Abstract:The resolution in nuclear magnetic resonance (NMR) microscopy is limited by a combination of the inherent low sensitivity of NMR, the destruction of spin magnetization gratings by molecular diffusion, and variations in the local magnetic field strength introduced by spatial variations of the bulk susceptibility. While none of these may be completely overcome, constant time imaging methods are the optimal approach to recording images when the above factors are important. This method aims to instantaneously create a spin magnetization grating and then to efficiently sample the spatially invariant portion of this - corresponding to a selected Fourier component of the sample distribution. The method is introduced, analyzed in the presence of molecular diffusion, and demonstrated to yield high-resolution images. To implement the method, a microscopy probe was constructed for a standard-bore 400-MHz NMR spectrometer (the latter was also implemented at 600 MHz), which is described. Abstract:The simultaneous effects of diffusion and coherent averaging by magic angle sample spinning (MASS) are investigated both theoretically and experimentally for spins diffusing in a spatially varying susceptibility field. The diffusion introduces a periodic modulation and a relaxation of the magnetization amplitude. The attenuation exponent for the nth echo at an early time is given by D(0)gamma (2)g(2)pin(4 omega (3)(r)), where omega (r) is the rotor frequency, D-0 the diffusion coefficient, gamma is the gyromagnetic factor, and a factor g(2) which is related to, but different from, the volume average of the gradient squared. Abstract:Nuclear magnetic resonance (NMR) technique presents a powerful analytical tool in the fields of chemistry, biology, and material science. The performance of the NMR technique highly relies on post-data processing methods, which are diverse according to different applications. In this article, we propose a general processing method that can cover a wide range of applications from molecular dynamics study to biomedical engineering. We analyze the computation performance of the method by comparing it to other well-known algorithms and demonstrate its application in various NMR data processing problems, i.e., diffusion-ordered spectroscopy processing, under-sampled magnetic resonance imaging (MRI) reconstruction, Laplace NMR (LNMR) inversion, and spectrum denoising. The results corroborate that the method performs effectively and efficiently compared to state-of-the-art methods. Moreover, we construct the method as a light graphic-based toolbox for convenient use. More detail instructions of the toolbox for chemists will be given in future works.

Monitoring Electron Spin Fluctuations with Paramagnetic Relaxation Enhancement

Abstract:The magnetic interactions between the spin of an unpaired electron and the surrounding nuclear spins can be exploited to gain structural information, to reduce nuclear relaxation times as well as to create nuclear hyperpolarization via dynamic nuclear polarization (DNP). A central aspect that determines how these interactions manifest from the point of view of NMR is the timescale of the fluctuations of the magnetic moment of the electron spins. These fluctuations, however, are elusive, particularly when electron relaxation times are short or interactions among electronic spins are strong. Here we map the fluctuations by analyzing the ratio between longitudinal and transverse nuclear relaxation times T1 and T2, a quantity which depends uniquely on the rate of the electron fluctuations and the Larmor frequency of the involved nuclei. This analysis enables rationalizing the evolution of NMR lineshapes, signal quenching as well as DNP enhancements as a function of the concentration of the paramagnetic species and the temperature, demonstrated here for LiMgMnPO4 and Fe(3+) doped Li4Ti5O12, respectively. For the latter, we observe a linear dependence of the DNP enhancement and the electron relaxation time within a temperature range between 100 and 300K.

Abstract:The function of biomolecular systems, including biological macromolecules, often crucially depends on their dynamics. Nuclear Magnetic Resonance (NMR) is one of the most informative methods used to study biomolecules and their internal mobility, with atomic resolution, in near-physiological conditions. NMR relaxation profiles, obtained from the field dependence of the nuclear relaxation rates, in particular, offer the possibility to probe dynamic processes over a wide range of time scales. Relaxation profiles are routinely acquired using field-cycling relaxometers operating at a maximum field of the order of 1 T. These measurements however suffer from a lack of resolution. On the other hand, relaxation rates measured at high magnetic fields contain poor information on motions on timescales longer than few nanoseconds. The possibility to acquire relaxation profiles extended to low fields but with high resolution, obtained by shuttling the sample back and forth in the stray field of a high-field spectrometer, is expected to dramatically improve the potentialities of NMR relaxometry. Here, we review investigations of relaxometry in a wide range of biomolecular systems, such as proteins, phospholipids, or biological fluids. Although multiple models of motions have been developed to describe the relaxation rates and their field dependence, most experimental investigations rely on the model-free approach. A variety of relaxation profiles of both diamagnetic and paramagnetic biomolecular systems are here reviewed and analysed using point dipole-point dipole interaction models.

Pitfalls in measurements of R1 relaxation rates of protein backbone 15N nuclei

Abstract:The dynamics of the backbone and side-chains of protein are routinely studied by interpreting experimentally determined 15N spin relaxation rates. R1(15N), the longitudinal relaxation rate, reports on fast motions and encodes, together with the transverse relaxation R2, structural information about the shape of the molecule and the orientation of the amide bond vectors in the internal diffusion frame. Determining error-free 15N longitudinal relaxation rates remains a challenge for small, disordered, and medium-sized proteins. Here, we show that mono-exponential fitting is sufficient, with no statistical preference for bi-exponential fitting up to 800 MHz. A detailed comparison of the TROSY and HSQC techniques at medium and high fields showed no statistically significant differences. The least error-prone DD/CSA interference removal technique is the selective inversion of amide signals while avoiding water resonance. The exchange of amide with solvent deuterons appears to affect the rate R1 of solvent-exposed amides in all fields tested and in each DD/CSA interference removal technique in a statistically significant manner. In summary, the most accurate R1(15N) rates in proteins are achieved by selective amide inversion, without the addition of D2O. Importantly, at high magnetic fields stronger than 800 MHz, when non-mono-exponential decay is involved, it is advisable to consider elimination of the shortest delays (typically up to 0.32 s) or bi-exponential fitting.

Abstract:Degenerate spin-systems consisting of magnetically equivalent nuclear spins, such as a 1H3 spin-system in selectively 13CH3-labeled proteins, present considerable challenges for the design of Carr-Purcell-Meiboom-Gill (CPMG) relaxation dispersion NMR experiments to characterize chemical exchange on the micro-to-millisecond time-scale. Several approaches have been previously proposed for the elimination of deleterious artifacts observed in methyl 1H CPMG relaxation dispersion profiles obtained for (13C)1H3 groups. We describe an alternative, experimentally simple solution and design a "steady-state" methyl 1H CPMG scheme, where 90° or acute-angle (<90°) 1H radiofrequency pulses are applied after each CPMG echo in-phase with methyl 1H magnetization, resulting in the establishment of a "steady-state" for effective rates of magnetization decay. A simple computational procedure for quantitative analysis of the "steady-state" CPMG relaxation dispersion profiles is developed. The "steady-state" CPMG methodology is applied to two protein systems where exchange between major and minor species occurs in different regimes on the chemical shift time-scale.

Abstract:Methods for T-1 relaxation and diffusion measurements based on magnetic resonance signals from laser-hyperpolarized Xe-129 nuclei are introduced. The methods involve optimum use of the perishable hyperpolarized magnetization of Xe-129. The necessary theoretical framework for the methods is developed, and then the methods are applied to measure the longitudinal relaxation constant, T-1, and the self-diffusion constant, D, of hyperpolarized Xe-129. In a cell containing natural abundance Xe-129 at 790 Torr, the T-1 value was determined to be 155 +/- 5 min at 20 degrees C and at 2.0 T field. For a second cell at 896 Torr, at the same field and temperature, the T-1 value was determined to be 66 +/- 2 min. At a higher field of 7.05 T, the T-1 values for the two cells were found to be 185 +/- 10 and 88 +/- 5 min, respectively. The Xe-129 self-diffusion constant for the first cell was measured to be 0.057 cm(2)/s and for the second cell it was 0.044 cm(2)/s. The methods were applied to Xe-129 in the gas phase, in vitro; however, they are, in principle, applicable for in vivo or ex vivo studies. The potential role of these methods in the development of newly emerging hyperpolarized Xe-129 MRI applications is discussed. (C) 1997 Academic Press.
A Non-Steady State NMR Effect Based Time-Varying Magnetic Field Measurement Method and Experimental Apparatus

Abstract:Accurate measuring magnetic induction intensity is important for some scientific research. In the paper, the conventional nuclear magnetic resonance (NMR) experiment under static magnetic field has been extended to longitudinal time-varying magnetic field (TVMF), and the time domain transient solution of the Bloch equations for spin nuclei exhibits the so-called non-steady state nuclear magnetic resonance (NSS-NMR) phenomenon. Based on the above effect and the precise gyromagnetic ratio of spin nucleus, a new magnetic field measurement method and corresponding experimental apparatus have been developed in longitudinal TVMF environment. The orthogonal dual-coil probe, radio frequency (RF) exciting and receiving hardware, key signal processing algorithms, host computer controlling and analysis software are deeply studied. In order to improve the signal-to-noise ratio of the NSS-NMR signal, the RF signal envelope shaping and the low-frequency pre-emphasis probe matching techniques are designed and verified by the invented circuit-field-nuclear multi-domain joint simulation subassembly (CFN-MDJS) which is integrated in the host computer software. Furthermore, the multi-tone RF excitation signal substituting the bandpass continuous spectrum signal for NSS-NMR experiment is also verified by the CFN-MDJS. The experimental results under longitudinal 50Hz TVMF environment has demonstrated the authentic existence of the NSS-NMR effect and simultaneously verified the effectiveness of the experimental scheme and signal processing algorithms proposed in the paper.

Abstract:We propose a novel method for measuring relaxation rates and Fermi-contact fields of the electron and nuclear spin ensembles rapidly and synchronously in spin-exchange relaxation-free (SERF) atomic comagnetometers based on the transient response. The transient response model of the comagnetometer is established by solving a set of Bloch equations. The decay rates and oscillation frequencies of the transient response as functions of the longitudinal magnetic field are investigated theoretically and experimentally. The total relaxation rate of the nuclear spin ensemble is obtained from its decay rate. A linear model is presented with a detailed mathematical derivation based on the decay rates and oscillation frequencies of the spin ensembles. The Fermi-contact fields of the spin ensembles and the slowing-down factor are extracted from the slope and intercept of the linear model. The total relaxation rate of the electron spins is obtained from the decay rates of the spin ensembles and the measured slowing-down factor. The validity of this method is further checked by the magnetic resonance (MR) method from a perspective of the electron resonance linewidth. Identification of these systematic parameters paves the way for operating the comagnetometers efficiently as well as the practical applications of the comagnetometers.

Longitudinal Relaxation Optimization Enhances 1h‐detected HSQC in Solid‐State NMR Spectroscopy on Challenging Biological Systems

Abstract:Solid-state (SS) NMR spectroscopy is a powerful technique for studying challenging biological systems, but it often suffers from low sensitivity. A longitudinal relaxation optimization scheme to enhance the signal sensitivity of HSQC experiments in SSNMR spectroscopy is reported. Under the proposed scheme, the H-1 spins of H-1-X (N-15 or C-13) are selected for signal acquisition, whereas other vast H-1 spins are flipped back to the axis of the static magnetic field to accelerate the spin recovery of the observed H-1 spins, resulting in enhanced sensitivity. Three biological systems are used to evaluate this strategy, including a seven-transmembrane protein, an RNA, and a whole-cell sample. For all three samples, the proposed scheme largely shortens the effective H-1 longitudinal relaxation time and results in a 1.3-2.5-fold gain in sensitivity. The selected systems are representative of challenging biological systems for observation by means of SSNMR spectroscopy; thus indicating the general applicability of this method, which is particularly important for biological samples with a short lifetime or with limited sample quantities.

Abstract:To determine the surface relaxivity of pores plays a vital role in the application of time-domain nuclear magnetic resonance (TD-NMR) technology to porous structure characterization for wood. Currently, the surface relaxivity of pores is calibrated using a standard sample with a pore size of the same order as the wood pore system. However, the uniformly distributed pore size of standard sample is unfit to accurately indicate the complexity of porous structure of wood, which significantly affects the accuracy of test results. By integrating the TD-NMR technology with mercury intrusion porosimetry (MIP), the surface relaxivity of macropores in the lumen of wood cells is calibrated in this study using the tested sample, so as to avoid the error in measurement as caused by existing method. Data processing is performed using several mathematical methods including interpolation arithmetic and least square principle. Notably, the node segmentation method is applied to identify the T 2 boundary of pores in cell lumen and to classify the porous structure of cell lumen into different pore systems. The approach proposed in this study is demonstrated to be effective in improving the accuracy of TD-NMR technology for characterizing the porous structure of wood. Also, it contributes a potential solution to accounting for the porous structure of wood based on the phenomenon of pore relaxation, which can improve the understanding of wood pore conformation.

Abstract:Deuterium rotating frame solid-state NMR relaxation measurements ( 2 H R1ρ ) are important tools in quantitative studies of molecular dynamics. We demonstrate how 2 H to 13 C cross-polarization (CP) approaches under 10–40 kHz magic angle spinning rates can be combined with the 2 H R1ρ blocks to allow for extension of deuterium rotating frame relaxation studies to methyl groups in biomolecules. This extension permits detection on the 13 C nuclei and, hence, for the achievement of site-specific resolution. The measurements are demonstrated using a nine-residue low complexity peptide with the sequence GGKGMGFGL, in which a single selective − 13 CD 3 label is placed at the methionine residue. Carbon-detected measurements are compared with the deuterium direct-detection results, which allows for fine-tuning of experimental approaches. In particular, we show how the adiabatic respiration CP scheme and the double adiabatic sweep on the 2 H and 13 C channels can be combined with the 2 H R1ρ relaxation rates measurement. Off-resonance 2 H R1ρ measurements are investigated in addition to the on-resonance condition, as they extent the range of effective spin-locking field.

Abstract:Diffusion-ordered spectroscopy (DOSY) NMR is based on a pulse-field gradient spin-echo NMR experiment, in which components experience diffusion. Consequently, the signal of each component decays with different diffusion rates as the gradient strength increases, constructing a bilinear NMR data set of a mixture. By calculating the diffusion coefficient for each component, it is possible to obtain a two-dimensional NMR spectrum: one dimension is for the conventional chemical shift and the other for the diffusion coefficient. The most interesting point is that this two-dimensional NMR allows non-invasive “chromatography” to obtain the pure spectrum for each component, providing a possible alternative for LC-NMR that is more expensive and time-consuming. Potential applications of DOSY NMR include identification of the components and impurities in complex mixtures, such as body fluids, or reaction mixtures, and technical or commercial products, e.g. comprising polymers or surfactants.Data processing is the most important step to interpret DOSY NMR. Single channel methods and multivariate methods have been proposed for the data processing but all of them have difficulties when applied to real-world cases. The big challenge appears when dealing with more complex samples, e.g. components with small differences in diffusion coefficients, or severely overlapping in the chemical shift dimension. Two single channel methods, including SPLMOD and continuous diffusion coefficient (CONTIN), and two multivariate methods, called direct exponential curve resolution algorithm (DECRA) and multivariate curve resolution (MCR), are critically evaluated by simulated and real DOSY data sets. The assessments in this paper indicate the possible improvement of the DOSY data processing by applying iterative principal component analysis (IPCA) followed by MCR-alternating least square (MCR-ALS).

Abstract:This work presents an approach to exploiting Nuclear Magnetic Resonance (NMR) relaxometry data (1H spin-lattice relaxation rates covering the frequency range from below 1 kHz to 10 MHz) for the purpose of differentiating between pathological and reference tissues. Characteristic quantities (markers) that can be obtained in a straightforward manner, not resorting to an advanced analysis of 1H spin-lattice relaxation data, have been identified and compared for pathological and reference colon tissues. Moreover, the relaxation data have been parametrised in terms of Lorentzian spectral densities and the possibility of using the obtained dipolar relaxation constants and correlation times as biomarkers to assess the state of tissues has been discussed. It has also been demonstrated that the relaxation data for the reference and the pathological tissues can be attributed to two groups (for each case). The studies are a step towards exploiting the potential of NMR relaxometry for characterisation of pathological changes in tissues.
Theory of spin-lattice relaxation in single-molecule magnets: Reasons for the slowness

Abstract:While exponential and power law relaxation-time-temperature dependences are widely used to fit experimental data, these known relations in bulk materials have not been well-foundedly validated in the context of single-molecule magnets. Based on pattern of the phonon density of states and spin-phonon coupling strength, we derived a theory showing how these dependences arise and how transition among them occurs with temperature variation. The theory also resolves the puzzle of lower-than-expected Orbach barrier and multiple barriers, and elucidates the presumed Orbach process as Raman process in disguise. Except for very high temperature at which real Orbach processes are accessible, the results justify the ground state tunnelling as the dominant relaxation channel, and identify small tunnelling rate and enclosure of the magnetic atom as reasons for the slowness. Our findings issue a call for shifting our research efforts from achieving super large Orbach barriers to magnetic engineering that diminishes the transverse and high order anisotropies, and to dynamic engineering that lifts vibrational modes possessing strong coupling with the spin.
Measuring Transverse Relaxation with a Single-Beam 894 nm VCSEL for Cs-Xe NMR Gyroscope Miniaturization

Abstract:The spin-exchange-pumped nuclear magnetic resonance gyroscope (NMRG) is a pivotal tool in quantum navigation. The transverse relaxation of atoms critically impacts the NMRG's performance parameters and is essential for judging normal operation. Conventional methods for measuring transverse relaxation typically use dual beams, which involves complex optical path and frequency stabilization systems, thereby complicating miniaturization and integration. This paper proposes a method to construct a 133Cs parametric resonance magnetometer using a single-beam vertical-cavity surface-emitting laser (VCSEL) to measure the transverse relaxation of 129Xe and 131Xe. Based on this method, the volume of the gyroscope probe is significantly reduced to 50 cm3. Experimental results demonstrate that the constructed Cs-Xe NMRG can achieve a transverse relaxation time (T2) of 8.1 s under static conditions. Within the cell temperature range of 70 °C to 110 °C, T2 decreases with increasing temperature, while the signal amplitude inversely increases. The research lays the foundation for continuous measurement operations of miniaturized NMRGs.
