Abstract:The rapid development of biotechnology accelerates modification of tree in molecular level.It is expected to have enormous benefits both for economy and ecology.This article presents an overview of the progress in genetic transformation of pear in the past twenty years.Particular attention is paid to the target genes,the explants,the methods for detecting transformants and the factors affecting pear transformation.The remaining problems were summarized and the key points for further research were suggested as well.

Abstract:Inheritable variation induced by plant grafting is a universal phenomenon; however, its mechanism has always been a controversial subject. In recent years, research on horizontal transfer of genetic materials between stock and scion has made great progress. The latest studies have found that genetic information in grafted plants is transported through plasmodesma and phloem to modulate the growth and development of the scion plants. Furthermore, non-coding RNAs could induce epigenetic modification, which facilitates plants to adapt to the grafting-caused stress. This article focuses on the review of recent advances in the induction and maintenance of the graft-induced genetic variation and could be helpful in thoroughly understanding the mechanism.

Abstract:Strawberry (Fragaria ananassa Duch.) is one of most important fruit crops cultivated widely in world. Genetic transformation has launched a new era in strawberry breeding and germplasm creativity. It offers a direct method of creating varieties that selectively targets gene or a few heterologous traits for introduction into the strawberry plant. Great advances have been made in strawberry genetic transformation in the past years. This paper reviews the recent progress in genetic transformation of strawberry on promoting resistance to viruses and fungi, insects, herbicides, stress and quality improvement. Problems and the prospects for application of genetic transformation in strawberry were discussed.

Abstract:The ability to efficiently genetically modify plant species is crucial, driving the need for innovative technologies in plant biotechnology. Existing plant genetic transformation systems include Agrobacterium -mediated transformation, biolistics, protoplast-based methods, and nanoparticle techniques. Despite these diverse methods, many species exhibit resistance to transformation, limiting the applicability of most published methods to specific species or genotypes. Tissue culture remains a significant barrier for most species, although other barriers exist. These include the infection and regeneration stages in Agrobacterium, cell death and genomic instability in biolistics, the creation and regeneration of protoplasts for protoplast-based methods, and the difficulty of achieving stable transformation with nanoparticles. To develop species-independent transformation methods, it is essential to address these transformation bottlenecks. This review examines recent advancements in plant biotechnology, highlighting both new and existing techniques that have improved the success rates of plant transformations. Additionally, several newly emerged plant model systems that have benefited from these technological advancements are also discussed.

Abstract:Rosaceae fruits are very important for fruit production in China and worldwide.Recently,significant advances in regeneration and genetic transformation of Rosaceae have been achieved and applied to various Rosaceae trees and fruits.However,the factors affecting the process of regeneration and transformation have not been reviewed in detail.This paper reviews recent progress in the regeneration pathway,factors affecting regeneration,genetically modified species and main traits,and the factors affecting Agrobacterium-mediated genetic transformation.The differences between species of Rosaceae fruits are emphasized,and the trends and areas for future studies are highlighted.

Abstract:In the past two decades, Chinese scientists have achieved significant progress on three aspects of wheat genetic transformation. First, the wheat transformation platform has been established and optimized to improve the transformation efficiency, shorten the time required from starting of transformation procedure to the fertile transgenic wheat plants obtained as well as to overcome the problem of genotype-dependent for wheat genetic transformation in wide range of wheat elite varieties. Second, with the help of many emerging techniques such as CRISPR/cas9 function of over 100 wheat genes has been investigated. Finally, modern technology has been combined with the traditional breeding technique such as crossing to accelerate the application of wheat transformation. Overall, the wheat end-use quality and the characteristics of wheat stress tolerance have been improved by wheat genetic engineering technique. So far, wheat transgenic lines integrated with quality-improved genes and stress tolerant genes have been on the way of Production Test stage in the field. The debates and the future studies on wheat transformation have been discussed, and the brief summary of Chinese wheat breeding research history has also been provided in this review.

Abstract:During the past two decades,substantial progress has been made in genetic modification of turfgrasses.The systems for genetic transformation have been established in most of the main turfgrass species,and the induction of resistant genes into the species has been becoming the hotspot.The factors affecting the genetic transformation of turfgrasses,and the problems existing in the course of transformation,i.e.,somaclonal variation and the stabilization in the transgenic plants,the efficiency of genetic transformation,the effect of foreign genes on the environment were summarized,the relationship between genetic transformation and traditional breeding was discussed and the research aims for turfgrass breeding in the future were pointed out.

Abstract:Transgenic technology is increasingly used in forest-tree breeding to overcome the disadvantages of traditional breeding methods, such as a long breeding cycle, complex cultivation environment, and complicated procedures. By introducing exogenous DNA, genes tightly related or contributed to ideal traits—including insect, disease, and herbicide resistance—were transferred into diverse forest trees, and genetically modified (GM) trees including poplars were cultivated. It is beneficial to develop new varieties of GM trees of high quality and promote the genetic improvement of forests. However, the low transformation efficiency has hampered the cultivation of GM trees and the identification of the molecular genetic mechanism in forest trees compared to annual herbaceous plants such as Oryza sativa . In this study, we reviewed advances in transgenic technology of forest trees, including the principles, advantages and disadvantages of diverse genetic transformation methods, and their application for trait improvement. The review provides insight into the establishment and improvement of genetic transformation systems for forest tree species. Challenges and perspectives pertaining to the genetic transformation of forest trees are also discussed.

Abstract:Great progress has been made in recent years in studies on the mechanism of Agrobacterium-mediated transformation and its application. Many details of the key molecular events within the bacterial cells involved in T-DNA transfer have been elucidated, and it is notable that some plant factors which were elusive before are purified and characterized. Vast kinds of species, which were either recalcitrant to or not included in the host range of Agrobacterium, can' now be transformed by this bacterium, and they include the very important cereal species, gymnosperms, yeast and many filamentous fungi. The simple in vivo transformation of tissue in intact plants and the 'agrolistic' methods to transform recalcitrant plants are the two novel technical achievements. Combined with other powerful techniques such as bacterial artificial chromosome, very large DNA fragment can be transformed into the plant genome by Agrobacterium. Abstract:Wheat is one of the most important stable crops in the world,feasible wheat transgenic technique has been advancing more slowly than that of other gramineous crops,including maize and rice,however,some gratifying results have been obtained in recent years.In the review,we made an analysis of various methods experimented in wheat transformation,such as pollen tube pathway,particle bombardment and agrobacterium-mediated transformation.We also made a classification analysis of the functional genes that have been transferred to wheat,e.g.DREB,LEA for salt/drought resistance,GNA,Bt for disease and insect resistance,aroA,Bar for herbicide resistance,and 1Dt10,1Ax1 for flour quality improvement.In the end,we gave a outlook of wheat transgenic techniques and its strategy,especially for the agrobacterium-mediated floral dip transformation,in order to promote the improvement of wheat transgenic techniques and their application in wheat breeding.

Abstract:Genetic transformation is an important strategy for enhancing plant biomass or resistance in response to adverse environments and population growth by imparting desirable genetic characteristics. Research on plant genetic transformation technology can promote the functional analysis of plant genes, the utilization of excellent traits, and precise breeding. Various technologies of genetic transformation have been continuously discovered and developed for convenient manipulation and high efficiency, mainly involving the delivery of exogenous genes and regeneration of transformed plants. Here, currently developed genetic transformation technologies were expounded and compared. Agrobacterium-mediated gene delivery methods are commonly used as direct genetic transformation, as well as external force-mediated ways such as particle bombardment, electroporation, silicon carbide whiskers, and pollen tubes as indirect ones. The regeneration of transformed plants usually involves the de novo organogenesis or somatic embryogenesis pathway of the explants. Ectopic expression of morphogenetic transcription factors (Bbm, Wus2, and GRF-GIF) can significantly improve plant regeneration efficiency and enable the transformation of some hard-to-transform plant genotypes. Meanwhile, some limitations in these gene transfer methods were compared including genotype dependence, low transformation efficiency, and plant tissue damage, and recently developed flexible approaches for plant genotype transformation are discussed regarding how gene delivery and regeneration strategies can be optimized to overcome species and genotype dependence. This review summarizes the principles of various techniques for plant genetic transformation and discusses their application scope and limiting factors, which can provide a reference for plant transgenic breeding.

Abstract:It was reviewed the rapeseed explant types,selection markers,transformation methods,the influence factors of regeneration systems and its applications in rapeseed breeding were discussed.It was also mentioned the problems and development in the future of rapeseed transformation.It was found that there were still limitations in the transformation of rapeseeds through the published papers and establishment of transformation system of rapeseeds.Firstly,the process of receptor regeneration was a long period and the frequency of gene transformation was low.Secondly,the introduced gene might be silent and the expression of transferred gene would unstable in the progeny.Thirdly,the scientific evaluation should be made about the bad influence of transgenic rapeseed to human heath and the natural environment.Moreover,more complete law and rule should be made to supervise the environment release of transgenic plants.At the same time,it should meet the needs of people by modifying the trait of rapeseeds.Furthermore,there would be a wide foreground to produce the substitute of energy source by introducing special gene into rapeseed plants.
