Abstract:A compound was isolated from Evodia rutaecarpa and identified to be Limonin. The NMR signals of the compound were assigned by 2D NMR. The results show that the assignment of some proton signals made in the previous studies was erroneous. Correction was made for the incorrect assignment.

Abstract:A new and highly oxygenated limonoids compound, evolimorutanin 2, was isolated from the unripe fruits of Evodia rutaecarpa (Juss.) Benth. var. bodinieri (Dode) Huang (fructus evodiae) as a traditional Chinese medicine together with the known limonin 1. The chemical structure was elucidated by IR, MS, ID NMR(H-1 NMR, C-13 NMR and distorsionless enhancement by polarization transfer (DEPT)) and 2D NMR (H-1-H-1 COSY, heteronuclear single quantum coherence (HSQC) and hetero nuclear multiple bond coherence (HMBC)). Both limonin and evolimorutanin belong to nortriterpenoids compounds on the chemical structure and their substituents and stereochemistry in the rings A, A', B, C, D were completely same, and their exclusive difference was the oxygenation in the ring E. In addition, the possible transformation route of evoli-morutanin from limonin via shihulimonin A is proposed.

Abstract:One new limonoid, named 19-hydroxy methyl isoobacunoate diosphenol (1); one new degraded limonoid, named 9α-methoxyl dictamdiol (9); two new quinolone alkaloids, 1-methyl-3-[(7E,9E,12Z)-7,9,12-pentadecadienyl]-4(1H)-quinolone (11) and 1-methyl-3-[(7E,9E,11E)-7,9,11-pentadecadienyl]-4(1H)-quinolone (12), along with eight known compounds, evodol (2), 7β-acetoxy-5-epilimonin (3), rutaevine (4), 6β-acetoxy-5-epilimonin (5), limonin (6), obacunone (7), clauemargine L (8), hiiranlactone E (10) were isolated from the fruits of Evodia rutaecarpa (Juss.) Benth.. Structures of the four new compounds were elucidated on the basis of extensive spectroscopic techniques, including 1D and 2D NMR techniques. Compounds 3, 5, 9, 11 and 12 showed obviously cytotoxic activity against six human tumor lines, while compounds 11, 12 displayed anti-platelet aggregation induced by ADP at 50 μM and 100 μM.

Simultaneous Quantification of Limonin, Two Indolequinazoline Alkaloids, and Four Quinolone Alkaloids in Evodia rutaecarpa (Juss.) Benth by HPLC-DAD Method.

Abstract:A simple and efficient HPLC-DAD (225 nm) method was developed and validated for the simultaneous determination of limonin and six key alkaloids (evodiamine, rutaecarpine, 1-methyl-2-undecyl-4(1H)-quinolone, evocarpine, 1-methy-2-[(6Z,9Z)]-6,9-pentadecadienyl-4-(1H)-quinolone, and dihydroevocarpine) in Evodia rutaecarpa (Juss.) Benth, which has been widely used as one of the Traditional Chinese Medicines. The chromatographic separation was carried out on a Hypersil BDS C18 column, and gradient elution was employed with a mobile phase containing acetonitrile and water. Contents of the analytes in 18 batches of samples were analyzed by ultrasonic extraction with ethanol and water mixture (80 : 20, v/v) followed by HPLC analysis. Separation of the seven analytes was achieved within 60 min with good linearity (r > 0.999). The RSD of both the intraday and interday precision was below 1.85%. The accuracy at different concentrations was within the range of 97.91 to 100.49%. Hierarchical clustering analysis was performed to differentiate and classify the samples based on the contents of the seven constituents. This study indicated that the quality control of E. rutaecarpa could be simplified to the measurement of four constituents, and that limonin, 1-methyl-2-undecyl-4(1H)-quinolone, and dihydroevocarpine should also be served as the chemical markers together with evodiamine for the quality control of Evodia rutaecarpa (Juss.) Benth.

Abstract:OBJECTIVE:To study Mass spectrometry of quinazoline alkaloid and limonin from Fructus Evodiae by an electrospray ionization (ESI) tandem mass spectrometry.METHODS:The ESI source was set at the positive ionization mode, and the MS1 and MS2 spectra of quinazoline alkaloid and limonin were acquired.RESULTS:The main fragments of quinazoline alkaloid and limonin from Fructus Evodiae were analyzed.CONCLUSIONS:The profiles of fragments will be usefull to set quality criteria and to study chemical composition of Fructus Evodiae.
Studies on Absorption and Tansport of Limoninoids from Fructus Evodiae in Caco-2 Cell Monolayer Model

Abstract:Objective To study the intestinal absorption and transepithelial transport of three limoninoids: evodol （EVO）, limonin （LIM）, and shihulimonin A （SHIA）, isolated from Fructus Evodiae [the unripe fruit of Evodia rutaecarpa and Evodia rutaecarpa var. bodinieri] in the human intestine. Methods The in vitro cultured human colon carcinoma cell line, Caco-2 cell monolayer model, was applied to studying the absorption and transepithelial transport of the three limoninoids from apical （AP） to basolateral （BL） side and from BL to AP side. The three limoninoids were measured by reversed-phase high performance liquid chromatography coupled with ultraviolet absorption detector. Transport parameters and apparent permeability coefficients （Papp） were then calculated and compared with those of Propranolol as a control substance of high permeability and Atenolol as a control substance of poor permeability. Results The Papp value of EVO and LIM from AP to BL side for absorption and transport were 1.78 × 10-5 cm/s and 1.16 × 10-5 cm/s, respectively, which was comparable to that of Propranolol with Papp 2.18 × 10-5 cm/s. Conclusion The absorption and transport of both EVO and LIM are main passive diffusion as the dominating process in Caco-2 cell monolayer model, and they were estimated to be high absorbed compounds. SHIA in Caco-2 cell monolayer model may be involved in metabolism in the transport processes.

Abstract:Limonin (LIM), a furan-containing limonoid, is one of the most abundant components of Dictamnus dasycarpus Turcz. Recent studies demonstrated that LIM has great potential for inhibiting the activity of drug-metabolizing enzymes. However, the mechanisms of LIM-induced enzyme inactivation processes remain unexplored. The main objective of this study was to identify the reactive metabolites of LIM using liquid chromatography-mass spectrometry. Three nucleophiles, glutathione (GSH), N-acetyl cysteine (NAC), and N-acetyl lysine (NAL), were used to trap the reactive metabolites of LIM in in vitro and in vivo models. Two different types of mass spectrometry, a hybrid quadrupole time-of-flight (Q-TOF) mass spectrometry and a LTQ velos Pro ion trap mass spectrometry, were employed to acquire structural information of nucleophile adducts of LIM. In total, six nucleophile adducts of LIM (M1-M6) with their isomers were identified; among them, M1 was a GSH and NAL conjugate of LIM, M2-M4 were glutathione adducts of LIM, M5 was a NAC and NAL conjugate of LIM, and M6 was a NAC adduct of LIM. Additionally, CYP3A4 was found to be the key enzyme responsible for the bioactivation of limonin. This metabolism study largely facilitates the understanding of mechanisms of limonin-induced enzyme inactivation processes.
Studies on the Chemical Constituents of Evodia Rutaecarpa and Evodia Rutaecarpa Passed with Liquorice and Biological Activities

Abstract:17 indole quinoline alkaloids, 8 limonins, 7 flavonoids, 7 triterpenoids and 11 other compounds were obtained from E. rutaecarpa and E. rutaecarpa passed with Liquorice. 6 of them are new compounds. About 19 compounds were tested by replanting tumor to mice, angioma of zebrafish model for antiangiogenic drug screening and human tumor cells in vitro. Thus compounds evodiagenine (2), evodialimonin (3), rhetsinine (4), dehydroevodiamine (5), wuchuyuamide I (7) and 1-methyl-2-undecyl-4(1H)-quinolone (8) showed moderate activity against human tumor cells, dievodiamine (1) and evodiamine (6) showed good activity. The inhibitory rate of 4 on sarcoma S180 was 57.78% at the dose of 5–10mg/kg·d in vivo. Moreover, the inhibition of the active selectivity of cycloxygenase 1 and 4 showed a stronger effect.
Isolation and Purification of Limonin,Evodiamine and Rutaecarpine from Evodia Rutaecarpa(Juss.) Benth by High-Speed Counter-Current Chromatography

Abstract:This study aimed to develop a preparative high-speed counter-current chromatography(HSCCC) for the isolation of alkalogenic constituents from Evodia rutaecarpa(Juss.) Benth.A two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water(5∶5∶5∶5) was used at a flow rate of 2.0mL·min-1,while the apparatus rotated at 855 rpm.And the effluent was detected at 254 nm.The obtained fractions were analyzed by high-performance liquid chromatography(HPLC).All 6.2 mg of limonin with the purity of 96.2%,10.4mg of evodiamine with the purity of 98.1%,and 10.5 mg of rutaecarpine with the purity of 95.4%,were obtained from 400mg of crude extract in a one-step elution within 5h.The results indicated that HSCCC is a powerful technique for the purification of natural products.

Abstract:Four new limonoids, tociliatonoid B-E (1–4) were obtained from the EtOAc extract of the twigs and leaves of Toona ciliata var. yunnanensis. Their structures were elucidated through comprehensive spectroscopic and X-ray crystallographic analysis. Compounds 1–4 are rare limonoids which feature unusual 3-substituted 1,5-dihydro-2H-pyrrol-2-one or maleimide ring moiety at C-17 and Compound 1 possess an aldehyde group at C-4.

Abstract:Limonoids are bioactive plant specialized metabolites found in the Meliaceae family. The basic limonoids, i.e., azadiradione, epoxyazadiradione, and gedunin have been exploited for various bioactivities and therefore are the potential drug leads for tomorrow. However, their low abundance, structural similarity, and lack of adequate mass fragmentation data have hampered their accurate identification and quantification from various sources. In the present study, basic limonoids such as azadirone, azadiradione, epoxyazadiradione, and gedunin isolated from Neem were utilized for the synthesis of their derivatives and isotopologs. A total of 30 one compounds were used in this study among which five were isolated, two were biotransformed, and 24 were synthesized. Among the synthesized compounds nine are novel compounds including six deuterated analogs/isotopologs which are (1,3-2H)-1,2-dihydro-3β-hydroxyazadiradione (9), (1,3,16-2H)-1,2-dihydro-3β-16β-dihydroxyazadiradione (10), 3β-hydroxyazadiradione (11), 3β-16β-dihydroxyazadiradione (12), (3-2H)-3β-hydroxyazadiradione (13), (3,16-2H)-3β-16β-dihydroxyazadiradione (14), (1,3,7-2H)-1,2-dihydro-3β-hydroxy-7-deacetylazadiradione (15), 1,2,20,21,22,23-hexahydroazadiradione (17), and (1,3-2H)-1,2-dihydro-3β-hydroxygedunin (29). These limonoids along with their semisynthesized derivatives were subjected to ultra high performance liquid chromatography mass spectrometry (UHPLC-MS/MS) and the fragmentation pathway was established based on structure-fragment relationships (SFRs), utilizing high resolution MS/MS data. We have developed a most reliable and easily reproducible protocol describing in depth analysis of SFRs based on the structural modifications and synthesis of isotopologs. Also, the MS/MS fragment library of these basic limonoids generated in this study acts as a fingerprint for accurate identification and quantification of limonoids by MS/MS analysis in various plant tissue extracts, phytopharmaceutical formulations and biological samples.

Abstract:Salviae Miltiorrhizae Radix et Rhizoma (Danshen in China) and its related preparations are widely used in clinical practice due to its high medicinal value. In recent years, H-1 NMR technology has made great progress and demonstrated its unique advantages in the field of botanical metabolomics. In this study, H-1 NMR-based metabolomics was used to investigate the dissolution of various metabolites in Danshen as a function of ethanol concentration. H-1 NMR spectroscopy of Danshen extract identified 28 metabolites including 6 sugars, 11 amino acids, 3 organic acids, 4 salvianolic acids, and 4 tanshinones. Multivariate statistical analysis was used to classify and compare various Danshen extracts. PCA and HCA were used to obtain a global overview of the similarity in the samples and two-class OPLS-DA models were established for identifying characteristic metabolites. Then, H-1-qNMR method was used to estimate the concentration of 22 metabolites, which is helpful to further describe the changes in metabolite ratios of various Danshen extracts. The result of this study laid the foundation for further biological activity research, and also provided an important reference for subsequent process research and quality control of Danshen related preparations.

Abstract:This study reports simultaneous isolation of three new limonoids (1-3), six known regio isomers (6, 7, 9-12), and three more known limonoids (4, 5, 8) from Swietenia macrophylla (S. macrophylla) seeds. Structures of these compounds were determined via extensive study of their 1D/2D-NMR and mass spectral data. Known limonoids (4-12) were identified by comparing their physical and spectroscopic data with literature values. A novel environmentally friendly supercritical fluid chromatography (SFC) technique facilitated simultaneous and rapid separation of these compounds. The pharmacological activities of the new limonoids were investigated.

Abstract:Limonin is a natural tetracyclic triterpenoid compound, which widely exists in Euodia rutaecarpa (Juss.) Benth., Phellodendron chinense Schneid., and Coptis chinensis Franch. Its extensive pharmacological effects have attracted considerable attention in recent years. However, there is no systematic review focusing on the pharmacology, toxicity, and pharmacokinetics of limonin. Therefore, this review aimed to provide the latest information on the pharmacology, toxicity, and pharmacokinetics of limonin, exploring the therapeutic potential of this compound and looking for ways to improve efficacy and bioavailability. Limonin has a wide spectrum of pharmacological effects, including anti-cancer, anti-inflammatory and analgesic, anti-bacterial and anti-virus, anti-oxidation, liver protection properties. However, limonin has also been shown to lead to hepatotoxicity, renal toxicity, and genetic damage. Moreover, limonin also has complex impacts on hepatic metabolic enzyme. Pharmacokinetic studies have demonstrated that limonin has poor bioavailability, and the reduction, hydrolysis, and methylation are the main metabolic pathways of limonin. We also found that the position and group of the substituents of limonin are key in affecting pharmacological activity and bioavailability. However, some issues still exist, such as the mechanism of antioxidant activity of limonin not being clear. In addition, there are few studies on the toxicity mechanism of limonin, and the effects of limonin concentration on pharmacological effects and toxicity are not clear, and no researchers have reported any ways in which to reduce the toxicity of limonin. Therefore, future research directions include the mechanism of antioxidant activity of limonin, how the concentration of limonin affects pharmacological effects and toxicity, finding ways to reduce the toxicity of limonin, and structural modification of limonin—one of the key methods necessary to enhance pharmacological activity and bioavailability.

Abstract:INTRODUCTION Melia toosendan Sieb. et Zucc. has been used as a Chinese folk medicine for roundworm treatment since ancient times. Many diverse limonoids have been isolated from Meliaceae plants, but it remains difficult to isolate and identify other limonoids because of their small natural concentrations. OBJECTIVE This study was performed to overcome the difficulties associated with fast and accurate identification of limonoids and establish a reliable and sensitive method for the analysis of minor limonoids in M. toosendan fruits. METHODS An efficient strategy for enrichment, detection, and identification of minor limonoids from M. toosendan fruits using solid-phase extraction with high-performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (SPE-HPLC-Q-TOF-MS/MS) was developed herein. RESULTS Characteristic fragmentations and fragmentation ions containing trichilin-, nimbin-, and vilasinin-class limonoid skeletons were initially studied, and characteristic diagnostic ions involved retro Diels-Alder (RDA) reactions or homolytic cleavages, which were used to identify minor limonoids. In total, 13 limonoids, including four new ones, were identified. CONCLUSION This is the first report on the analysis of M. toosendan fruits to identify limonoids. This novel analysis method may stimulate further research regarding the identification of limonoids in other plant species.
[Flavonoid Glycosides from Dried and Nearly Ripe Fruits of Evodia Rutaecarpa].

Abstract:OBJECTIVE:To study the chemical constituents from the dried and nearly ripe fruits of Evodia (Euodia) rutaecarpa.METHOD:The compounds were separated and purified by solvent and chromatographic methods. Their structures were identified by spectroscopic techniques.RESULT:Fifteen compounds were separated from the normal butanol extracts of the 70% aqueous ethanol extract of the dried and nearly ripe fruits of E. rutaecarpa. Among of them, four compounds were reported in the essay and identified as diosmetin-7-O-beta-D-glucopyranoside (1), isorhamnetin-3-O-rutinoside (2), diosmin (3) and chrysoeriol-7-O-rutinoside (4).CONCLUSION:Compounds 1, 3 and 4 were separated from the dried and nearly ripe fruits of E. rutaecarpa for the first time.

Abstract:Leonurine, a major bioactive component from Herba Leonuri, shows therapeutic potential for cardiovascular disease and stroke prevention in some preclinical experiments. The aim of this study is to characterize metabolites of leonurine in rats using high performance liquid chromatography coupled with tandem mass spectrometry (HPLC/MS/MS). The chromatographic separation was performed on an Agilent ZORBAX SB-C18 column using a gradient elution with acetonitrile/ammonium acetate buffer (10 mM, pH 4.0) solvent system. An information dependent acquisition (IDA) method was developed for screening and identifying metabolites of leonurine under positive ion mode. Compared with control, the interesting compound in the extracted ion chromatogram (XIC) of the in vivo samples was chosen and further identified by analyzing their retention times, changes in observed mass (Δm/z), and spectral patterns of product ion utilizing advanced software tool. For the first time, a total of three metabolites were identified, including two phase II metabolites generated by glucuronidation (M1) and sulfation (M2) and one phase I metabolite formed by O-demethylation (M3). Finally, the lead metabolite M1 was isolated from urine and its structure was characterized as leonurine-10-O- β-D-glucuronide by NMR spectroscopy (¹H, ¹³C, HMBC, and HSQC).

Abstract:Objective To isolate and elucidate the constituents of Evodia rutaecarpa.Methods Various chromatographic techniques,phytochemistry and the spectra methods were used to identify the structures of these compounds from Evodia rutaecarpa.Results Six compounds are elucidated as evodiamine(Ⅰ),dehydroevodiamine(Ⅱ),limonin(Ⅲ),p-hydroxy-benzoic acid ethyl ester(Ⅳ),berberine(Ⅴ),oleanolic acid(Ⅵ).Conclusion Among them,compounds Ⅳ,Ⅴ,Ⅵ are isolated from this genus for the first time.

Abstract:Eight vilasinin-class limonoids, including the unusually chlorinated rubescins K–M (1–3), the 2,3-epoxylated rubescin N (4), and rubescins O–R (5–8), were newly isolated from Trichilia rubescens. The structures of the isolated compounds were determined through spectroscopic and spectrometric analyses, as well as ECD calculations. The natural occurrence of chlorinated limonoids 1–3 was confirmed by chemical methods and HPLC analysis of a roughly fractionated portion of the plant extract. Eight selected limonoids, including previously known and new compounds, were evaluated for antiproliferative activity against five human tumor cell lines. All tested limonoids, except 8, exhibited significant potency, with IC50 values of <10 μM; in particular, limonoid 14 strongly inhibited tumor cell growth, with IC50 values of 0.54–2.06 μM against all tumor cell lines, including multi-drug-resistant cells.
