Abstract:A concise total synthesis of rac-alsmaphorazine D has been described for the first time. The efficient synthetic strategy features four key transformations: 1) a catalytic intramolecular oxidative cyclization for the delta-lactamindole backbone; 2) an oxidative cyclic aminal formation for the hexahydropyrrolo[2,3-b]pyrrole framework; 3) a transannular radical cyclization for the construction of the diazabicyclo-[3.3.1]nonane structure; and 4) a one-pot desilylation/double epimerization reaction that affirms the relative stereochemistry.

Abstract:A concise total synthesis of amphidinin B, a cytotoxic linear dicarboxylic acid associated with amphidinolide T marine macrolides, has been accomplished from the 19-membered cycloalkene intermediates designed for diverted total synthesis of amphidinolide T1 and T3T5. The 1H and 13C NMR spectra of the synthesized amphidinin B in C6D6 and CDCl3 were compared with those for the natural and synthetic forms reported in the literature, showing solvent-dependence of the 1H and 13C NMR signals of amphidinin B in C6D6 and CDCl3.

Abstract:The Daphniphyllum alkaloids are a large class of natural products isolated from a genus of evergreen plants widely used in Chinese herbal medicine. They display a remarkable range of biological activities, including anticancer, antioxidant, and vasorelaxation properties as well as elevation of nerve growth factor. Daphenylline is a structurally unique member among the predominately aliphatic Daphniphyllum alkaloids, and contains a tetrasubstituted arene moiety mounted on a sterically compact hexacyclic scaffold. Herein, we describe the first total synthesis of daphenylline. A gold-catalysed 6- exo -dig cyclization reaction and a subsequent intramolecular Michael addition reaction, inspired by Dixon's seminal work, were exploited to construct the bridged 6,6,5-tricyclic motif of the natural product at an early stage, and the aromatic moiety was forged through a photoinduced olefin isomerization/6 π -electrocyclization cascade followed by an oxidative aromatization process.

Abstract:A new method for synthesis of synthesis 6-hydroxy-3, 4-dihydro-2(lH) quinolinone was investigated with higher yield and simpler operation. The title compound was prepared starting from 3-chloropropionyl chloride, via cyclizing, nitration, reduction, diazotization and hydrolysis. The total yield is 67.7%, and the yield of each step is between 86.5% -99.6%. 6-Hydroxy-3, 4-dihydro-2(1H)quinolinone was characterized by MS and 1H NMR.

Abstract:A unified modular synthetic strategy has been developed for the first total synthesis of dictyodendrins G and synthesis of dictyodendrin F, H and I in 11 steps. The synthesis features consecutive functionalization of the core aminoquinone by palladium-mediated Suzuki Miyaura coupling reaction, 1,4-addition, acylation and base mediated formation of a pyrrolinone, and the formation of carbazolequinone moiety through a formal [3 + 2] cydoaddition using arynes generated in situ. Several dictyodendrin analogues were also synthesized using this strategy.

Abstract:1-Allyl-6-chloro-7-methyl-1,4-dihydro-quinoxaline-2,3-dione,which was designed to prepare a series of N-methyl-D-aspartate(NMDA) receptor antagonists,was synthesized from 1-chloro-4-fluoro-2-methylbenzene.1-Chloro-4-fluoro-2-methyl-5-nitrobenzene was obtained in 94.0% yield by nitration of the starting material in concentrated sulfuric acid.Amination with allylamine under refluxing gave allyl-(4-chloro-5-methyl-2-nitrophenyl)-amine in 81.0% yield and triethylamine was served as base in this reaction.Reduction of the nitro group by iron powder afforded amino compound in 95.0% yield,which by cyclization with oxalic acid dihydrate gave the target compound in 90.6% yield.The total yield from the starting material to the target product is 65.5%.

Abstract:Six undescribed quinoline alkaloids, named dasycarines A-E, and 18 known ones were isolated from the root bark of Dictamnus dasycarpus. All the structures were elucidated on the basis of comprehensive analysis of UV, IR, NMR, and HRMS spectroscopic data, and the absolute configurations were assigned via comparison of the calculated and experimental ECD data. (+)-Dasycarine A (1a) and (-)-Dasycarine A (1b) are a pair of enantiomers of dimeric furoquinoline alkaloid, which are the first dimeric via [2 + 2] cycloaddition of furan. The structure and absolute configuration of ( - )-dasycarine A was determined via X-ray crystallography. Additionally, all the isolated compounds were tested for their inhibitory effects on NO production stimulated by LPS in BV-2 microglial cells. Three compounds showed strong inhibition with IC50 values below 5.0 mu M; nine compounds exhibited inhibition with IC50 values in the range of 7.8-28.4 mu M. Furthermore, we demonstrated that preskimmianine suppressed the levels of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6), and nuclear factor kappa B (NF-kappa B) in LPS-induced BV-2 microglial cells.

Abstract:Daphenylline, a structurally unique triterpenoid Daphniphyllum alkaloid, has attracted considerable attention from the synthetic community since its isolation. Herein, we delineate an enantioselective total synthesis of (‐)‐daphenylline, which is based on a gold‐catalyzed dearomatization reaction of β‐methoxynaphthalene. Other features include installation of the initial chiral stereocenter via an enantioselective rhodium‐catalyzed hydrogenation of an indene derivative; construction of the other vital stereocenter by organocatalytic asymmetric Mukaiyama–Michael reaction of a structurally complex benzofused cyclohexanone; and a tandem reductive amination/amidation double cyclization reaction to assemble the distinctive azapolycyclic cage‐like architecture.

Abstract:Efforts toward the enantioselective synthesis of Daphniphyllum alkaloid calyciphylline N which leads to efficient preparation of the ABCF tetracyclic framework containing three bridgehead all-carbon quaternary stereocenters are described. This synthetic work features the utilization of an asymmetric conjugate addition to install the C5 all-carbon quaternary center, an efficient successive inter/intramolecular aldol sequence to build the critical bicyclo[2.2.2]octanone BC core, and a ring closing metathesis reaction followed by stereoselective Nagata conjugate cyanation to deliver the functionalized F ring.

Abstract:One of the largest subfamilies within the famous Daphniphyllum alkaloid family is made up of the yuzurimine-type (or macrodaphniphyllamine-type) alkaloids. Their complex aza-polycyclic caged structures, several contiguous stereogenic centers, and vicinal all-carbon quaternary centers make these alkaloids formidable challenge for synthetic chemists. Recently, synthesis of these alkaloids has received extensive attention from our community. Herein, we wish to report the total synthesis of C14-epi-deoxycalyciphylline H, a putative member of yuzurimine-type alkaloid subfamily. Key transformations employed in our approach include an intramolecular Prins reaction and a Pd-catalyzed enyne cycloisomerization. In addition, synthesis of a daphnezomine L-type alkaloid, paxdaphnidine A, was also studied.

Abstract:N-methyl-4-nitro aniline was subjected to a sequential chloroacetylation , nucleophilic sub-stitution and catalytic hydrogenation sequence achieving a key intermediate N-( 4-aminophenyl )-N-methyl-2-( 4-metrylpiperazin-1-yl ) chloroacetamide ( 4 ) .6-Methoxycarbonyl-2-oxindole ( 8 ) was pre-pared from 4-chlorine-3-nitro benzoic acid via a four-step reaction of esterification , nucleophilic substi-tution, catalytic hydrogenation and cyclization .Then, another key intermediate , methyl-1-( acetyl)-3-[ methoxy ( phenyl ) methylene ]-2-oxoindoline-6-carboxylate ( 9 ) was prepared by a "one-pot" process of compound 8 in the presence of triethyl orthobenzoate and acetic anhydride .Nintedanib with an over-all yield of 57 .2％was synthesized via a"one-pot" process of substitution of 4 with 9 and then depro-tection.The structure was confirmed by 1H NMR, 13C NMR and MS(ESI).

Abstract:A simple and efficient synthetic route for preparing the benzonaphthyridine framework is reported. Only seven steps are needed for the assembly of 3-alkylamino aaptamine from inexpensive isoquinoline 6 by this route with about 20% overall yield. The two key steps are a novel palladium-catalyzed reductive cyclization with Mo(CO)6 as reductant to form aaptamine and demethyloxyaaptamine and a hydrogen-bond-mediated oxidative alkylamination to account for the complete regioselectivity.

Abstract:Two new quinoline alkaloids ( 1 – 2 ) together with twenty-two known alkaloids ( 3 – 24 ) were isolated and identified from Dictamnus dasycarpus Turcz. Compounds 6 – 7 , 9 , 11 , 15 – 16 , 19 and 24 were isolated from D. dasycarpus for the first time. The structures of all compounds were characterised by spectroscopic methods (1D, 2D NMR and HRESIMS). The anti-proliferative activity was mediated by the arrest of three human cancer cell lines (SW982, HepG2 and A549) of all the compounds that were evaluated by CCK-8 assay.

Abstract:5,7-Dichloro-2,3-dihydroquinolin-4-(1H)-one is an important intermediate for the synthesis of 4,5,7-trichloroquinoline, which can be synthesized from 3,5-dichloroaniline and acrylic acid. The obtained condensa-tion 3-(3,5-dichloroanilino)-propionic acid via cyclization in polyphosphoric acid for 3 hours to give 5,7-dichloro-2,3-dihydroquinolin-4-(1H)-one, yield 85%.
