Studies of Modulating Immune Function of T Lymphocyte by Granulocyte Colony-Stimulating Factor

Abstract:Objective To study the modulation of T lymphocyte immune function by granulocyte colony stimulating factor. Me- thods Peripheral blood samples from 15 donors were harvested before and after G-CSF treatment. Cell immune markers, AnnexinV, and intracellular cytokines were measured by flow cytometry (FCM). T-cell activity test was performed in vitro. Results Our data showed that G-CSF treatment induced an increasing expression of IL-4, and a significant reduction in the expression of IFN-γas well as the proliferative response of T-cell to mitogenic stimulation. There is no significant change in CD4 +, CD8 + cell counts after G-CSF treatment. No significant difference of AnnexinV expression was fund between Th1 and Th2 cells. Conclusion G-CSF suppresses the mitogen-stimulated T-cell proliferative response and polarizes the donor T cells toward the production of type 2 cytokines. Cell apoptosis might not be the mechanism of Th1/Th2 paradigm.
