Abstract:Equilibrium geometries and electronic structures of complexes between β -cyclodextrin ( β -CD) and some small molecules as well as monosaccharides were investigated by Austin Model 1 (AM1) to obtain binding energy of the complexes. It was indicated that β -CD could bind the structurally similar solvent molecules and monosaccharides because of the negative binding energy of the complexes, and especially could show the chiral binding ability to monosaccharides with more hydroxyl groups, due to its chiral characteristics. The complexes were stabilized by the hydrogen bonding between β -CD and guests. Based on the AM1 optimized geometries, the IR spectra were calculated by AM1 method. Vibration frequencies of O-H bonds in the guests were red-shifted owing to the weakening of the O-H bonds with the formation of the complexes.

Abstract:Studies of cyclodextrin chemistry by quantum chemical methods are briefly surveyed. Emphases are put on what types of quantum chemical methods can be used for cyclodextrin chemistry, how to use quantum chemical methods to find the global minimum, to study the structures, binding energies, driving forces for cyclodextrin complexes, as well as chemical reactions occurring inside cyclodextrin cavities. Problems associated with the application of quantum chemical methods in cyclodextrin chemistry are also discussed.
A Quantum-Chemical Study on the Molecular Recognition of Β-Cyclodextrin with Ground and Excited Xanthones

Abstract:PM3 and density function theory B3LYP/3-21G(d) calculations in vacuo and in water were performed on the inclusion complexation of β-cyclodextrin (CD) with the ground singlet and excited triplet xanthones. It revealed that the complex of β-CD with the singlet xanthone was significantly more stable than that with the triplet one, which agreed with the experimental observation. Calculations on the model system at the level of B3LYP/6-311G(p, d) supported the above result, which indicated that the repulsion between the oxygens of xanthone and the oxygens of the secondary hydroxyls of β-CD constituted the origin for the above behavior. Hence, caution should be given when extrapolating excited state behavior to the supramolecular systems in their ground state.

Abstract:This article aims to review the application of various quantum chemical methods (semi-empirical, density functional theory (DFT), second order Møller–Plesset perturbation theory (MP2)) in the studies of cyclodextrin host–guest complexes. The details of applied approaches such as functionals, basis sets, dispersion corrections or solvent treatment methods are analyzed, pointing to the best possible options for such theoretical studies. Apart from reviewing the ways that the computations are usually performed, the reasons for such studies are presented and discussed. The successful applications of theoretical calculations are not limited to the determination of stable conformations but also include the prediction of thermodynamic properties as well as UV–Vis, IR, and NMR spectra. It has been shown that quantum chemical calculations, when applied to the studies of CD complexes, can provide results unobtainable by any other methods, both experimental and computational.

Abstract:For the excellent complex and recognition function,cyclodextrins have attracted wide research interest. In order to utilize these special properties of cyclodextrins more effectively, immobilization of cyclodextrins,with specific orientation,upon solid substrate is necessary in many potential applications. This paper summarizes the common methods to prepare monolayer/multilayers of cyclodextrins on solid substrate,including the Langmuir-Blodgett method,Layer-by-Layer technique,and chemical adsorption. These methods are compared and evaluated. The application situation of these systems is introduced. Finally,based on preparations and applications,the future research directions of cyclodextrins monolayer/multilayers membranes are discussed. Improving routine methods and developing simpler methods will be the key research of this area in the future.
Studies on Molecular Recongnition of (A)-Cyclodextrin with Diphenyl Compounds

Abstract:The cyclodextrins(CDs) are a class of cyclic oligosaccharides made up of six(a), seven(a)or more [a-(1,4)-linked] D-glucopyranose units, and shaped like truncated cones with primary and secondary hydroxyl groups crowning the narrower rim and wider rim respectively. As they have a hydrophobic cavity of appropriate dimension, they can bind with various guest moleculars, such as hydrocarbon, cyclohexane, aromatic compounds, to form inclusion complexes. The cyclodextins inclusion complexation has been considered an ideal model mimicking the enzyme-substrate interaction and a lot of effect has been devoted to it. In this paper, we report our investigation on the inclusion complexation behavior of a-cyclodextrin(a-CD) with diphenyl compounds in order to further explore the molecular recongnition mechanism of 2:1 inclusion complexation of a-CD with aromatic compounds.Figure 1: Possible structures of the inclusion compounds.The inclusion complexation behavior of a-CD with sym-diphenyl-urea, sym-diphenyl-thiourea and diphenyl kotone as respective guest moleclars was studied by ultraviolet spectrometric titrations.The absorption spectral changes observed for the compounds in the absence and presence of a-CD are used to draw the corresponding Benesi-Hildebrand plots and caculate the complex stability constant value (Ks) for the inclusion compounds.The 2:1 inclusion complexations show higher binding constants by cooperative binding of one guest molecular in the closely two hydrophobic cyclodextrin cavites as compared with 1:1 inclusion complexations.The highest value observed for sym-diphenyl-urea could be due to the formation of a hydrogen bond between the carbonyl group and the hydrogen in the hydroxyl group of CD and this is not possible with sym-tiphenyl-thiourea. The lowest value observed for diphenyl kotone indicate the hydrophobic interaction is one of the binding force of cyclodextrin inclusion complex.

Abstract:As a versatile tool in separation science, cyclodextrins and their derivatives, known as emerging functional monomers, have been used extensively in molecular imprinting techniques. The attributes of cyclodextrins and their derivatives are widely known to form host-guest inclusion complex processes between the polymer and template. The exploitation of the imprinting technique could produce a product of molecularly imprinted polymers, which are very robust with long-term stability, reliability, cost-efficiency, and selectivity. Hence, molecularly imprinted polymers have gained popularity in chemical separation and analysis. Molecularly imprinted polymers containing either cyclodextrin or its derivatives demonstrate superior binding effects for a target molecule. As noted in the previous studies, the functional monomers of cyclodextrins and their derivatives have been used in molecular imprinting for selective separation with a wide range of chemical compounds, including steroidals, amino acids, polysaccharides, drugs, plant hormones, proteins, pesticides, and plastic additives. Therefore, the main goal of this review is to illustrate the exotic applications of imprinting techniques employing cyclodextrins and their derivatives as single or binary functional monomers in synthesizing molecularly imprinted polymers in areas of separation science by reviewing some of the latest studies reported in the literature.

Abstract:Cyclodextrins are macrocyclic rings composed of glucose residues. Due to their remarkable structural properties, they can form host-guest inclusion complexes, which is why they are frequently used in the pharmaceutical, cosmetic, and food industries, as well as in environmental and analytical chemistry. This review presents the reports from 2011 to 2023 on the quantitative structure-activity/property relationship (QSAR/QSPR) approach, which is primarily employed to predict the thermodynamic stability of inclusion complexes. This article extensively discusses the significant developments related to the size of available experimental data, the available sets of descriptors, and the machine learning (ML) algorithms used, such as support vector machines, random forests, artificial neural networks, and gradient boosting. As QSAR/QPR analysis only requires molecular structures of guests and experimental values of stability constants, this approach may be particularly useful for predicting these values for complexes with randomly substituted cyclodextrins, as well as for estimating their dependence on pH. This work proposes solutions on how to effectively use this knowledge, which is especially important for researchers who will deal with this topic in the future. This review also presents other applications of ML in relation to CD complexes, including the prediction of physicochemical properties of CD complexes, the development of analytical methods based on complexation with CDs, and the optimisation of experimental conditions for the preparation of the complexes.

Abstract:In this paper, the structure and properties of cyclodextrins (CDs) with hydrophilic external and hydrophobic internal cavities are presented. For the special molecular structure of CDs, the characteristics and performances of membranes will be improved by introducing CDs and their derivatives into the membrane. The preparation methods and applications of membranes with CDs are reviewed, including separation of isomers, transfer of metal ions, controlled release of drugs, drug detection and analysis, hydrolyzation of esters etc. The study on molecular conduit of CDs is summarized. Based on these reviews, the prospect of membrane with CDs is discussed.

This review comprehensively explores the latest advancements in cyclodextrin chemistry, focusing on the synthesis and characterization of cyclodextrin derivatives and their inclusion complexes. This review examines the biological activities of cyclodextrins, highlighting their pharmacological properties and pharmacokinetics, and discussing their promising applications in drug delivery systems. Furthermore, the industrial utilization of cyclodextrins, including their role in nanomaterials and nanostructured coatings, as well as their potential in environmental remediation, are explored. The present research also addresses the critical aspect of toxicity, particularly concerning cyclodextrin inclusion complexes, providing an overview of the current understanding and safety considerations. Abstract:Cyclodextrins (CDs) are highly respected for their ability to form inclusion complexes via host–guest noncovalent interactions and, thus, ensofance other molecular properties. Various molecular modeling methods have found their applications in the analysis of those complexes. However, as showed in this review, molecular dynamics (MD) simulations could provide the information unobtainable by any other means. It is therefore not surprising that published works on MD simulations used in this field have rapidly increased since the early 2010s. This review provides an overview of the successful applications of MD simulations in the studies on CD complexes. Information that is crucial for MD simulations, such as application of force fields, the length of the simulation, or solvent treatment method, are thoroughly discussed. Therefore, this work can serve as a guide to properly set up such calculations and analyze their results.

Abstract:Cyclodextrins (CDs) are a class of macrocyclic molecules that have exhibited many promising applications in various fields. The knowledge of the complexation modes and recognition mechanisms of CDs with their guests are of paramount importance for rational design of more variants with controlled properties. Herein we investigated the binding conformations and the structural characteristics of α-/β-CD with three amino acids (AA, AA=Gly, L-Leu, L-Phe) in the gas phase by a combined experimental and computational approach. Electrospray ionization-mass spectrometry suggested the formation of 1:1 anionic complexes between CDs and AAs and the complex anions were further identified by tandem mass spectrometry. Moreover, ion mobility-mass spectrometry experiments revealed the inclusion complexation adopted for [α-CD+Gly]- as well as β-CD with either amino acid, whereas [α-CD+Leu]- and [α-CD+Phe]- favored an exclusion conformation, indicating size-dependent binding modes. The association is primarily driven by polar interactions via the formation of hydrogen bonds. Furthermore, the relative dynamic stabilities of the complex ions were observed to be in correlation with the gas-phase basicities of the deprotonated amino acid and CD anions. These above findings are well in line with our atomistic molecular dynamics simulation results. This study advances our understanding of the mechanisms underlying CD host-guest recognition.

Due to their excellent biologic and optical properties, cyclodextrins have recently attracted increasingly more attention. In this review the newest development of the supramolecular self-assembly based on cyclodextrin is summarized. The article provided detail information about the synthesis and applications of both of rotaxanes formed by cyclodextrin ring and polymer axes, and supramolecular polymer without polymer backbone.

Abstract:This study involves initial Hartree-Fock and Density Functional theory calculations on the molecular recognition of the cyclodextrins. The alpha-cyclodextrin-acetophenone complexation system was investigated with PM3, HF/3-21G* and B3LYP/3-21G* methods. The results indicated that the inclusion orientation in which the acetyl group of the acetophenone points towards the secondary hydroxyls of the a-cyclodextrin was preferable in energy. The steric effect was supposed as the physical reason of such a behavior. Hence, the simple rule the anti-parallel arrangement of the dipoles of the host and guest molecules in the cyclodextrin complexation is not generally applicable.

Abstract:Cyclodextrins (CDs) are a series of cyclic oligosaccharides formed by amylose under the action of CD glucosyltransferase that is produced by Bacillus. After being modified by polymerization, substitution and grafting, high molecular weight cyclodextrin polymers (pCDs) containing multiple CD units can be obtained. pCDs retain the internal hydrophobic-external hydrophilic cavity structure characteristic of CDs, while also possessing the stability of polymer. They are a class of functional polymer materials with strong development potential and have been applied in many fields. This review introduces the research progress of pCDs, including the synthesis of pCDs and their applications in analytical separation science, materials science, and biomedicine.

Abstract:Cyclodextrin (CD) derivatives have recently gained worldwide attention, which have versatile advantages and restrained the defects of parent CDs. The superior properties of CD derivatives in encapsulation, stabilization, and solubilization facilitate their application in food, biomedicine, daily chemicals, and textiles. In this review, the preparation, classification, and main benefits of CD derivatives are systematically introduced. By introducing targeted groups into the parent CD molecule, they exhibit significant improvement in their required characteristic. Besides, the important point closely related to application, the safety assessment, has also been highlighted. Most tested CD derivatives have been verified to be relatively safe in a limited dosage. Then, the applications of CD derivatives have been described in detail from the food to its related field. In food field, CD derivatives play an important role in the stability and bioavailability of bioactive compounds, control flavor release, and improve the antimicrobial and antioxidant properties of packaging materials. These advantages can also be expanded to the related field, offering innovative solutions that enhance product quality, human health, and environmental sustainability. This review highlights the broad applications and potential of CD derivatives, underscoring their role in driving advancements across multiple industries.

Abstract:Highly stable inclusion complexes between cyclodextrin (CD) and guest molecules form due to their host-guest interaction. Polymer systems based on the complexes have been extensively reported in recent years. In this review, we highlight some recent advances from polymer systems with adamantane (Ada), azobenzene (Azo), ferrocene (Fc) and so on as guest molecules in terms of their synthesis, properties and functions. Various polymer topologies ranging from linear, branched, comb-like and hyper-branched are discussed in detail.
