We introduced briefly the essential characteristics and superiority of heavy ion beams applied in plant breeding,and reviewed further the mutagenic research in three levels(e.g.,molecular,cell and individual) of plants.At last,we summarized artificial mutants obtained in plants irradiated by heavy ion beams.

Abstract:Radio-frequency atmospheric pressure glow discharge (RF APGD) plasmas have outstanding features of no need of expensive and complicated vacuum system, low gas temperatures, high concentrations of reactive species, good discharge uniformity, strong controllability and diverse interactions with various biomolecules, which have attracted more and more attentions for their applications in biotechnology. Our research group introduced the RF APGD plasma jet into the mutation breeding field. Based on the study on the physical characteristics of RF APGD plasmas and their interaction mechanisms with bio-macromolecules and whole cells, a novel mutation breeding system, named as atmospheric and room temperature plasma (ARTP) mutation breeding machine was invented. The ARTP mutation breeding system has the features of high safety for the operators and environment, easy operation, rapid mutation capability, high mutation rate and genetic stability of targeted mutants,as demonstrated by successful mutation for more than 40 different microbes including bacteria, fungi and algae. This paper reviews the recent research progress of ARTP mutation breeding technology, hopefully contributing to the development of the life evolution study and the industrial strain modification as a powerful mutation tool.

Abstract:Bio-effects of non-thermal plasma include lethality of microorganisms and stimulation of mammalian cells,which brings up a series of techniques in disinfection and clinical medicine.In this paper,the applications of non-thermal plasma in medical device sterilization,water and air cleaning,as well as food and package treatment are reviewed.Commercialized plasma sterilizer for medical devices can be classified into vacuum and atmospheric pressure,the volume of the former is much larger while the latter is more simple and easy to operate.For disinfection of water,air and food,it is not yet commercialized due to its energy consumption,efficiency or chemical residues.In the last decade,the developed plasma sources and techniques for medicine are successfully applied to treatment of decayed tooth,skin,wound,and cancer,with some of them approved for clinical practice.In the future,the research focus will be identifying the interactions between plasmas and cells,and developing more effective and practical equipment for different applications.

Transgenic plants testing system can not only evaluate the level of genotoxicity, but also provide information on the genetic effects at molecular level. Abstract:At present, atmospheric and room‐temperature plasma (ARTP) is regarded as a new and powerful mutagenesis technology with the advantages of environment‐friendliness, operation under mild conditions, and fast mutagenesis speed. Compared with traditional mutagenesis strategies, ARTP is used mainly to change the structure of microbial DNA, enzymes, and proteins through a series of physical, chemical, and electromagnetic effects with the organisms, leading to nucleotide breakage, conversion or inversion, causing various DNA damages, so as to screen out the microbial mutants with better biological characteristics. As a result, in recent years, ARTP mutagenesis and the combination of ARTP with traditional mutagenesis have been widely used in microbiology, showing great potential for application. In this review, the recent progress of ARTP mutagenesis in different application fields and bottlenecks of this technology are systematically summarized, with a view to providing a theoretical basis and technical support for better application. Finally, the outlook of ARTP mutagenesis is presented, and we identify the challenges in the field of microbial mutagenesis by ARTP.

Abstract:Irradiation of energetic particles on microbes can induce varied biological effects,therefore, ion-beam biotechnology has been applied for mutation breeding. But the interaction between the energetic particles and biological systems is very complicated,and the mechanism for the physical and chemical changes is still elusive. A new trend is to apply various spectroscopic tools to monitor the detailed processes at the micro-scale because they can provide rapid and noninvasive measurements. In this work,the author applied α-particles from241Am to irradiate Rhizopus oryzae and employed Raman microspectroscopy and imaging tool to observe and record the changes in the microorganism. Based on the spectral observation and analysis the involved biological effect was revealed and discussed.

Current Issues in Molecular Biology

Abstract:Atmospheric and room-temperature plasma (ARTP) is an efficient microbial mutagenesis method with broad application prospects. Compared to traditional methods, ARTP technology can more effectively induce DNA damage and generate stable mutant strains. It is characterized by its simplicity, cost-effectiveness, and avoidance of hazardous chemicals, presenting a vast potential for application. The ARTP technology is widely used in bacterial, fungal, and microalgal mutagenesis for increasing productivity and improving characteristics. In conclusion, ARTP technology holds significant promise in the field of microbial breeding. Through ARTP technology, we can create mutant strains with specific genetic traits and improved performance, thereby increasing yield, improving quality, and meeting market demands. The field of microbial breeding will witness further innovation and progress with continuous refinement and optimization of ARTP technology.

Abstract:Femtosecond laser induced micro-organisms mutation has become a new microbial technology,because of its characteristics of ultrafast pulse,high peak power and focusing on a small size.In this paper,compared to the advantages and disadvantages of the traditional low-power He-Ne laser irradiation,the technology of femtosecond laser induced micro-organisms mutation is discussed.Meanwhile,compared to traditional laser mutation mechanism of micro-organisms,the mechanisms of multi-photon absorption,formation of plasma,formation of reactive oxygen species,DNA damage and self-repair are summarized.Finally,the prospect for technology of femtosecond laser induced micro-organisms mutation is given,and the combination of key-enzyme activity measurement and dynamics parameters determination is important in choosing the technological parameters of femtosecond laser induced micro-organisms mutation.
Biological Effects on Fruit Fly by N~+ ion Beam Implantation

Abstract:Mutation induced by low energy ion beam implantation has been applied widely both in plants and microbes. However, due to the vacuum limitation, such ion implantation into animals was never studied except for silkworm. In this study, Pupae of fruit fly were irradiated with different dosage N+ ions at energy 20 KeV to study the biological effect of ion beam on animal. The results showed a saddle-like curve exists between incubate rate and dosage. Damage of pupae by ion beam implantation was observed using scanning electron microscope. Some individuals with incomplete wing were obtained after implantation but no similar character was observed in their offspring. Furthermore, about 5.47% mutants with wide variation appeared in M1 generation. Therefore, ion beam implantation could be widely used for mutation breeding.

Abstract:In this paper, both simulations and experiments are conducted to investigate the physical characteristics of the radio-frequency atmospheric-pressure glow discharges (RF APGDs) first. The experimental and modeling results show that there are abundant chemically reactive species, e.g., electrons, helium metastables and ions, with high number densities in the discharge region. The corresponding plasma jet temperatures are in the range of 27°C - 39°C with the power input lower than 120 W, which is suitable for the treatment of the biomolecules or cells. Then, a prototype, which is the so-called atmospheric and room-temperature plasma mutation system, is designed with the RF APGD plasma generator as the core component. As an example, it is used for the mutation breeding of Methylosinus trichosporium OB3b. The experimental results show that the RF APGD plasma source is an environmentally friendly and efficient tool for the genome mutation of microorganisms to construct mutant library with large diversity.

Abstract:Developing rapid and diverse microbial mutation tool is of importance to strain modification. In this review, a new mutagenesis method for microbial mutation breeding using the radio-frequency atmospheric-pressure glow discharge (RF APGD) plasma jets is summarized. Based on the experimental study, the helium RF APGD plasma jet has been found to be able to change the DNA sequences significantly, indicating that the RF APGD plasma jet would be a powerful tool for the microbial mutagenesis with its outstanding features, such as the low and controllable gas temperatures, abundant chemically reactive species, rapid mutation, high operation flexibility, etc. Then, with the RF APGD plasma generator as the core component, a mutation machine named as atmospheric and room temperature plasma (ARTP) mutation system has been developed and successfully employed for the mutation breeding of more than 40 kinds of microorganisms including bacteria, fungi, and microalgae. Finally, the prospect of the ARTP mutagenesis is discussed.

Abstract:Microbial mutation breeding has been widely used because it is one of the most efficient and practical breeding strategies in the fermentation industry. However, different mutagenesis methods cause various degrees of DNA damage to individual microorganisms, which lead to diverse characteristics of the mutants. In this study, the effects of four different mutagenesis methods on the mutation breeding of Streptomyces avermitilis for improving avermectin B1a production were investigated with an optimized liquid microtiter plate (MTP) culture system. First, an effective and feasible MTP system for mutant strain screening was evaluated through the optimization of the oxygen transfer rate and rapid titer determination. Then, high energy carbon heavy ion irradiation, diethyl sulfate, ultraviolet- (UV) irradiation combined with lithium chloride, and sodium nitrite were used as the mutagens for mutation breeding, respectively. Results showed that carbon heavy ion irradiation had the advantages of possessing the highest positive mutation rate and mean-production of positive mutant strains in the first generation. Sodium nitrite treatment resulted in mutant strains with better inherited stability than the other three methods. Through the combined treatment of carbon heavy ion irradiation and sodium nitrite treatment, an inheritstable mutant S. avermitilis S-233 with high avermectin B1a production was successfully obtained. The fermentation verification in a 500-liter (L) bioreactor demonstrated that the avermectin B1a produced by mutant S. avermitilis S-233 reached 6818 μg/mL, which was 23.8% higher than that of parent strains.

Abstract:Microorganisms are indispensable in the food industry, but wild-type strains hardly meet the current industrial demands due to several undesirable traits. Therefore, microbial strain improvement offers a critical solution to enhance the food industry. Traditional techniques for food microbial improvement, such as the use of chemical mutagens and manual isolation/purification, are inefficient, time-consuming, and laborious, restricting further progress in the area of food fermentation. In this review, the applications of novel mutagenesis and screening technologies used for the improvement of food microbes were summarized, including random mutagenesis based on physical irradiation, microbial screening facilitated by a microtiter plate, fluorescence-activated cell or droplet sorting, and microscaled fermentation in a microtiter plate or microbioreactor. In comparison with conventional methods, these new tools have the potential in accelerating microbial strain improvement and their combined applications could create a new trend for strain development. However, several problems that could affect its potential application may include the following: the lack of specific mutagenesis devices and biosensing systems, the insufficient improvement of the mixed culture system, the low efficiency when using filamentous fungi and flocculating bacteria, and the insufficient safety assessment on harnessing genome-editing technology. Therefore, future works on strain improvement remain challenging for the food industry.
