Abstract:Calixarenes are highly versatile scaffolds for the design of 3-D cavities, and have been regarded as the third generation of host molecules after crown ethers and cyclodextrins in supermolecular chemistry, which can selectively form complexes with guest molecules or ions. Recently, calixarenes containing polymers have attracted much attention. Due to the good stability and machinability of polymer materials, calixarenes containing polymers are expected to be developed as novel functional materials, which have been widely studied in ion selective electrodes and membranes I chemical and biochemical sensors and selective extraction of ions and neutral molecules. Based on the chain structures, there are three kinds of calixarenes containing polymers: calixarene in polymer backbone, calixarene as polymer side group and star polymer with a calixarene core. In this paper, the recent progress on the synthesis and application of these calixarenes containing polymers is reviewed.

Abstract:Calixarenes are "chalice like" phenol-based macrocycles that are one of the most fascinating studied scaffolds in supramolecular chemistry. Their preorganized nonpolar cavities and ion binding sites, and their well-defined conformations all lay important foundations for forming host–guest complexes. Conjugation of calixarene scaffolds with various fluorophores at either upper or lower rims has led to the development of smart fluorescent probes for inorganic molecules or ions, aliphatic or aromatic compounds, biomolecules, temperature and hypoxia, even multi-component traditional Chinese medicine (TCM). Moreover, significant advancements have been made for biological applications. This review critically summarizes the recent advances made in these areas.

Abstract:Natural products can generally exhibit a variety of biological activities, but most show mediocre performance in preliminary activity evaluation. Natural products often require structural modification to obtain promising lead compounds. Cinnamic acid (CA) is readily available and has diverse biological activities and low cytotoxicity. Introducing CA into natural products may improve their performance, enhance biological activity, and reduce toxic side effect. Herein, we aimed to discuss related applications of CA in the structural modification of natural products and provide a theoretical basis for future derivatization and drug development of natural products. Published articles, web databases (PubMed, Science Direct, SCI Finder, and CNKI), and clinical trial websites (https://clinicaltrials.gov/) related to natural products and CA derivatives were included in the discussion. Based on the inclusion criteria, 128 studies were selected and discussed herein. Screening natural products of CA de-rivatives allowed for classification by their biological activities. The full text is organized according to the biological activities of the derivatives, with the following categories: anti-tumor, neuroprotective, anti-diabetic, anti-microbial, anti-parasitic, anti-oxidative, anti-inflammatory, and other activities. The biological activity of each CA derivative is discussed in detail. Notably, most derivatives exhibited enhanced biological activity and reduced cytotoxicity compared with the lead compound. CA has various advantages and can be widely used in the synthesis of natural product derivatives to enhance the properties of drug candidates or lead compounds.
Molecular recognition and "on-off" switching of calix[4]arenes to monosaccharides and pyrimidine derivatives

Abstract:The AM1 and INDO/SCI method were used to, study supra-molecular complexes composed of octahydroxy calix[4]arenes and monosaccharides and pyrimidine derivatives. It was shown that the host performs a better molecular recognition to mono-saccharides. especially to the erythrose by the CH-pi, interaction but there is almost no pi-pi interaction between the host and the pyrimidine which exists in the pyrimidine derivative when H is substituted by -COOH, and the "on-off" molecular switching can be designed by this. The reason of red-shift for UV absorption peaks of the complexes compared to that of the host was discussed.

Abstract:Calixarenes are capvity-shipped cyclic molecules made up of phenol units linked by methylene groups.Calixarenes posses great potentials in the molecular recognition,which are to be counted as the third generation of supramolecules after cyclodextrins and crown ethers.They are widely used in modern separation science like chromatography and capillary electrophoresis.Their applications in chromatography and capillary electrophoresis are reviewed with 54 references,while some problems and strategies are pointed out.

Abstract:A series of novel calix[4]arenes containing acetal, amid and cyano group respectively have been prepared by the O - alkylation. In reactions of O - alkylation, mono,1,3 - di and tetra ethers can be obtained respectively as main products by changing reaction conditions. The regio - selectivity depends greatly on the type of bases used and the reaction time.

Abstract:Supramolecular chemistry is a central topic in modern chemistry. It touches on many traditional disciplines, such as organic chemistry, inorganic chemistry, physical chemistry, materials chemistry, environmental chemistry, and biological chemistry. Supramolecular hosts, inter alia macrocyclic hosts, play critical roles in supramolecular chemistry. Calix[4]pyrroles, non-aromatic tetrapyrrolic macrocycles defined by sp3 hybridized meso bridges, have proved to be versatile receptors for neutral species, anions, and cations, as well as ion pairs. Compared to the parent system, octamethylcalix[4]pyrrole and its derivatives bearing simple appended functionalities, strapped calix[4]pyrroles typically display enhanced binding affinities and selectivities. In this review, we summarize advances in the design and synthesis of strapped calix[4]pyrroles, as well as their broad utility in molecular recognition, supramolecular extraction, separation technology, ion transport, and as agents capable of inhibiting cancer cell proliferation. Future challenges within this sub-field are also discussed.

Abstract:Because they hold together molecules by means of non-covalent interactions - relatively weak and thus, potentially reversible - the anionic calixarenes have become an interesting tool for efficiently binding a large range of ligands - from gases to large organic molecules. Being highly water soluble and conveniently biocompatible, they showed growing interest for many interdisciplinary fields, particularly in biology and medicine. Thanks to their intrinsic conical shape, they provide suitable platforms, from vesicles to bilayers. This is a valuable characteristic, as so they mimic the biologically functional architectures. The anionic calixarenes propose efficient alternatives for overcoming the limitations linked to drug delivery and bioavailability, as well as drug resistance along with limiting the undesirable side effects. Moreover, the dynamic non-covalent binding with the drugs enables predictable and on demand drug release, controlled by the stimuli present in the targeted environment. This particular feature instigated the use of these versatile, stimuli-responsive compounds for sensing biomarkers of diverse pathologies. The present review describes the recent achievements of the anionic calixarenes in the field of life science, from drug carriers to biomedical engineering, with a particular outlook on their applications for the diagnosis and treatment of different pathologies.

Abstract:Due to their structural features, macrocyclic compounds such as calixarenes, conjugated with a variety of fluorophores have led to the development of fluorescent probes for numerous applications. This review covers the recent advances (from 2009 to date) made in calixarene-based fluorescent sensors and their biological applications. In addition to the fluorescence mechanisms used to signal the analyte binding, this article focuses mainly on the detection of biological relevant ions, on the selective sensing of biomolecules, such as amino acids, enzymes, drugs and other organic compounds, and on intracellular imaging. Calixarene-containing fluorescent nanoparticles and nanoaggregates for imaging and drug delivery are also described. Finally, this review presents some conclusions and future perspectives in this field.

Abstract:The use of calixarenes in asymmetric catalysis was described. The current progress of asymmetric catalysts based on calixarene scaffold for the enantioselective reactions including allylation, alkylation, addition of diethylzinc to benzaldehyde, Micheal-type addition, aldol reaction, hydrogenation, Friedal Crafts reaction, epoxidation, hydroformylation, and Biginelli reaction was illustrated. In these cases, calixarenes were used as organocatalysts or ligands in metal-catalyzed asymmetric reactions.

Abstract:The applications of calixarenes in polymer synthesis have been reviewed. Calixarenes have been used as ligands to prepare rare earth calixarene complexes. A series of rare earth calixarene complexes have been synthesized and employed as efficient catalysts for the polymerization of ethylene, styrene, butadiene, propylene oxide, styrene oxide, trimethylene carbonate, and 5,5-dimethyl trimethylene carbonate. on the other hand, the synthesis and characterization of star-shaped polymers with calixarene as core molecules are also described.

Abstract:Four tetraalkylethers 3a similar to 3d(alkyl: n - C4H9, n - C8H17, n - C12H25, n - C16H33) of calix [4]arene and their corresponding p - tetraacetyl derivatives 4a similar to 4d were prepared by the etheration with NaH as base in DMF and acetylation on phenyl. H-1 NMR indicated that ail new calix [4] arene derivatives have cone conformation.

Abstract:A series of p-tetra-tert-butyl calix[4]arene derivatives with alkyl guanidinium or chiral bicyclic guanidinium functionalities were synthesized as the receptors of organic anions. Mono- and di- alkyl guanidinium derivatives were prepared by the reaction of 5,11,17,23-tetra-tert-butyl-25,27-(2-aminoethoxy)-26,28-dihydroxy calix[4]arene (1) with methyl isothiourea sulphate. While mono- and di- chiral bicyclic guanicinium derivatives were all obtained via an O-alkylation reaction of p-tetra-tert-butyl calix[4]arene with chloromethyl chiral bicyclic guanidinium (6 or 7) in the presence of anhydrous K2CO3 in acetonitrile, good yields were only observed for mono- substituted derivatives.

Abstract:Chiral macrocyclic arenes possess confined three-dimensional asymmetric cavity, electron-rich structures, chiral luminescent properties and excellent enantioselective recognition properties, which have represented a frontier and hotspot of macrocyclic chemistry and supramolecular chemistry. In recent years, there has been growing interest in the development of novel chiral macrocyclic arenes, which have found applications in various research areas. In this review, the construction, properties and functional applications in enantioselective recognition, chiral sorting and chiral luminescent materials of novel chiral macrocyclic arenes according to their chiral types including central, axial, planar, and inherent chirality are summarized. It is expected that this review will be helpful to the research of supramolecular chemistry and promote the development of synthetic chemistry, material chemistry and biochemistry.

Abstract:Cancer is one of the leading causes of death worldwide, and the limited success of currently used anticancer drugs is a driving force for the search for new compounds with anticancer potential. Calixarenes make up a fascinating class of macrocycles that find a wide range of biological applications in supramolecular chemistry. Especially in recent years, the anticancer activity of various calixarene derivatives has been reported by many researchers. The aim of this review article is to find relatively simple, efficient, and synthetic methods for the preparation of macrocyclic compounds containing various pharmacophoric groups to produce new and effective anticancer materials in cancer research and to investigate the usability of calixarene-based supramolecular in cancer research.
