Abstract:Allenes represent a distinctive class of organic molecules characterized by their unique physical and chemical properties. Among them, chiral allenes play a significant role in modern chemistry. They not only serve as pivotal frameworks for natural products and drug molecules but also are essential building blocks in organic synthesis. Over the years, numerous remarkable and efficient methods have been developed for the synthesis of chiral allenes. Notably, the catalytic synthesis of chiral allenes using transition metals in conjunction with chiral ligands has garnered considerable attention. This review paper aims to provide an overview of recent advancements in the synthesis of chiral allenes through asymmetric 1,4‐difunctionalization of 1,3‐enynes, in which two distinct strategies including non‐radical and radical processes are highlighted.

Abstract:As a structural analog of oxazoline, imidazoline (4,5-dihydroimidazole) has received much attention in the rational design of chiral ligands. The additional N-substituent provides broader space for fine-tuning of electronic and steric effects, and it offers a good handle for immobilizing onto solid supports. In the past decades, imidazoline ring has emerged as a powerful candidate for the design of chiral nitrogen-containing ligands, as well as a significant alternative for oxazoline ring. Various chiral imidazoline ligands have been designed and utilized in asymmetric organic reactions. These new catalysts can not only be applied in classical reactions, but also be employed to develop new organic reactions with high enantioselectivities. This review provides an overview of chiral imidazoline ligands. Their applications in asymmetric synthesis are also summarized.

Abstract:Asymmetric synthesis of chiral allenes starting from prochiral substrates under mild reaction conditions promoted by Pd-SYNPHOS catalyst is reported. This protocol provides an efficient access to various enantioenriched aryl- and alkyl- substituted allenes, which are versatile building blocks of high utility to both organic and medicinal chemists, in excellent isolated yields (up to 96%) and high enatiomeric ratio values (up to 95:5). In addition, a comparative study using several C-2-symmetric atropisomeric diphosphine ligands revealed the overwhelming impact of the steric and electronic properties of the ligands for the catalytic efficiency of this process.

Abstract:In the last few years, the research area of chiral β-aminophosphines capable of promoting a wide range of diverse organic transformations has attracted more attention. Their derivatives constitute attractive reagents towards this end, due to their stability, low toxicity and ease of handling. These novel β-aminophosphine derivatives are potentially useful as organocatalysts and ligands in metal-catalysed reactions. In particular, chiral β-aminophosphines have recently emerged as powerful phosphine catalysts in asymmetric synthesis and catalysis. The asymmetric transformations to which metal complexes of these ligands have been applied include palladium-catalysed allylic substitutions and copper-catalysed 1,4-additions to enones among others. This review summarizes the most significant developments in the area of synthesis and application of chiral β-aminophosphine derivatives, providing a detailed overview of the current state of the art and including future aspects.

Sharpless J. Am. Chem. Soc. 1998, 120, 1207-1217), amination of enolates (D. A. Evans, S. G. Nelson J. Am. Chem. Soc. 1997, 119, 6452-6453) or reductive amination (R. Kadyrov, T. H. Riermeier, U. Dingerdissen, V. Tararov, A. Börner J. Org. Chem. 2003, 68, 4067-4070) but in some cases low regioselectivity, high catalyst loading or poor reactivity is observed. The authors applied their recently developed TangPhos ligand (Angew. Chem. Int. Ed. 2002, 41, 1612-1614) to the enantioselective hydrogenation of a variety of α-imino esters.

Abstract:In recent decades, the chiral allylation of imines emerged as a key methodology in the synthesis of alkaloids and natural products with 4-, 5- and 6-membered cyclic amine motifs. Initially reliant on stoichiometric reagents, synthetic chemists predominantly used N-substituted chiral imines, organometallic chiral reagents and achiral reagents with an equimolar chiral controller. However, recent years have witnessed the rise of asymmetric transition-metal catalysts and, importantly, organocatalytic allylation, reshaping the landscape of modern synthetic chemistry. This review explores the latest developments in the asymmetric allylation of imines, encompassing cutting-edge advances in hydrogen-bond catalysis and non-classical approaches. Furthermore, practical examples showcasing the application of these innovative methodologies in total synthesis are presented.

Abstract:Due to their importance in natural products, organic synthesis, life science, and pharmaceuticals, reliable methods for the synthesis of chiral amines are urgently required. Here in this contribution, the dual chiral catalytic method has been established for the enantioselective reaction of terminal alkynes, aldehydes, and amines affording optically active propargylic amines with a decent enantioselectivity and a broad substrate scope. These propargylic amines have been demonstrated as platform molecules for the syntheses of different types of chiral amines. By applying this protocol, the asymmetric total syntheses of two linear monoamine natural products, (S)-5 and (S)-11, which were isolated from marine cyanobacteria Microcoleus lyngbyaceus, have been realized. The envisioned chiral ion pair, which has boosted the enantioselectivity greatly, and other key intermediates have been successfully characterized by MS studies.

Abstract:Nitrogen containing compounds like chiral amines have been widely used in synthetic medicines, biomass, and fine chemical products because of their particularly valuable motifs and exceedingly broad range of functionality. Enantioconvergent synthesis of chiral amines has high atomic economy and enantioselectivity with theoretically 100 % yield of enantioenriched products. In this concept article, we hope to provide a further understanding of chiral amines enantioconvergent synthesis methodology with an introduction of recent progresses in enantioconvergent amination reactions catalyzed by early transition metals and facilitate the development of enantioconvergent C(sp(3))-N coupling for benign and efficient preparation of highly valuable chiral nitrogen-containing units.

Abstract:Chiral α-branched amines are common structural motifs in functional materials, pharmaceuticals, and chiral catalysts. Therefore, developing efficient methods for preparing compounds with these privileged scaffolds is an important endeavor in synthetic chemistry. Herein, we describe an atom-economical, modular method for a nickel-catalyzed enantioselective α-alkenylation of readily available linear N-sulfonyl amines with alkynes to afford a wide variety of allylic amines without the need for exogenous oxidants, reductants, or activating reagents. The method provides a platform for constructing chiral α-branched amines as well as derivatives such as α-amino amides and β-amino alcohols, which can be conveniently accessed from the newly introduced alkene. Abstract:Ugi's amine has become one type of privileged chiral skeleton for chiral ligand design bearing central/planar chirality, and such ligands have exhibited tremendous success in various asymmetric catalysis. However, the current access to enantiopure Ugi's amine is quite tedious and relies heavily on optical resolution, which impedes its practical applications, to some extent. Herein, we present a facile asymmetric synthesis of enantioenriched Ugi-type amines bearing a long-carbon chain through Ir-catalyzed cascade allylation/2-aza-Cope rearrangement, followed by amino exchange and Pd/C-catalyzed one-pot hydrogenation/reductive amination. The protocol could be readily scaled up, and it has been conducted in 20-g-scale asymmetric synthesis of (S)-Ugi-type amine from commercially available reagents, in >99% ee and >70% overall yield in four steps with one short silica gel-plug purification. Abstract:Catalytic enantioselective α-C–H functionalization of widely available achiral alkyl amines could provide an ideal synthetic approach towards chiral amines. However, the inert nature of the α-C–H of alkyl amines renders their activation as carbanionic nucleophiles for catalytic asymmetric reactions an important yet unmet challenge. Here we describe how N -arylidene-protected alkyl amines could be activated as carbanions for asymmetric conjugate addition and the Mannich reaction. These results represent an intriguing and generally useful approach to the synthesis of chiral α,α-dialkyl amines. More importantly, they highlight the enormous potential of N -arylidene-protected amines as readily available and widely applicable synthons for the asymmetric synthesis of chiral amines.

Abstract:Chiral allenes and styrenes are essential components in fields like medicinal chemistry, materials science, and organic synthesis. They serve a crucial role as chiral ligands and catalysts in asymmetric synthesis. Over the past decade, there has been a significant advancement in the development of practical methods utilizing organocatalytic strategies for the synthesis of chiral allenes and styrenes. It is noteworthy that despite extensive studies on the formation of allenes and styrenes, existing reviews on their construction confined to a specific organocatalysis, called chiral phosphoric acid catalysis, are less documented. This review aims to explore different conceptual approaches based on the electrophilic species involved in the reaction to produce stereoselective chiral allenes and styrenes, providing insights into recent advancements in the field. Emphasis is placed on works published since 2017, with detailed discussions on reaction mechanisms and examples from recent literature.

Abstract:Catalytic enantioselective synthesis of chiral helicenes and helicenoids has emerged as an important area of research. In the efforts toward efficient and enantioselective synthesis of chiral helicenes and helicenoids, chemists have developed several catalytic asymmetric reactions, primarily transition metal-catalyzed asymmetric intra- or intermolecular [2 + 2 + 2] cycloaddition, hydroarylation, olefin metathesis, and oxidative coupling reactions. In addition, asymmetric organocatalysis has shown potential for use in the enantioselective synthesis of chiral helicenes and helicenoids, including chiral phosphoric acid- and chiral amide-catalyzed enantioselective [3 + 2] and [4 + 2] cyclizations. In this review, advancements in the catalytic enantioselective synthesis of chiral helicenes and helicenoids from 1999 to the beginning of 2022 have been classified based on asymmetric transition metal catalysis and asymmetric organocatalysis. In addition, this review highlights the remaining challenges in this research field and sheds light on the future of this field.
