Effects of Ethylene on Cut Flower Opening and Senescence and Expression of Ethylene Receptor Genes in Wintersweet (Chimonanthus praecox)

Abstract:To investigate whether ethylene involved in the regulation of lfower opening and senescence in win-tersweet, ethylene production of lfowers at different stages was measured with gas chromatography. Meanwhile, cut lfowers were treated with ethylene and 1-methylcyclopropene (1-MCP), an inhibitor of ethylene receptor, respectively. Then the process of lfower opening and senescence was observed and the expression of ethylene receptor genes was analyzed. The results showed that endogenous ethylene was produced slightly, with a maxi-mum amount at full bloom stage of lfowers. Exogenous ethylene treatment signiifcantly accelerated the process of lfower opening and senescence, resulted in a shortened vase life by 1.9 days compared to the control. In con-trast, treatment with 1-MCP extended the vase life by 2.4 days compared to control. Three ethylene receptor genes,CpETR-1, CpETR-2 andCpETR-3, were detected in wintersweet lfowers. Their expressions were en-hanced by exogenous ethylene and inhibited by 1-MCP treatment, and the changes in the expression of tran-scriptional level were closely related to the process of lfower opening and senescence. These results above indi-cated that although ethylene production in wintersweet lfowers was slight, ethylene also involved in regulation of lfower opening and senescence, and inlfuenced the expression of ethylene receptor genes.
