Abstract:Objective:Chromatographic method using amylose chiral stationary phase for separation of fluoxetine hydrochloride enantiomers has been developed.Tris (3,5-dimethylphenyl carbamate)amylose chiral stationary phase was used in the normal-phase mode.The effects of mobile phase composition,flow rate and column tempera- ture on the ehiral separation were discussed systematically.Method:The column used was Chiralpak ADH (250mm ×4.6mm,5μm).The mobile phase studied was hexane alcohol(include isopropanol,ethanol and 1-propanol). UV detector was used at 226 nm.Result:The optimal composition of mobile phase was hexane isopropanol diethyl amine(98:2:0.2,v/v/v).The optimal flow rate was about 0.4mL min~(-1)from Van Deemter equation,however, baseline separation was achieved (R_S1.5)at flow rate lower than 0.8mL·min~(-1).Within all the temperature range examined,the fluoxetine enantiomers were baseline separated.Conclusion:Using this method,the enantiomers of fluoxetine could be separated quickly and easily.
Direct Enantiomeric Separation of Β -Aminoalcohols and Β -Hydroxy Sulfides Using Polysaccharide Derivatives As Chiral Stationary Phases

Abstract:Direct enantiomeric separation of racemic mixtures of 13 β - amino alcohols and β -hydroxy sulfides was achieved by HPLC using Chiralcel OD, Chiralcel OJ and Chiralpak AD as chiral stationary phases and hexane/2 - propanol mixture as eluent.These racemates were completely separated into enantiomers at least on one of these stationary phases. The chromatographic parameters of these racemates on AD, OD and OJ columns at various ratios of hexane/2 - propanol were examined. The results showed that the properties of substituents of the compounds obviously had effects on their resolution on columns. The hydrogen bonding and π -π interactions between the chiral stationary phase and the polar group of racemates may be responsible for the chiral recognition. This method had been applied to determination of the optical purity of the product from asymmetric ring opening of meso -epoxides.

Abstract:Amylose-tris(cyclohexylcarbamate) (ACHC) coated on a kind of small particle silica gel was prepared. On this chiral stationary phase (CSP), the direct enantiomeric separation of nine chiral compounds has been achieved using n-hexane as the mobile phase containing various alcohols as modifiers. The effect of mobile phase modifiers on their resolution and retention was investigated. Chromatographic behavior of chiral compounds on ACHC-CSP was compared with that on amylose-tris(3,5-dimethylphenyicarbamate) by hexane/isopropanol as the mobile phase. The result suggests that ACHC showed high chiral recognition abilities as well as amylose-tris(3,5-dimethylphenylcarbamate) in HPLC. ACHC-CSP is a suitable CSP for the optical resolution of a lot of racemates by HPLC.

Abstract:In this work, a series of chiral phenethylamine synergistic tricarboxylic acid modified ,B-cyclodextrin bonded stationary phase for high performance liquid chromatography (HPLC) were synthesized via a sim-ple one-pot synthesis approach. Various racemates (aryl alcohols, flavanones, triazoles, benzoin, etc. ) were well separated on the tricarboxylic acid modified chiral stationary phases in both normal and reversed modes with good reproducibility and stability, and the influence of mobile phase composition on reso-lution (Rs) were deeply investigated. The RSD values of Rs for repeatability and column-to-column were below 1.28% and 3.05%, respectively. Hence, the fabrication of tricarboxylic acid modified chiral stationary phase (CSPs) is a new efficient strategy to improve the application of ,B-cyclodextrin as CSPs in the field of chromatography.(c) 2023 Published by Elsevier B.V. on behalf of Chinese Chemical Society and Institute of Materia Medica, Chinese Academy of Medical Sciences.

Abstract:Abstract This paper reported the enantioseparation of uniconazole and imazalil using four different self‐prepared cellulose derivative columns, namely, CTB, CTMB, CTPC, and CDMPC. The mobile phase composition including the type of the alcohol modifier and the ethanol concentration in hexane changed, and the influence on the retention factor (k′); for each enantiomer, the separation factor (α) and the resolution (RS) was studied. The influence of the structures of the analytes on the chiral separation was investigated. The results showed that uniconazole obtained the best resolution of 2.16 on CTPC, while imazalil obtained the best separation on CTMB, with the maximal resolution of 4.83. It was concluded that the structure of solutes and CSP play a key role in the chiral recognition. And the chiral attraction between them is the predominant factor.

Abstract:Enantioseparation of nineteen liquid crystalline racemic mixtures obtained based on (R,S)-2-octanol was studied in reversed-phase mode on an amylose tris(3-chloro-5-methylphenylcarbamate) (ReproSil Chiral-MIG) and a cellulose tris(3,5-dichlorophenylcarbamate) (ReproSil Chiral-MIC). These polysaccharide-based chiral stationary phase (CSP) columns for High-Performance Liquid Chromatography (HPLC) were highly effective in recognizing isomers of minor structural differences. The mobile phase (MP), which consists of acetonitrile (ACN)/water (H2O) at different volume ratios, was used. The mobile phases were pumped at a flow rate of 0.3, 0.5, or 1 mL·min−1 with a column temperature of 25 °C, using a UV detector at 254 nm. The order of the elution was also determined. The chromatographic parameters, such as resolution (Rs), selectivity (α), and the number of theoretical plates, i.e., column efficiency (N), were determined. The polysaccharide-based CSP columns have unique advantages in separation technology, and this study has shown the potential usefulness of the CSP columns in separating liquid crystalline racemic mixtures belonging to the same homologous series.

Abstract:A novel chiral stationary phase (CSP) consisting of a silica-based chitosan tris(3-chlorophenylcarbamate) derivative was used for the enantioseparation of nine racemic compounds in the normal phase mode by high-performance liquid chromatography (LC). The influence of the type and percentage of modifier in the mobile phase on the optimization of the resolution was investigated. The resolution of epoxiconazole using this system was compared with those achieved using commercially available Chiralcel® OD-H and Chiralpak® AD-H columns. Among the tested racemates, seven racemic compounds were separated most quickly on the CSP of chitosan tris(3-chlorophenylcarbamate). Baseline or near-baseline separation was achieved for benzoin, penconazole, hexaconazole, and epoxiconazole, while the others were partially separated. The enantioseparation results for epoxiconazole were not inferior to those of commercially available Chiralcel® OD-H and Chiralpak® AD-H columns.

Abstract:The chiral separation of racemic tetrahedral metal clusters was directly performed on a self-made cellulose-tris(3,5-dimethylphenylcarbamate)(CDMPC-CSP)chiral stationary phase with satisfactory reˉsults.The influences of the alcoholic type mobile phase modifier in the mobile phase and the structures of chiral tetrahedral type metal clusters on the chiral separation and retention were studied.It was found that the structure and alcohol concentration in mobile phase and the subtle structural differences in racemic could have a pronounced effect on enantiometric separation and retention.The chiral separation of racemic tetraheˉdral metal clusters was most effective when using n-butanol as mobile phase modifier.
Enantioseparation of Three Isomeric Α-(Chlorophenyl)propanoic Acid by Countercurrent Chromatography and Investigation of Chlorine Substituent Through Characterization of Inclusion Interaction

Abstract:The influence of chlorine substituents in chiral separation of three racemic 2-(chlorophenyl)propanoic acids by countercurrent chromatography using hydroxypropyl-β-cyclodextrin as a chiral additive were mainly investigated in the present paper, including 2-(2-chlorophenyl)propanoic acids, 2-(3-chlorophenyl)propanoic acids and 2-(4-chlorophenyl)propanoic acids. The influences of chromatographic conditions on the retention behavior were studied by enantioselective liquid-liquid extraction experiments using the methodology of response surface It was found that 2-(3-chlorophenyl)propanoic acids could be successfully chiral separated by countercurrent chromatography, while no resolution was achieved for racemic 2-(2-chlorophenyl)propanoic acids and 2-(4-chlorophenyl)propanoic acids under optimized separation conditions. The formation of 1:1 stoichiometric inclusion compounds between 2-(3-chlorophenyl)propanoic acids and HP-β-CD was determined by UV spectra measurements. The inclusion constants for 2-(3-chlorophenyl)propanoic acids and HP-β-CD were determined by the Benesi-Hildebrand equation. Meanwhile, the inclusion constants of 2-(3-chlorophenyl)-propanoic acid enantiomer and HP-β-CD were obtained by the pesudophase retention equation in countercurrent chromatography. Furthermore, the inclusion interactions of the three racemates with HP-β-CD were also investigated by the molecular docking. The results obtained from UV spectra measurements and molecular docking showed that the racemate with chlorine substituents in meta-position presented the highest enantiorecognition while the racemates with chlorine substituents in ortho-position had the lowest enantiorecognition. The above results further indicated that forming a stable inclusion complex between racemate and chiral selector is a prerequisite for a successful enantioseparation and at the same time, the difference in inclusion capacity between the two enantiomers is also essential for the enantioseparation.

Abstract:Optical resolution of a range of racemic compounds was investigated using cellulose tribenzate and trisphenylcarbamate coated onto a small-partial spherical organosilane-modified silica. The influence on chiral discrimination of factors such as coated loadings and mobile phase was performed. The loadings of 15% on the support gave the best results. The regioselectivity of an asymmetric hydroesterification of styrene was determined by HPLC using a chiral stationary phase(CTPC-CSP). A satisfactory result had been obtained on CTPC chiral column with n-hexane/2-propanol (98/2 in volume ratio) as mobile phase.

Abstract:The chiral selector 6-azido-2, 3-di(p-chlorophenylcarbamoylated) cellulose was synthesized and further chemically immobilized onto 5-μm amino functionalized spherical porous silica gel. It was used as chiral stationary phase in high-performance liquid chromatography. Thirty racemates were successfully separated into enantiomers in either normal phase mode or reversed-phase mode. Good reproducibility and stability of the chiral stationary phase have been demonstrated.

Abstract:This paper reported the chiral separation of substituted cyclohexanone and substituted cyclohexanol using self-prepared cellulose derivative CTB, CTMB, CTPC, CDMPC whose structures were shown in Fig. 2 of the text and (S, S)-Whelk-O 1 columns. The influence of the structures of the analytes on the chiral separation was studied. The difference of chiral recognition mechanism on cellulose derivative chiral columns and (S, S) -Whelk-O 1 columns was compared. The results indicate that on (S, S) -Whelk-O 1 the attraction between solute and stationary phase is primary to the chiral recognition, while on cellulose derivative chiral columns the solutes' stereo structure must play a chief role in meeting the requirement for a stereoseletive fit into the cellulose chiral cavity, which may be the key to the chiral recognition on cellulose derivatives chiral stationary phase.

Abstract:Cellulose trisphenylcarbamate was regioselectively bonded to 3-aminopropyl silica gel at 6-position of the primary hydroxyl group on the glucose unit of cellulose with 4,4'-diphenylmethane diisocyanate as a spacer. Nine racemates were resolved on the prepared stationary phase under normal-HPLC mobile phases. It was observed that enantioseparations, on the bonded-phase showed higher efficiency by using tetrahydrofuran as a component of the mobile phase, and the structure of the derivative agent reacted with the amino acids played a role for the enantioseparation. Chiral separation on the bonded phase under reversed-HPLC mobile phase was also performed successfully. It has been shown that the retention factors of the racemates decreased appreciably with the increment of the concentration of acetonitrile in the mobile phase, while no significant change was observed for the enantioselectivity. Separation of enantiomer for the acidic racemate could be improved with a lower pH value of the mobile phase since the dissociation of acidic compound was impressed, but it did not affect significantly the resolution of neutral compounds.

Abstract:Allethrin is an important insecticide and its two enantiomers have different bioactivities. The allyl4hydroxy3methyl2cyclopenten1one is an important intermediate in chiral synthesis of allethrin, and its optical purity is closely related to that of the final product. So it is very important to establish a method to separate the racemates of 2allyl4hydroxy3methyl2cyclopenten1one. Amylpectin tris (3,5dimethylphenylcarbamate) was synthesized and coated on spherical silica gel as a chiral stationary phase (CSP),with which a practical and useful chiral column was packed for high enantiomeric resolution. The effects of isopropanol concentration in the mobile phase on the retention and resolution were investigated. The retention factors of the racemates decreased appreciably with the increment of the concentration of isopropanol and so was the resolution factor.

Abstract:Enantioseparation of lysine derivatives by chiral high-performance liquid chromatography (HPLC) was accomplished using two chiral stationary phases (CSPs, Chiralpak IA and Chiralpak IC) based on polysaccharides under normal phase (NP) conditions. All analytes were completely separated. The impacts of polar modifiers, analyte structure and column temperature on chiral separation were discussed. Moreover, the relationship between structure and retention was investigated. The van't Hoff equation was employed to evaluate the thermodynamic parameters of the chiral separation process. The data suggest that the chiral separation process was enthalpy-driven. Surprisingly, two uncommon phenomena were observed: (1) high separation factors on Chiralpak IA and (2) different binding mechanisms with CSP and the two enantiomers.
In situ growth preparation of a new chiral covalent triazine framework core-shell microspheres used for HPLC enantioseparation

Abstract:The manufacturing of chiral covalent triazine framework core-shell microspheres CC-MP CCTF@SiO 2 composite is reported as stationary phase for HPLC enantioseparation. The CC-MP CCTF@SiO 2 core-shell microspheres were prepared by immobilizing chiral COF CC-MP CCTF constructed using cyanuric chloride and (S)-2-methylpiperazine on the surface of activated SiO 2 through an in-situ growth approach. Various racemates as analytes were separated on the CC-MP CCTF@SiO 2 -packed column. The experimental results indicate that 19 pairs of enantiomers were well separated on the CC-MP CCTF@SiO 2 -packed column, including alcohols, phenols, amines, ketones, and organic acids. Among them, there are 17 pairs of enantiomers that can achieve baseline separation with good peak shapes. Their resolution values on this chiral column are between 0.4 and 5.61. The influences of analyte mass, column temperature, and composition of the mobile phase on the resolution of enantiomers were studied. In addition, the chiral resolution ability of CC-MP CCTF@SiO 2 -packed column was compared with the commercial chiral chromatographic columns (Chiralpak AD-H and Chiralcel OD-H columns) and some CCOF@SiO 2 chiral columns (β-CD-COF@SiO 2 , CTpBD@SiO 2 , and MDI-β-CD-modified COF@SiO 2 ). The CC-MP CCTF@SiO 2 -packed column exhibited some unique advantages and can complement these chiral columns in chiral separations. The research results show that the CC-MP CCTF@SiO 2 chiral column offered high column efficiency (e.g., 17680 plates m −1 for ethyl mandelate), low column backpressure (5–9 bar), high enantioselectivity, and excellent chiral resolution ability for HPLC enantioseparation with good stability and reproducibility. The relative standard deviations (RSD) ( n = 5) of the retention time, and peak areas by repeated separation of ethyl mandelate are 0.23% and 0.67%, respectively. It demonstrates that the CC-MP CCTF@SiO 2 core-shell microsphere composite has great potential in enantiomeric separation by HPLC. Graphical abstract

On this ADMPC chiral stationary phase (CSP), the direct enantiomeric separation of six novel chiral transition metal tetrahedral clusters has firstly been achieved using n-hexane as the mobile phase containing various alcohols as modifiers. The effect of mobile phase modifiers and the structural variation of the solutes on their retention factors (k') and resolutions (Rs) were investigated. The result suggests that not only the structure and concentration of alcohol in mobile phase, but also the structural differences in racemates can have a pronounced effect on enantiomeric separation. ADMPC-CSP is a suitable CSP for the optical resolution of chiral tetrahedral cluster by HPLC.

Abstract:D-Phenylglycine derivative chiral stationary phase(CSP) was synthesized with D-3,5dinitrobenzoylphenylglycine and 3-aminopropyl silica gel and the chiral column was prepared.Ten nonderivatives racemates,which were amino acids,amines,alcohols and carboxylic acids,were separated on the CSP using hexane and isopropanol as mobile phase by high-performance liquid chromatography(HPLC). The results showed that seven of the ten enantiomers could be baseline separated and the best value of resolution factors(Rs) was 5.56.The evaluation of column efficiency was carried out by benzene and the number of theoretical plate was 80 000 /m.

Abstract:Material with a core–shell structure based on a porous silica core and Ag nanoparticles (AgNPs) as shell (SiO2@Ag) was prepared with the seed growth method. Cellulose derivatives as chiral selectors were coated on the surfaces of SiO2@Ag particles by means of coating and intermolecular polycondensation as chiral stationary phases (CSPs), respectively. Fifteen racemates were used to evaluate their chiral recognition ability by high-performance liquid chromatography (HPLC). The results implied that the CSPs based on SiO2@Ag particles obtained stronger interaction with the analytes leading to better enantioseparation due to the existence of AgNPs on the surfaces of silica gel, compared with a cellulose derivative-modified silica column. In addition, the new CSPs showed special selectivity for ketone enantiomers, and AgNPs played a synergistic effect with cellulose derivatives on enantioseparation.Graphical abstractSchematic of SiO2@Ag stationary phase
