Establishment and evaluation of mouse model of atherosclerosis in systemic lupus erythematosus

Abstract:Cardiovascular complications related to atherosclerosis are major causes of morbidity and mortality in patients with systemic lupus erythematosus (SLE). However the underlying mechanisms are not fully understood.The mouse model of atherosclerosis (AS) in systemic lupus erythematosus (SLE) was established and evaluated for the purpose to provide mice for further studies of the pathogenesis and treatment of AS in SLE. The Fasl-/-(SLE) mice were cross-bred apoE-/-(atherosclerosis) mice, and the resulting heterozygous mice were further crossed to produce homozygous animals (Fasl-/-apoE-/-). The polymerase chain reaction was performed to determine Fasl-/-and apoE-/-genotypes. The renal function and lipid profiles in serum were detected. The histological changes of kidney and heart were assessed. The results of PCR showed that double-mutant apoE-/-Fasl-/-were generated. Compared with wild type mice, the serum creatinine, triglycerides, total cholesterol and low density lipoprotein cholesterol were increased, while the high density lipoprotein cholesterol was decreased in apoE-/-Fasl-/-mice. The apoE-/-Fasl-/-mice had high titer of anti-ds DNA antibody in serum. Histopathological examination of kidneys from apoE-/-Fasl-/-mice displayed a pattern of glomerulonephritis typically found in systemic lupus erythematosus. The heart of apoE-/-Fasl-/-mice demonstrated visible lipid deposits after Oil red O staining. The generated double-mutant apoE-/-Fasl-/-mice displayed typical characteristics of SLE and atherosclerosis and could be used for further studies on the athero-sclerosis in SLE.
