An amphiphilic fluorescent probe-based method for detection of dental biofilm in vitro

Abstract:It is valuable to develop a new method to detect to dental biofilm since it is closely related to the oral disease. In this paper, a new detection method of dental biofilm in vitro based on an amphiphilic fluorescent probe (TR4) was developed. This TR4 consists of tetraphenylethylene ( TPE) , palmitic acid and polypeptide R4 corresponding to the fluorescence emitting, particle assembly and electrostatic adsorption. The whole test process can be completed within 1 min through simple mixing of TR4 and the sample and the TR4 fluorescent probe provides a rapid and convenient detection of dental biofilm in vitro which can help preventing and controlling oral disease. The fluorescence intensity of TR4 molecules was significantly enhanced after adhering with dental biofilm, which suggested that the TR4 molecules, as a potential fluorescent probe, can be employed to detect dental biofilm. By means of transmission electron microscope, fluorescence spectrometer and confocal laser scanning microscopy techniques, we found that TR4 molecules can assemble into nanoparticles after adhering to dental biofilm surface and emitting fluorescence due to the AIE behaviors of tetraphenylethylene ( TPE) molecules.
