Abstract:There are two major problems need to be solved urgently in artificial synthetic small diameter vascular grafts:anticoagulation and rapid endothelialization.Organoseleniumimmobilized polyethyleneimine (SePEI) catalyzes the generation of nitric oxide (NO) in situ,thereby effectively inhibiting thrombosis.The laminin-derived peptide Tyr-Ile-Gly-Ser-Arg (YIGSR) has an endothelial cell selectivity.The rapid formation of endothelial layer can be achieved by introducing YIGSR on the grafts' surface.In this study,polycaprolactone (PCL) gratis were constructed by electrospinning.SePEI and hyaluronic acid were deposited layer-by-layer to catalyze NO generation,and the outmost HA layer was modified with YIGSR to promote endothelialization.The physical and chemical properties of prepared materials before and after modification were investigated.The results show that the modification process has no obvious effect on the fiber morphology,and the stability of layer-by-layer assembly can be maintained above 99% over 65 days.After modification,the mechanical properties of the electrospun PCL grafts have been enhanced,which can meet the requirements of vascular grafts.The constructed materials will be further examined with cell culture and in vivo experiments.
