Abstract:Molecular machines can convert the chemical energy into the mechanical work through intra- or intermolecular motions at the single-molecule scale under external stimuli, such as heating, illumination, and so on. In recent decades, the working mechanisms of various molecular machines, such as molecular shuttles, molecular motors, molecular walkers, etc. have been extensively studied through different ensemble experiments, where the unique and unexpected behaviors of a single molecular machine may be overwhelmed by the average effect caused by a large number of molecules. Thanks to the development of single-molecule characterization technologies, the intrinsic properties of single molecular machines at the single-molecule scale are being explored, which is essential for future applications at the molecular scale. This Review offers a timely and systematical survey on the structural designs, characterization methods, working mechanisms, and influencing factors of molecular machines at the single-molecule scale, providing the comparison among different studies and in-depth comprehension of the intrinsic principles of individual molecular machines.This article has not yet been cited by other publications.

Abstract:Molecular machines are key to cellular activity where they are involved in converting chemical and light energy into efficient mechanical work. During the last 60 years, designing molecular structures capable of generating unidirectional mechanical motion at the nanoscale has been the topic of intense research. Effective progress has been made, attributed to advances in various fields such as supramolecular chemistry, biology and nanotechnology, and informatics. However, individual molecular machines are only capable of producing nanometer work and generally have only a single functionality. In order to address these problems, collective behaviors realized by integrating several or more of these individual mechanical units in space and time have become a new paradigm. In this review, we comprehensively discuss recent developments in the collective behaviors of molecular machines. In particular, collective behavior is divided into two paradigms. One is the appropriate integration of molecular machines to efficiently amplify molecular motions and deformations to construct novel functional materials. The other is the construction of swarming modes at the supramolecular level to perform nanoscale or microscale operations. We discuss design strategies for both modes and focus on the modulation of features and properties. Subsequently, in order to address existing challenges, the idea of transferring experience gained in the field of micro/nano robotics is presented, offering prospects for future developments in the collective behavior of molecular machines.

Abstract:Artificial molecular machine is one of the most brilliant areas in the last half a century. They represent people's control abilities to the microscopic molecules in solution. Beside the hard works in the proof-of-concept research on the exploration of fundamental principles, the wide hybrid of them with diverse materials was confirmed. Through the coupling of these movable molecules with various materials, the microscopic motion of molecular machines could be easily translated and amplified to macroscopic world to produce useful work. The integration further improves the ability of us to control the molecules in solid state. Among the various materials, polymers have the widest applications in many areas. The incorporation of artificial molecular machines into the bulk polymer materials would produce more influence on our life in the near future. In this review, we summarized the proofed hybrid strategies of polymers with different molecular machines. We hope that the summarization could provide a research roadmap for the related researchers. Abstract:Biomolecular machines are widely present in nature, especially in complex living organisms, and are involved in important biological processes. Inspired by nature and increasingly mature synthetic technologies, a series of artificial molecular machines (AMMs) that exhibit similar processes and functions as biomolecular counterparts have been developed, and to date, some dramatic achievements have been obtained. Herein, the use of AMMs in smart systems and materials with controllable regulations and interesting functions are summarized, presenting the specific micro‐ to macroscale applications in solid surface modification, transmembrane transport, smart catalysts, liquid crystals, artificial molecular muscles, and stimuli‐responsive polymers. The challenges of developing novel complex AMMs with intelligent functions are discussed, and some potential solutions are proposed.

Abstract:As one of the important parts of the molecular machines, molecular machines driven by acid-base chemistry play the crucial role in molecular machines, and have attracted more and more attentions for the applications in the design and construction of controllable molecular machines. At the same time, owing to the great progresses in theory studies of the molecular machine, several different kinds of functional and practical molecular machines driven by acid-base stimuli have been reported and utilized in supramolecular gels, supramolecular catalysts, supramolecular vesicles, supramolecular nanovalves, supramolecular polymers, and so on. In this paper, the design and applications of acid-base controllable molecular machines based on pseudorotaxanes, rotaxanes, and catenanes are reviewed, and the prospects of such molecular machines are also described.

Abstract:In order to fabricate highly sophisticated nanostructures, various research fields must be fused into one unified concept. In a recently emerging nanoarchitectonics, nanotechnology is combined with materials science, supramolecular self-assembly, and biological science to construct functional materials from nanoscale units. This review article focuses onto dynamic features of nanoarchitectonics. In terms of this concept, molecular machines as one of the forefront research targets are produced from DNA and RNA. Fundamental designs of DNA/RNA molecular machines are first described with an emphasis on the significant roles of interlocked structures such as rings, catenanes, rotaxanes, and connectors of these parts. In the latter part, DNA/RNA molecular machines including shuttles, molecular transporters, walkers, nano pumps, robots, amplifiers, and logic gates are presented, together with related functions such as sensitive sensing and catalysis regulation. The exemplified strategy should be universally useful to fabricate various functional materials which fulfill the social demands at present as well as in the future.This review describes the dynamic features of supramolecular DNA/RNA nanoarchitectonics leading to molecular machines. Significant roles of DNA/RNA rings, catenanes, rotaxanes, and their connectors are emphasized. Applications of these parts to molecular machines (e.g., shuttles, walkers, pumps, robots, and logic gates) are widely covered.

Abstract:The rise of DNA nanotechnology has driven the development of DNA-based molecular machines, which are capable of performing specific operations and tasks at the nanoscale. Benefitting from the programmability of DNA molecules and the predictability of DNA hybridization and strand displacement, DNA-based molecular machines can be designed with various structures and dynamic behaviors and have been implemented for wide applications in the field of biosensing due to their unique advantages. This review summarizes the reported controlling mechanisms of DNA-based molecular machines and introduces biosensing applications of DNA-based molecular machines in amplified detection, multiplex detection, real-time monitoring, spatial recognition detection, and single-molecule detection of biomarkers. The challenges and future directions of DNA-based molecular machines in biosensing are also discussed.

Abstract:As a post-nanotechnology concept, nanoarchitectonics combines nanotechnology with advanced materials science. Molecular machines made by assembling molecular units and their organizational bodies are also products of nanoarchitectonics. They can be regarded as the smallest functional materials. Originally, studies on molecular machines have analyzed the average properties of objects dispersed in solution by spectroscopic methods. Their playgrounds partially shifted to solid interfaces, because high-resolution image observation of molecular machines is usually done on solid interfaces under high vacuum and cryogenic conditions. It is further considered that practical uses of molecular machines need to function under ambient conditions. The latter conditions are met in dynamic interfacial environments such as the surface of water at room temperature. According to these backgrounds, this review summarizes the trends of molecular machines that continue to evolve under the concept of nanoarchitectonics in the interfacial environment. Some recent examples of molecular machines in solution are briefly introduced first, which is followed by an overview of studies of molecular machines and similar supramolecular structures in various interfacial environments. The interfacial environments are classified into (i) solid interfaces, (ii) liquid interfaces, and (iii) various material and biological interfaces. Molecular machines are expanding their activities from the static environment of the solid interface to the more dynamic environment of the liquid interface. Molecular machines change their field of activity while maintaining their basic functions and induce the accumulation of individual molecular machines into macroscopic physical properties and, conversely, to control molecular machines through macroscopic mechanical motions. Finally, their active fields reach even at the interface of living organisms.

Abstract:Molecular machines have attracted significant attentions as one of the most promising aspects of chemistry for their potential applications ever since receiving the 2016 Nobel Prize in Chemistry.The molecular assembler, also called the nanofactory, is a novel type of molecular machines that are capable of controlling the chemical reactions precisely at the microscopic level.As an analog to the macroscopic factories, nanofactories are comprised of a "transporting" part, the molecular walkers, and an "assembling" part, the molecular robotic arms.In this review, we provide a brief introduction of the research progress in recent years together with analysis on the principles of designing, constructing and operating molecular assemblers.We also summarize the prospects and challenges in the research area of molecular assemblers.

Abstract:Molecular machines are essential dynamic components for fuel production, cargo delivery, information storage and processing in living systems. Scientists have demonstrated that they can design and synthesize artificial molecular machines that operate efficiently in isolation — for example, at high dilution in solution — fuelled by chemicals, electricity or light. To organize the spatial arrangement and motion of these machines within close proximity to one another in solid frameworks, such that useful macroscopic work can be performed, remains a challenge in both chemical and materials science. In this Review, we summarize the progress that has been made during the past decade in organizing dynamic molecular entities in such solid frameworks. Emerging applications of these dynamic smart materials in the contexts of molecular recognition, optoelectronics, drug delivery, photodynamic therapy and water desalination are highlighted. Finally, we review recent work on a new non-equilibrium adsorption phenomenon for which we have coined the term mechanisorption. The ability to use external energy to drive directional processes in mechanized extended frameworks augurs well for the future development of artificial molecular factories.

Abstract:Molecular machines are nanoscale devices capable of performing mechanical works at molecular level. These systems could be a single molecule or a collection of component molecules that interrelate with one another to produce nanomechanical movements and resulting performances. The design of the components of molecular machine with bioinspired traits results in various nanomechanical motions. Some known molecular machines are rotors, motors, nanocars, gears, elevators, and so on based on their nanomechanical motion. The conversion of these individual nanomechanical motions to collective motions via integration into suitable platforms yields impressive macroscopic output at varied sizes. Instead of limited experimental acquaintances, the researchers demonstrated several applications of molecular machines in chemical transformation, energy conversion, gas/liquid separation, biomedical use, and soft material fabrication. As a result, the development of new molecular machines and their applications has accelerated over the previous two decades. This review highlights the design principles and application scopes of several rotors and rotary motor systems because these machines are used in real applications. This review also offers a systematic and thorough overview of current advancements in rotary motors, providing in-depth knowledge and predicting future problems and goals in this area.

Abstract:An overview of recent developments on the fabrication, locomotion characterization, motion manipulation, and biomedical applications of nanomotors has been presented. With continued attention and innovation, it is believed that, in the future, nanomotors will innovate biomedical applications.Artificial nanomotors are nanoscale machines capable of converting surrounding other energy into mechanical motion and thus entering the tissues and cells of organisms. They hold great potential to revolutionize the diagnosis and treatment of diseases by actively targeting the lesion location, though there are many new challenges that arise with decreasing the size to nanoscale. This review summarizes and comments on the state‐of‐the‐art artificial nanomotors with advantages and limitations. It starts with the fabrication methods, including common physical vapor deposition and colloidal chemistry methods, followed by the locomotion characterization and motion manipulation. Then, the in vitro and in vivo biomedical applications are introduced in detail. The challenges and future prospects are discussed at the end.

Abstract:Artificial molecular machines have received significant attention from chemists because of their unique ability to mimic the behaviors of biological systems. Artificial molecular machines can be easily modified with functional groups to construct new types of functional molecular switches. However, practical applications of artificial molecular machines are still challenging, because the working platform of artificial molecular machines is mostly in solution. Artificial molecular machine immobilized surfaces (AMMISs) are considered a promising platform to construct functional materials. Herein, we provide a minireview of some recent advances of functional AMMISs. The functions of AMMISs are highlighted and strategies for their construction are also discussed. Furthermore, a brief perspective of the development of artificial molecular machines towards functional materials is given.

Abstract:Mechanically interlocked molecules are perhaps best known as components of molecular machines, a view further reinforced by the Nobel Prize in 2016 to Stoddart and Sauvage. Despite amazing progress since these pioneers of the field reported the first examples of molecular shuttles, genuine applications of interlocked molecular machines remain elusive, and many barriers remain to be overcome before such molecular devices make the transition from impressive prototypes on the laboratory bench to useful products. Here, we discuss simplicity as a design principle that could be applied in the development of the next generation of molecular machines with a view to moving toward real-world applications of these intriguing systems in the longer term.Simplicity is discussed as a design principle that could be applied in the development of the next generation of molecular machines.

Abstract:The designability, functionalization, and diverse secondary structures of DNA enable the construction of DNA motors with stimuli-responsiveness. Therefore, it has been widely used to fabricate functional systems or generate mechanical power under external stimuli, such as pH, light, heat, electrical, and chemical molecular signals. Furthermore, the DNA motor has also been demonstrated to promote the applications of smart devices and materials, particularly in controllable drug delivery and reversible molecular switching. In this review, we have summarized and discussed recent progress of the construction and applications of DNA motor-based functional systems, such as responsive nanodevices, modified surfaces, and hydrogels.

Abstract:Molecular shuttles are typical molecular machines that could be applied in various fields. The motion modes of wheel components in rotaxanes could be strategically modulated by external stimuli, such as pH, ions, solvent, light, and so on. Light is particularly attractive because it is harmless and can be operated in a remote mode and usually no byproducts are formed. Over the past decade, many examples of light-driven molecular shuttles are emerging. Accordingly, this review summarizes the recent research progress of light-driven molecular shuttles. First, the light-driven mechanisms of molecular motions with different functional groups are discussed in detail, which show how to drive photoresponsive or non-photoresponsive molecular shuttles. Subsequently, the practical applications of molecular shuttles in different fields, such as optical information storage, catalysis for organic reactions, drug delivery, and so on, are demonstrated. Finally, the future development of light-driven molecular shuttle is briefly prospected.

Abstract:Since the establishment of the Watson-Crick model more than five decades ago, the understandings of DNA structures are well sufficient to enable applications of DNA in designing and assembling two-dimensional (2D) and three-dimensional (3D) structures at the nanoscale. Furthermore, the conformational switchability of DNA also enables the fabrication of nanoscale molecular machines, which can perform movements upon stimuli. In this article, we will summarize the present efforts on constructions of DNA nanomachines based on different driven mechanisms, and further discuss their evolutional processes, in order to find applications and future development directions.

Abstract:Background: The DNA strand displacement reaction, which uses flexible and programmable DNA molecules as reaction components, is the basis of dynamic DNA nanotechnology, and has been widely used in the design of complex autonomous behaviors. Results: In this review, we first briefly introduce the concept of toehold-mediated strand displacement reaction and its kinetics regulation in pure solution. Thereafter, we review the recent progresses in DNA complex circuit, the assembly of AuNPs driven by DNA molecular machines, and the detection of single nucleotide polymorphism (SNP) using DNA toehold exchange probes in pure solution and in interface state. Lastly, the applications of toehold-mediated strand displacement in the genetic regulation and silencing through combining gene circuit with RNA interference systems are reviewed. Conclusions: The toehold-mediated strand displacement reaction makes DNA an excellent material for the fabrication of molecular machines and complex circuit, and may potentially be used in the disease diagnosis and the regulation of gene silencing in the near future.
