Abstract:Abstract A reversed-phase high performance liquid chromatography (RP-HPLC) method was established for the determination of isoflavones (daidzein, glycitein, equol, and genistein) in human urine. After the addition of flavone as internal standard and the enzymatic hydrolysis in urine sample, the isoflavones were extracted with ethyl acetate (1:1) in medium of pH 7.0 and then separated by gradient elution with a ternary eluant of 0.02% trifluoroacetic acid-methanol-acetonitrile. The detection limits were 12.9 nM, 13.9 nM, 71.6 nM, and 11.8 nM for daidzein, glycitein, equol, and genistein, respectively. The recovery of the isoflavones was more than 89%. The proposed method is easier to carry out, accurate, reproducible, and suitable for the analysis of a large number of samples.
THE SIMULTANEOUS ANALYSIS OF SIX ISOFLAVONE METABOLITES IN HUMAN URINE BY HPLC

Abstract:Objective To establish a high performance liquid chromatographic(HPLC) method for simul-taneous analysis of 6 compounds including isoflavone metabolites of equol etc.in human urines.Method The samples were hydrolyzed with β-glucuronidase enzyme solution,and extracted with ethylacetate.The analytes were separated using ethylacetate/methanol/phosphate system as mobile phase with a linear gradient elution program.Wave length was set at 254,280nm by UV detection.Results Linear range was 0.05～10 μg/ml.The limits of detection were 0.03～0.05μg/ml;and the limits of quantification were 0.1～0.17μg/ml.The mean recoveries for each analyte ranged from 71.4% to 102.1% with relative standard deviation below 7.9%.The method was applied to the analysis of ten human urine samples.Conclusion The method is accurate,and can be used for evaluation of isoflavone metabolites in human urine.
Determination of Oxalate,phosphate and Citrate in Human Urine by Ion Chromatography with Suppressor Conductivity Detection

Abstract:Objective:To develop a simple method for the determination of oxalate,phosphate,citrate in human urine by ion chromatography with gradient elution and conductivity detection.Methods:Fresh collective urine samples were centrifuged for 15 min at 5000 r·min-1 and then the above solution was filtered through a 0.2 μm syringe membrane filter,On-Guard H column and C 18 cleanup column before injection.The chromatographic separation was performed on Dionex IonPac AG23(50 mm×4 mm) guard column,and an Dionex IonPac AS23(250 mm×4 mm) analytical column using potassium hydroxide as eluent with a self-regeneration suppressor.Results:This novel method exhibited excellent regression(r≥0.9998)in the range of 0.5-50 μg·mL-1,satisfactory precision(RSD less than 3.6% within one day and less than 6.9%day to day).The recovery obtained in the accuracy test was in the range of 96.0%-100.2%.Conclusion:The method is simple,accurate,stable and precise.It can be easily and conveniently adopted for the routine quality control analysis.
Development and validation of an HPLC-MS/MS method for the simultaneous analysis of volatile organic compound metabolites, hydroxylated polycyclic aromatic hydrocarbons, and 8-hydroxy-2'-deoxyguanosine in human urine

Abstract:Humans are widely and concurrently exposed to volatile organic compounds (VOCs) and polycyclic aromatic hydrocarbons (PAHs). However, few studies have reported the internal co-exposure levels of these chemicals in occupational and general populations. Specifically, the associations revealed between the urinary levels of metabolites of VOCs (mVOCs), hydroxylated PAHs (OH-PAHs), and oxidative stress biomarkers for humans remain limited. In this study, a method based on solid-phase extraction (SPE) and liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) was developed for the simultaneous analysis of 22 mVOCs, 12 OH-PAHs, and 8-hydroxy-2'-deoxyguanosine (8-OHdG) in human urine samples. The method was validated with all target analyte accuracies and precisions in the range of 76 %-120 % and 1 %-14 % at three levels of spiked urine samples, respectively. The limit of detection (LOD) and limit of quantification (LOQ) of the target analytes were 0.01-0.34 ng/mL and 0.01-7.57 ng/mL, respectively. And the method was applied to measure urinary levels of target analytes from 38 petrochemical workers in Guangzhou, South China. Except for 3-hydroxy-benzo[a]pyrene, all target analytes were detected in the urine samples. The average levels were 0.05-12.6 ng/mL for individual OH-PAHs, 0.20-73620 ng/mL for individual mVOCs, and 1.00 ng/mL for 8-OHdG. Additionally, 3-hydroxy-phenanthrene, 1-hydroxy-pyrene, 6-hydroxy-chrysene, N-acetyl-S-(trichlorovinyl)-L-cysteine, 2-methylhippuric acid, thiodiacetic acid, trans, trans-Muconic acid, and N-acetyl-S-(3,4-dihydroxybutyl)-L-cysteine had statistically significant positive effects on 8-OHdG levels, while 1-hydroxy-naphthalene, 1,2-dihydroxybenzene, and hippuric acid showed a negative effect on 8-OHdG, indicating these metabolites could lead to synergistic or antagonistic oxidative DNA damage. This study provides a robust analytical method that permits a comprehensive assessment of co-exposure to PAHs and VOCs and their potential adverse health effects.

Abstract:This work described the utilization of ion-pair reversed phase liquid chromatography coupled to inductively coupled plasma mass spectrometry (RP-LC-ICP-MS) for iodine speciation analysis in urine. Considering the requirements of green analytical chemistry and the Ar ICP-MS instrument, three aqueous mobile phases were employed for the separation of seven iodine species including iodide, iodate and five iodo amino acids (monoiodotyrosine - MIT, di-iodotyrosine - DIT, tri-iodothyronine - T-3, reversed tri-iodothyronine - rT(3), and thyroxine - T-4). The aqueous mobile phases were composed of an ion-pair reagent (tetrabutylammonium hydroxide - TBAH) and an eluent (ammonium chloride, L-phenylalanine or deoxycholic acid) at low concentrations in ultrapure water. Owing to tremendous difference in retention behavior between these iodinated forms, a gradient elution mode was performed for the rapid separation of IO3- and I-, MIT and DIT, and T-3, rT(3) and T-4, respectively. Iodine species separation was achieved with a 12.5 mm C-18 guard column in 7 min. The detection limits for IO3-, I-, MIT, DIT, T-3, rT(3) and T-4 were 0.047, 0.046, 0.057, 0.072, 0.093, 0.094 and 0.081 mu g L-1, respectively. Application of the proposed method was demonstrated by the speciation analysis of iodine in four real urine samples. The developed method offered satisfactory recoveries in the 93-106% range and good repeatability, showing great potential in routine analysis of iodine speciation in environmental, food and biological samples.

Abstract:The determination of α-ketoacid concentration is demanded to evaluate the absorption and metabolic behavior of compound α-ketoacid tablets taken by chronic kidney disease patients. To eliminate the interference of endogenous substance of urine and enrich the analytes, a three-phase hollow-fiber liquid-phase microextraction combined with ion-pair high-performance liquid chromatography method was established for the determination of d,l-α-hydroxymethionine calcium, d,l-α-ketoisoleucine calcium, α-ketovaline calcium, α-ketoleucine calcium, and α-ketophenylalanine calcium of compound α-ketoacid tablets in human urine samples. The extraction parameters, such as organic solvent, pH of donor phase and acceptor phase, stirring rate, and extraction time were optimized. Under the optimal conditions, the obtained enrichment factors were up to 11-, 110-, 198-, 202-, and 50-fold, respectively. The calibration curves for these analytes were linear over the range of 0.1-10 mg/L for α-ketovaline calcium, d,l-α-ketoisoleucine calcium, and α-ketoleucine calcium, 0.5-10 mg/L for d,l-α-hydroxymethionine calcium, and α-ketophenylalanine calcium with r > 0.99. The relative standard deviations (n = 5) were less than 6.27% and the LODs were 100.7, 10.0, 5.8, 7.8, and 8.6 μg/L (based on S/N = 3), respectively. Good recoveries from spiked urine samples (92-118%) were obtained. The proposed method demonstrated excellent sample clean-up and analytes enrichment to determine the five components in human urine.

Abstract:A syringe-dispersive solid-phase extraction method was developed for the determination of seven nitroimidazoles and nine steroids in manure-based fertilizers by ultra-high performance liquid chromatography with tandem mass spectrometry. Methanol and acetonitrile were used to extract the sample, and mixed dispersive sorbents dispersed in the syringe were used for purification. The extract was separated with an HSS-T3 column and detected in positive or negative multiple reaction monitoring mode. Under the optimal conditions, the recoveries of the 16 compounds ranged from 70.3 to 112.3% at the four spiked levels (3, 10, 20, and 50 mu g/kg) and the relative standard deviations ranged from 1.0 to 12.4%. The limits of detection and quantification were 0.22-0.86 and 0.73-2.87 mu g/kg, respectively. This method is simple, fast, and reliable, and can be used to simultaneously screen and determine nitroimidazoles and steroids in manure-based fertilizers.

Abstract:Liquid-liquid extraction and solid phase extraction followed by high-performance liquid chromatography (HPLC) connected with ultraviolet (UV) detection were used for the determination of phytohormones. The parameters influencing the performance of the HPLC-UV method, including composition of the mobile phase for gradient elution, column temperature, flow rate, and detection wavelength, were optimized. This method can simultaneously determine 11 phytohormones, including nine cytokinins, indole-3-acetic acid, and abscisic acid. The limit of detection of this method is 0.22 to 1.1 μg L−1, and the coefficient factors of linear regression are >0.998. The recoveries of the target phytohormones ranged between 62.1~109.4%, and the relative standard deviations were <10%. This method is suitable for determining phytohormones, especially cytokinins, in young panicles, roots, and xylem sap of rice plants.

Method Validation for Determination of 5-Hydroxyindoleacetic Acid in Human Urine with Ultra-performance Liquid Chromatography-tandem Mass Spectrometry

Abstract:Objective: An efficient analytical method for the quantitative determination of 5-hydroxyindoleacetic acid (5-HIAA) in human urine by ultra-performance liquid chromatography (UPLC) -tandem mass spectrometry (MS/MS) was developed. Methods: The artificial human urine UR-N-CONTROL LEVEL 2 NP was used for calibration curve preparation, artificial urine and natural urine were used for quality control samples preparation. 5-Hydroxyindoleacetic acid-d5 (5-HIAA-d5) was used as internal standard, urine samples were diluted with acetonitrile. The supernatants were detected by UPLC-MS/MS. Separation was carried out on a Waters HSS T3 analytical column (100×2.1 mm, 1.8 μm). Mobile phase consisting A: 2 m M ammonium acetate (containing 0.02 ％ acetic acid) and B: acetonitrile (containing 0.02 ％ acetic acid). Gradient elution using the proportion of mobile phase B ranged from 5 ％ to 100 ％ in 2.5 min at a flow rate of 0.5 m L/min. The column temperature was 50 ℃ and the injection volume was 3.0 μL. The electrospray source and negative ion scan were used. 5-HIAA and the internal standard (5-HIAA-d5) were monitored at m/z 190.1 146.2 and m/z 195.1 151.0, respectively. Results: The linear calibration curve was observed in the concentration range from 0.0500 to 50.0 μg/m L, with the r ≥ 0.9948.The lower limit of quantification was 0.0500 μg/m L. The accuracy (intra-assay and inter-assay) for 5-HIAA was between 89.3 ％ and99.8 ％, with the precision (CV) ≤ 8.4 ％. The mean recoveries ranged from 100.3 ％ to 102.3 ％. Conclusions: The method is proved to be highly selective, sensitive and rapid. Artificial urine and natural urine are no differences in quantitative analysis. They are suitable for determination of 5-HIAA in human urine.

Abstract:OBJECTIVE To establish a method of simultaneously determining the contents of oxalate and citrate in human urine by reversed phase high performance liquid chromatography.METHODS To observe the nomal value of oxalate and citrate in human urine,they were determined by Agilent SB C18(250mm×4.6mm,5μm)column and Agilent zorbax extend-C18(4.6mm×12.5mm,5μm)guard column;an aqueous solution containing 0.018mol·L-1potassium dihydrogenphosphate,0.010mol·L-1tetrabutylammonium hydrogensulphate and 2.69×10-5mol/L EDTA at pH 2.4～2.5 was used as mobile phase.Flow rate was 1.5mL·min-1 at 25℃.Injection volume was 20μL.All separation was at UV 210nm.Deproteinization of urine samples was carried out by well mixing 0.5 mL urine sample with 25 mg crystalline sulfosalicylic acid at 4℃ for 30 min. RESULTS The detection limit of oxalate and citrate was 0.4μg·mL1 and 0.8μg·mL-1,respectively.The linear range of oxalate was 1.563～100.00μg·mL-1,its average recovery was 97.66% and RSD of intra-day and inter-day were less than 13.50%.The linear range of citrate was 3.125～200.00μg·mL-1,its average recovery was 97.71% and RSD of intra-day and inter-day were less than 8.25%.CONCLUSION The method is simple,sensitive and reproducible for quantifying oxalate and citrate in human urine and the research of formation of uric calcium oxalate stone.
Target-based metabolomics for fast and sensitive quantification of eight small molecules in human urine using HPLC-DAD and chemometrics tools resolving of highly overlapping peaks

Abstract:Chemometrics multivariate calibration coupled with high performance liquid chromatography-diode array detection (HPLC-DAD) analytical strategy was applied for fast and sensitive quantification of the eight small molecules (uric acid, creatinine, tyrosine, homovanillic acid, hippuric acid, indole-3-acetic acid, tryptophan and 2-methylhippuric acid) in human urine. The objective of this work was to get the successful resolution of the complex matrix with minimum experimental time in the presence of highly overlapping peaks, of distortions in the time and baseline aspects among chromatograms, and of the presence of unknown and background interferences. All the analysis were based on a short C18 column with the chromatographic system operating in isocratic mode and all analytes can be successfully quantified within 6 min. The second-order HPLC-DAD data acquired were handled intelligently by two typical chemometrics tools including alternating trilinear decomposition (ATLD) and multivariate curve resolution-alternating least squares (MCR-ALS). Reasonable resolution and satisfactory quantification results were obtained regardless of the complex matrix interferences from the urine samples and the second-order advantage was fully exploited. With the validation by classic HPLC method, the proposed strategy could take extra advantages such as increased selectivity and sensitivity, shorter analysis time, undemanding elution conditions and sufficiency of lower limit of quantification benefit from multivariate calibration. The method was shown as a promising means for fast and sensitive determination of small molecules in human urine and also for fast diagnosis or surveillance in related diseases.
Simple Determination of Betaine, L-carnitine and Choline in Human Urine Using Self-Packed Column and Column-Switching Ion Chromatography with Nonsuppressed Conductivity Detection

Abstract:A sequential online extraction, clean-up and separation system for the determination of betaine, L-carnitine and choline in human urine using column-switching ion chromatography with nonsuppressed conductivity detection was developed in this work. A self-packed pretreatment column (50 x 4.6mm, i.d.) was used for the extraction and clean-up of betaine, l-carnitine and choline. The separation was achieved using self-packed cationic exchange column (150 x 4.6 mm, i.d.), followed by nonsuppressed conductivity detection. Under optimized experimental conditions, the developed method presented good analytical performance, with excellent linearity in the range of 0.60-100 mu g mL(-1) for betaine, 0.75-100 mu g mL(-1) for l-carnitine and 0.50-100 mu g mL(-1) for choline, with all correlation coefficients (R-2) >0.99 in urine. The limits of detection were 0.15 mu g mL(-1) for betaine, 0.20 mu g mL(-1) for l-carnitine and 0.09 mu g mL(-1) for choline. The intra- and inter-day accuracy and precision for all quality controls were within +/- 10.32 and +/- 9.05%, respectively. Satisfactory recovery was observed between 92.8 and 102.0%. The validated method was successfully applied to the detection of urinary samples from 10 healthy people. The values detected in human urine using the proposed method showed good agreement with the measurement reported previously.

Abstract:Objective:To quickly determine 15 endogenous carbohydrates in human urine by hydrophilic interaction liquid chromatography coupled with tandem mass spectrometry.Methods:Chromatography conditions,mass spectrometry conditions and sample preparation method were all optimized.The methanol-acetonitrile (50 ∶ 50,v/v) mixture was used to effectively remove the protein and impurities in urine,with the ratio of 5 times volume of organic solvents to 1 volume of urine sample.The chromatographic separation for endogenous carbohydrates in urine was accomplished by using an ACQUITY UPLC BEH Amide column (2.1 mm × 100 mm,1.7 μm).Mobile phase A was composed of 10 mmol· L-1 ammonium acetate in water-acetonitrile (95 ∶ 5,v/v),and mobile phase B consisted of 10 mmol · L-1 ammonium acetate in acetonitrile-water (95 ∶ 5,v/v),and the gradient elution mode was applied to chromatographic separation.The compounds of interest were detected by MS/MS in negative ion multiple reaction monitoring (MRM) mode.Results:Method validation including linearity,precision,matrix effects,recovery and stability was performed.The calibration curves showed a good linearity (correlation coefficient of r＞0.99) with the LOD in the range of 0.075-30 nmol· L-1 for most of compounds.The intra-and inter-day precision (RSD)were befow 15％,ranging from 2.5％-7.6％ and 3.3％-7.3％,respectively.The recovery was in the range of 85.5％-115％.The matrix effect of most chemicals was in the range of 80％-120％.The stability data in the range of 93.1％-106％ suggesting that post-preparative samples stored at 4 ℃ for 72 h was stable under routine laboratory analysis.Finally,concentrations of fifteen carbohydrates in human urine(mean ± SE):melibiose (2 668.1± 134.4)nmol · L-1;sucrose(5 070.6± 277.6)nmol · L-1;maltose (15 214.5 ± 442.5)nmol· L-1;maltotriose (849.7 ± 42.6) nmol · L-1;glucose (173 965.0 ± 4 918.9) nmol · L-1;fructose (22 172.5 ± 1 463.6)nmol · L-1;raffinose (292.1 ± 17.6) nmol · L-1;inosine (2 083.5 ± 93.4) nmol · L-1;adenosine (2 645.5 ±78.2) nmol · L-1;mannitol (83 925.0 ± 3 938.6) nmol · L-1;erythritol (375 805.0 ± 12 094.6) nmol · L-1;arabitol (192 921.1 ± 5 689.2) nmol · L-1;maltotetraose,xylose and ribose were below the limit of quantitation (60,600 and 3000 nmol· L-1,respectively).Conclusion:LC-MS/MS method for determination of 15 endogenous carbohydrates in human urine was developed and validated.The method was applied to the analysis of normal human urine,and could be used in the validation of biomarker and metabolic mechanism research in the future.
Doping Control Analysis of Small Peptides in Human Urine Using LC-HRMS with Parallel Reaction Monitoring Mode: Screening and Confirmation.

Abstract:This study described a reliable analytical method, which combines solid-phase extraction (SPE) with liquid chromatography-high resolution mass spectrometry (LC-HRMS) employing the parallel reaction monitoring (PRM) mode, for screening 41 small peptides and 3 non-peptide growth hormone secretagogues in human urine. Additionally 36 small peptides and 3 non-peptide growth hormone secretagogues were also confirmed in the same way. For the whole screening procedure, the PRM mode was applied to the HRMS detection of small peptides, which reduces the background noise from matrix compounds to a large extent and thus improves the selectivity and reliability of the peptide analytes. Meanwhile, competent chromatographic separation was achieved within a total runtime of 14 minutes, indicating an improvement in the detection efficiency. Moreover, the PRM mode could also be applied to the confirmation procedure due to its strong identification power with a low risk of generating false positives or negatives and good selectivity. Validation was performed according to the relevant World Anti-Doping Agency (WADA) criteria, including selectivity and reliability, limit of detection (LOD), limit of identification (LOI), recovery, extraction stability and carryover. The LODs of the peptide analytes ranged between 0.20 ng mL-1 and 0.92 ng mL-1 in urine, while their LOIs ranged between 0.20 ng mL-1 and 2.00 ng mL-1, which met the corresponding Minimum Required Performance Levels (MRPLs) as defined by WADA. The developed method furnished the rapid and sensitive detection of small peptides in urine for more than 5000 samples with no false-positive or false-negative, indicating that it is an eligible method for doping control analysis.

Abstract:A simple method for the determination of betaine, L-carnitine, and choline in human urine was developed based on column-switching ion chromatography coupled with nonsuppressed conductivity detection by using a self-packed column. A pretreatment column (50 mm x 4.6 mm, id) packed with poly(glycidyl methacrylate-divinylbenzene) microspheres was used for the extraction and cleanup of analytes. Chromatographic separation was achieved within 10 min on a cationic exchange column (150 mm x 4.6 mm, id) using maleic anhydride modified poly(glycidyl methacrylate-divinylbenzene) as the particles for packing. The detection was performed by ion chromatography with nonsuppressed conductivity detection. Parameters including column-switching time, eluent type, flow rates of eluent, and interfering effects were optimized. Linearity (r(2) >= 0.99) was obtained for the concentration range of 0.50-100, 0.75-100, and 0.25-100 mu g/mL for betaine, L-carnitine, and choline, respectively. Detection limits were 0.12, 0.20, and 0.05 mu g/mL for betaine, L-carnitine, and choline, respectively. The intra-and interday accuracy and precision for all quality controls were within +/- 10.11%. Satisfactory recovery was observed between 92.5 and 105.0%. The validated method was successfully applied for the determination of betaine, L-carnitine, and choline in urine samples from healthy people.
Accurate Quantification of EVT201 and Its Two Metabolites in Human Urine Using UHPLC-MS/MS Method with Solid Phase Extraction

Abstract:A sensitive and accurate ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was developed and validated for the determination of EVT201 and its two metabolites, Ro46-1927 and Ro18-5528, in human urine. Different sample preparation methods were compared, and solid-phase extraction (SPE) was finally employed. Separation conditions and mass spectrometry parameters were optimized to achieve complete separation and enough sensitivity. Finally, the three analytes were separated on an Acquity BEH C18 column (2.1 mm × 50 mm, 1.7 μm) with a gradient mobile phase. The gradient elution consisted of acetonitrile (containing 0.1% formic acid) and 10 mM ammonium formate (containing 1% acetonitrile and 0.1% formic acid) at a flow rate of 0.50 mL/min. Detection was performed on an electrospray ionization (ESI) source in positive mode with multiple reaction monitoring (MRM). The method was fully validated in accordance with the bioanalysis guidance in Chinese Pharmacopoeia. It showed satisfying linearity, accuracy, and precision in the range of 0.2–200 ng/mL for all the three analytes. The mean extraction recoveries were 85.2%, 65.6%, 87.9%, and 86.4% for EVT201, Ro46-1927, Ro18-5528, and the IS, respectively. The method was successfully applied to the bioanalysis of 833 urine samples to determine the concentration of EVT201 and its two metabolites simultaneously.
Optimization of Analysis Method of Kudzu Root Crude Extract by High Performance Liquid Chromatography

Abstract:Kudzu root is an important traditional Chinese herb. Its crude extract has been used in the treatment of hypertension and angina pectoris in China. Several parameters for high performance liquid chromatography (HPLC) analytical method for kudzu root extract, including HPLC model, mobile phase, additive and gradient conditions have been optimized. The HPLC retention parameters a and c, and the peak shape parameters sigma and tau of 25 chromatographic peaks were obtained accurately and rapidly using five linear gradients and were calculated using CSASS software. The CSASS software was then used to simulate the gradient conditions in the experiments, and the optimized condition was obtained. The comparison of the simulated and real chromatogram profiles showed that the potential of simulation using the CSSASS software as compared to that of the real experimental conditions was precise. Reversed-phase HPLC model and the mobile phase of (A) acetonitrile containing 0.1% (v/v) acetic acid and (B) water containing 0.1% (v/v) acetic acid were used. The optimized gradient profile showed a linear increase as follows: from 5% A to 35% A at 0 - 33 min; from 35% A to 100% A at 33 - 50 min, and then the HPLC system was held for 5 min. The reproducibility and precision of the method were investigated. The relative standard deviations (RSDs) of retention time, peak height, and peak area were less than 0.25%, 11%, and 4.9%, respectively, which showed that the method was stable, reliable, and reproducible.

Abstract:This study developed a new method for the analysis of 20 phenolic compounds in human urine. The urine samples were prepared by hydrochloric acid (HCl) hydrolysis, liquid–liquid extraction (LLE), and solid-phase extraction (SPE) cleanup. We found that HCl hydrolysis is of similar effectiveness to, and much cheaper than, the traditional enzymatic method. Vanillic acid was co-eluted with butyl paraben and interfered with the determination of butyl paraben in urine. K 2 CO 3 -treated-silica-gel SPE was designed to efficiently eliminate interference from the endogenous organic acids (especially vanillic acid) in urine. After derivatization, the samples were analyzed by large-volume-injection gas chromatography–tandem mass spectrometry (LVI-GC–MS–MS). Good linearity ( R 2 ≥ 0.996) was established in the range 0.1–100 ng mL −1 for all analytes. Method detection limits (MDLs) were 0.7–9.8 pg mL −1 . Intraday ( n = 5) and interday ( n = 5 days) validation was performed, with satisfactory accuracy (recovery: 70–126 % and 73–107 %, respectively) and precision (RSD ≤ 19 %) at two levels (low: 0.1 and 0.5 ng mL −1 ; high: 5 and 10 ng mL −1 ). The method was used in a population study and achieved more than 85 % detection for most analytes; mean analyte concentrations were in the range 0.01–185 ng mL −1 . The method is suitable for the analysis of multiple phenolic metabolites in human urine.

Abstract:Objective:Using ion chromatography to measure the content of Oxalic acid and citric acid in the urine was correctly and fastly.Methods:The test of electrical conductance is to measure the content,the leacheate is 8.0mmol/L sodium carbonate and 10.0mmol/L sodiumhydroxide.After high speed centrifugation by C18Pretreatment column and 0.25μm millipore filter,all samples were to be added into Metrosep A SUPP 5/150anion chromatographic column.Results:The correlation coefficients of Oxalic acid ion and citric acid standard curve were 0.999898and 0.999927respectively,the detection limit was 0.97μg/L.The labeled recovery rate of oxalate was 98.9%-101.6% and of citric acid was 98.8%-101.8%,the analysis of ion chromatography had high Degree of precision(RSD%<2.0).Conclusions:Comparing the traditional analysis,the advantages of ion chromatography measuring the content of Oxalic acid and citric acid in the urine were accurate,simple,sensitive and rapid.This way could be used to analyze the risk factors of urine stone for the patients in clinic.

Abstract:OBJECTIVE To establish a method for determining oxalate in human urine by HPLC.METHODS To observe the normal value of oxalate in human urine,oxalate was determined by using Agilent SB C_(18)(250 mm×(4.6) mm,5μm)column and Agilent zorbax extend-C_(18)((4.6) mm×(12.5) mm,5μm)guard column;An aqueous solution containing(0.018) mol·L~(-1)potassium dihydrogenphosphate and(0.010) mol·L~(-1)tetrabutylammonium hydrogensulphate and(2.69)×10~(-5) mol·L~(-1) EDTA at pH(2.4)-(2.5) was used as mobile phase.Flow rate was(1.5) mL·min~(-1) at 25 ℃.Injection volume was 20 μL.All separation was detected at UV 210 nm.Deproteinization of urine samples was carried out by well mixing(0.5) mL urine sample with 25 mg crystalline sulfosalicylic acid at 4 ℃ for 30 min.RESULTS The detection limit was(0.4) mg·L~(-1).The linear range was(3.125)-(100.00) mg·L~(-1).The average recovery was(97.35%),RSDs of intra-day and inter-day were less than(10.06%).CONCLUSION The method is simple,sensitive and producible for quantifying oxalate in human urine and the research of formation of uric calcium oxalate stone.
