Abstract:The chemical stability of L?glutamine was studied when ion exchanging took place on the four kinds of strong acidic ion exchange columns (H form).Under the described conditions,L?glutamine was fairly stable.100 ml centrifugal supernatant from glutamine broth contained Gln 312mmol/L,Glu 67.0 mmol/L,0.75 mol/L(NH 4) 2SO 4 etc.;the column's bed volume and supernatant loading rate was 200 ml and 1.0 BV/h respectively;elution rate by 0.5 mol/L ammonium solution was 0.5 BV/h.
Separation of glutathione by aqueous two-phase extraction combined with temperature-induced phase separation

Abstract:The techniques for separation of glutathione (GSH) by aqueous two-phase extraction combined with temperature-induced phase separation were studied. The partition behavior of GSH in ethylene oxide-propylene oxide random copolymer (EOPO)/hydroxypropyl starch (PES)/water systems was investigated. The effects of such factors, as concentration of polymer, pH, on the partition coefficients in two separation processes (aqueous two-phase extraction and temperature-induced phase separation) were studied. The optimum conditions are EOPO4000 13%, PES100 10%, pH=10.5. The separation technique for GSH from yeast cell was also investigated. A feasible separation scheme was suggested, and the results show that the total extraction yield can reach over 80%.

Abstract:Molecular interactions between ligands and target biomolecules are crucial in the development of chromatographic techniques for the separation and purification of biotherapeutics. In this study, the role of functional moieties on a mixed-mode ligand (phenylalanine-tyrosine-glutamate-5-aminobenzimidazole) for human immunoglobulin G purification was investigated and a detailed mechanism was discussed. A similar ligand with glutamic acid substituted by glutamine (phenylalanine-tyrosine-glutamine-5-aminobenzimidazole) together with other resins including a commercial resin (CM Bestarose Fast Flow), phenylalanine-tyrosine-glutamate, glutamate-5-aminobenzimidazole, and 5-aminobenzimidazole resins were prepared for comparison. Molecular dynamics simulation and experimental studies were used to analyze the difference between these ligands. The results showed that the carboxyl group of phenylalanine-tyrosine-glutamate-5-aminobenzimidazole contributed 70% of the electrostatic interaction during human immunoglobulin G binding, and 5-aminobenzimidazole provided electrostatic repulsion for desorption, which showed low selectivity and binding capacities at pH 4.0 (dynamic binding capacities at 10% breakthrough of human immunoglobulin G = 1.0 mg/ml resin, dynamic binding capacities at 10% breakthrough of human serum albumin = 1.2 mg/ml resin) when used as an individual resin ligand. The results showed in this study demonstrated that it is possible to achieve optimal antibody separation and purification through reasonable ligand design by understanding the performance of key functional moieties in binding and elution processes.

Abstract:Optically pure D-glutamine is initially prepared by biotransformations of D,L-glutamine in the presence of L-glutamine decarboxylase in Escherichia coli AS1.505.Factors that affected the enzymatic activity such as converting temperature,pH value,etc.,were studied.The results show that the specific enzyme's activity can come up to 4200U,and L-glutamine can be completely degraded by the decarboxylase for 8 h under the optical conditions as follows:37℃,4.8 of pH value,5 g/L of cell concentration,0.15 g/L of tween-80,14 h of strain age,and 40 g/L of the substrate concentration.

Abstract:In the overall alkaline stability for guanidinium cations, the substituted mode has been proven to play a very important role. Free and bulky alkyl substituents such as ethyl groups in guanidinium cations can provide large steric hindrance to resist the OH- attack and achieve much higher stability than conventional methyl-substituted ones. To obtain high-dimensional and good alkaline stabilities of anion exchange membranes (AEMs), linking the stable guanidiniums through suitable crosslinked structures (with different spacer structures or lengths) is a feasible strategy. In this study, bi-guanidinium-based crosslinked AEMs were prepared by Menshutkin reactions using brominated poly(2,6-dimethyl-phenylene oxide) and bi-guanidine crosslinkers followed by a complete quaternization of the remaining guanidine. The prepared membranes possess a low swelling ratio (<15% even at 80 degrees C), good thermal stability, and excellent mechanical properties. Alkaline stability test results showed that the bi-guanidinium crosslinked AEMs have excellent resistance to alkaline solutions, and more than 90% cations remained after immersing in 1 mol L-1 of NaOH solution at 60 degrees C for 30 days. These results suggest that crosslinking structures are effective in restraining the excess swelling and that delocalized guanidiniums are promising cations for application in AEMs.
[The Chemical Decomposition of Glutamine and Its Effect on Hybridoma Cell Culture].

Abstract:The chemical decomposition of glutamine is a first-order reaction. Its reaction constants under storage and culture conditions were determined as 0.0009 h-1 and 0.0032 h-1 respectively. Batch culture of hybridoma cell C50 with different initial contents of glutamine helped to understand its real metabolic characteristic. The results show that when the initial concentration of glutamine is lower than 5 mmol/L, more than 80% is used by cells. And the lower the initial content, the more being used. As the initial glutamine concentration increases, the ratio of its utilization decreases. When it reaches 10 nmol/L, the ratio decreases dramatically.
A general strategy to enhance the alkaline stability of anion exchange membranes

Abstract:Quaternary ammonium (QA)-type polysulfone (PSf) anion exchange membranes (AEMs) with pendant carbonyl groups were synthesized by an environmentally friendly method. Low toxicity 3-chloro-2,2dimethylpropionyl chloride (CDPC) was chosen as an electrophilic reagent to introduce chloroalkyl groups to PSf, instead of highly toxic chloromethyl methyl ether. The experimental conditions were carefully optimized to avoid the Friedel-Crafts alkylation side reaction on the chloroalkyl group. After quaternizing with trimethylamine (TMA), AEMs were prepared by solution casting. The obtained AEMs showed good ionic conductivity and mechanical stability. The alkaline stability of these AEMs was excellent; the AEMs can maintain 95% of their initial conductivity after immersion in a 1 M NaOH aqueous solution at 80 degrees C for 10 days. The reason for this outstanding alkaline stability was evaluated by NMR and density functional theory calculations. The most severe degradation pathway, the S(N)2 reaction, can be weakened by the competing reaction between OH- and carbonyl groups. The formed intermediate oxygen anion can decrease the possibility of SN2 by reducing the electro-static potential of the QA groups. Because of the reversibility of the reaction between OH- and carbonyl groups, QA groups can survive longer in strong alkaline conditions at high temperature. Also, the possibility of S(N)2 reaction on the aromatic polymer chain can be decreased because of the positive charge carrier, QA groups, which are not directly attached to benzyl groups. Hoffmann elimination can not happen because there was no hydrogen atom on the beta-position of QA group. These structural advantages of the synthesized AEMs suggest another strategy that the potential for generalisation to other AEM systems and to improve the alkaline stability of QA-type AEMs.

Abstract:The mole fraction solubility of L-glutamine in 12 pure solvents (methanol, n-propanol, sec-butanol, 2-butanone, ethanol, acetonitrile, n-butanol, ethyl acetate, n-pentanol, water, n-hexane, and isobutanol) was determined by the static gravimetric method at a temperature range of 283.15–323.15 K under a pressure of 98.8 kPa. The equilibrium solid phase of L-glutamine in the solvent systems was characterized by powder X-ray diffraction analysis. The solubility in the 12 monosolvents was found to increase with absolute temperature. The maximum and minimum values were in water and ethyl acetate, respectively. The empirical solvent polarity parameters (E T(30)), cohesive energy density, and hydrogen bond donor/acceptor (HBD/HBA) tendencies were employed to analyze the solubility behavior and solvent effects. The solubility data were fitted using the modified Apelblat model and the Yaws model, at the same time, the model parameters and data deviation values were calculated. These two solubility models were evaluated using the Akaike Information Criteria and Akaike weights. The results showed that the modified Apelblat model had better correlation results. The solubility data could be used in the preparation and optimization of L-glutamine crystallization processes.

Abstract:A new technique for separation of glutamine from its fermentation broth was developed by using one-step anion exchange, based on the difference of the ionization constants of glutamic acid (Glu) and glutamine (Gln). When the solution of crude glutamine crystal from the fermentation was separated by single anion exchange column, the highest recovery of Gln was 83%, and the removal ratio of Glu reached 90%. These results show that this new process is effective in separation of glutamine from the fermentation broth and reduction of the cost.

Abstract:This research focused on the unique aggregation behavior of l-Lysine (L-Lys) and l-Arginine (L-Arg) on quinoa protein (QP) gelation under microwave (MW) irradiation. The results of pH and conductivity manifested that the discrepancy of the influence of two alkaline amino acids on protein was due to the diversity of their electrochemical characteristics rather than to aggregation induced by pH-dominated acid-base transitions. Solubility, turbidity and particle size, ζ-potential results exhibited that L-Lys enhanced the ordered aggregation of protein induced by MW heating more than L-Arg. Circular dichroism (CD), UV absorption spectra, surface hydrophobicity (H0) and sulfhydryl groups demonstrated that L-Lys denaturized and unfolded proteins more vigorously than L-Arg, exposing more hydrophobic residues and boosting the hydrophobicity of the protein's surface. These findings emphasized that L-Lys and MW had a more pronounced synergistic effect on the strength and water holding capacity (WHC) of protein gels than L-Arg. Scanning electron microscopy (SEM) also revealed a denser porous network microstructure in the QP gels with L-Lys added.

Abstract:Imidazolium cations with various N3-substituents (including methyl, butyl, heptyl, dodecyl, isopropyl, and diphenylmethyl groups) were synthesized and investigated in terms of their alkaline stability. The effect of the N3-substituent on the alkaline stability was studied by quantitative 1H NMR spectra and density functional theory (GGA-BLYP) calculations. The isopropyl substituted imidazolium cation ([DMIIm]+) with the highest LUMO energy value exhibited the highest alkaline stability in aqueous NaOH. The [DMIIm]+ cation also exhibited higher alkaline stability than that of a quaternary ammonium cation, benzyltrimethyl-ammonium ([BTMA]+), in CD3OD/D2O NaOH solution at elevated temperatures. This observation inspired the preparation of [DMIIm]+-based alkaline anion exchange membranes (AEMs) which showed high alkaline stability in alkaline solution.
Application of Electrodialysis Desalination to the Separation of Glutamine from Fermentation Broth

Abstract:A new technique of one-step anion exchange for purification of glutamine from its fermentation broth was developed by using electrodialysis desalination to remove (NH4)2SO4 existing in the broth. Adjusting the pH of broth to the the isoelectric point of glutamine, most (NH4)2SO4 and only a little glutamine would transfer from the broth to the adjacent chamber through ion membrane during electrodialysis operation. When the desalination ratio reached 95%, the recovery ratio of glutamine and the current efficiency were 76% and 85%, respectively. After electrodialysis desalination, glutamine could be separated successfully from glutamic acid by passing the retained broth through a single anion exchange column. Experimental results show that this new process is effective in decreasing the amount of resins, preventing the decomposing of glutamine in strong acid and alkaline environment, and in the reduction of the production cost. The total yield of glutamine reaches about 60% and the purity of product satisfies the pharmaceutical standard.

Abstract:The sulfate in the ion-exchange wastewater from monosodium L-glutamate production was removed with flocculation and precipitation process.The orthogonal experiment showed that the optimum levels of influence factors were as follows: original concentration(SO_4~(2-) 35.526 g/L),pH 3.2,Ca~(2+)/SO~(2-) ratio of 1.5,addition of neither poly aluminium chloride nor polyacrylamide.The contribution of influence factors to sulfate removal was,in decreasing order,wastewater concentration,Ca~(2+) dosage,pH,dosing sequence and coagulant dosage.On the condition that the Ca~(2+) dosage was equal to the SO_4~(2-) amount in the original wastewater,the sulfate removal efficiency was above 90% and the supernatant was clean after reaction for 15 min.The water content of the CaSO_4 sediment was about(81.4%) after being settled for 24 h.It is highly recommended to remove the sulfate from the wastewater at its original concentration rather than using the diluted wastewater,because the sulfate removal efficiency was lower,the supernatant was turbid and the water content of the CaSO_4 sediment was higher if the diluted wastewater was used.

Abstract:The solubility behavior of Boc-l-glutamine in water, ethanol, n-propanol, isopropanol, n-butanol, isobutanol, n-pentanol, acetone, 2-butanone, acetonitrile, methyl acetate, and ethyl acetate was investigated by the static gravimetric method at 98.8 kPa and multiple temperatures from 283.15 to 323.15 K. The equilibrium solid-phase characterization of Boc-l-glutamine in different solvent systems was carried out by the powder X-ray diffraction analysis. In 12 pure solvents, the solubility increased with absolute temperature, and the maximum value was recorded in ethanol and the minimum in ethyl acetate. The solubility behavior and solvent effects in different solvents were first analyzed by the empirical solvent polarity parameters (ET (30)) as the main factor and then interpreted by hydrogen bond acceptor tendencies, cohesive energy density, structural similarity, steric effect, and solvent viscosity for some exceptions respectively. Furthermore, the Yaws model and the modified Apelblat model were employed to fit the solubility data. Akaike Information Criterion and Akaike weights were calculated to evaluate the two solubility models. The results show that the modified Apelblat model has better correlation results than the Yaws model. The results could provide a theoretical basis for the production and application of Boc-l-glutamine in organic or biological process development.

Effects of Broth Components on Separation of L-glutamine by Nanofiltration

Abstract:The effects of salt ions and other components in L-glutamine fermentation broth on the separation of L-glutamine and L-glutamic acid by nanofiltration were investigated and the effects of process operation parameters on permeation properties were studied systematically. The optimum dilution times of fermentation broth and temperature was established. The study also showed that bivalent cations could affect the permeation properties of L-glutamine and L-glutamic acid, and decreased observed rejection of glutamic acid by 8%. Higher concentration of (NH4)2SO4 in broth reduced the observed rejection of L-glutamine and L-glutamic acid and permeation flux greatly, but higher concentration of NH4Cl did not, which demonstrated different repulsion forces of membrane to different ions. When the broth was diluted to 1% for L-glutamine concentration and adjusted to pH=7, other salt ions and residual glucose in broth would not significantly influence L-glutamine separation by nanofiltration due to their low concentrations.

Abstract:A guanidinium-functionalized anion exchange membrane (AEM) was prepared and characterized. The AEM stability in an alkaline environment and transition structures during the degradation process were studied with DFT (density functional theory)/B3LYP method, 6-31 + G (d) basis set. Experimental results and theoretical analysis showed that the second step (Step 2′) of degradation reaction was the control procedure; guanidinium cation was unstable under alkaline condition. It had lower energy barriers, which decided it was easier for the degradation reaction to occur in high pH environment. The ionic conductivity of AEM was 2.4 × 10−2 S cm−1 at 80 °C. However, the AEM soaked in 1 M NaOH solution, fragments were found after 10 days. The FTIR analysis showed that the structure of the membrane had been changed due to the attack of hydroxide ion. A new substance, tetramethylurea, had produced.

Abstract:Ion exclusion method was used to purify sodium chloride-containing β-alanine.Different types of strong-acid cation exchange resins and operation conditions were studied.The suitable parameters were:50～100 mesh exchange resin of model 001×8,feeding volume 15%～20% BV(bed volume),flow rate 1/120～1/60 BV/min,concentration of feeding solution:β-alanine 4%～8%,NaCl 7%～14%.

Abstract:Imidazolium cations with butyl groups at various substitution positions (N1-, C2-, and N3-), 1-butyl-2,3-dimethylimidazolium ([N1-BDMIm](+)), 2-butyl-1,3-dimethylimidazolium ([C2-BDMIm](+)), and 3-butyl-1,2-dimethylimidazolium ([N3-BDMIm](+)), were synthesized. Quantitative (1)H NMR spectra and density functional theory calculation were applied to investigate the chemical stability of the imidazolium cations in alkaline solutions. The results suggested that the alkaline stability of the imidazolium cations was drastically affected by the C2-substitution groups. The alkaline stability of imidazolium cations with various substitution groups at the C2-position, including 2-ethyl-1-butyl-3-methylimidazolium ([C2-EBMIm](+)), 1,2-dibutyl-3-methylimidazolium ([C2-BBMIm](+)), and 2-hydroxymethyl-1-butyl-3-methylimidazolium ([C2-HMBMIm](+)), was further studied. The butyl group substituted imidazolium cation ([C2-BBMIm](+)) exhibited the highest alkaline stability at the elevated temperatures. The synthesized anion-exchange membranes based on the [C2-BBMIm](+) cation showed promising alkaline stability. These observations should pave the way to the practical application of imidazolium-based anion exchange membrane fuel cells.

Abstract:The adsorption capacities of ion exchangers with N-methyl-D-glucamine (NMDG) groups (Amberlite IRA 743, Lewatit MK 51, Purolite S110 and Purolite S108) relative to V(V) ions were tested in a batch system, taking into account the influence of various parameters, such as the adsorbent mass (0.05–0.20 g), phase contact time (1–240 min), initial concentration (10–150 mg/L), and temperature (293–333 K), as well as in a column system where the variable operating parameters were initial concentration (50, 100 mg/L), bed volume (10, 100 mL) and flow rate (0.6, 6 mL/min). Pseudo-first order, pseudo-second order, intraparticle diffusion and Boyd models were used to describe the kinetic studies. The best fit was obtained for the pseudo-second order model. The Langmuir, Freundlich and Temkin adsorption models were used to describe the equilibrium data to acquire better knowledge about the adsorption mechanism. The thermodynamic parameters were also calculated, which showed that the studied processes are endothermic, spontaneous and thermodynamically favorable. The physicochemical properties of the ion exchangers were characterized by nitrogen adsorption/desorption analyses, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR) and X-ray photo electron spectroscopy (XPS). The point of zero charge (pHPZC) was also determined.
