Abstract:Antennal surface structure of Pteromalus puparum L. was observed by scanning electron microscope. Seven types of sensilla, namely, thick-walled chemoreceptor, thin-walled chemoreceptor, plate organ, short-curved tactile hair, sharp-tipped tactile hair, basiconic capitate peg and chaeticum sensillum were found on the antennae both of female and male. The shape of these sensilla and their distribution were described, and their difference between sexes was discussed.

Abstract:A modified method,which is simpler and faster than most of the traditional methods,for fixing and processing plant parasitic nematodes during their transform from water to glycerin,was provided on the basis of Seinhorst's work(1959).It was found that some of the important morphological characters,such as the shape of stylet,the incisures in lateral field and the bursa of males of tylenchoid nematodes,and the position of guiding ring,morphology of base of odontostyle, and the shape of tail of longidorid nematodes etc.,were successfully maintained when specimens from Heterodera,Helicotylenchus,Hirschmanniella,Xiphinema and Longidorus were processed with the improved method.

Abstract:Abstract Museum collections contain enormous quantities of insect specimens collected over the past century, covering a period of increased and varied insecticide usage. These historic collections are therefore incredibly valuable as genomic snapshots of organisms before, during, and after exposure to novel selective pressures. However, these samples come with their own challenges compared with present-day collections, as they are fragile and retrievable DNA is low yield and fragmented. In this article, we tested several DNA extraction procedures across pinned historic Diptera specimens from four disease vector genera: Anopheles, Aedes, Culex, and Glossina. We identify an approach that minimizes morphological damage while maximizing DNA retrieval for Illumina library preparation and sequencing that can accommodate the fragmented and low yield nature of historic DNA. We identify several key points in retrieving sufficient DNA while keeping morphological damage to a minimum: an initial rehydration step, a short incubation without agitation in a modified low salt Proteinase K buffer (referred to as “lysis buffer C” throughout), and critical point drying of samples post-extraction to prevent tissue collapse caused by air drying. The suggested method presented here provides a solid foundation for exploring the genomes and morphology of historic Diptera collections.

A practical approach to the sampling, fixation, softening, and sectioning of whole honey bees for histologic evaluation

Abstract:The Western honey bee (Apis mellifera) is economically important as the primary managed pollinator of many agricultural crops and for the production of various hive-related commodities. Honey bees are not classically or thoroughly covered in veterinary pathology training programs. Given their unique anatomic and biological differences from the other species more traditionally evaluated by veterinary pathologists, establishing routine and consistent methods for processing samples for histology ensures accurate diagnostic and research conclusions. We developed and tested several field protocols for the sampling of honey bees. We compared the tissue-quality outcomes for worker bees fixed, collected, and/or softened under the following protocols: 1) routine formalin fixation; 2) softening chitin via exposure to Nair for 2 d or 3) 5 d; 4) shortened times between formalin submersion and trimming of body segments to enhance penetration of formalin into internal tissues; 5) ethanol submersion of specimen prior to formalin fixation; 6) indirect dry ice exposure; and 7) prolonged -80°C storage. Routine formalin fixation, exposure to Nair for 2 d, indirect dry ice exposure, and trimming body segments within 2 h of formalin submersion resulted in the highest quality histologic tissue sections. The poorest quality sections resulted from softening of chitin by exposure to Nair for 5 d, submersion in ethanol for 3 d before formalin fixation, and prolonged storage at -80°C. Our results indicate that routine formalin fixation is adequate, and that immobilizing bees with indirect dry ice exposure aids in sample collection without negatively impacting the quality of histologic sections.

Abstract:Thrips (Insecta: Thysanoptera) constitute a well-derined order of insects that has considerable economic importance. Because of the small size of thrips, it requires accurate microscopic slides and careful examination under a compound microscope at high magnification to examine their minute details clearly. Thrips species usually cannot be recognized without mounting specimens onto microscope slides.The techniques and materials used in preparing specimens for high-power microscopic examination are variable in different studies. A new method is described here, which needs fewer chemical materials and is especially suitable for preparing dark specimen, and allows us to identify thrips specimens rapidly; furthermore, specimens mounted by this new method are clear and retain their original colors, which is often necessary for identification. This new method has the shortest steps in preparing specimens, including maceration by KOH, dehydration by absolute ethanol, mounting by Hoyer's medium, desiccation and labeling, A discussion is provided for comprising this method with others.

Abstract:Ooencyrtus phongi (Hymenoptera: Encyrtidae) is an important egg parasitoid of the litchi stink bug, Tessaratoma papillosa (Drury) (Hemiptera: Tessaratomidae). Antennae of parasitic Hymenoptera are important sensory organs and play an important role in host location, host discrimination, courtship, and mating behavior. In this article, we examined the external morphology of the antennal sensilla of female and male of O. phongi using scanning electron microscopy. Twelve morphological sensillar types were recognized in both sexes, including the placoid sensilla, basiconic sensilla, two types of sensilla trichodea, and eight types of sensilla chaetica. Major differences were found between the sexes, in number, distribution, shape, structure, and size of the identified sensilla. These results are discussed in relation to the possible role of these sensilla in the host location behavior of O. phongi.

Abstract:The woodwasp Sirex noctilio Fabricius is a major quarantine pest that was reported in China in 2013 and mostly damages Pinus sylvestris var. mongolica. Reverse chemical ecology, which uses chemical lures to catch or block insects from mating is the classic way to control forestry pests. This indicates that insect sensilla play a crucial role in detecting external chemical and physical stimuli. Nonetheless, the categorization and distribution of sensilla on the antennae and ovipositor of S. noctilio are insufficiently specific. In this paper, scanning electron microscopy (SEM) was used to observe the ultrastructure of the sensilla of S. noctilio on the antenna and ovipositor. It was found that the types and distribution of sensilla on the antennae of S. noctilio male and female are consistent, and six types of sensilla are found: sensilla trichodea (ST), sensilla chaetica (SC), Böhm bristles (BB), sensilla basiconica (SB), sensilla ampullacea (SA), and contact chemoreceptors (CC). Besides, there are five types of sensilla on the female ovipositor. In addition to ST, SC and BB, two more types of sensilla are also found: sensilla cavity (SCa) and sensilla coeloconica (SCo). Through identification of the morphology and distribution of the sensilla, the functions of different sensilla in the mating and host selection mechanisms of S. noctilio are proposed, thereby establishing a foundation for S. noctilio chemical communication research.

Abstract:[Objectives]Transmission electron microscopy is playing a more and more important role in biological research, including tracking extended structures such as neural networks and three dimensional reconstruction. Consequently, the demand for continuous sections of biological tissues for transmission electron microscopy has greatly increased. However, limitations of the current fishing method make the acquisition of continuous sections difficult, especially when samples are comprised of dense insect exoskeleton, such as continuous sections of the antennae. Insufficient penetration of the agent usually causes poor polymerization, which undoubtedly increases the difficulty of collecting intact serial sections. To solve these problems, we developed a kind of combined device for collecting sections with slot grids.[Methods] A hybrid device comprised of a special fishing loop and a lofting platform was developed and its ability to fish for serial sections and place them onto slot grids was evaluated.[Results] No partial detachment and folding occurred in the serial sections obtained by this combined device; specimen sections neither separated completely, nor slid to the side of the resin section. Sample sections were intact without any damage or wrinkles, and serial sections clung to the slot area in a line.[Conclusion]The new device is an improvement on previous methods of obtaining continuous sections of insect antennae for electron microscopy.
Observation of the fine structure of antennal sensilla of the stink bug, Eocanthecona furcellata (Hemiptera: Pentatomidae)

Abstract:In order to explore the mechanism underlying chemosensation in Eocanthecona furcellata, the external morphology of its antennae and the type, quantity, distribution and ultrastructure of the sensilla were observed on both sexes of adults and 5th-instar nymphs using scanning electron microscopy. The results showed that the antennae of E. furcellata consisted of three parts: scape, pedicel and flagellum. There were five types of sensilla on the antennae, which included sensilla trichoidea (ST), sensilla chaetica (SCh), sensilla coeloconica (SCo), sensilla basiconica (SB) and squamifornia denticles (SD). Further, there were 4 subtypes of ST and SB and 2 subtypes of SCo and SCh. The number of sensilla on nymphs was significantly lower than that on adults. The antennae of adults showed sexual dimorphism, as the number of sensilla on female adults was higher than that on male adults. SB4 was found only on females and SCo2 was found only on males. These inter-sexual differences may be related to chemoreception of sex pheromone and chemical predation location. The morphology and putative functions of each sensilla were compared and discussed. These results provide a reference for further study of the behavioral biology, chemical ecology and electrophysiology of insects, and also provides a scientific basis for new ways of biological control.
Morphological analysis of sensilla on different organs in Pachyneuron aphidis, a hyperparasitoid of Myzus persicae

Abstract:Aphidius gifuensis is the main enemy of Myzus persieae. While its parasitic rate can be influenced by the hyperparasitoid, Pachyneuron aphidis. As important parts of insects to sense odors from various environments, study of sensilla can lay the foundation of the further study about the parasitic mechanisms, reduce the hyperparasitic rate, and make the most effect usage of A. gifuensis. Here, we give a fundamental study about the morphology of the sensilla on the whole body of male and female P. aphidis. We observed seven main types of sensilla on them totally by using scanning electron microscopy. Including Böhm bristle (BB), chaetica sensilla (ChS), basiconic sensilla (BS), trichoid sensilla (TS), and placodea sensilla (PS), coeleoconica sensilla (CoS), basiconic capitate peg sensilla (BCPS). In addition, TS on antennae can be divided into four subtypes, on wings can be divided into two subtypes. Sensilla were most abundant on the antennae. We observed all types of sensilla on antennae. TS4 was uniporous and PS was multiporous. The other sensilla were nonporous. We did not find sexual dimorphism with regards to sensilla on the antennae except for the location of CoS. In male, CoS situated on the fourth subsegment of flagellum, but on the eighth subsegment in female. In other organs, TS has the largest number. We also found BS on compound eyes and ovipositor, BB on thoracic legs. The possible roles of these sensilla played in life activities are discussed. Our study makes a contribution of the parasitic mechanism of hyperparasitoids.

Abstract:ABSTRACT Through natural selection, many animal organs with similar functions have evolved different macroscopic morphologies and microscopic structures. Here, we comparatively investigate the structures, properties and functions of honey bee stings and paper wasp stings. Their elegant structures were systematically observed. To examine their behaviors of penetrating into different materials, we performed penetration–extraction tests and slow motion analyses of their insertion process. In comparison, the barbed stings of honey bees are relatively difficult to be withdrawn from fibrous tissues (e.g. skin), while the removal of paper wasp stings is easier due to their different structures and insertion skills. The similarities and differences of the two kinds of stings are summarized on the basis of the experiments and observations.

Abstract:Objective To observe the microstructure of Pthirus pubis and its eggs.Methods Obtain adult and eggs of P.pubis,dealing through irrigation,fixation,dryness and plating,and observe under scanning electron microscope(SEM).Results The observation proved that palpus were located on either side of haustellum;a pair of concavity nests are newly discovered at the base of first segment of first foot,in each of which there was ecptoma,probably sense organ,shaped like semi-dumbbell;a pair of pits were found on the dorsal surface of chest.Conclusion The microstructure of P.pubis are observed and described detailedly,providing more information on morphology study of P.pubis.

Abstract:The millions of specimens stored in entomological collections provide a unique opportunity to study historical insect diversity. Current technologies allow to sequence entire genomes of historical specimens and estimate past genetic diversity of present-day endangered species, advancing our understanding of anthropogenic impact on genetic diversity and enabling the implementation of conservation strategies. A limiting challenge is the extraction of historical DNA (hDNA) of adequate quality for sequencing platforms. We tested four hDNA extraction protocols on five body parts of pinned false heath fritillary butterflies, Melitaea diamina , aiming to minimise specimen damage, preserve their scientific value to the collections, and maximise DNA quality and yield for whole-genome re-sequencing. We developed a very effective approach that successfully recovers hDNA appropriate for short-read sequencing from a single leg of pinned specimens using silica-based DNA extraction columns and an extraction buffer that includes SDS, Tris, Proteinase K, EDTA, NaCl, PTB, and DTT. We observed substantial variation in the ratio of nuclear to mitochondrial DNA in extractions from different tissues, indicating that optimal tissue choice depends on project aims and anticipated downstream analyses. We found that sufficient DNA for whole genome re-sequencing can reliably be extracted from a single leg, opening the possibility to monitor changes in genetic diversity maintaining the scientific value of specimens while supporting current and future conservation strategies.

Abstract:The pre-overwintering nest structure and the immature stages of the hornet Vespa fumida van der Vecht are for the first time reported. All three adult forms (queen, worker and drone) are briefly described. The eggs have many micropyles concentrated at the larger end of the egg. The mature larvae can be identified by having the ecdysial sulcus obliterated ventrally; the parietal band distinctly margined ventrally on the outer side; the palate with a median patch and a pair of dorsal patches; the collar processes of the spiracle long and thick, with many branches over almost all the surface. Morphology and distribution of the different types of sensilla (setae, conical papillae, campaniform sensilla, basiconic sensilla, etc.) and the spicules are illustrated in detail. Close relationship between the Vespa affinis-group and V. fumida previously suggested by adult morphology is briefly discussed in terms of the morphology of the mature larvae.

Abstract:Technical competence exerts a substantial influence on morphological assessment. This chapter provides an overview of morphological analysis for laboratory technologists and advanced students in medicine and biology. The focus is on two major issues of tissue preparation for morphology, namely appropriate tissue preparation and processing, in routine paraffin section (fixation, dehydration, and impregnation), frozen section, and electron microscopy. A variety of common techniques are discussed, including routine hematoxylin and eosin (H&E) stain, special stains (histochemical staining), immunohistochemical staining, DNA/RNA hybridization in situ, and electron microscopy. The authors introduce not only principal chemical/biochemical theories, but also technical essentials in these techniques. The high-throughput tissue microarray technique is also briefly reviewed. Initial tissue processing is critical for morphology analysis, particularly for molecular analysis such as immunohistochemical staining and DNA/RNA hybridization in situ.

Abstract:Morphological diversity of seminal vesicles and spermatozoa play an important role in the taxonomic and phylogenetic analysis,but it has been scarcely studied in the subfamily Polistinae,even in the whole Vespidae.The male reproductive system of the paper wasp Polistes chinensis antennalis Perez was dissected for the first time,with emphasis on the ultrastructure of the seminal vesicle.The morphology of the sperm was also briefly reported.Seminal vesicle and accessory gland are not surrounded by a common sheath.The epithelium of seminal vesicle consists of a monolayer of cubical cells,adhering to a thick basal lamina,and these epithelial cells are polarized;the apical half of the epithelial cells is especially rich in mitochondria with the apical membrane being differentiated into microvilli,while the basal half contains the usual cellular organelles,including some mitochondria,the nucleus and prominent inclusions.The spermatozoa of P.chinensis antennalis are about 94μm in length with the head about 21.4μm in length,so it is the species with the shortest sperm and the longest relative head length of sperm described for the family Vespidae.The results may provide a theoretical foundation for phylogenetic analysis and reproductive physiology of Vespidae.
